Abstract:The application of immobilized lipase in the enzymatic production of biodiesel has shown numerous advantages. In this study, surface of Polyacrylonitrile (PAN) hollow membrane was first modified using nitrile-click chemistry in order to fit for interaction with enzyme proteins. Then sodium alginate (SA) was introduced and the membrane was post-treated by CaCl2. When the prepared PAN-PEI-SA-CaCl2 was used for lipase immobilization, the protein loading was 36.90 mg/g, and the enzyme activity reached up to 54.47 U/g, which was 2.5 times as much as that of Novozym® 435. As a result, the constructed immobilized lipase obtained a maximum biodiesel yield of 78.5%, which was 2.4 times that of the Novozym® 435 in transesterification reactions. Moreover, the biodiesel yield decreased by only 11% after the immobilized enzyme was continuously used for 20 times. This study exhibits that this technic has broad application prospects in the field of conversion of biomass resources.
Effect of Immobilization by Modified Ultrastable-Y Molecular Sieve on the Resolution of (R,s)-2-octanol in Nonaqueous Media by Lipase

Abstract:The lipase from Penicillium expansum PED-03 (PEL) was immobilized into a modified ultrastable-Y molecular sieve and the resolution of (R,S)-2-octanol was carried out in a bioreactor in nonaqueous media by the immobilized lipase to investigate the effect of immobilization on the catalytic property of PEL duing the enzymatic reaction. It was found that the conversion (c), enantiomeric excess (ee), and enantioselectivity (E) of the resolution catalyzed by the PEL immobilized on the modified ultrastable-Y molecular sieve were much higher than those of the reaction catalyzed by free PEL and PEL immobilized on other supports. Being immobilized on the modified Ultrastable-Y molecular sieve, the lipase exhibited obvious activity in a wider pH range and at a much higher temperature than the free PEL, and showed a marked enhancement of stability against thermal inactivation, by which the suitable pH value of the buffer used for immobilization could be "memorized". The conversion of the reaction catalyzed by PEL immobilized on the modified ultrastable-Y molecular sieve reached 97.68% of the theoretical value with excellent enantioselectivity (Average E of 8 batches >460) in nonaqueous media at "memorial" pH 9.5, 50 degrees C for 24 h, which showed a good application potential to the production of optically pure (R,S)-2-octanol.

Abstract:Various hydrophobic supports have been used for lipase immobilization since the active site of lipase can be opened in a hydrophobic environment. Nevertheless, the increase of lipase activity is still limited. This study demonstrates a hyperactivation-protection strategy of lipase after immobilization on poly(n-butyl acrylate)-polyaldehyde dextran (PBA-PAD) core-shell nanoparticles. The inner hydrophobic PBA domain helps to rearrange lipase conformation to a more active form after immobilization into the PAD shell. More importantly, the outer PAD shell with dense polysaccharide chains prevents the immobilized lipase from contact with outside aqueous medium and revert its conformation back to an inactive form. As a result, under optimal conditions the activity of lipase immobilized in PBA-PAD nanoparticles was enhanced 40 times over the free one, much higher than in any previous report. Furthermore, the immobilized lipase retained more than 80 % of its activity after 10 reaction cycles.

Abstract:An excellent oil–water interface is one of the prerequisites for effective lipase catalysis. Therefore, this study aimed to improve lipase activity in terms of catalytic interface optimization. A novel approach for constructing oil–water interfaces was proposed. The structural similarity and the hydrophilic differences between polyvinyl pyrrolidone gel–fiber membranes (GFMs) and poly(lauryl methacrylate) (PLMA) organogel inspired us to hybridize the two to form PVP/PLMA hybrid gel–fiber membranes (HGFMs) based on electrospinning and gelation. The prepared PVP/PLMA-HGFMs were capable of being adopted as novel carriers for lipase catalysis due to their ability to swell both in the aqueous phase (swelling ratio = 187.5%) and the organic phase (swelling ratio = 40.5%). Additionally, Confocal laser scanning microscopy (CLSM) results showed that abundant network pores inside the carriers enabled numerous effective microscopic oil–water interfaces. The catalytic activity of Burkholderia cepacia lipase (BCL) in PVP/PLMA-HGFMs ranged between 1.21 and 8.70 times that of the control ("oil-up/water-down" system) under different experimental conditions. Meanwhile, PVP/PLMA-HGFMs increased lipase activity by about eight times at −20 °C and had good application characteristics at extreme pH conditions.

