Gut Bacteria Variation of Human Flora-associated Mice under High-fat Diet

Abstract:To study the body weight gain and fat accumulation of different human flora-associated mice (HFA-mice)under high-fat diet,the changes of their gut bacteria and their relevance to fat accumulation process were investigated.A couple of mother and daughter volunteers with higher and normal BMI were recruited to construct a HFA-mice model using C57BL/6 germ-free mice.The germ-free mice were divided into 2 groups,5 mice in each group.Group H1 were colonized with mother's gut flora while group H2 with daughter's.After colonization,a six-week high-fat diet feeding experiment was carried out.Body weights were measured once a week and feces were collected once every two weeks.High throughput sequencing was used for gut bacteria analysis.Abdominal fat of the mice was measured afterward,and the ratio of fat mass to body weight was used for express the fat accumulation process.Data showed that HFA-mice in group H1,colonized by gut flora from the overweight mother,gained body weight faster.The ratio of fat mass to body weight of group H1 was 1.32%,significantly higher than that of group H2,which was 0.70%.Gut bacteria of the two groups of mice showed different variation at genus level,including Bacteroidesm,Akkermansia,Clostridium,Parabacteroides,Escherichia,Paraprevotella,and Phascolarcto bacterium,among which,Escherichia,Paraprevotella and Phascolarcto bacterium demonstrated negative correlation to body weight gain and fat accumulation of the host.Results indicated that gut flora from different volunteers was closely associated with body weight change and fat accumulation of mice.Gut flora from people with normal BMI was considered to be able to slow down the process of body weight gain and fat accumulation of mice under high-fat diet.
