Abstract:We present a scalable droplet generator with capillaries embedded in a microchannel, assembled with 3D printed T-connector and fittings. We first analyze the influences of geometric and operating parameters on droplet characteristics using a high-speed camera in a single-capillary device. We then scale up the device by parallel arranging three capillaries with axial misalignment to mitigate the interaction between droplets, which addresses the limitations of droplet productivity. We classify the droplets formed in the three-capillary device into different size distribution intervals and elucidate the underlying mechanism, enabling the manipulation of high-frequency generation of tiny droplets. At a velocity ratio of 12.0, the droplet generation frequency reaches 1260 Hz, in which the similar to 30 mu m droplets account for a large proportion of 65 %. Our low-cost and fast-fabricated capillary embedded devices have a high potential for producing high-throughput tiny droplets and handling liquid-liquid reactions requiring precise control of droplet size.

Abstract:Microparticle-templated droplets or dropicles have recently gained interest in the fields of diagnostic immunoassays, single-cell analysis, and digital molecular biology. Amphiphilic particles have been shown to spontaneously capture aqueous droplets within their cavities upon mixing with an immiscible oil phase, where each particle templates a single droplet. Here, we fabricated an amphiphilic microparticle with four discrete hydrophilic patches embedded at the inner corners of a square-shaped hydrophobic outer ring of the particle (4C particle). 3D computational fluid dynamics simulations predicted droplet formation dynamics and differing equilibrium conditions depending on the patterning configuration. Experiments recapitulated equilibrium conditions enabling tunable dropicle configurations with reproducible volumes down to ~200pL templated by the amphiphilic particles. The dropicle configurations depended predominantly on the size of the hydrophilic patches of the 4C particles. This validated modeling approach can inform the design of dropicles with varying volumes and numbers per particle, which can be harnessed in new amplified bioassays for greater sensitivity, dynamic range, and statistical confidence.

Abstract:We proposed a new flow-focusing technique for generation of monodisperse femtoliter droplets, based on the capillary micro-cross. A funnel-shaped interface of two phase system is observed in a capillary cross for mass production of uniform drops, where a tapered exit orifice is extruded into the dispersed feeding capillary. The droplets, down to 2 μm in size at frequency of 20 kHz, are controllable in size when choosing orifice and capillary sizes, as well as flow rates of inner and outer fluids. For a specific diameter of exit orifice, there is a maximal flow rate of outer fluid, beyond which the interface will be penetrated. Until then, the interface is in steady state and all droplets are highly uniform (<3%), implicating an absolute instability in the whole process.

Abstract:The efficient generation of monodispersed droplets holds great promise for micro-/nanoparticle synthesis and biochemical analysis. However, it remains a challenge to achieve high-throughput generation of monodispersed droplets in a portable and/or parallel manner in nonexpert biomedical laboratories. Herein, a versatile microchannel array (mu CA) device is reported that is portable, multifaceted, reliable, and mass-manufacturable, supporting the high-throughput generation of monodispersed droplets in different ways. This device consists of a silicon-based mu CA chip based on the step emulsification principle, as well as two matching plastic containers, both of which can be mass-manufactured by traditional microfabrication methods at low material costs. With the mu CA device, aqueous solution can be dispersed into emulsion droplets by various modes, such as mechanical pump-based large-scale, handheld syringe-based portable, and gas pump-based highly parallel droplet generation. Furthermore, it is demonstrated that our cost-effective device can be applied for digital polymerase chain reaction analysis, supporting the need for more accessible microfluidic systems. Thus, the present device is expected to have a significant impact on both benchside and bedside applications. Abstract:Droplet microfluidics enables the generation of highly uniform emulsions with excellent stability, precise control over droplet volume, and morphology, which offer superior platforms over conventional technologies for material synthesis and biological assays. However, it remains a challenge to scale up the production of the microfluidic devices due to their complicated geometry and long-term reliability. In this study, we present a high-throughput droplet generator by parallelization of high aspect ratio rectangular structures, which enables facile and scalable generation of uniform droplets without the need to precisely control external flow conditions. A multilayer device is formed by stacking layer-by-layer of the polydimethylsiloxane (PDMS) replica patterned with parallelized generators. By feeding the sample fluid into the device immersed in the carrying fluid, we used the multilayer device with 1200 parallelized generators to generate monodisperse droplets (~45 μm in diameter with a coefficient of variation <3%) at a frequency of 25 kHz. We demonstrate this approach is versatile for a wide range of materials by synthesis of polyacrylamide hydrogel and Poly (l-lactide-co-glycolide) (PLGA) through water-in-oil (W/O) and oil-in-water (O/W) emulsion templates, respectively. The combined scalability and robustness of such droplet emulsion technology is promising for production of monodisperse functional materials for large-scale applications.