Abstract:Efficient lipase catalysis requires sufficient oil–water interface engineered through structural design. Inspired by the architectural features of fabrics, a novel lipase-membrane catalytic system with interwoven polyacrylic acid (PAA) gel fibers and polyvinyl pyrrolidone (PVP) gel fibers was developed in this study by using double-needle electrospinning and gelation. It has been demonstrated that PAA/PVP hybrid gel fiber membranes (HGFMs) have a high swelling capacity for both water and oil phases, which created numerous discontinuous oil–water contact surface units in limited space of HGFMs, consequently forming effective interfacial catalytic systems. Volume competition between the water and oil phases suggests that balancing the proportions of these phases is very important for effective construction of oil–water interfaces and conditioning catalysis. Regulation of multiple factors of PAA/PVP HGFMs resulted in a catalytic efficiency of up to 2.1 times that of a macroscopic "oil-up/water-down" system (room temperature, pH = 7), and 2.9 times when three membranes are superimposed, as well as excellent pH and temperature stability. HGFMs were stacked to build a high-performing catalytic performance reactor. We expect that this study will be a beneficial exploration for expanding the lipase catalytic system.

Abstract:Enzymatic membrane bioreactors (EMBRs) possess the characteristic of combining catalysis with separation, and therefore have promising application potentials. In order to achieve a high-performance EMBR, membrane property, as well as operating parameters, should give special cause for concerns. In this work, an EMBR based on hollow fiber polysulfone microfiltration membranes with radial gradient pore structure was fabricated and enzyme immobilization was achieved through pressure-driven filtration. Lipase from Candida rugosa was used for immobilization and EMBR performance was studied with the enzymatic hydrolysis of glycerol triacetate as a model reaction. The influences of membrane pore diameter, substrate feed direction as well as operational parameters of operation pressure, substrate concentration, and temperature on the EMBR activity were investigated with the production of hydrolysates kinetically fitted. The complete EMBR system showed the highest activity of 1.07 × 104 U⋅g−1. The results in this work indicate future efforts for improvement in EMBR.

Abstract:The work reported in this paper is aimed at exploring the feasibility of immobilizing alkali lipase from Penicillium expansum on a bimodal ceramic foam, which has both macro- and micro-pore structures. After being chemically modified with a silane coupling agent, the ceramic foam was used as a support for lipase immobilization. To determine the preferable immobilization conditions, the effects of the amount of enzyme for loading, immobilization time, temperature, and pH on enzyme activity were investigated. The results showed that the chemically modified ceramic foam has a high loading capacity and a strong binding ability for the lipase. Thanks to the low internal mass transfer resistance, the ceramic foam has greatly enhanced the rate of immobilization. As a comparison, the immobilized-lipase activity was much higher than that on many frequently used porous materials like diatomite, alumina and activated carbon.

Abstract:In this study, a biphasic enzymatic membrane reactor was made by immobilizing Candida Rugosa lipase onto the dense surface of polysulfone ultrafiltration membrane by filtration and then cross-linking with glutaraldehyde solution. The reactor was further applied for the hydrolysis of olive oil, the performance of which was evaluated in respect of apparent reaction rate based on the amount of fatty acids extracted into the aqueous phase per minute and per membrane surface. It was found that the ultrafiltration and cross-linking process greatly improved the reaction rate per unit membrane area and the enzyme lifetime. The highest reaction rate reached 0.089μmol FFA/mincm2 when the enzyme loading density was 0.098mg/cm2. The results also indicated that the performance of lipase immobilized on the membrane surface was superior to that immobilized in the pores, and the apparent reaction rate and stability of immobilized lipases were improved greatly after cross-linking. It suggested that immobilization of enzymes by filtration and then cross-linking the enzymes onto the membrane surface is a simple and convenient way to prepare a high-activity immobilized enzyme membrane.

Abstract:A composite membrane made of a hydrophobic synthetic material, PTFE, and a biocompatible material, PVA, is proposed to immobilize lipase. The structure of this composite membrane is specially designed and includes a porous PTFE layer and an ultrafiltration PVA layer. In the present study, enzymes were absorbed in pores of PTFE layer and deposited on the interface between the two layers by a filtration process. The ultrafiltration PVA layer rejects enzymes to control the enzyme loading and prevents enzymes from being dissolved into the aqueous phase. The porous PTFE layer with micrometer pores supplies a hydrophobic environment for immobilized lipases which is propitious to the activation of lipase. An immobilized lipase membrane was employed in a biphasic membrane reactor (BMR) for the hydrolysis of olive oil. The volumetric reaction rate of the BMR was used to evaluate the catalytic ability of immobilized lipase membrane. The effect of the structure of the PTFE support layer and the PVA layer, the lipase loading density and the BMR stability was investigated, and The maximum reaction rate per unit of membrane area (9.25μmol/hcm2) in experimental conditions was found to be higher than the value reported in some literature. The leakage of immobilized lipases was small and did not have a great effect on the catalytic ability when an immobilized lipase membrane was employed in the BMR.
Immobilization of Lipase Onto Functional Cyclomatrix Polyphosphazene Microspheres

