Effects of ketamine on behavior and hippocampal nitric oxide synthase in depressed rats induced by chronic stress

Abstract:Objective To observe the effects of ketamine on behavior and the expression of hippocampal nitric oxide synthase(NOS)in the depressed rats induced by chronic stress. Methods Thirty-two male adult Wistar rats were randomized into 4groups(n=8):saline 1hgroup(group S1),saline 24 hgroup(group S24),ketamine 1 hgroup(group K1),and ketamine 24hgroup(group K24).The chronic unpredicted stress was used as a depression model.Sucrose preference test(SPT)and forced swimming test(FST)were performed to evaluate the behavioral changes of rats. The rats were decapitated immediately after the behavioral tests and the hippocampus was harvested for the determination of NOS activity.Results The immobility time during the FST,the total NOS(tNOS)activity,and the inducible NOS(iNOS)activity were decreased in the group K1compared with the group S1(P<0.05).Compared with the group S24,the group K24had increased sucrose consumption during the SPT,decreased immobility time during the FST,and decreased tNOS and iNOS activities(P<0.05). No significant difference was observed respect to cNOS.Conclusion Ketamine may rapidly improve the depressive behaviors in rats with the chronic stress,which might be potentially attributed to the inhibition of hippocampal iNOS activity.
