Abstract:Significance Optical microresonators have emerged as promising platforms for label-free detection of molecules. However, approaching optimum sensitivity is hindered due to the weak tail of evanescent fields. Here, we report the implementation of the interface modes for ultrasensitive sensing in a microbubble resonator. With the electromagnetic field peaked at the interface between the optical resonator and the analyte solution, interface modes enable sensing of biomolecules with a detection limit of 0.3 pg/cm 2 . Single-molecule detection is further demonstrated using the plasmonic-enhanced interface modes. In addition, intrinsically integrated into a microfluidic channel, the sensor exhibits ultrasmall sample consumption down to 10 pL, providing an automatic platform for biomedical analysis.

Abstract:Surface plasmon resonance (SPR) and localized SPR (LSPR) effects have been shown as the principles of some highlysensitive sensors in recent decades. Due to the advances in nano-fabrication technology, the plasmon nano-array sensors based on SPR and LSPR phenomena have been widely used in chemical and bioloical analysis. Sensing with surface-enhanced field and sensing for refractive index changes are able to identify the analytes quantitatively and qualitatively. With the newly developed ultrasensitive plasmonic biosensors, platforms with excellent performance have been built for various biomedical applications, including point-of-care diagnosis and personalized medicine. In addition, flexible integration of plasmonics nano-arrays and combining them with electrochemical sensing have significantly enlarged the application scenarios of the plasmonic nano-array sensors, as well as improved the sensing accuracy.

Abstract:In this work, we propose a MgF2/Au/MgF2/Analyte based, angle-interrogation surface plasmon resonance sensor. This symmetrical structure results in higher surface electric field strengths, and longer surface propagation lengths and depths. A refractive index resolution of 8.1×10−8RIU in fluid protocol and 3.5×10−7RIU in atmosphere protocol is acquired by a simple SPR imaging system. In order to demonstrate the biomedical applications of this system, we detect Escherichia coli with different concentrations using dopamine film on the sensor surface to immobilize antibodies. A detection limit of 103cfu/mL of E. coli bacteria is acquired from this system.

Abstract:We present the fabrication and performance of an ultra-sensitive, high-absorbing, yet simple-to-fabricate plasmonic nanocavity antenna structure fabricated inside fluidic channels. The novel fabrication is based on nanoimprint lithography and multiple resist layer etching. And large fluorescence enhancement (~20X) of DNA molecules (YOYO-1 dye labeled) on the plasmonic antenna was observed, demonstrating its potential in fast and real-time biochemical sensing applications.

Abstract:Molecular binding kinetics quantification is pivotal in the design of antibodies, small-molecule drugs, and early diagnosis of complex disease. This study presents a portable microfluidic system, featuring a nanoplasmonic sensor with cavity reflection enhancement (NSCRE), designed for the ultrasensitive detection and analysis of molecular interactions. This device comprises liquid flow paths, generic fiber spectrometers, a versatile NSCRE sensor chip, and a novel data analysis system. Furthermore, a neural network data processing system is developed based on a deep learning model to decrease significant data fluctuations caused by air segments. This "all-in-one" platform, integrating multifunctional, easy-to-use, and renewable NSCRE biosensors, boasts detection capabilities for both slow and rapid kinetics. It can automatically monitor interactions between molecules across a broad range of molecular weights (0.259–150 kDa), allowing for real-time, ultra-sensitive, and highly selective analysis. In addition, the platform with CRE effect facilitates a 7.2-fold increase in reflection intensity signals compared to that of a sensor without CRE. The immunoglobulin G detection with a limit of detection of 73 pM based on this device, showing a 50-fold increase in sensitivity compared to previously reported values. The miniaturized automatic detection system paves a new path for the use of molecular interaction analysis in life science research, drug discovery, early diagnosis, and other fields.

