Application and Effectiveness Evaluation of Electrostatic Spinning Plga-Silk Fibroin-Collagen Nerve Conduits for Peripheral Nerve Regeneration

Abstract:Peripheral nerve regeneration remains a significant clinical challenge to researchers. With advances in technical methodology, the grafting of biocompatible conduits may become a viable alternative for the reconstruction of nerve gaps. In this study, electrospinning combined with rotor drum receiving method were used to fabricating nerve conduits (NCs) with PLGA-silk fibroin-collagen (PLGA-SF-CL) and PLGA. NCs with PLGA-SF-CL and PLGA alone or nerve autograft were employed to bridge 10 mm defects in the sciatic nerves of Sprague Dawley rats. The nerve conduction velocity (NCV) and compound muscle action potentials (CMAPs) were used to evaluate the recovery of the nerve function at 6 and 12 weeks after the operation. The histomorphology of regenerated nerves, number and diameter of regenerated nerve fibers, thickness of regenerated myelin sheath, anti-NF200 immunochemical staining of regenerated axons, atrophy of rat gastrocnemius muscle and other indices were observed at 12 weeks after the operation implantation. All the results of electrophysiological evaluation and histological assessment and TEM studies demonstrated that the NC with PLGA-SF-CL promoted the regeneration of peripheral nerves, the repair effects of the NC with PLGA-SF-CL which performed similar to the autograft controls and better than the NC with PLGA alone. This method offers a promising alternative for the use of nerve autograft.
