THE EXPRESSION AND CLINICAL SIGNIFICANCE OF CYTOKERATIN 20 mRNA IN CENTRAL VEIN BLOOD OF PATIENTS WITH GASTRIC CANCER BY RT-PCR

Abstract:Objective: The nested reverse transcriptional polymerase chain reaction(RT-PCR) method was used to investigate the expression of cytokeratin 20(CK-20) in central vein blood of patients with gastric cancer.Methods: Central vein blood was taken from 16 benign diseases without a history of gastric cancer and from 64 patients with gastric cancer,a comparison and analysis was made between CK 20mRNA and pathological information.Results:CK20mRNA was positive in 34 of 64 central vein blood specimens of patients with gastric cancer,positive rate was 53.1%;negative results were obtained in 16 central vein blood specimens of patients with benign diseases;micrometastasis rate was relevant to clinical staging,metastasis of liver and abdomen cavity,tumor size(P<0.05),but was not relevant to histological type(P<0.05).CK20 mRNA was positive in 2 of 8 central vein blood specimens of patients with early gastric cancer.Conclusion:Through amplifying CK20 mRNA,nest RT-PCR could be a sensitive and specific method to examine the micrometastasis in central vein blood of patients with gastric cancer.