Abstract:DNA detection based on DNA hybridization has wide applications in various fields, such as clinical diagnostics, food safety, and environmental monitoring, etc. At present, common DNA detection approaches include fluorescence-based microarray technique, electrochemical method and surface plasmon resonance (SPR), etc. However, these methods require additional precise equipments and relatively complicated detection process. In this work, we developed a biosensor platform based on a liquid crystal (LC)-amplified optofluidic whispering gallery mode (WGM) resonator to achieve ultra-sensitive, label-free, and quick-response DNA hybridization detection. Liquid crystal is a material with high sensitivity, rapid response, and low cost. It exhibits significant directional and positional ordering, and it is sensitive and responsive to external stimuli. LC molecules exhibit a uniform orientation on the surface of the resonator, when the surface is covered with an appropriate amount of ssDNA. Once complementary DNA and ssDNA hybridize on the surface, the homotropic orientation of the LCs will be destroyed. Due to the simultaneous interaction of the WGM and the LCs in the optofluidic resonator, changes caused by the DNA hybridization can be amplified, resulting in a shift in the resonance wavelength. In this experiment, we used the spectral wavelength shift as a sensing parameter to achieve the detection of target DNA, and a lower detection limit compared to traditional DNA detection methods was obtained. At the same time, this biosensor platform also shows good selectivity. Our research results suggest that the LC based WGM optical microcavity sensing platform can provide an ultra-sensitive, label free solution for DNA detection.

Abstract:We developed a nanoplasmonic sensor platform employing the extraordinary optical properties of one-dimensional nanogratings patterned on 30nm-thick ultrathin Ag films. Excitation of Fano resonances in the ultrathin Ag nanogratings results in transmission spectra with high amplitude, large contrast, and narrow bandwidth, making them well-suited for rapid and highly-sensitive sensing applications. The ultrathin nanoplasmonic sensor chip was integrated with a polydimethylsiloxane (PDMS) microfluidic channel, and the measured refractive index resolution was found to be 1.46x10-6 refractive index units (RIU) with a high temporal resolution of 1 sec. This compares favorably with commercial prism-based surface plasmon resonance sensors, but is achieved using a more convenient collinear transmission geometry and a significantly smaller sensor footprint of 50x50um2. In addition, an order-of-magnitude improvement in the temporal and spatial resolutions was achieved relative to state-of-the-art nanoplasmonic sensors, for comparable detection resolutions.

Abstract:DNA is fundamental for storing and transmitting genetic information. Analyzing DNA or RNA base sequences enables the identification of genetic disorders, monitoring gene expression, and detecting pathogens. Traditional detection techniques like polymerase chain reaction (PCR) and next-generation sequencing (NGS) have limitations, including complexity, high cost, and the need for advanced computational skills. Therefore, there is a significant demand for enzyme-free and amplification-free strategies for rapid, low-cost, and sensitive DNA detection. DNA biosensors, especially those utilizing plasmonic nanomaterials, offer a promising solution. This study introduces a novel DNA-functionalized waveguide-enhanced nanoplasmonic optofluidic biosensor using a nanogold-linked sorbent assay for enzyme-free and amplification-free DNA detection. Integrating plasmonic gold nanoparticles (AuNPs) with a glass planar waveguide (WG) and a microfluidic channel, fabricated through cost-effective, vacuum-free methods, the biosensor achieves specific detection of complementary target DNA sequences. Utilizing a sandwich architecture, AuNPs labeled with detection DNA probes enhance sensitivity by altering evanescent wave distribution and inducing plasmon resonance modes. The biosensor demonstrated exceptional performance in DNA detection, achieving a limit of detection (LOD) of 33.1 fg/mL (4.36 fM) with a rapid response time of approximately 8 min. This ultrasensitive, rapid, and cost-effective biosensor exhibits minimal background nonspecific adsorption, making it highly suitable for clinical applications and early disease diagnosis. The innovative design and fabrication processes offer significant advantages for mass production, presenting a viable tool for precise disease diagnostics and improved clinical outcomes.

Abstract:Due to the specific cofactor dependence and the potent catalytic capability, DNAzymes have shown great promise in the application of biosensors. However, it poses significant challenges for researchers to enhance the sensitivities of DNAzyme-based biosensing platforms. In this work, an ultra-sensitive, label-free, and quick-responsive DNAzyme biosensor is developed based on whispering gallery mode (WGM) optofluidic resonator with a hybrid amplification from liquid crystals (LCs) and Au nanoparticles (AuNPs). When a DNAzyme is cleaved by biotargets, the event of DNA hybridization is triggered and will result in the orientation transition of LC molecules (in the resonator core). According to the quadruple amplification from the disturbance of AuNPs, the excess polarizability of adhering biomolecules, the orientation variation of LC molecules, and the WGM resonance, the spectral wavelength shift can be employed as the sensing parameter to indicate the information of biotarget. L-histidine molecules are applied as the analyte. An ultra-low detection limit at a sub-femtomole level (~ 5 × 10−16 M) has been achieved using this highly-selective biosensor, which is eight orders of magnitude lower compared to the reported LC-based L-histidine sensor. In addition to L-histidine, this proposed optofluidic scheme with LC-AuNP hybrid amplification can also extend the applicability of DNAzyme-based biosensors for the detection of various molecules.

