Abstract:Nisin, a 34-residue antibacterial peptide, is widely utilized in food industry as highly effective, safe and natural food preservative. It contains cyclic lanthionine rings and unusual dehydrated amino acids, which are formed by posttranslational modification. The strucure of nisin is usually used to study on the relationship of structure-function. In this paper the molecule structure and the interaction mechanism with target membrane of nisin, and the mode for nisin pore formation were described.

Abstract:Small pentacyclic peptides, represented by nisin, have been successfully utilized as preservatives in the food industry and have evolved into a paradigm for understanding the genetic structure, expression, and control of genes created by lantibiotics. Due to the ever-increasing antibiotic resistance, nisin-relevant antimicrobial peptides have received much attention, which calls for a summarization on their synthesis, modification and applications. In this review, we first provided a timeline of select highlights in nisin biosynthesizing and engineering. Then, we outlined the current developments in nisin synthesis. We also provided an overview of engineering, screening, and producing nisin-relevant antimicrobial peptides based on enzyme alteration, substrate modification, and sequence mining. Furthermore, an updated summarization for applications of nisin-relevant antimicrobial peptides was developed on food applications. Finally, this study offered insights into emerging technologies, limitation and future development of nisin-relevant antimicrobial peptides in pathogen inhibition, food preservatives, and improved health.

Abstract:Nisin is a kind of lantibiotis produced by certain strains of Lactococcus lactis, It has been used in food industry because of its efficient antimicrobial activity against a wide range of gram-positive organisms such as bacillus, streptococcus,and Listeria species. Studies showed that nisin biosynthesis gene cluster which included nis ABTCIPRKFEG locates in a chromosomally located conjugative transposon,together with the genes for sucrose-6-phosphate hydrolase,one or several phage-resistant systems and N5-(L-1-carboxyethyl)-L-ornithine. Nisin gene cluster included three operons and could be regulated by a mechanism of autoregulation through signal transduction via two-component regulators. The structure,gene cluster of nisin and its autoregulation mechanism were reviewed comprehensively.

Abstract:Nisin is a kind of lantibiotis produced by certain strains of Lactococcus Lactis. It has been employed as a food preservative as it has a high antibacterial activity with a relatively low toxicity for human. Pore formation is considered the primary mode of antimicrobial action of nisin. Nisin forms wedge-like pore in the membrane by electrostatic interaction between the positively charged C-terminus of the peptide and anionic phospholipids. Nisin can also form lipid II-mediated pore via a targeted mechanism by using lipid II,the bactoprenol-bound precursor of the bacterial cell wall,as a docking molecule. Another consequence of binding with lipid II is the inhibition of peptidoglycan biosynthesis. Nisin utilizes two distinct mechanisms of antibacterial activity:a high-salt-sensitive mechanism for Escherichia coli and a high-salt-insensitive mechanism for Staphylococcus aureus. Antibacterial mechanism of nisin was reviewed comprehensively in this article.

Abstract:Nisin, one kind of natural antimicrobial peptide, is produced by certain Lactococcus lactis strains, which generally require expensive high-quality nitrogen sources due to limited ability of amino acids biosynthesis. Here we use defatted soybean meal (DSM) as sole nitrogen source to support L. lactis growth and nisin production. DSM medium composition and fermentation conditions were optimized using the methods of Plackett-Burman design and central composite design. The highest nisin production of 3879.58 IU/ml was obtained in DSM medium, which was 21.3% higher than that of commercial medium. To further increase the utilization ability of nitrogen sources, we enhanced the proteolytic function in L. lactis through rationally expressing the related enzymes, which were selected according to the compositions of amino acids and molecular weight of peptides in DSM medium. Significantly, an artificial proteolytic system consisting of a heterologous protease (NprB), an oligopeptides transporter subunit (OppA) and two peptidases (PepF and PepM) was introduced into L.lactis. The constructed strain BAFM was capable of achieving efficient biomass accumulation and nisin yield with 30% decreased amount of DSM hydrolysates, which further reduced the cost of nisin production. The strategy described here offers opportunities for low-cost L. lactis fermentation and large-scale nisin production in industry.