Abstract:Functional polyphosphazene cyclomatrix microspheres (PMS) containing glucose and dodecyl moieties were utilized for physical adsorption of lipase. In this regard, Candia Rugosa lipase was immobilized onto the above microspheres through physical adsorption, which exhibited catalytic hydrolysis of glycerol triacetate as the model reaction. As observed under transmission electron microscope (TEM) and scanning electron microscope (SEM), the size of all the microspheres ranged from 400 nm to 1 mu m with a mean diameter of 680 nm. Zeta potential analysis demonstrated that the microspheres were negatively charged. The influences of buffer pH, substrate concentration and temperature were also studied along with reusability. The obtained results demonstrated that the immobilized lipase on deprotected glycosylated PMS (D-GPMS) showed the best performance and possessed the highest enzymatic loading activity around 1.0884 mmol L-1 min(-1) g(-1), and retained nearly 50% of its initial activity even after 10 cycles of application. Moreover, the lipase-microsphere complex displayed high pH adaptability from 7 to 9 and temperature stability above 40 degrees C. From the viewpoint of biocompatible PMS support, the GPMS-lipase conjugate have potential applications in field such as food, medicine and environment. (C) 2016 Elsevier B.V. All rights reserved.

Abstract:Practical application of biphasic enzyme-immobilized membrane bioreactors (EMBR) requires efficient loading of the enzyme with retention of enzymatic activity. Here, we report a method to fabricate an ultrafine fiber membrane conjugated to lipase with high levels of enzyme loading and activity retention. A cellulose acetate (CA) non-woven ultrafine fiber membrane was prepared with 200 nm nominal fiber diameter by electrospinning, followed by alkaline hydrolysis to obtain regenerated cellulose (RC). The RC ultrafine fiber membrane was oxidized by exposure to NaIO4, simultaneously generating aldehyde groups to couple with pentaethylenehexamine (PEHA) as a spacer for lipase immobilization. A biphasic EMBR was assembled with the PEHA-modified and lipase-immobilized membranes. The effect of operation variables, namely aqueous-phase system, reaction pH, accelerant (sodium taurocholate) content, reaction temperature, and membrane usage on the performance of this bioreactor was investigated with the hydrolysis of olive oil. A bioreactor activity as high as 9.83 × 104 U/m2 was obtained under optimum operational conditions.
In-situ immobilization of lipase on α-alumina membrane for oil fouling control and cleaning

Abstract:Enzyme immobilization on inorganic membranes is typically a difficult, time-consuming, multi-step process carried out ex-situ (outside the filtration system). To enable process scaling, an in-situ immobilization technique is essential. Therefore, an in-situ method to immobilize the lipase Eversa Transform 2.0 (ET2) on a ceramic membrane was developed using polydopamine as a bonding agent. The in-situ immobilization was successful, and optimum dopamine hydrochloride and ET2 concentrations (0.3 mg·mL −1 and 4 mg·mL −1 , respectively) resulted in an enzyme loading of 10 g·m −2 that increased the membrane water affinity and hydrolytic activity towards soybean oil (38 mmol·min −1 ·m −2 ). The enzymatic membrane demonstrated great self-cleaning capability (pure water permeance recovery of 97 % after cleaning with deionized water at 40 °C for 6 h) and strong fouling resistance (pure water permeance decline limited to 43 % following oil emulsion filtration). To reuse the membrane after it lost its hydrolytic activity, calcination or in-situ chemical cleaning were tested. There were no morphological or chemical alterations on the membrane surface after five cycles of modification-regeneration. When compared to ex-situ immobilization techniques, in-situ enzyme immobilization and chemical regeneration can save costs, improve the sustainability of the process, and ease scale-up.