Abstract:We demonstrated a temperature-compensated optofluidic DNA biosensor available for microfluidic chip. The optofluidic sensor was composed of an interferometer and a fiber Bragg grating (FBG) by femtosecond laser direct writing micro/nano processing technology. The sensing arm of the interferometer was suspended on the inner wall of the microchannel and could directly interact with the microfluid. With the immobilization of the single stranded probe DNA (pDNA), this optofluidic biosensor could achieve specific detection of single stranded complementary DNA (scDNA). The experimental results indicated that a linear response within 50 nM and the detection limit of 1.87 nM were achieved. In addition, the optofluidic biosensor could simultaneously monitor temperature to avoid temperature fluctuations interfering with the DNA hybridization detection process. And, the optofluidic detection channel could achieve fast sample replacement within 10 s at a flow rate of 2 μL/min and sample consumption only required nanoliters. This optofluidic DNA biosensor had the advantages of label-free, good specificity, dual parameter detection, low sample consumption, fast response, and easy repeatable preparation, which was of great significance for the field of DNA hybridization research and solving the temperature sensitivity problem of biosensors and had good prospects in biological analysis.

Abstract:Practical applications of chemical and biological detections through surface-enhanced Raman scattering (SERS) require high reproducibility, sensitivity, and efficiency, along with low-cost, straightforward fabrication. In this work, we integrated a poly-(dimethylsiloxane) (PDMS) chip with quasi-3D gold plasmonic nanostructure arrays (Q3D-PNAs), which serve as SERS-active substrates, into an optofluidic microsystem for online sensitive and reproducible SERS detections. The Q3D-PNA PDMS chip was fabricated through soft lithography to ensure both precision and low-cost fabrication. The optimal dimension of the Q3D-PNA in PDMS was designed using finite-difference time-domain (FDTD) electromagnetic simulations with a simulated enhancement factor (EF) of 1.6×10(6). The real-time monitoring capability of the SERS-based optofluidic microsystem was investigated by kinetic on/off experiments through alternatively flowing Rhodamine 6G (R6G) and ethanol in the microfluidic channel. A switch-off time of ∼2 min at a flow rate of 0.3 mL min(-1) was demonstrated. When applied to the detection of low concentration malathion, the SERS-based optofluidic microsystem with Q3D-PNAs showed high reproducibility, significantly improved efficiency and higher detection sensitivity via increasing the flow rate. The optofluidic microsystem presented in this paper offers a simple and low-cost approach for online, label-free chemical and biological analysis and sensing with high sensitivity, reproducibility, efficiency, and molecular specificity.

Abstract:We developed a micro-nanofluidic bioreactor-detector system for isothermal DNA amplification and sensitive real-time detection of the amplified products for sequence-specific molecular identification. Aluminum (Al) chips with a range of volumes from 7.07 μL to 39 nL and an associated real-time confocal optical detector are described. The detector provided highly sensitive fluorescence detection and low background noise. One of the important aspects of the system was the development of a surface processing technique that afforded chips with an inert surface to improve amplification stability in micro-nanoliter reaction assays. The micro-nanofluidic system exhibited more sensitive exponential DNA amplification characteristics than a standard PCR tube amplification system with a volume of 25 μL, the response time was clearly reduced at the same DNA template concentration, and the sensitivity in the number of copies of the DNA template was improved by >600-fold. Efficient amplification of nucleic acid was achieved with as few as three copies of the DNA template. This system may be useful for the development of novel lab-on-a-chip devices and shows promise for single-molecule amplification in droplet assays, with potential applications in nanobiotechnology, nanomedicine, and clinical molecular diagnostics.
Microsphere-enhanced Label-Free High-Throughput Molecular Detection

