Fabrication and Characterization of Silk/Magnesium Wire Braided Porous Nerve Guidance Conduits

Abstract:Peripheral nerve repair is a common but challenging operation conducted by surgeons. Autologous tissue transplant is the "Golden Standard" for repairing peripheral nerve defects; however, its application is limited by the scarcity of available donor nerves, donor area morbidity, and neuroma formation. To combat the limited ability for peripheral nerves to regenerate, biomaterials for engineering artificial nerves display remarkable promising prospects. In this study, a composite porous scaffold fabricated from silk/magnesium wire with three-way braided tube, silk fibroin and chitosan mixed solution (SF/CS) was made. Its structure was evaluated and characterized through several techniques such as SEM, Texture analyzer and Tensile compression anti-suture tension. The mechanical properties of the scaffold and its effect on the viability of Schwann cells were studied. The Mg/Silk-SF/CS scaffold demonstrated excellent mechanical properties and biocompatibility, as well as physical guidance. These results suggest that Mg/Silk-SFCS scaffold is a potential cell carrier in nerve tissue engineering.