Abstract:Enzyme-immobilized membrane bioreactors combine biocatalysis with separation, and thus have generated interest among applied researchers. Despite the advantages of enzyme-immobilized membrane bioreactors, some issues associated with the membrane and operation parameters still remain to be addressed in order to achieve scale-up of such systems. We report the fabrication of a biphasic lipase-immobilized nanofiber membrane bioreactor with high catalytic performance. A poly(acrylonitrile-co-acrylic acid) non-woven nanofiber membrane was prepared by electrospinning and lipase from Candida rugosa was covalently coupled to it via an activation process by N-(3-dimethylaminopropyl)-N'-ethyl-carbodiimide hydrochloride/N-hydroxy-succinimide. The influences of membrane diameters and operation variables on bioreactor efficiency were studied using the catalytic hydrolysis of olive oil as the model reaction. Furthermore, a pseudo first order model contributed to the evaluation of lipase performance in a more standardized way. A bioreactor activity of 1.85 x 10(4) U g(-1) was obtained under optimum operation conditions; also, the catalytic system exhibited good operational and recycling stability.

Abstract:In this work, poly(n-butylamino)(allylamino) phosphazene (PBAP) was synthesized and tethered on polypropylene microporous membrane (PPMM) with the aim of offering a biocompatible and, at the same time, moderately hydrophobic microenvironment to lipase for the first time. Lipase from Candida rugosa was used and the influence of membrane surface conditions on the activities of immobilized lipases was evaluated. Water contact angle measurement as well as field emission scanning electron microscopy were used to characterize the morphology of the modified membranes. The results showed an improvement in the adsorption capacity (26.0 mg/m(2)) and activity retention (68.2%) of the immobilized lipases on the PBAP-modified PPMM. Moreover, the lipases immobilized on the modified PPMM showed better thermal and pH stability.

Abstract:A specially designed microstructure in the composite membrane with a porous hydrophobic layer and a relatively dense hydrophilic layer is proposed for lipase immobilization. Firstly, the composite membrane was prepared by coating the cellulose acetate layer with the average pore size of 1.40nm on the hydrophobic PTFE layer with the average pore size of 76.3nm. Then, enzymes were absorbed in pores of PTFE layer and deposited on the interface between the two layers by the filtration process. The relatively dense CA layer was used to reject the enzymes controlling the enzyme loading which prevented enzymes from being dissolved into the aqueous phase. The porous PTFE layer supplied a hydrophobic environment and a large specific surface area for the immobilization of lipases which were propitious to the activation of lipase. The activity of immobilized lipase membrane based on hydrolysis of olive oil was assayed in the biphasic membrane reactor and the maximum specific activity (1.20±0.04μmol-FFAmin−1cm−2) was found to be higher than the value reported in some literature. The kinetic parameters of the immobilized lipases, Km and kmax were fitted with the Michaelis–Menten equation. The Thiele modulus (φ) was used to evaluate the effect of the mass transfer through the membrane on the performance of reaction systems. The optimum enzyme loading (0.020±0.002mg-proteincm−2) was obtained with the highest activity and without the diffusion-limited. Furthermore, the immobilized lipases retained 80% residual activity after ten hydrolysis cycles. The composite membrane was easily regenerated and lipases immobilized in the regenerated membrane remained a high activity.
Lipase Immobilized on Layer-by-Layer Polysaccharide-Coated Fe3O4@SiO2 Microspheres As a Reusable Biocatalyst for the Production of Structured Lipids

Abstract:A promising strategy for lipase immobilization based on the natural polymers of polysaccharides (hyaluronan (HA) and chitosan (CHI)) functionalized magnetic microspheres (Fe3O4@SiO2@{CHI/HA}(3)) was developed. First, Fe3O4 magnetic microspheres and Fe3O4@SiO2 core/shell microspheres were synthesized by hydrothermal reaction and sol gel method, respectively. Owing to the abundant carboxyl groups in HA chains, the lipase was covalently bonded on the surface of the polysaccharide-functionalized magnetic supports by ultilizing 1-ethyl-3-(3-(dimethylamino)-propyl) carbodiimide and N-hydroxysuccinimide chemistry to produce robust biocatalysts of Fe3O4@SiO2@{CHI/HA}(3)@lipase. The morphology, core shell structure, and magnetic property of the supports and immobilized lipase were investigated through various analytical techniques, including FT-IR analysis, transmission electron microscopy, scanning electron microscopy equipped with energy dispersive spectrometer, elemental analysis, vibrating sample magnetometer, thermogravimetric analysis, and X-ray diffraction. Consequently, the magnetic Fe3O4@SiO2@{CHI/HA}(3) microspheres exhibited a superior performance in terms of immobilizing lipase. The magnetic immobilized lipase showed good thermal and long-term stability, reusability, and catalytic activity for the synthesis of the structured lipid of 1,3-dioleoyl-2-palmitoylglycerol (OPO), which has recently received much interest as a healthy component of food, oil, and pharmaceutical intermediates. The magnetic immobilized lipase could be considered a green and sustainable biocatalyst for the highly efficient synthesis of OPO.