Abstract:In this paper, we developed a microsphere enhanced and label free high throughput molecular detection based on SPRI and microarray chips, and a self-built surface plasmon resonance imaging instrument was set. One label-free protein chip forming a 7× probe array was designed and fabricated using a commercial microarray robot spotter on chemical modified gold-coated glass slide. Antibody molecules were successfully detected in label-free and high-throughput method by using this chip. The detection signals on the chip was successfully enhanced by using microspheres with a diameter of 1 μm.
A micro-nano interface integrated SERS-based microfluidic sensor for miRNA detection using DNAzyme walker amplification

Abstract:Background: Precise and specific miRNA detection plays a vital role in exploring development mechanisms of cancer disease, thereby it can significantly improve relevant prevention and treatment strategies. Results: In this work, a surface-enhanced Raman spectroscopy (SERS)-based microfluidic chip has been devised with a microcone array SERS substrate (MCASS) for the miR-141 detection. This substrate excels in unique SERS activity and large surface area for DNA oligonucleotide modification. As the presence of miR-141, the DNAzyme walker induced cleavage reaction took place on the finely designed and prepared dual DNA conjugated SERS nanoprobes. The SERS nanoprobes can anchor on MCASS by the DNA hybridization that achieved an impressive detection limit in the femtomolar level. Significance: With this integrated SERS-based microfluidic chip, we provided a miRNA detection strategy using DNAzyme walker amplification technology. It is believed that this strategy could be a powerful tool for miRNA detection and related cancer screening test.

Abstract:The robust identification and quantification of various biomarkers is of utmost significance in clinical diagnostics and precision medicine. Fluorescent immunoassays are widely used and considered as a gold standard for biomarker detection due to their high specificity and accuracy. However, current commercial immunoassay tests suffer from limited detection sensitivity and complicated, labor-intensive operation procedures, making them impractical for point-of-care diagnosis, particularly in resource-limited regions. Recently, microfluidic immunoassay devices integrated with plasmonic nanostructures have emerged as a powerful tool for sensitive detection of biomarkers, addressing specific issues, such as integration schemes, easy operation, multiplexed detection, and sensitivity enhancement. In this paper, we provide a discussion on the recent advances in the plasmonic nanostructures integrated with microfluidic devices for fluorescent immunoassays. We shed light on the nanofabrication strategies and various fluidic designs for rapid, sensitive, and highly efficient sensing of antigens. Finally, we share our perspectives on the potential directions of these integrated devices for practical applications.

Abstract:The detection of small molecules has increasingly attracted the attention of researchers because of its important physiological function. In this manuscript, we propose a novel optical sensor which uses an optofluidic microbubble resonator (OFMBR) for the highly sensitive detection of small molecules. This paper demonstrates the binding of the small molecule biotin to surface-immobilized streptavidin with a detection limit reduced to 0.41 pM. Furthermore, binding specificity of four additional small molecules to surface-immobilized streptavidin is shown. A label-free OFMBR-based optical sensor has great potential in small molecule detection and drug screening because of its high sensitivity, low detection limit, and minimal sample consumption.

Abstract:A highly-sensitive optical waveguide biosensor integrated with microfluidic channels based on silicon-on-insulator (SOI) was investigated in this paper. Experimental results of the label-free detection exhibits this novel biosensor with the superior reliability for quantitative and kinetic measurement of the interaction between biological molecules, dramatically improving the sensitivity due to the Vernier effect induced by cascaded double-microring resonators.

Abstract:MicroRNAs (miRNAs), a class of small, endogenous, noncoding RNA molecules, can serve as biomarkers for potential applications in cancer diagnosis, prognosis, and prediction due to its abnormal expression. As a result, a novel label-free biosensor with nanometer scale was prepared and employed in the detection of trace oligonucleotides based on the localized surface plasmon resonance (LSPR). The dielectric constant on the surface of DNA modified gold nanoparticle would change when probe single-strand DNA hybridized with target oligonucleotides, which resulted in the notable red shift of scattering peak position. The biosensor with excellent selectivity can be used in a real-time monitoring hybridization process. Notably, this method provided label-free detection of DNA and miRNA at single nanoparticle level with limit of detection up to 3 nM. Due to the advantages of LSPR scattering spectra, single nanoparticle biosensor can be designed for trace cancer-relevant miRNAs detection in the future.