Abstract:Twenty isolates of Staphylococcus aureus and Streptococcus agalactiae were obtained from mastitic cases in dairies in suburb of Hangzhou.Sensitive test of the bacteria to some conventional antibacterial drugs was done.The results indicated that more than 50% of Staphylococcus aureus isolates were resistant to penicillin,norflo-(xacin),paediatric compound sulfamethoxazole and streptomycin.Nisin had an effective inhibitory effect on Streptococcus agalactiae as well as Staphylococcus aureus both resistant and sensitive to penicillin.In addition,a microplate TTC assay showed that nisin was effective to inhibit the bacteria in milk but less effective than in culture(medium.)

Nisin inhibits pathogenic food borne bacteria such as Listeria monocytogenes and many other Gram-positive food spoilage microorganisms. Nisin can be used alone or in combination with other preservatives or also with several physical treatments. This paper reviews physicochemical and biological properties of nisin, the main factors affecting its antimicrobial effectiveness, and its food applications as an additive directly incorporated into food matrices.

Abstract:Nisin is used as a natural food preservative because of its broad-spectrum antimicrobial activity against Gram-positive bacteria. However, free nisin is susceptible to various factors that reduce its antimicrobial activity. Milk protein, a protein derived from milk, has self-assembly properties and is a good carrier of bioactive substances. In this study, lactoferrin-nisin nanoparticles (L-N), bovine serum albumin-nisin nanoparticles (B-N), and casein-nisin nanoparticles (C-N) were successfully prepared by a self-assembly technique, and then their properties were investigated. The studies revealed that lactoferrin (LF) and nisin formed L-N mainly through hydrophobic interactions and hydrogen bonding, and L-N had the best performance. The small particle size (29.83 ± 2.42 nm), dense reticular structure, and good thermal stability, storage stability, and emulsification of L-N laid a certain foundation for its application in food. Further bacteriostatic studies showed that L-N enhanced the bacteriostatic activity of nisin, with prominent inhibitory properties against Listeria monocytogenes, Staphylococcus aureus, and Bacillus cereus, which mainly disrupted the cell membrane of the bacteria. The above results broaden our understanding of milk protein-nisin nanoparticles, while the excellent antibacterial activity of L-N makes it promising for application as a novel food preservative, which will help to improve the bioavailability of nisin in food systems.

Abstract:SlpB, a surface layer protein isolated from Lactobacillus crispatus, has the potential to enhance the antimicrobial activity of nisin. Previous research indicated that, when combined with nisin, SlpB acted synergistically to inhibit Staphylococcus saprophyticus growth, thus extending the shelf life of chicken meat. In order to understand how SlpB enhances the antibacterial activity of nisin, electron microscopy, confocal laser scanning microscopy, flow cytometry and transmembrane electrical potential analysis were used to study cell wall organization and cell membrane integrity. No remarkable bacteriolytic effects were observed, indicating that cell death could not be attributed to cell lysis, although SlpB caused dramatic modifications of cell wall, thereby altering cell shape. The combination of SlpB and nisin also induced the release of ATP or UV-absorbing materials, as well as sudden dissipation of the transmembrane electrical potential by compromising membrane integrity. Considering that SlpB led to structural disorganization of the cell wall, and nisin access is enhanced to form a stable pore, cell death is a predictable outcome. SlpB significantly enhanced the effect of nisin at half of the minimum inhibitory concentration, which resulted in cell death by destroying the cell wall and cell membrane, therefore providing a new, feasible approach in food preservation.

