Determination of Ketoprofen and Sodium Acetate in High Concentration of Ketoprofen Injection by HPLC

Abstract:The aim of this article was to establish the HPLC methods for determination of ketoprofen and sodium acetate (NaAc) content in ketoprofen injection of high concentration (5%). Ketoprofen content in the injection was higher than marketing preparations for NaAc was added as the cosolvent. A Shim-pack VP-ODS C18 reversed phase column (250 mm x 4.6 mm, 5 mu m) was applied in the determination of ketoprofen and NaAc. The HPLC chromatography conditions for detecting ketoprofen were as follows: the mobile phase consisted of purified water, methyl cyanide and potassium dihydrogen phosphate buffer solution (adjusted to pH 3.5 with phosphoric acid) (55: 43: 2, v/v/v), with a flow rate of 1.2 mL/min. The injection volume was 10 mu L and the detection wavelength was fixed at 255 nm, separation was performed at 25 degrees C. For the determination of sodium acetate, the mobile phase was composed of 0.07% phosphoric acid solution (adjusted to pH 3.0 with sodium hydroxide) and methanol (95: 5, v/v), the flow rate was 1.0 mL/min and 20 mu L sample was injected into the chromatography system, the detection was carried out at 210 nm under the column temperature of 25 degrees C. The related substances were well separated under the chromatogram conditions, the standard curves of ketoprofen and NaAc were linear in the range of 60 to 160 mu g/mL (r = 0.9991) and 180 to 420 mu g/mL (r = 0.9991), respectively. The average recoveries (n = 9) were 98.11% for ketoprofen (RSD = 1.79%), and 98.92% for NaAc (RSD = 0.89%). The LOD for ketoprofen and NaAc were 0.81 mu g/mL and 0.31 mu g/mL, and LOQ of them were 2.53 mu g/mL and 1.05 mu g/mL separately. In conclusion, the methods are sensitive, repeatable, easy to operate, and suitable for determining ketoprofen and NaAc content in ketoprofen injection of high concentration.