Abstract:BACKGROUND:During lipase-mediated biodiesel production, by-product glycerol adsorbing on immobilized lipase is a common trouble that hinders enzymatic catalytic activity in biodiesel production process. In this work, we built a hydrophobic pore space in macroporous ZIF-8 (named as M-ZIF-8) to accommodate lipase so that the generated glycerol would be hard to be adsorbed in such hydrophobic environment. The performance of the immobilized lipase in biodiesel production as well as its characteristics for glycerol adsorption were systematically studied. The PDMS (polydimethylsiloxane) CVD (chemical vapor deposition) method was utilized to get hydrophobic M-ZIF-8-PDMS with hydrophobic macropore space and then ANL (Aspergillus niger lipase) was immobilized on M-ZIF-8 and M-ZIF-8-PDMS by diffusion into the macropores.RESULTS:ANL@M-ZIF-8-PDMS presented higher enzymatic activity recovery and better biodiesel production catalytic performance compared to ANL@M-ZIF-8. Further study revealed that less glycerol adsorption was observed through the hydrophobic modification, which may attribute to the improved immobilized lipase performance during biodiesel production and ANL@M-ZIF-8-PDMS remained more than 96% activity after five cycles' reuse. Through secondary structure and kinetic parameters' analysis, we found that ANL@M-ZIF-8-PDMS had lower extent of protein aggregation and twice catalytic efficiency (V max/K m) than ANL@M-ZIF-8.CONCLUSIONS:Hydrophobic pore space constituted in macroporous ZIF-8 for lipase immobilization greatly improved lipase catalytic performance in biodiesel preparation. The hydrophobic modification time showed negligible influence on the reusability of the immobilized lipase. This work broadened the prospect of immobilization of enzyme on MOFs with some inspiration.
Lipase-Entrapped Colloidosomes with Tunable Positioning at the Oil-Water Interface for Pickering Emulsion-Enhanced Biocatalysis

Abstract:Pickering interfacial biocatalysis (PIB) paves the way for efficient enzymatic catalysis in the biphasic system. However, the Pickering interfacial biocatalysts located on the oil-water interface still face the inevitable deactivation when one of the phases contains the reactant that inactivates the enzyme. Herein, the positioning of lipase-entrapped colloidosomes (LECs) at the emulsion interface is rationally designed by physically tuning the wettability, which allows LECs to protrude into the selected phase to protect the lipase away from the damage of the reactant. As a proof of concept, LECs with different positioning at the interface are used as Pickering interfacial biocatalysts to produce biodiesel by esterification of lauric acid and methanol. Impressively, the LECs that protrude into the oil phase possess an optimal catalytic performance to protect more lipases away from the damage of the reactant of short-chain alcohol, which shows an 8.18-fold enhancement in specific activity relative to the free lipase, reach a biodiesel yield of 80.37% after 8 h, and retain the 96.44% of relative activity after 10 cycles. This study provides a novel and robust platform for Pickering emulsion-enhanced biocatalysis.
Preparation and Characterization of Magnetic Metal–Organic Frameworks Functionalized by Ionic Liquid as Supports for Immobilization of Pancreatic Lipase

Abstract:Enzymes are difficult to recycle, which limits their large-scale industrial applications. In this work, an ionic liquid-modified magnetic metal–organic framework composite, IL-Fe3O4@UiO-66-NH2, was prepared and used as a support for enzyme immobilization. The properties of the support were characterized with X-ray powder diffraction (XRD), Fourier-transform infrared (FTIR) spectra, transmission electron microscopy (TEM), scanning electronic microscopy (SEM), and so on. The catalytic performance of the immobilized enzyme was also investigated in the hydrolysis reaction of glyceryl triacetate. Compared with soluble porcine pancreatic lipase (PPL), immobilized lipase (PPL-IL-Fe3O4@UiO-66-NH2) had greater catalytic activity under reaction conditions. It also showed better thermal stability and anti-denaturant properties. The specific activity of PPL-IL-Fe3O4@UiO-66-NH2 was 2.3 times higher than that of soluble PPL. After 10 repeated catalytic cycles, the residual activity of PPL-IL-Fe3O4@UiO-66-NH2 reached 74.4%, which was higher than that of PPL-Fe3O4@UiO-66-NH2 (62.3%). In addition, kinetic parameter tests revealed that PPL-IL-Fe3O4@UiO-66-NH2 had a stronger affinity to the substrate and, thus, exhibited higher catalytic efficiency. The results demonstrated that Fe3O4@UiO-66-NH2 modified by ionic liquids has great potential for immobilized enzymes.
