Determination of Main Drug Content and Related Substances of New Compound Analgesic-Ketoprofen and Lansoprazole Sustained-Release Pellets

Abstract:Based on our recent established optimal formulation of ketoprofen/lansoprazole (K/L) sustained-release pellets, the present study is aimed to develop a method for determining the two main contents and related substances in preparation. The two drugs were detected by high performance liquid chromatography using the following conditions: BDS C18 (250 mm x 4.6 mm, 5 mu m) column; mobile phases consisting of water: acetonitrile: triethylamine (65: 32: 3, V/V/V), 10% phosphoric acid was used to adjust the pH to 7.0 and a flow rate of 1 mL/min was used; A detection wavelength of 285 nm was applied and the column temperature was 25 degrees C. The related substance was determined by a self-control method without a correction factor. Ketoprofen, lansoprazole and the related substances could be completely separated. A ketoprofen concentration in the range of 40 similar to 160 mu g/mL exhibited a good linear relationship. The rate of recovery was 99.92% (n = 9), the detection limit was 3.4 ng/mL and the limit of quantitation was 20.0 ng/mL. The concentration of lansoprazole (6 similar to 24 mu g/mL) exhibited a good linear relationship in the peak area and the rate of recovery was 99.93% (n = 9). The detection limit was 6.7 ng/mL and the limit of quantitation was 60.0 ng/mL. The average content of the ketoprofen and lansoprazole was 100.82 and 98.93%, respectively. All related substances were determined to be less than 2.0%. This developed method proved to be simple, accurate, rapid, and highly sensitive. Therefore, the method may be suitable for the determination of K/L sustained-release pellets and related substances.
