Abstract:Nisin is a natural preservative for many food products. This bacteriocin is mainly used in dairy and meat products. Nisin inhibits pathogenic food borne bacteria such as Listeria monocytogenes and many other Gram-positive food spoilage microorganisms. Nisin can be used alone or in combination with other preservatives or also with several physical treatments. Abstract:Abstract This review examines the biosynthesis of nisin, its antimicrobial mechanism, and its applications in food preservation. Nisin is synthesized withinside the ribosome as a pre-peptide which undergoes amendment withinside the post-translational level as a number one metabolite from lactic acid bacteria. Nisin secretion occurs in the exponential phase and is maximally produced at the end of the exponential phase or the beginning of the stationary phase. As an antibiotic in the form of a peptide molecule, nisin is characterized by the presence of lanthionine-type thioester bonds. Nisin has been reported to have the ability to inhibit bacteria, especially Gram-positive bacteria such as Bacillus subtilis, Bacillus stearothermophilus, Clostridium botulinum, Streptococcus hemolyticus, Staphylococcus aureus , and Listeria monocytogenes . Nevertheless, nisin is less effective against Gram-negative bacteria, yeast, and molds. Based on its ability to inhibit bacteria plus the non-toxic nature of nisin, Nisin can be used as bio preservation in food products. Besides being non-toxic, several other factors that can be considered for nisin as bio preservation are that nisin is readily biodegradable. It is a protein compound that does not harm the intestinal flora as it is easily digested by enzymes in the gastrointestinal tract and does not cause bacterial resistance as therapeutic antibiotics are safe for humans.

In this paper the molecule structure and the interaction mechanism with target membrane of nisin, and the mode for nisin pore formation were described.

Abstract:Small pentacyclic peptides, represented by nisin, have been successfully utilized as preservatives in the food industry and have evolved into a paradigm for understanding the genetic structure, expression, and control of genes created by lantibiotics. Due to the ever-increasing antibiotic resistance, nisin-relevant antimicrobial peptides have received much attention, which calls for a summarization on their synthesis, modification and applications. In this review, we first provided a timeline of select highlights in nisin biosynthesizing and engineering. Then, we outlined the current developments in nisin synthesis. We also provided an overview of engineering, screening, and producing nisin-relevant antimicrobial peptides based on enzyme alteration, substrate modification, and sequence mining. Furthermore, an updated summarization for applications of nisin-relevant antimicrobial peptides was developed on food applications. Finally, this study offered insights into emerging technologies, limitation and future development of nisin-relevant antimicrobial peptides in pathogen inhibition, food preservatives, and improved health.

Abstract:Nisin is a typical representative of class I bacteriocins belonging to subtype A1 lantibiotics. This lantibiotic is produced by gram-positive bacteria of the genera Lactococcus, Streptococcus, Staphylococcus, and Blautia. Currently, at least twelve natural variants of nisin are known. They differ in some physicochemical properties and biological activity. Nisin is generally recognized as safe (GRAS) and has been used as a food additive for seven decades. The current review presents the data from scientific reports over the past decade supporting the value of this lantibiotic as a therapeutic agent. Most research works demonstrate a high inhibitory potential of nisin against antibiotic-resistant bacteria when using alone and in combination with antibiotics. A number of studies prove the ability of nisin to modulate the composition of oral and intestinal microbiomes, to inhibit the growth of both bacteria causing infectious processes and bacterial pathogens associated with cancer. Nisin has been shown to have its own antitumor potential and to be able to enhance the therapeutic effect of chemotherapeutic drugs. There is convincing evidence of its immunomodulatory and anti-inflammatory activity. The reasons limiting its widespread application in clinical practice can be successfully eliminated via modern bioengineering technologies that improve the functional characteristics and stability of nisin in various physiological conditions.

Abstract:The global scenario is changing toward the safety of foods. Under this concern, pathogens that can be targeted in foods using antimicrobial peptides of lactic acid bacteria (LAB) along with other newer modified small peptides are receiving much attention to mitigate the adverse effects of pathogens. Various strategies to use bacteriocin nisin as a preservative in foods and packagings are under intensive research. Since additive nisin is very costly, it has limited use in developing countries, for the cost of producing it fermentatively. In striking a balance, however, the increase in chemical preservation is receiving much attention, and use of alternative additives like nisin (GRAS approved) is claimed, as safe to replace chemicals instead as per need in natural (direct) and in minimally processed foods. Some of the variants (structural) of nisin are produced by the LAB Lactococcus lactis that also produces other known bacteriocins like lacticins 3145 and 481 and lactocin 140. The focus here is on understanding its structure-function relationships along with the properties, its purification, to trace developments of nisin in quantification and its uses. Many constraints are associated with nisin fermentative production along with its assay and purification. Pure nisin for wider application in foods and packagings is a far-fetched target and needs a very thorough investigation into production, assay, purification, and its application in food.

Abstract:Nisin, is a class of low molecular weight polypeptides or proteins, produced by most lactic acid bacteria, Nisin A was first identified in milk cultures in 1928 and is the only bacteriocin approved by the FDA as a food biopreservative and has been extensively studied. The antimicrobial is not limited to food-derived microorganisms, but also includes bacteria, fungi and treponemes associated with human disease. The oral cavity harbors >1000 microorganisms, where homeostasis is maintained through dynamic host-microbe interactions. With the safety of nisin in food approved for >60 years, scientists and dentists are beginning to focus on its potential application in oral health maintenance and disease control. In fact, nisin, nisin-containing dental materials, and nisin-producing probiotics show promising performance against dental caries, endodontic and periodontal diseases, and even oral cancer. In this review, we will discuss nisin, its current use, and future directions for improving oral health.

