Abstract:An air-stable supported Cu(I) catalyst, CuI@PS-Phen, was designed and synthesized. CuI@PS-Phen can efficiently catalyze the click polymerization of diynes a and diazides b to produce soluble and thermally stable polytriazoles with high molecular weights ( M w up to 30800), and low copper residue content (down to 190 ppm) in high yields (up to 94.2%) under mild reaction conditions without the exclusion of oxygen.

Abstract:Cu-II-catalysed diazo transfer and Cu-I-catalysed azide-alkyne 1,3-dipolar cycloaddition (CuAAC) "click chemistry" were used to synthesis C7N aminocyclitol-derivatised 1,2,3-triazoles. In the course of this work, the -N=N- moiety was transferred onto C7N aminocyclitols such as validamine, valienamine and valiolamine by employing imidazole-1-sulfonyl azide as the diazo transfer reagent with catalysis by Cu-II, Zn-II and Ni-II, in moderate to good yields. The obtained azidocyclitols were coupled with various terminal alkynes under modified Meldal's conditions with good to excellent yields. The stereo- and regiochemistry of the products were confirmed by 2D-NMR (NOESY and HMBC). One-pot syntheses of the corresponding 1,2,3-triazoles, as safer and more efficient procedures, were also investigated and gave moderate to good yields.

Abstract:The Cu(I)-catalyzed azide-alkyne click polymerization has become a powerful tool for the preparation of functional polytriazoles. However, copper residues in the products could harm the performance of optoelectronic devices and be cytotoxic in biological applications. The complete removal of these residues after reaction from the polymers is therefore necessary but difficult. The metal-free azide-alkyne click polymerization (MFCP) provides a solution to this problem. Currently, the MFCP is facilitated by activated alkynes, and no activated comonomer, i.e. azide, has been reported. In this paper, an activated azide, 4,4'-diazidoperfluorobenzophenone , was designed and synthesized. Compared to the ordinary azide, such a monomer can remarkably accelerate the MFCP with alkynes. Thus, a new MFCP of activated azide and alkyne was established, and polytriazoles with high regioregularity (fraction of 1,4-isomer higher than 82%) and satisfactory molecular weights (M-w up to 34 000) were produced in high yields (up to 95.1%).

Abstract:The "thio-click" polymerization is a well-expanded concept of click polymerization. Among the click polymerizations, the thiol-yne click polymerization is less developed and still in its infancy stage. In general, UV light, elevated temperature, amine, or transition metal complexes is needed to catalyze the thiol-yne click polymerization, which greatly complicates the experimental operation and limits its application. In this work, a facile and powerful thiol-yne click polymerization was developed, which could be carried out under very mild conditions without using external catalyst. Simply mixing the aromatic diynes (1a-1e) and dithiols (2-4) with equivalent molar ratio in THF at 30 degrees C will readily produce soluble and regioregular functional poly(vinylene sulfide)s (PIa-PIe, PII, and PIII) with high molecular weights (M-w, up to 85 200) in excellent yields (up to 97%) after as short as 2 h. Furthermore, no double addition product of an ethynyl group was found. This catalyst-free thiol-yne click polymerization has remarkably simplified the reaction conditions and will facilitate the preparation of functional materials applied in diverse areas.

Abstract:Click chemistry focuses on the development of highly selective reactions using simple precursors for the exquisite synthesis of molecules. Undisputedly, the Cu-I-catalyzed azide-alkyne cycloaddition (CuAAC) is one of the most valuable examples of click chemistry, but it suffers from some limitations as it requires additional reducing agents and ligands as well as cytotoxic copper. Here, we demonstrate a novel strategy for the azide-alkyne cycloaddition reaction that involves a photoredox electron-transfer radical mechanism instead of the traditional metal-catalyzed coordination process. This newly developed photocatalyzed azide-alkyne cycloaddition reaction can be performed under mild conditions at room temperature in the presence of air and visible light and shows good functional group tolerance, excellent atom economy, high yields of up to 99 %, and absolute regioselectivity, affording a variety of 1,4-disubstituted 1,2,3-triazole derivatives, including bioactive molecules and pharmaceuticals. The use of a recyclable photocatalyst, solar energy, and water as solvent makes this photocatalytic system sustainable and environmentally friendly. Moreover, the azide-alkyne cycloaddition reaction could be photocatalyzed in the presence of a metal-free catalyst with excellent regioselectivity, which represents an important development for click chemistry and should find versatile applications in organic synthesis, chemical biology, and materials science.