Abstract:Nisin is a typical representative of class I bacteriocins belonging to subtype A1 lantibiotics. This lantibiotic is produced by gram-positive bacteria of the genera Lactococcus, Streptococcus, Staphylococcus, and Blautia. Currently, at least twelve natural variants of nisin are known. They differ in some physicochemical properties and biological activity. Nisin is generally recognized as safe (GRAS) and has been used as a food additive for seven decades. The current review presents the data from scientific reports over the past decade supporting the value of this lantibiotic as a therapeutic agent. Most research works demonstrate a high inhibitory potential of nisin against antibiotic-resistant bacteria when using alone and in combination with antibiotics. A number of studies prove the ability of nisin to modulate the composition of oral and intestinal microbiomes, to inhibit the growth of both bacteria causing infectious processes and bacterial pathogens associated with cancer. Nisin has been shown to have its own antitumor potential and to be able to enhance the therapeutic effect of chemotherapeutic drugs. There is convincing evidence of its immunomodulatory and anti-inflammatory activity. The reasons limiting its widespread application in clinical practice can be successfully eliminated via modern bioengineering technologies that improve the functional characteristics and stability of nisin in various physiological conditions.

Abstract:Aims: This study investigates the activity of the broad-spectrum bacteriocin nisin against a large panel of Gram-negative bacterial isolates, including relevant plant, animal, and human pathogens. The aim is to generate supportive evidence towards the use/inclusion of bacteriocin-based therapeutics and open avenues for their continued development. Methods and Results: Nisin inhibitory activity was screened against a panel of 575 strains of Gram-negative bacteria, encompassing 17 genera. Nisin inhibition was observed in 309 out of 575 strains, challenging the prevailing belief that nisin lacks effectiveness against Gram-negative bacteria. The genera Acinetobacter, Helicobacter, Erwinia, and Xanthomonas exhibited particularly high nisin sensitivity. Conclusions: The findings of this study highlight the promising potential of nisin as a therapeutic agent for several key Gram-negative plant, animal, and human pathogens. These results challenge the prevailing notion that nisin is less effective or ineffective against Gram-negative pathogens when compared to Gram-positive pathogens and support future pursuits of nisin as a complementary therapy to existing antibiotics. Significance and Impact of Study: This research supports further exploration of nisin as a promising therapeutic agent for numerous human, animal, and plant health applications, offering a complementary tool for infection control in the face of multidrug-resistant bacteria.

Abstract:The global scenario is changing toward the safety of foods. Under this concern, pathogens that can be targeted in foods using antimicrobial peptides of lactic acid bacteria (LAB) along with other newer modified small peptides are receiving much attention to mitigate the adverse effects of pathogens. Various strategies to use bacteriocin nisin as a preservative in foods and packagings are under intensive research. Since additive nisin is very costly, it has limited use in developing countries, for the cost of producing it fermentatively. In striking a balance, however, the increase in chemical preservation is receiving much attention, and use of alternative additives like nisin (GRAS approved) is claimed, as safe to replace chemicals instead as per need in natural (direct) and in minimally processed foods. Some of the variants (structural) of nisin are produced by the LAB Lactococcus lactis that also produces other known bacteriocins like lacticins 3145 and 481 and lactocin 140. The focus here is on understanding its structure-function relationships along with the properties, its purification, to trace developments of nisin in quantification and its uses. Many constraints are associated with nisin fermentative production along with its assay and purification. Pure nisin for wider application in foods and packagings is a far-fetched target and needs a very thorough investigation into production, assay, purification, and its application in food.

