Preparation of Oleic Acid Modified Superparamagnetic Iron Oxide Nanoparticles

Abstract:Firstly,hydropHilic superparamagnetic iron oxide nanoparticles (SPION) were synthesized from ferrous chloride with ferric chloride via a coprecipitation method.Then,the carboxyl groups of oleic acid were activated.Finally,the activated oleic acid was used as modifier and superparamagnetic iron oxide nanoparticles modified with oleic acid (O-SPION) were obtained by a chemical method.The samples were characterized by X-ray diffraction (XRD),Fourier transform infrared (FTIR) spectroscopy,dynamic light scattering (DLS),and transmission electron microscope (TEM) techniques.Additionally,the cytotoxicity of O-SPION on HepG2 cells was assessed by MTT assay,and the cell uptake ability was measured by Prussian blue staining.The results showed that the resulting O-SPION had regular morpHology with the core particle size of (12±1.5) nm and the Zeta potential of (36±1.5) mV.Moreover,the O-SPION could form stable magnetic fluid in organic solvent.The in vitro tests demonstrated that O-SPION showed no toxicity to HepG2 cells even if the concentration of O-SPION reached 800 mg/L.Meanwhile,the result of Prussian blue staining revealed that the intracellular uptake of O-SPION was higher than that of SPION.
