A study of enhancing effect of mesenchymal stem cells in healing of experimental epidermal "wound" in vitro

Abstract:Objective:To investigate the enhancing effect of human mesenchymal stem cells (hMSCs) on the healing of experimental epidermal defect. Methods:hMSCs were isolated and cultured from normal human marrow, and the second generation of hMSCs were identified with flow cytometry after being marked with CD14, CD29, CD34, CD44, CD45 and CD106 antigens. hMSCs were randomly divided into 3 groups.hMSCs (1×105/ml) were mixed and co-cultured with confluent human epidermal keratinocyte (HEKa) in direct co-cultured group(n=8). hMSCs [(2—3)×105 cells) was seeded in the upper chamber and indirectly co-cultured with HEKa cells in lower transwell chamber in the indirect co-cultured group (n=7). HEKa cells were cultured in HEKa culture group(n=7). Then a 100 μm "wound" of epidermis was reproduced by scraping confluent HEKa cells under inverted microscope. The number of cells migrating across the edge of the "wound" was counted, and the rate of "wound healing" was calculated by SigmaScan Pro 5 software. The proliferating effect of hMSCs on HEKa in vitro was determined by incorporation of [3H] thymidine.Results:Flow cytometry showed hMSCs were positive for CD29 , CD44 and CD106 (94.81%, 95.38%, 97.40% ), negative for CD14, CD34, CD45 (1.23%, 3.05%, 2.64%). In the direct co-cultured group, indirect co-cultured group and HEKa cultured group, the number of cells crossing the edge of wound was (10.00±0.25) , (5.50±0.20) and (5.20±0.35)cells/HP at 24 hours, (55.30±0.22),(27.00±0.34) and(26.50±0.25) cells/HP at 48 hours, ( 110.50±0.45) ,(42.50±0.50) and (40.20±0.38) cells/HP at 72 hours after culturing (P<0.01), respectively. The rate of wound healing was (60.00±0.04) %, (35.00 ±0.01)%、(33.00±0.05) %(P<0.01), and the proliferation speed of HEKa was (1 650±270), (1 240±210 ), (1180±220) cpm/105 cell in the three groups respectively. There was significant difference between the direct co-cultured group and indirect co-cultured group(P<0.01), but there was no significant difference between the indirect co-cultured group and HEKa cultured group(P>0.05). Conclusion: The hMSCs can be used to help repair the wound of epidermis by enhancing process of wound healing.