Abstract:Nisin serves as the prototype within the lantibiotic group of antimicrobial peptides, exhibiting a broad-spectrum inhibition against Gram-positive bacteria, including important food-borne pathogens and clinically relevant antibiotic-resistant strains. The gene-encoded nature of nisin allows for gene-based bioengineering, enabling the generation of novel derivatives. It has been demonstrated that nisin mutants can be produced with improved functional properties. Here, we particularly focus on the uncommon amino acid residues dehydroalanine (Dha) and dehydrobutyrin (Dhb), whose functions are not yet fully elucidated. Prior to this study, we developed a new expression system that utilizes the nisin modification machinery NisBTC to advance expression, resulting in enhanced peptide dehydration efficiency. Through this approach, we discovered that the dehydrated amino acid Dhb at position 18 in the peptide rombocin, a short variant of nisin, displayed four times higher activity compared to the non-dehydrated peptide against the strain Lactococcus lactis. Furthermore, we observed that in the peptides nisin and rombocin, the dehydrated amino acid Dha at residue positon 18 exhibited superior activity compared to the dehydrated amino acid Dhb. Upon purifying the wild-type nisin and its variant nisinG18/Dha to homogeneity, the minimum inhibitory concentration (MIC) indicated that the variant exhibited activity similar to that of wild-type nisin in inhibiting the growth of Bacillus cereus but showed twice the MIC values against the other four tested gram-positive strains. Further stability tests demonstrated that the dehydrated peptide exhibited properties similar to wild-type nisin under different temperatures but displayed higher resistance to proteolytic enzymes compared to wild-type nisin.

Study on the antibacterial activity of lysozyme hydrolysate

Abstract:Objective：Provide the theoretical foundation for broadening the application of lysozyme in food industry through researching on the bacteriostatic ability of Lysozyme hydrolysate. Methods：Lysozyme hydrolysate（LH） was obtained by using pepsin. The antibacterial activity of LH was tested by antibacterial circle experiment; meanwhile,lysozyme and nisin were also tested. The minimum bactericide concentration of LH against common food bacteria was determined with the two-fold dilution method. Results：LH showed great inhibition effect on positive bacteria,especially staphylococcus aureus. Without heat treatment,the bacteriostatic ability of LH on staphylococcus aureus was weaker than lysozyme,but much stronger than nisin. After heat treatment,LH was more antibacterial than lysozyme and nisin. LH was promising to be a natural food preservative.

Abstract:Lantibiotics are potential alternatives to antibiotics because of their broad-range killing spectrum. The producer strain is immune against its own synthesized lantibiotic via the expression of two proteins LanI and LanFEG. Recently, gene operons are found in mainly human pathogenic strains, which confer resistance against lantibiotics. Of all the lantibiotics discovered till date, nisin produced by some Lactococcus lactis strains is the most prominent member. Nisin has multiple mode of actions of which binding to the cell wall precursor lipid II and subsequent insertion into the bacterial membrane to form pores are the most effective. The nisin producing strains express the lipoprotein NisI to prevent a suicidal effect. NisI binds nisin, inducing a reversible cell clustering to prevent nisin from reaching the membrane. Importantly NisI does not modify nisin and releases it as soon as the concentration in the media drops below a certain level. The human pathogen Streptococcus agalactiae is naturally resistant against nisin by expressing a resistance protein called SaNSR, which is a nisin degrading enzyme. By cleaving off the last six amino acids of nisin, its effectiveness is 100-fold reduced. This cleavage reaction appears to be specific for nisin since SaNSR recognizes the C-terminal located lanthionine rings. Recently, the structures of both NisI and SaNSR were determined by NMR and X-ray crystallography, respectively. Furthermore, for both proteins the binding site for nisin was determined. Within this review, the structures of both proteins and their different defense mechanisms are described.

Abstract:Background Food preservation is important in increasing the shelf life of food and ensuring food safety. Investigations on food preservation have focused on meeting consumer demands for more natural and healthier food. Bacteriocins produced by lactic acid bacteria have caught the attention of food scientists to be used as natural food biopreservatives due to their antimicrobial activity against food spoilage and pathogenic bacteria. Class II bacteriocins are the largest bacteriocin family of lactic acid bacteria but have received disproportionately less attention than class I bacteriocins. Scope and approach This article systematically compared the sequence property, 3-D structure, and mode of action of the four categories of class II bacteriocins of lactic acid bacteria. The current and potential applications of class II bacteriocins in the food industry have also been reviewed. A SWOT (Strength, Weaknesses, Opportunities and Threats) analysis of the different approaches and development on class II bacteriocins was performed. Key findings and conclusions The class II bacteriocins produced by lactic acid bacteria have a variety of sequences and diverse properties, structures, and modes of action. Some of class II bacteriocins, e.g. pediocin, enterocin AS-48, plantaricin EF, are considered as attractive compounds in the food industry to prevent food spoilage and pathogenic bacterial growth. Elucidation of their characteristics and modes of action provides fundamental information in designing novel application strategies. The SWOT analysis will be helpful for current and future researches on class II bacteriocins.