Abstract:The azide-alkyne click polymerization (AACP) has emerged as a powerful tool for the synthesis of functional polytriazoles. While, for the Cu(I)-catalyzed AACP, the removal of the catalytic Cu(I) species from the resulting polytriazoles is difficult, and the research on the recyclability and reusability of the catalyst remains intact. Herein, we reported the first example of using recyclable and reusable supported Cu(I) catalyst of CuI@A-21 for the AACP. CuI@A-21 could not only efficiently catalyze the AACP but also be reused for at least 4 cycles. Moreover, pronounced reduction of copper residues in the products was achieved. Apart from being a green and cost-effective polymer synthesis strategy, this method will also broaden the application of AACP in material and biological sciences and provide guidelines for other polymerizations with metal catalysts.

Abstract:Abstract Click reactions, with remarkable merits including high efficiency, atom economy, mild reaction conditions, good functionality tolerance, simple isolation process, and regio‐ and/or stereoselectivity, have drawn intense attention of polymer chemists and found widespread applications in polymer synthesis. Besides the famous Cu(I)‐catalyzed azide–alkyne click reaction, plentiful click reactions, such as Diels–Alder, thiol‐based, nonaldol carbonyl, amino‐yne, hydroxyl–yne, and sulfur(VI) fluoride‐exchange click reactions, have also been introduced into the field of polymer chemistry. With these efficient click reactions in hand, diverse functional groups can be readily incorporated into polymer chains to endow them with desired functionalities. Copolymers with complex structures, such as block, graft, star, and cyclic polymers, can be synthesized using click reaction as polymer conjugation method. These click reactions have also been developed into facile polymerization techniques, that is, click polymerization, for the direct synthesis of linear, hyperbranched, and cross‐linked polymers with various properties. This article intends to give a brief summary of click reactions applied in polymer synthesis, including preparation of polymers and postpolymerization modification of preformed polymers.

Abstract:The copper-catalyzed azide-alkyne cycloaddition (CuAAC) reaction is regarded as a prime example of "click chemistry", but the asymmetric click cycloaddition of internal alkynes still remains challenging. A new asymmetric Rh-catalyzed click cycloaddition of N-alkynylindoles with azides was developed, providing atroposelective access to C-N axially chiral triazolyl indoles, a new type of heterobiaryl, with excellent yields and enantioselectivity. This asymmetric approach is efficient, mild, robust and atom-economic, and features very broad substrate scope with easily available Tol-BINAP ligands.

Abstract:The transition-metal catalyzed and metal-free click polymerizations have been developed as powerful tools for the construction of functional polymers with linear and hyperbranched structures. The latter provides a thorough solution for the completely removing metallic residues from the products encountered in the former. Compared to the activated alkyne-azide metal-free click polymerization, the activated azide-alkyne one is rarely studied. In this Communication, a perfluorophenyl-activated azide of hexane-1,6-diyl-bis(4-azido-2,3,5,6-tetrafluorobenzoate) is rationally designed and facilely prepared. Through systematical optimization of the reaction conditions, an efficient metal-free perfluorophenylazide-alkyne polycycloaddition is established, and polytriazoles with high molecular weights (up to 166 000) and excellent solubility are obtained in excellent yields (up to 93%) under mild reaction conditions. Interestingly, the regioselectivity of the reaction could be fine-tuned by the solvents and diyne monomers. Therefore, this work provides not only a powerful tool for the preparation of functional polytriazoles, but also an attractive method for fine-tuning their regioregularity.

Abstract:The hydroamination reaction of alkynes has been widely studied, but has not been developed as a powerful tool for polymer synthesis. Herein, a novel Cu(I)-catalyzed polyhydroamination has been developed for the facile preparation of nitrogen-containing polymers. This polymerization proceeded smoothly in bulk with 100% atom utilization in a regio-specific and stereo-selective fashion, producing polyenamines with well-defined structures and high molecular weights (up to 13 470) in high yields (up to 97%). Thus, this polymerization is a kind of amino-yne click polymerization. The resulting polymers possess excellent solubility, high thermal stability, strong UV-light shielding efficiency, and good visible light transparency. Considering the high efficiency, ready monomer availability and excellent performance of the products, this Cu(I)-catalyzed amino-yne click polymerization opens up enormous opportunities for preparing functional polymers.

