Abstract:Thiopetheride antibiotics are a class of ribosomal peptides that are produced by microbial secondary metabolism, rich in sulfur, and highly modified amino acid residues. Thiopeptide antibiotics have a series of very important biological activities, including anti-infection, anti-tumor and immunosuppression, and their mechanism of action targeting ribosomes is different from those commonly used in clinical practice, which makes thiopeptide antibiotics have great development potential, but their poor water solubility and low bioavailability limit their clinical application. In order to improve the physicochemical properties of thiopeptide antibiotics, researchers tried to modified the structure of thiopeptide antibiotics by means of chemosemi-synthesis, combinatorial biosynthesis and precursors directed mutation biosynthesis. The complex structure of thiopeptide antibiotics provides many modification sites for its chemical semi-synthetic modification. In recent years, research on the chemical semi-synthetic modification of thiopeptide antibiotics has developed rapidly. In this paper, the research progress of thiopeptide antibiotic analogues obtained by chemical semi-synthetic modification in recent ten years is reviewed.

Abstract:Thiophene, as an important sulfur-containing heterocyclic, is widely used in pharmaceuticals, functional materials, and natural products. Furthermore, thiophene is of great interest to synthetic chemists due to its diverse reactivities. Over the past few decades, many synthetic strategies are developed for functionalized thiophenes. Several reviews have been reported. However, there is no review on thiophene synthesis summarized based on the sulfur sources. The current review envisioned to summary the synthesis methods for thiophene with the respect of sulfur sources and give support for choose the appropriate tools to obtain thiophene substrates. This review provides an overview of the recent synthesis methods developed and made available for thiophene products preparation. Thiophene being an important sulfur-containing heterocyclic motif finds use in pharmaceuticals, functional materials, organic chemistry and natural products, its efficient synthesis is equally important.+ image

Abstract:Compounds possessing a thioamide function play a crucial role in organic synthesis, serving as key building blocks. They are also important in the pharmaceutical chemistry and drug design, owing to their ability to mimic the amide function in biomolecules while retaining or developing biological activity. From the synthetic viewpoint, several methods have been developed for preparing thioamides using sulfuration agents. The purpose of this review is to give an update of the last decade of contributions focusing on the formation of thioamides employing different sulfur sources. When appropriate, the cleanness and practicality of the new methods are highlighted.

Abstract:The construction of N-aryl sulfonamides is usually an important reaction in organic synthesis because these compounds are privileged in the structure of many natural products and pharmaceutical molecules. Also, N-aryl sulfonamides are well-known due to their antibacterial, antitumor, and anti-HIV biological activities. Therefore, research on the laboratory synthesis of N-aryl sulfonamides is an important challenge in organic synthesis. In this review, we discussed on methods reported up to now for the synthesis of N-aryl sulfonamides with the goal of stimulating further progresses in this field. We believe that this review will open up a new road for the preparation of pharmaceutically and biologically active sulfonamides.

Abstract:The widespread applications of sulfur-containing molecules, especially thioethers, in drug synthesis and as important intermediates have garnered significant attention for these compounds. Initially, the formation of C-S bonds primarily relied on transition metal catalysis. However, in recent years, there has been remarkable progress in photocatalytic systems, making the construction of such chemical bonds through photocatalysis particularly attractive. This reaction often proceeds under mild conditions and allows for the highly regioselective construction of functionalized sulfones and thioethers. This article focuses on the photocatalytic preparation processes of various thioethers and systematically summarizes the reaction mechanisms involved.

Abstract:Thietanes are important aliphatic four-membered thiaheterocycles that are found in the pharmaceutical core and structural motifs of some biological compounds. They are also useful intermediates in organic synthesis. Various synthetic methods of thietanes have been developed, including inter- and intramolecular nucleophilic thioetherifications, photochemical [2 + 2] cycloadditions, ring expansions and contractions, nucleophilic cyclizations, and some miscellaneous methods. The recently developed methods provide some new strategies for the efficient preparation of thietanes and their derivatives. This review focuses on the synthetic methods to construct thietane backbones developed during 1966 to 2019.

Abstract:An efficient method for the synthesis of disubstituted thioureas via the reaction of N,N'-di-Boc-substituted thiourea 5 with alkyl and aryl amines under mild conditions has been developed. In the presence of NaH as a base, trifluoroacetic anhydride (TFAA) reacted with 5 providing intermediate 6, which then reacted with amines giving thioureas 7 in excellent yields. This reaction conditions tolerated other functional groups such as amide, ester, enol ether and hydroxyl groups. (C) 2008 Elsevier Ltd. All rights reserved.

Abstract:Benzothiazoles have played an important role in the field of biochemistry and medicinal chemistry due to their highly pharmaceutical and biological activity. The development of synthetic processes is undoubtedly one of the most significant problems facing researchers. In this review paper, we provided recent advances in the synthesis of benzothiazole compounds related to green chemistry from condensation of 2-aminobenzenethiol with aldehydes/ketones/acids/acyl chlorides and the cyclization of thioamide or carbon dioxide (CO2) as raw materials, and the future development trend and prospect of the synthesis of benzothiazoles were anticipated.