Abstract:We describe new developments for controlled fabrication of monodisperse non-spherical particles using droplet microfluidics. The high degree of control afforded by microfluidic technologies enables generation of single and multiple emulsion droplets. We show that these droplets can be transformed to non-spherical particles through further simple, spontaneous processing steps, including arrested coalescence, asymmetric polymer solidification, polymerization in microfluidic flow, and evaporation-driven clustering. These versatile and scalable microfluidic approaches can be used for producing large quantities of non-spherical particles that are monodisperse in both size and shape; these have great potential for commercial applications.

Abstract:This paper reports a microfluidic reactor for synthesizing micro- and nano-droplets using fluid shearing instability of immiscible flow in microchannels. Three factors, flow rate between the two inputs, spout geometry and surface tension are numerically studied. Based on the simulation, a device with the optimal parameters was fabricated and tested. Both computational fluid dynamics (CFD) simulation results and experimental results suggested that this technique is capable of conveniently controlling the droplets sizes from hundreds of micrometers down to several micrometers or even nanometers. This pilot research offers a proof-of-concept demonstration using instabilities in the microchannels to synthesize large quantity of microdroplets. Although the microdroplets in this study were formed by DI water containing Tween20 and silicon oil, the design with modification could be applied for synthesis of other types of micro- and nano-droplets.

Abstract:<p>Many uses of emulsion droplets require precise control over droplet size and shape. Here we report a 'shape-memorable' micro-droplet formulation stabilized by a polyethylene glycol (PEG)-modified protein-surfactant, the droplets are stable against coalescence for months and can maintain non-spherical shapes for hours, depending on the surface coverage of PEGylated protein. Monodisperse droplets with aspect ratios ranging from 1.0 to 3.4 were controllably synthesized with a flow-focusing microfluidic device. Mechanical properties of the interfacial protein network were explored to elucidate the mechanism behind the droplet shape conservation phenomenon. Characterization of the protein film revealed that the presence of a PEG layer at interfaces alters the mechanical responses of the protein film, resulting in interfacial networks with improved strength. Taking advantage of the prolonged stabilization of non-spherical droplets, we demonstrate functionalization of the droplet interface with accessible biotin moieties. The stabilization of micro-droplet shape with surface-active proteins that also serve as an anchor for integrating functional moieties, provides a tailorable interface for diverse biomimetic applications.</p>

Abstract:Crescent‐shaped hydrogel microparticles have been shown to template uniform volume aqueous droplets upon simple mixing with aqueous and oil media for various bioassays. This emerging "lab on a particle" technique requires hydrogel particles with tunable material properties and dimensions. The crescent shape of the particles is attained by aqueous two‐phase separation of polymers inside a spherical droplet followed by photopolymerization of the curable precursor. In this work, we have investigated the phase separation of photo‐curable poly(ethylene glycol) diacrylate (PEGDA, Mw 700) and dextran (Mw 40,000) for tunable manufacturing of crescent‐shaped particles. The particles' morphology was precisely tuned by following a phase diagram, varying the UV intensity, and adjusting the flow rate ratio between the three streams, containing PEGDA, dextran, and oil, within a microfluidic droplet generation device. The fabricated particles with variable cavity sizes and outer diameters encapsulated uniform aqueous droplets upon mixing with a continuous oil phase. The particles were fluorescently labeled with red and blue emitting dyes at variable concentrations to produce six color‐coded particles. The blue fluorescent dye showed a moderate response to the pH change from 1 to 7 in terms of an increase in emitted intensity. The fluorescently labeled particles were able to tolerate an extremely acidic solution (pH 1) but disintegrated within an extremely basic solution (pH 14). The particle‐templated droplets were able to effectively retain the disintegrating particle and the fluorescent signal at pH 14, indicating completely segregated compartments. Abstract:Overview of latest advances in droplet microfluidic formation of highly monodispersed microparticles, involving a plethora of types including spherical, nonspherical, and Janus type, as well as core–shell, hole–shell, and controllable multicompartment particles, is provided. The critical analysis of the current status and existing challenges is also conducted, and the future development in the emerging fields by microfluidic emulsification is envisioned. The remarkable control function over the functional material formation process enabled by droplet microfluidic emulsification approaches can lead to the efficient and one‐step encapsulation of active substances in microparticles, with the microparticle characteristics well regulated. In comparison to the conventional fabrication methods, droplet microfluidic technology can not only construct microparticles with various shapes, but also provide excellent templates, which enrich and expand the application fields of microparticles. For instance, intersection with disciplines in pharmacy, life sciences, and others, modifying the structure of microspheres and appending functional materials can be completed in the preparation of microparticles. The as‐prepared polymer particles have great potential in a wide range of applications for chemical analysis, heavy metal adsorption, and detection. This review systematically introduces the devices and basic principles of particle preparation using droplet microfluidic technology and discusses the research of functional microparticle formation with high monodispersity, involving a plethora of types including spherical, nonspherical, and Janus type, as well as core–shell, hole–shell, and controllable multicompartment particles. Moreover, this review paper also exhibits a critical analysis of the current status and existing challenges, and outlook of the future development in the emerging fields has been discussed.

