Three deprotection pathways quantitatively removing the Nδ-phenoxycarbonyl group, i.e., aminolysis, deprotection and guanidination, produce poly-L-citrulline, poly-L-ornithine and poly-L-arginine, respectively.

Abstract:Ethyl and phenyl oxanilates were readily prepared from N-(2-bromobenzyl)anilines and oxalyl chloride monoethyl and monophenyl esters, respectively. It was found that the ethyl oxanilate survived in the presence of K2CO3 in DMA at 120 degrees C and underwent an intramolecular direct arylation using Pd(OAc)(2)-dppf, furnishing the 5,6-dihydrophenanthridine derivative. In contrast, the corresponding phenyl oxanilates decomposed upon exposure to K2CO3 in DMA at 120 degrees C and were transformed into N-arylisoindolin-1-ones via Pd(OAc)(2)-dppf-catalyzed intramolecular decarbonylative coupling. Except for the 4-methoxy-substituted oxanilic acid phenyl ester, other phenyl oxanilates possessing electron-withdrawing (NO2, Cl) and weak electron-donating (Me) substituents provided the N-arylisoindolin-1-ones in 43-80% yields.

Abstract:As an important nitrogen source, isocyanides have been involved in numerous organic reactions. As a result, many complicated compounds have been successfully synthesized through isocyanide chemistry. However, compared with its popular research in organic reactions, the application of isocyanides in polymerization is less investigated. In this work, a new polymerization based on isocyanide monomers is established. By simply mixing diisocyanoacetates and dialdehydes in the presence of a catalytic system of CuCl/PPh3 /organobase in dichloromethane at room temperature readily produces soluble and thermally stable oxazoline-containing polymers with moderate weight-averaged molecular weights (Mw up to 11 200) in excellent yields (up to 97%) after 6 h. Furthermore, introducing the tetraphenylethene moiety into the main chains endows the resultant polymers with aggregation-induced emission, which can function as fluorescent probes for Fe3+ ion detection with high sensitivity and selectivity. This work not only enriches the family of isocyanide-based polymerizations but also provides an efficient tool for the preparation of functional heterocycle-containing polymers.

Abstract:Transition metal-catalyzed directed C-H functionalization promises a straightforward route to polymerization but remains highly underexplored. We report herein the development of a polymer synthesis protocol based on Rh(III)-catalyzed N-nitroso-directed C-H olefination reaction. The single bifunctional monomer approach allows the high-yield generation of polymers with relatively narrow PDI under mild conditions. Further convenient denitrosation transformation furnishes responsive fluorescent polymers with metal ion-selective quenching behaviors. The high Hg2+ selectivity observed in one system provides a handy enabling tool for detecting this hazardous species.

Abstract:Cu(II)-modified nitrogen-rich covalent organic polymer (Cu/N-COP-1) was fabricated and employed to catalyze the cross-dehydrogenative ortho-aminomethylation of phenols. The Cu/N-COP-1 catalyst displayed excellent catalytic activity under mild conditions without any ligand. Various substituted phenols and N,N-dimethylanilines were successfully converted to the corresponding products. Moreover, the catalyst can be readily reused for at least five consecutive runs without obvious loss of catalytic activity. This can be attributed to the interaction between the nitrogen-containing group of the covalent organic polymer and Cu species, which makes Cu species steadily immobilized on the surface of polymer.

Abstract:A general and regiospecific transformation of substituted phenols into the related o-nitrophenols has been achieved via a three-step process involving the palladium-catalyzed chelation-assisted ortho-C-H bond nitration as the key step. In the process, 2-pyridinyloxy groups act as removable directing groups for the palladium-catalyzed ortho-nitration of substituted 2-phenoxypridines, and they can be readily removed in the subsequent conversion of the resulting 2-(2-nitrophenoxy)pyridines into 2-nitrophenols.

Abstract:An efficient one-pot three-component palladium-catalyzed domino reaction of aryl iodide, 2-bromophenylboronic acid, and norbornadiene to produce phenanthrenes has been developed. Norbornadiene serves both as the activator of ortho-C-H bond and the source of ethylene via a retro-Diels-Alder reaction. The method features inexpensive and readily available substrates, a broad range of functional groups, and good yields.