Abstract:Nisin is used as a natural food preservative because of its broad-spectrum antimicrobial activity against Gram-positive bacteria. However, free nisin is susceptible to various factors that reduce its antimicrobial activity. Milk protein, a protein derived from milk, has self-assembly properties and is a good carrier of bioactive substances. In this study, lactoferrin-nisin nanoparticles (L-N), bovine serum albumin-nisin nanoparticles (B-N), and casein-nisin nanoparticles (C-N) were successfully prepared by a self-assembly technique, and then their properties were investigated. The studies revealed that lactoferrin (LF) and nisin formed L-N mainly through hydrophobic interactions and hydrogen bonding, and L-N had the best performance. The small particle size (29.83 ± 2.42 nm), dense reticular structure, and good thermal stability, storage stability, and emulsification of L-N laid a certain foundation for its application in food. Further bacteriostatic studies showed that L-N enhanced the bacteriostatic activity of nisin, with prominent inhibitory properties against Listeria monocytogenes, Staphylococcus aureus, and Bacillus cereus, which mainly disrupted the cell membrane of the bacteria. The above results broaden our understanding of milk protein-nisin nanoparticles, while the excellent antibacterial activity of L-N makes it promising for application as a novel food preservative, which will help to improve the bioavailability of nisin in food systems.

Abstract:With the continuous improvement of living standards and growing concern on health,people put forward higher requirements on food safety. Natural food preservatives are a class of antimicrobial materials,directly extracted from plants,animals and microorganisms. Natural food preservatives have the advantage of abundant resources,strong antimicrobial activity,water-soluble,non-toxic,and wide antimicrobial spectrum are becoming a research hotspot focus in recent years. However,natural food preservatives have not been widely used yet. The natural property,antibacterial mechanism and application of the five kinds of animal antimicrobial materials were reviewed in this article,and their future development and problems encountered in use were also discussed.

Abstract:Nisin is a small peptide produced by Lactococcus lactis ssp lactis that is currently industrially produced. This preservative is often used for growth prevention of pathogenic bacteria contaminating the food products. However, the use of nisin as a food preservative is limited by its low production during fermentation. This low production is mainly attributed to the multitude of parameters influencing the fermentation progress such as bacterial cells activity, growth medium composition (namely carbon and nitrogen sources), pH, ionic strength, temperature, and aeration. This review article focuses on the main parameters that affect nisin production by Lactococcus lactis bacteria. Moreover, nisin applications as a food preservative and the main strategies generally used are also discussed.

Abstract:Nisin and other similar food preservatives of natural origins have seen a substantial upsurge in global interest in recent years. The use of nanotechnology to regulate and manipulate nisin for enhanced capacities in the food and nutrition sector is expanding dramatically. Nanotechnology has significant food science applications in nanoparticle delivery systems, packaging, food security, and safety. However, there have been considerable issues regarding the use of nisin in the food sector, including its uncontrolled interactions with various food components, its degradation and electrostatic repulsion. These issues potentially limit its use. Alternate strategies, including a variety of nanoparticle systems such as nanoemulsions, polymeric nanoparticles, nanofibers, and combinations of nisin with other technologies, are employed to enhance the utility of nisin in the food industry. This review highlights the recent developments and new perspectives related to the uses of nisin in the food industry.Industrial relevanceThis review highlights the current status of nanotechnology in the food industry. The issues concerning use of nisin by the food industry are addressed. Nisin shows enhanced efficacy in combination with other current technologies for improved food safety.

Abstract:It is almost a century since nisin was discovered in fermented milk cultures, coincidentally in the same year that penicillin was first described. Over the last 100 years this small, highly modified pentacyclic peptide has not only found success in the food industry as a preservative but has also served as the paradigm for our understanding of the genetic organization, expression, and regulation of genes involved in lantibiotic biosynthesis-one of the few cases of extensive post-translation modification in prokaryotes. Recent developments in understanding the complex biosynthesis of nisin have shed light on the cellular location of the modification and transport machinery and the co-ordinated series of spatio-temporal events required to produce active nisin and provide resistance and immunity. The continued unearthing of new natural variants from within human and animal gastrointestinal tracts has sparked interest in the potential application of nisin to influence the microbiome, given the growing recognition of the role the gastrointestinal microbiota plays in health and disease. Moreover, interdisciplinary approaches have taken advantage of biotechnological advancements to bioengineer nisin to produce novel variants and expand nisin functionality for applications in the biomedical field. This review will discuss the latest progress in these aspects of nisin research.

Abstract:A growing awareness among consumers towards the health aspects has increased their interest on natural products.As an alternative to certain disadvantages associated with chemical preservatives,industries are paying more emphasis on the use of natural preservatives.Utilization of natural preservatives has rendered foods with high nutritional value,free from chemical preservatives and adequate microbiological safety.Natural preservatives are the chemical agents derived from plants,animals,microbes and their metabolites that prevent the decomposition of products by any means.The mode of action of these natural preservatives is inhibition of microbial growth,oxidation and certain enzymatic reactions occurring in the foodstuffs.This review focuses on variouses naturally derived constituents,their mechanisms of antimicrobial action,and the application research status in food preservation.The application result in food hurdle technology is introduced.The future application prospect of natural biological preservative and the main problems to be solved in the research and development are outlooked in food systems.