Abstract:Hierarchical emulsions are interesting for both scientific researches and practical applications. Hierarchical emulsions prepared by microfluidics require complicated device geometry and delicate control of flow rates. Here, a versatile method is developed to design hierarchical emulsions using microfluidic 3D droplet printing in droplet. The process of droplet printing in droplet mimics the dragonfly laying eggs and has advantages of easy processing and flexible design. To demonstrate the capability of the method, double emulsions and triple emulsions with tunable core number, core size, and core composition are prepared. The hierarchical emulsions are excellent templates for the developments of functional materials. Flattened crescent-moon-shaped particles are then fabricated using double emulsions printed in confined 2D space as templates. The particles are excellent delivery vehicles for 2D interfaces, which can load and transport cargos through a well-defined trajectory under external magnetic steering. Microfluidic 3D droplet printing in droplet provides a powerful platform with improved simplicity and flexibility for the design of hierarchical emulsions and functional materials.

Abstract:Droplet microfluidics, which can generate monodisperse droplets or bubbles in unlimited numbers, at high speed and with complex structures, have been extensively investigated in chemical and biological fields. However, most current methods for fabricating microfluidic devices, such as glass etching, soft lithography in polydimethylsiloxane (PDMS) or assembly of glass capillaries, are usually either expensive or complicated. Here we report the fabrication of simple and cheap microfluidic devices based on patterned coverslips and microscope glass slides. The advantages of our approach for fabricating microfluidic devices lie in a simple process, inexpensive processing equipment and economical laboratory supplies. The fabricated microfluidic devices feature a flexible design of microchannels, easy spatial patterning of surface wettability, and good chemical compatibility and optical properties. We demonstrate their utilities for generation of monodisperse single and double emulsions with highly controllable flexibility.

Abstract:We propose a novel method to obtain versatile droplets arrays on a regional hydrophilic chip that is fabricated by PDMS soft lithography and regional plasma treatment. It enables rapid liquid dispensation and droplets array formation just making the chip surface in contact with solution. By combining this chip with a special Christmas Tree structure, the droplets array with concentrations in gradient is generated. It possesses the greatly improved performance of convenience and versatility in bioscreening and biosensing. For example, high throughput condition screening of toxic tests of CdSe quantum dots on HL-60 cells are conducted and cell death rates are successfully counted quickly and efficiently. Furthermore, a rapid biosensing approach for cancer biomarkers carcinoma embryonic antigen (CEA) is developed via magnetic beads (MBs)-based sandwich immunoassay methods.
Picoliter Droplet Generation and Dense Bead-in-Droplet Encapsulation via Microfluidic Devices Fabricated via 3D Printed Molds

Abstract:Picoliter-scale droplets have many applications in chemistry and biology, such as biomolecule synthesis, drug discovery, nucleic acid quantification, and single cell analysis. However, due to the complicated processes used to fabricate microfluidic channels, most picoliter (pL) droplet generation methods are limited to research in laboratories with cleanroom facilities and complex instrumentation. The purpose of this work is to investigate a method that uses 3D printing to fabricate microfluidic devices that can generate droplets with sizes <100 pL and encapsulate single dense beads mechanistically. Our device generated monodisperse droplets as small as ~48 pL and we demonstrated the usefulness of this droplet generation technique in biomolecule analysis by detecting Lactobacillus acidophillus 16s rRNA via digital loop-mediated isothermal amplification (dLAMP). We also designed a mixer that can be integrated into a syringe to overcome dense bead sedimentation and found that the bead-in-droplet (BiD) emulsions created from our device had <2% of the droplets populated with more than 1 bead. This study will enable researchers to create devices that generate pL-scale droplets and encapsulate dense beads with inexpensive and simple instrumentation (3D printer and syringe pump). The rapid prototyping and integration ability of this module with other components or processes can accelerate the development of point-of-care microfluidic devices that use droplet-bead emulsions to analyze biological or chemical samples with high throughput and precision.