Abstract:Nisin produced by certain Lactococcus lactis strains is commercially used in meat and dairy industries because of its effective antibacterial activity and food safety characteristics. It has been proved that the antibacterial activity could be enhanced when combined with other antimicrobial agents. In this study, we demonstrated that nisin and 3-phenyllactic acid (PLA) in combination displayed excellent combinational antibacterial activity against foodborne pathogens including S. xylosus and M. luteus . The potential application in food preservation was further verified via microbial analysis during the storage of meat and milk, and determination of strawberry rot rate. Scanning electron microscopy observation indicated a distinct mode of PLA with nisin, which may target at the dividing cell, contributing to their combinational antibacterial effect of nisin and PLA. Considering the positive results, a nisin-PLA co-producing strain was constructed based on the food-grade strain L. lactis F44, a nisin Z producer. By the knockout of two L-lactate dehydrogenase (LDH) and overexpression of D-LDH Y 25A , the yield of PLA was significantly increased 1.77-fold in comparison with the wild type. Anti-bacterial assays demonstrated that the fermentation product of the recombinant strain performed highly effective antibacterial activity. These results provided a promising prospect for the nisin-PLA co-expressing L. lactis in food preservation on account of its considerable antibacterial activity and cost-effective performance.

Observasion of Nisin for Bovine Mastitis Therapy

Abstract:Nisin is an antibacterial peptide of 34 amino acids produced by Lactococcus lactis,and is harmfulness for human.In this study,92 cows with 107 clinically mastitis quarters and 90 cows with 90 recessive mastitic quarters were randomly divided into nisin-treated and control groups.In nisin-treated groups,intramammary infusion of a nisin-based preparation was administered.In the control groups,intramammary infusion of gentamycin was used for clinical mastitis and no treatment was used for recessive mastitis.The results indicated that bacteriological and clinical cure rates were 60.8% and 90.2% in nisin-treated clinical mastitis cases respectively,while 44.6% and 91.2% in gentamycin-treated group respectively.There was no statistical difference between two groups.After nisin treatment in recessive mastitis cases,cows with high milk somatic cell count and milk NAGase activity were significantly decreased,and milk samples negative to bacteriological examination were increased.Inhibitory effect on fermentation was found until 36 hours after nisin treatment and 72 hours after gentamycin treatment.
After a century of nisin research - where are we now?

Abstract:It is almost a century since nisin was discovered in fermented milk cultures, coincidentally in the same year that penicillin was first described. Over the last 100 years this small, highly modified pentacyclic peptide has not only found success in the food industry as a preservative but has also served as the paradigm for our understanding of the genetic organization, expression, and regulation of genes involved in lantibiotic biosynthesis-one of the few cases of extensive post-translation modification in prokaryotes. Recent developments in understanding the complex biosynthesis of nisin have shed light on the cellular location of the modification and transport machinery and the co-ordinated series of spatio-temporal events required to produce active nisin and provide resistance and immunity. The continued unearthing of new natural variants from within human and animal gastrointestinal tracts has sparked interest in the potential application of nisin to influence the microbiome, given the growing recognition of the role the gastrointestinal microbiota plays in health and disease. Moreover, interdisciplinary approaches have taken advantage of biotechnological advancements to bioengineer nisin to produce novel variants and expand nisin functionality for applications in the biomedical field. This review will discuss the latest progress in these aspects of nisin research.