Abstract:Olefin isomerization has an important impact on organic synthesis, fine chemicals, and polymer chemistry. However, due to the small energy differences between various potential isomers, achieving regioselective isomerization remains a challenge. We report on the nickel-catalyzed divergent isomerization of olefins, which leads to the selective formation of either a kinetic or thermodynamic isomer, with the specific product determined by the choice of phosphine ligand. This double-bond chain-walking reaction is characterized by its mild conditions, broad substrate scope, good regioselectivity, high yields, gram-scale synthesis, and the availability of materials from commercial sources. The pairing of this isomerization with ethenolysis led to the formation of formal one-carbon-deletion products. Mechanistic studies emphasize the critical role of the phosphine ligand in governing the regioselectivity of the double-bond chain-walking reaction, and a distinctive nickel-hydrogen-bonding intermediate was pointed out to be key to the success of this transformation.

Abstract:[Objective]To synthesis Nδ-protective ornithine under microwave irradiation.[Method]L-ornithine monohydrochloride was first reacted with basic curpic carbonate to give L-ornithine copper complexes.Subsequently,the δ-amino was protected with tert-butoxycarbonyl(Boc),benzyloxycarbonyl(Z),fluorenylmethyloxycarbonyl(Fmoc),acetyl(Acetyl) and phthaloyl(Phthalyl),after the use of EDTA-2Na for copper removal,the Nδ-benzyloxycarbonyl ornithine(H-Orn(Z)-OH),Nδ-tert-butoxycarbonyl ornithine(H-Orn(Boc)-OH),Nδ-fluorenylmethyloxycarbonyl ornithine(H-Orn(Fmoc)-OH),Nδ-acetyl ornithine(H-Orn(Acetyl)-OH) and Nδ-phthaloyl ornithine(H-Orn(Phthalyl)-OH) were obtained.Finally,the microwave irradiation technology was applied for the removal of copper to explore the effects of microwave radiation power,time and the proportion of materials on yield.[Result]The optimal conditions were obtained as follows: n(Nδ-protective ornithine copper complexes)∶ n(EDTA)=1∶ 1.1-1∶ 1.2,microwave power was 250-300 W,reaction time was 4-6 min.The yield of Nδ-benzyloxycarbonyl-L-Ornithine,Nδ-tert-butyloxycarbonyl-L-Ornithine,Nδ-fluorenylmethyl-oxycarbonyl-L-Ornithine,Nδ-acetyl-L-Ornithine and Nδ-phthalyl-L-Ornithine were 93.2%,86.2%,88.7%,86.3% and 86.4% respectively.The structure of aimed compounds was confirmed by elementary analysis and 1H NMR.[Conclusion] The improved synthetic route was environment friendly and convenient in operation with better product yield and purity.

Abstract:The development of a catalytic multicomponent reaction by orthogonal activation of readily available substrates for the streamlined di-functionalization of alkynes is a compelling objective in organic chemistry. Alkyne carboalkynylation, in particular, offers a direct entry to valuable 1,3-enynes with different substitution patterns. Here, we show that the synthesis of stereodefined 1,3-enynes featuring a trisubstituted olefin is achieved by merging alkynes, alkynyl bromides, and redox-active N-(acyloxy)phthalimides through nickel-catalyzed reductive alkylalkynylation. Products are generated in up to an 89% yield as single regio- and E isomers. Transformations are tolerant of diverse functional groups and the resulting 1,3-enynes are amenable to further elaboration to synthetically useful building blocks. With olefin-tethered N-(acyloxy)phthalimides, a cascade radical addition/cyclization/alkynylation process can be implemented to obtain 1,5-enynes. This study underscores the crucial role of redox-active esters as superior alkyl group donors compared with haloalkanes in reductive alkyne dicarbofunctionalizations.

Abstract:Unsymmetrical N-benzyloxycarbonyl-protected 1-amino-1-arylalkylphosphonate mixed diesters were synthesized using a one-pot reaction involving benzyl carbamate, aromatic aldehydes and alkoxydichlorophosphine, followed by treatment with alcohols in the presence of triethylamine. The reactions were followed by 31P NMR and a mechanism is proposed.

Abstract:Palladium-catalyzed C-H activation reactions have attracted the attention of organic researchers due to their unique high selectivity, broad functional group tolerance, and high efficiency, and they are widely used in natural products and asymmetric synthesis. Here, we report an example of enantioselective C-H alkenylation between β-alkyl phenylethylamine compounds and styrenes with Boc-L-lle-OH as the ligand and nosylamide as the directing group. This reaction is applicable to styrene containing various electron-deficient and electron-donating substitutions and may be utilized for the synthesis of benzoazepine compounds.

Abstract:A palladium-catalyzed three-component reaction enabled the synthesis of N-formylanilines from aryl iodides, NaN(3), and oxalic acid. This one-pot process involves aminocarbonylation, the Curtius rearrangement, and reduction using oxalic acid as both a carbon monoxide and hydrogen donor. The method has a broad substrate scope, tolerates various functional groups, and delivers N-formylanilines in high yields, making it a green and useful synthetic approach.