Abstract:Thietanes are important structural motifs of some biological compounds and useful intermediates in organic synthesis. Various synthetic methods of thietanes have been developed recently with readily available saturated three‐membered heterocycles as starting materials or key intermediates. Both saturated three membered heterocycle‐2‐methyl halides and sulfonates and thiiranes are widely utilized materials. Both nucleophilic and electrophilic ring expansions of thiiranes are efficient methods for synthesis of thietanes. In this Focus Review, we have described various strategies for the synthesis of strained four‐membered thiaheterocyclic thietanes from readily available strained and saturated three‐membered heterocycles.

Abstract:The use of sulfonium salts in organic synthesis has experienced a dramatic increase during the last years that can arguably be attributed to three main factors; the development of more direct and efficient synthetic methods that make easily available sulfonium reagents of a wide structural variety, their intrinsic thermal stability, which facilitates their structural modification, handling and purification even on large scale, and the recognition that their reactivity resembles that of hypervalent iodine compounds and therefore, they can be used as replacement of such reagents for most of their uses. This renewed interest has led to the improvement of already existing reactions, as well as to the discovery of unprecedented transformations; in particular, by the implementation of photocatalytic protocols. This review aims to summarize the most recent advancements on the area focusing on the work published during and after 2020. The scope of the methods developed will be highlighted and their limitations critically evaluated.

Abstract:Sulfur‐containing alkynes, are important starting materials and intermediates in organic reactions. Some of sulfur‐containing alkynes display interesting bioactivities and are potentially applied as drugs. Various synthetic methodologies towards sulfur‐containing alkynes have been developed, including C(sp)–S coupling, thiolation, S‐alkynylation, trifluoromethylthiolation, etc. In this review, the approaches towards alkynyl sulfides in recent 3 years, as well as to form alkynyl trifluoromethyl sulfides and alkynylthiocyanates were highlighted. Firstly, the property and application of alkynes were introduced. After presenting the superiority of sulfur‐containing alkynes, their synthetic methods were classified and presented in details. According to different kinds of sulfur‐containing alkynes, synthetic methodologies for alkynyl sulfides, alkynyl trifluoromethyl sulfides and alkynylthiocyanates were summarized and the proposed reaction mechanisms were demonstrated if available.

Abstract:A one‐pot method has been developed for synthesizing diverse substituted 5‐aminotetrazoles via the electrochemically induced desulfurizative addition‐cyclization of thioureas with azidotrimethylsilane. This methodology enables the construction of C–N and N–N bonds under mild reaction conditions without transition metal catalysts or external oxidizing agents. The applicability of this approach is demonstrated by its broad substrate scope, compatibility with various functional groups, scalability to gram‐scale synthesis, and practicality evidenced by the synthesis of an antibacterial agent

Abstract:Umpolung of thiols has become a common strategy for the synthesis of various S-containing compounds and thus a series of sulfur electrophilies have been designed and applied for diverse thiolation. To meet different demands for the incorporation of thio groups with regio- and stereo-selectivity, new catalyst systems and methodologies based on the utility of sulfur electrophilies have been well developed. In this review, achievements on various thiolation methodologies with sulfur electrophilies have been summarized, mainly including thiofunctionalization of alkenes or alkynes, insertion of carbene precursors, C-S cross coupling and electrophilic substitution, aiming to give a comprehensive insights for the application of electrophilic sulfur reagents.

Abstract:Thiocyanates are common in natural products, synthetic drugs and bioactive molecules. Many thiocyanate derivatives show excellent antibacterial, antiparasitic and anticancer activities. Thiocyanation can introduce SCN groups into parent molecules for constructing SCN-containing small organic molecules. Among them, the direct introduction method mainly includes nucleophilic reaction, electrophilic reaction and free radical reaction, which can simply and quickly introduce SCN groups at the target sites to construct thiocyanates, and has broad application prospects. In this review, we summarize the research progress of direct thiocyanation in recent years.

Abstract:In light of the important biological activities and widespread applications of organic disulfides, dithiocarbamates, xanthates, thiocarbamates and thiocarbonates, the continual persuit of efficient methods for their synthesis remains crucial. Traditionally, the preparation of such compounds heavily relied on intricate multi‐step syntheses and the use of highly prefunctionalized starting materials. Over the past two decades, the direct sulfuration of C–H bonds has evolved into a straightforward, atom‐ and step‐economical method for the preparation of organosulfur compounds. This review aims to provide an up‐to‐date discussion on direct C–H disulfuration, dithiocarbamation, xanthylation, thiocarbamation and thiocarbonation, with a special focus on describing scopes and mechanistic aspects. Moreover, the synthetic limitations and applications of some of these methodologies, along with the key unsolved challenges to be addressed in the future are also discussed. The majority of examples covered in this review are accomplished via metal‐free, photochemical or electrochemical approaches, which are in alignment with the overraching objectives of green and sustainable chemistry. This comprehensive review aims to consolidate recent advancements, providing valuable insights into the dynamic landscape of efficient and sustainable synthetic strategies for these crucial classes of organosulfur compounds.