Abstract:A novel 3D‐printed microfluidic device is reported to fabricate a range of bar‐coded amphiphilic particles by configuring it with multiple outlet tubing and photomasks. The fabricated particles are characterized based on their dimensional parameters and their ability to hold uniform droplet volumes. Compartmentalizing an aqueous media into numerous nanoliter‐scale droplets has substantially improved the performance of amplification assays. Particle‐templated droplets or dropicles offer a user‐friendly workflow for creating uniform volume compartments upon simple mixing of reagents and particles using common laboratory apparatus. Amphiphilic shape‐coded particles are demonstrated to spontaneously hold aqueous droplets within hydrophilic cavities for multiplexed diagnostic assays. Here, a configurable 3D‐printed microfluidic device is proposed for the tunable fabrication of amphiphilic size‐coded particles. The device is configured with multiple outlet tubings of different diameters and photomasks of variable slit lengths to fabricate a wide range of size‐coded particles. a range of unique particle codes are fabricated using a single reconfigurable device. The cross‐sectional profile of the particles is further engineered by tuning the flow rate ratios of precursor streams to vary the inner and outer diameters of the particles and the thicknesses of the inner hydrophilic and outer hydrophobic layers. A range of cavity diameters and particle lengths enabled dropicle volumes of ≈1 nL to ≈30 nL. The fabricated particles are characterized by their ability to hold uniform droplet volumes and to orient themselves facing upward or sideways in a well plate based on their aspect ratios.

Abstract:The compartmentalization of reactions in monodispersed droplets is valuable for applications across biology. However, the requirement of microfluidics to partition the sample into monodispersed droplets is a significant barrier that impedes implementation. Here, we introduce particle-templated emulsification, a method to encapsulate samples in monodispersed emulsions without microfluidics. By vortexing a mixture of hydrogel particles and sample solution, we encapsulate the sample in monodispersed emulsions that are useful for most droplet applications. We illustrate the method with ddPCR and single cell culture. The ability to encapsulate samples in monodispersed droplets without microfluidics should facilitate the implementation of compartmentalized reactions in biology.

Abstract:Hydrogel microspheres are biocompatible materials widely used in biological and medical fields. Emulsification and stirring are the commonly used methods to prepare hydrogels. However, the size distribution is considerably wide, the monodispersity and the mechanical intensity are poor, and the stable operation conditions are comparatively narrow to meet some sophisticated applications. In this paper, a T-shaped stepwise microchannel combined with a simple side microchannel structure is developed to explore the liquid-liquid dispersion mechanism, interfacial evolution behavior, satellite droplet formation mechanism and separation, and the eventual successful synthesis of dextran hydrogel microspheres. The effect of the operation parameters on droplet and microsphere size is comprehensively studied. The flow pattern and the stable operation condition range are given, and mathematical prediction models are developed under three different flow regimes for droplet size prediction. Based on the stable operating conditions, a microdroplet-based method combined with UV light curing is developed to synthesize the dextran hydrogel microsphere. The highly uniform and monodispersed dextran microspheres with good mechanical intensity are synthesized in the developed microfluidic platform. The size of the microsphere could be tuned from 50 to 300 μm with a capillary number in the range of 0.006-0.742. This work not only provides a facile method for functional polymeric microsphere preparation but also offers important design guidelines for the development of a robust microreactor.
Droplet Pattern Formation and Translation in New Microfluidic Flow-Focusing Devices

Abstract:We conducted experiments on specially designed microfluidic chips that generate droplets through a microfluidic flow-focusing approach. The fluid flow in the microfluidic channel produced a shear flow field at low Reynolds numbers. The droplets in the microfluidic system exhibited special droplet pattern formations similar to periodic crystal-like lattices because of the competition between shear forces and surface tension. By adjusting the flow rate ratio of the water (droplet phase) to oil (continuous phase) phases and changing the outlet channel widths, the droplets formed monolayer dispersion to double-layer formation to monolayer squeezing when the outlet channel widths were 250 or 300 mu m. We also obtained droplets with monolayer dispersion, three-layer arrangements, double-layer squeezing, and monolayer squeezing when the outlet channel width was 350 mu m. The outlet channel width was increased to 400 mu m, and four-layer arrangements were observed. We also studied the translation of droplet formation, which resulted in a detailed strategy to control drop size and droplet pattern formation for emulsification in microfluidic devices. We expect that our strategy can provide theoretical guidance to synthesize dispersion or polydisperse colloid particles.