Abstract:Bis/trisubstituted alkenylboronates are one of the most important motifs in organic synthesis. Herein, the synthesized series of copper metallized porous carbene ligand polymers are reported. Using CuCl@POL‐NHC‐Ph as the optimal nanocatalyst, the highly selective β‐regio‐ and syn‐stereoselective 1,2‐carboboration of alkynes are achieved to obtain bis/trisubstituted alkenylboronates. Alkenylboronates are highly versatile building blocks and valuable reagents in the synthesis of complex molecules. Compared with that of monosubstituted alkenylboronates, the synthesis of multisubstituted alkenylboronates is challenging. The copper‐catalyzed carboboration of alkynes is an operationally simple and straightforward method for synthesizing bis/trisubstituted alkenylboronates. In this work, a series of copper‐metallized N‐Heterocyclic Carbene (NHC) ligand porous polymer catalysts are designed and synthesized in accordance with the mechanism of carboboration. By using CuCl@POL‐NHC‐Ph as the optimal nanocatalyst, this study realizes the β‐regio‐ and stereoselective (syn‐addition) 1,2‐carboboration of alkynes (regioselectivity up to >99:1) with satisfactory yields and a wide range of substrates. This work not only overcomes the selectivity of carboboration but also provides a new strategy for the design of nanocatalysts and their application in organic synthesis.

Abstract:Differing from the moisture-sensitive alpha-amino acid N-carboxyanhydrides (AA-NCAs) monomers, N-phenoxycarbonyl alpha-amino acids (AA-NPCs) can be prepared and stored in open air. In this contribution, we report that the controlled polymerizations of AA-NPC monomers of O-tert-butyl-dl-serine (BRS-NPC), N-epsilon-benzyloxycarbonyl-l-lysine (ZLL-NPC) and N-epsilon-trifluoroacetyl-l-lysine (FLL-NPC) initiated by amines are surprisingly able to tolerate common nucleophilic impurities such as water and alcohols at a level of monomer concentration. The structures of polypeptides synthesized in the presence of water or alcohols agree well with the designed ones in the case of repeated chain extensions. Detailed mechanism study and density functional theory calculation reveal that the low concentration of AA-NCA and the high activity of amines are the key factors to the controllability of AA-NPC polymerizations. The water- and alcohol-tolerant property in polymerizations of AA-NPCs encourages the following studies on unprotected (phenolic) hydroxyl groups containing AA-NPCs. The controllable polymerizations of N-phenoxycarbonyl l-tyrosine (LT-NPC) and N-phenoxycarbonyl S-(3-hydroxypropyl)-l-cysteine (HLC-NPC) initiated by amines are confirmed and reported for the first time, which extends the library of AA-NPCs and polypeptides as well. All the universality of library, the convenience of monomer preparation, and the controllability and water- and alcohol-tolerant property of polymerization of AA-NPCs significantly enhance the feasibility of polypeptide synthesis, making AA-NPC approach a promising synthetic method of polypeptides. Abstract:Palladium-catalyzed carbonylation reactions of ortho-phenylene dihalides were studied using aminoethanols as heterobifunctional N,O-nucleophiles. The activity of aryl-iodide and –bromide as well as the chemoselective transformation of amine and hydroxyl functionalities were studied systematically under carbonylation conditions. Aminocarbonylation can be selectively realized under optimized conditions, enabling the formation of amide alcohols, and the challenging alkoxycarbonylation can also be proved feasible, enabling amide-ester production. Intramolecular double carbonylation reaction can be achieved using 1,2-diiodobenzene and amino alcohols featuring secondary amine groups, giving oxazocine derivatives. Useful reaction scope with various amino alcohols was performed with good isolated yields of the targeted compounds. Intramolecular C-O coupling of amide alcohols possessing bromo substituent in adjacent ortho position is also demonstrated as a potential next step in benzoxazepine heterocycle formation.

Abstract:With the vastly increasing applications of chiral phosphine-oxazoline (PHOX) hybrid ligands in various transition metal-catalyzed reactions, novel PHOX ligands bearing innovative backbones are highly valuable and in great demand. This study describes the development of a new type of chiral PHOX ligands based on a hexamethyl-1,1'-spirobiindane scaffold and incorporating both a phosphine and an oxazoline moiety. The optimal ligand provided high yields and excellent enantioselectivities for the Ni-catalyzed asymmetric arylation of cyclic N-sulfonyl imines with arylboronic acids leading to chiral amines.
