Clinical diagnosis value of PAX1 methylation for detecting patients with cervical intraepithelial neoplasia

Abstract:Objective To explore the application value of methylation quantitative detection of PAX1 gene in the diagnosis of cervical precancerous lesions. Method From January 2013 to October 2015, 121 patients with high risk human papilloma virus (HPV) infection in the 6th People's Hospital A?liated to Shanghai Jiao Tong University were selected as the study objects. According to the pathological results, they were divided into normal cervical group (28 cases), low grade lesion group [cervical intraepi-thelial neoplasia (CIN) Ⅰ , 32 cases], high grade lesion group (CIN Ⅱ~Ⅲ, 34 cases), cervical cancer group [invasive cervical cancers (ICC), 27 cases]. Collected cervical tissue and cervical smear samples in each group, used reverse transcription polymerase chain reaction (RT-PCR) and pyrosequencing technique quantitatively to analyze PAX1 gene methylation at 9 sites and calculated the relevance of PAX1 gene methylation and all levels of the cervix associated lesions by ROC curve. Result Cervical 9 methylation sites PAX1 average gene was 4.66% in cervical cancer group, 3.55%in high grade lesion group, 2.10%in low grade lesion group, and 1.90%in normal cervical group. There were significant differences among four groups (P<0.05). Further, methylation level of PAX1 gene was associated with higher in cervical cancer group than other groups (P<0.05). The area under the ROC curve detection PAX1 gene methylation level identification of cervical cancer were 0.88, and associated&nbsp;with statistically significant (P<0.001). Conclusion Quantification of the levels PAX1 gene methylation of cervical tissue can effectively diagnose the high-level cervical lesions before cervical cancer, and it related to the expression of disease staging.
