Effects of bone marrow mesenchymal stem cells co cultured with skin tissue on wound healing

Abstract:ObjectiveTo compare the effect of single bone marrow mesenchymal stem cells (BMSCs) to wound healing, to observe whether the BMSCs co cultured with newborn rat skin tissue block can accelerate the wound healing rate and inquiry into its mechanism.MethodsIn vitro, the neonatal rat dorsal skin was isolated, cultured and purified, and co cultured with rat BMSCs. Totally 72 wounds were made in12 rats. Each rat back from the head to the tail was divided into three groups: BMSCs co culture with the skin tissue group (Group A), single BMSCs group (Group B) and aseptic PBS control group (Group C). The two wounds in each group are symmetrically distributed along the spine. The time point to observe the wound healing was 3, 7, 14 and 21 days after injury. Then the statistical and immunohistochemical methods were carried to probe into the wound healing mechanism.Results The wound healing rate in 3 groups was statistically difference. Wound healing speed from fast to slow was followed by group A, group B and group C. Through immunohistochemical detection the positive expression cells of proliferating cell nuclear antigen (PCNA) were the most in group A, intermediate in group B, the least in group C. The positive expression cells of integrinβ1 were the least in Group A, intermediate in group B, the most in group C.Conclusion Compared with single BMSCs, the BMSCs co cultured with the skin tissue block can be differentiated into the skin tissue cells faster and promote the wound healing better.
