Expression of human telomerase reverse transcriptase gene and c-myc protein in gastric carcinogenesis

Abstract:AIM: To investigate the expression of human telomerase reverse transcriptase (hTERT) gene and c-myc protein in gastric carcinogenesis. METHODS: Total 41 cases of primary gastric tumors, 15 dysplasia, 23 intestinal metaplasia and 10 normal mucosa were examined. Expression of hTERTmRNA, hTERT protein and c-myc protein were determined by in situ hybridization and immunohistochemistry. RESULTS. In normal mucosa, intestinal metaplasia, dysplasia and neoplasms, c-myc expression were 0, 30 %, 53 % and 59 %, hTERTmRNA were 10%, 39%, 67% and 85 % and hTERT protein were 0, 30%, 60% and 78%, respectively. All the three parameters were significantly increased in dysplasia and neoplasm, compared to normal mucosa. The expression were also significantly higher in neoplasia than that in intestinal metaplasia (P < 0.05). Expression of c-myc and hTERT protein were significantly increased in the poorly differentiated group, lymph node-positive group, advanced carcinoma and those which had invaded to serosa (P < 0.01 ∼ 0.05). HTERTmRNA expression was higher only in advanced carcinoma and had no statistic relations with other clinical and histological factors. There were significant relationships among the three parameters in intestinal metaplasia, dysplasia and neoplasm groups, respectively (rs = 0.504 ∼ 0.696, P < 0.05). CONCLUSION: Expressions of hTERT gene and c-myc protein are up-regulated at an early stage of the gastric carcinogenesis. It is likely that c-myc protein has an ability to stimulate the expression of hTERT and thereby enhance telomerase activity, which may be an important step in carcinogenesis.