Abstract:Introduction: An effective Cu-complex, [Cu(NH3)4SO4 • H2O] was prepared conveniently from the inexpensive and easily available starting reagents in a simple route. Materials and methods: Excellent reactivity of the catalyst was observed towards two competent clickcycloadditions: (a) oxidative cycloaddition of azides with electron-poor olefins and (b) one-pot cycloaddition of alkynes with boronic acid and sodium azide under "click-appropriate" conditions. Results: No external oxidant, short reaction time, high product yield, wide substrate scope, and aqueous solvent media make the azide-olefin cycloaddition approach a greener route in contrast to the reported methods. Conclusion: The newly developed mild, green, and rapid three-component strategy shows product diversity with superb yields at room temperature by reducing the synthetic process time and using only 1 mol % of the synthesized copper complex.

Abstract:Click polymerization, a powerful synthetic technique to construct polymers with unique structures and advanced functions, is of crucial importance in the areas of polymer and material sciences. A variety of click polymerizations such as azide-alkyne, thiol-yne, amino-yne, and hydroxyl-yne reactions have been established, wherein the catalytic systems play an indispensable role in realizing these highly practical reactions based on triple-bond building blocks, as they directly influence the efficiencies of the click polymerizations and the performances of the resultant polymers. The vital employment of catalysts is reviewed and their developments from innovative discoveries to the eminent position are outlined. Moreover, the challenges and perspectives in this area are also briefly discussed.

Abstract:One-pot three-component regioselective azide-alkyne cycloadditions are central reactions for synthesizing pharmaceuticals and fine chemicals and are also applied for in vivo metabolic labeling biotechnology. Homogeneous catalysts based on copper species coordinated with ancillary ligands are regularly used to perform this reaction, offering superior catalytic activity and selectivity compared to conventional heterogeneous counterparts based on supported copper nanoparticles. However, the challenge of catalyst recovery limits the use of these homogeneous compounds in many large-scale applications. In this work, we report the high catalytic performance of a family of Cu-based single-atom catalysts for triazole synthesis, with an emphasis on the fundamental understanding of the structure and function of the catalyst. The catalysts were prepared via tricyanomethanide polymerization to create a joint electronic structure where the mesoporous graphitic carbon nitride carrier acts as a ligand for the atomically dispersed copper species. The material properties and the precise metal location/coordination (i.e., deposited in the heptazine pore of carbon nitride, substituted in the framework of carbon nitride, hosted in a vacancy, or entrapped in sandwich-like arrangement) were characterized through a battery of spectroscopic and theoretical methods. The catalysts were employed in the synthesis of 1,2,3-triazoles employing azide-alkyne click reaction under base-free conditions. The single-atom Cu catalysts demonstrated improved activity and selectivity compared to the homogeneous reference catalyst. Density functional theory calculations corroborated the results and showed that the reaction proceeds through a barrier given by the activation of the acetylenic moiety on Cu-1. The activity of this step was primarily affected by the coordination of the metal with the support. Therefore, understanding the metal coordination in single-atom catalysts is critical to further optimizing single-atom catalysts and greening synthetic chemistry.

Abstract:Click chemistry is a concept in which modular synthesis is used to rapidly find new molecules with desirable properties<a href="/articles/s41586-019-1589-1#ref-CR1">1</a>. Copper(i)-catalysed azide–alkyne cycloaddition (CuAAC) triazole annulation and sulfur(vi) fluoride exchange (SuFEx) catalysis are widely regarded as click reactions<a href="#ref-CR2">2</a>,<a href="#ref-CR3">3</a>,<a href="/articles/s41586-019-1589-1#ref-CR4">4</a>, providing rapid access to their products in yields approaching 100% while being largely orthogonal to other reactions. However, in the case of CuAAC reactions, the availability of azide reagents is limited owing to their potential toxicity and the risk of explosion involved in their preparation. Here we report another reaction to add to the click reaction family: the formation of azides from primary amines, one of the most abundant functional groups<a href="/articles/s41586-019-1589-1#ref-CR5">5</a>. The reaction uses just one equivalent of a simple diazotizing species, fluorosulfuryl azide<a href="#ref-CR6">6</a>,<a href="#ref-CR7">7</a>,<a href="#ref-CR8">8</a>,<a href="#ref-CR9">9</a>,<a href="#ref-CR10">10</a>,<a href="/articles/s41586-019-1589-1#ref-CR11">11</a> (FSO2N3), and enables the preparation of over 1,200 azides on 96-well plates in a safe and practical manner. This reliable transformation is a powerful tool for the CuAAC triazole annulation, the most widely used click reaction at present. This method greatly expands the number of accessible azides and 1,2,3-triazoles and, given the ubiquity of the CuAAC reaction, it should find application in organic synthesis, medicinal chemistry, chemical biology and materials science.
