Abstract:Four novel cycloartanes, named sphaerophysone A (1), B (2), C (3) and D (4), were isolated from the ethanol extract of Sphaerophysa salsula (Pall.) DC. The structures were elucidated on the basis of spectral evidence, and the stereochemistry of compound 1 was defined by X-ray crystallographic analysis. Sphaerophysone B (2) may be an artifact formed in the isolation procedure.

Abstract:Three new triterpenoid derivatives, named (15 alpha)-15-hydroxysoyasapogenol B (1), (7 beta,15 alpha)-7,15-dihydroxysoyasapogenol B (2), and (7 beta)-7,29-dihydroxysoyasapogenol B (3), were isolated from cultures of the plant endophytic fungus Pestalotiopsis clavispora. Their structures and relative configurations were elucidated by extensive spectroscopic analysis and X-ray crystallography.

The structure was elucidated on the basis of spectral evidences and confirmed by X-ray analysis, the stereochemistry of the compound was also defined by X-ray analysis.

Abstract:Seven new pentacyclic triterpenoid saponins, named dianversicosides A-G (1-7), together with nine known compounds, were isolated from the aerial parts of Dianthus versicolor. The structures of 1-7 were elucidated on the basis of spectroscopic data and chemical evidence. The absolute configuration of the 3-hydroxyl-3-methylglutaryl (HMG) group in 1-4 was ascertained by chemical analysis combined with a chiral HPLC method. Cytotoxic activities of the isolated compounds were evaluated against a small panel of cancer and other cell lines.

Abstract:A new steroidal glycoside (1), (25R)-14α, 17α-hydroxyspirost-5-en-3β-yl 3-O-α-L-rhamnpyranosyl-(1 → 2)-β-D-glucopyranosyl-(1 → 3)-β-D-glucopyranoside, together with three known steroidal glycosides, (25R)-3β-hydroxyspirost-5-en-1β-yl-3-O-α-L-rhamnopyranosyl-(1 → 2)-O-β-D-xylopyranosyl-(1 → 3)-α-L-arabinopyranoside (2), Cixi-ophiopogon B (3) and Cixi-ophiopogon A (4), were obtained from the tuberous roots of Ophiopogon japonicus (Liliaceae). Compound 2 was isolated from the Ophiopogon genus for the first time. Their structures were identified on the basis of extensive mass and nuclear magnetic resonance spectroscopic analysis.

Abstract:Bipolarolides A-G (1-7), seven novel ophiobolin-derived sesterterpenes representing three new types of skeletons, were characterized from fungus Bipolaris sp. TJ403-B1. Their structures were determined via spectroscopic analyses, X-ray crystallography, and quantum chemical 13C NMR and electronic circular dichroism (ECD) calculations. Compounds 1 and 2 were uniquely defined by a multicyclic caged oxapentacyclo[9.3.0.01,6.05,9.18,12]pentadecane-bridged system; compounds 3 and 4 featured an unprecedented 5-5-5-5-fused core skeleton, while 3 also contained an unexpected C-3-C-14 oxygen bridge to construct the caged architecture; compounds 5-7 represented a new class of highly modified pentacyclic oxaspiro[4.4]nonane-containing sesterterpene-alkaloid hybrids. Their biosynthetic pathways and potential HMGR inhibitory and antimicrobial activities are also discussed.

Abstract:Two new triterpenoid saponins (1, 2) and a known saponin (3) were isolated from the husks of Xanthoceras sorbifolia Bunge., and their structures were elucidated as 3-O-β-D-glucopyranosyl(1 → 6)-[angeloyl(1 → 2)]-β-D-glucopyranosyl-28-O-α-L-rhamnopyranosyl(1 → 2)-[β-D-glucopyranosyl(1 → 6)]-β-D-glucopyranosyl-21β,22α-dihydroxyl-olean-12-ene (1), 3-O-β-D-glucopyranosyl-28-O-[β-D-glucopyranosyl(1 → 2)]-β-D-glucopyranosyl-21β,22α-dihydroxyl-olean-12-ene (2), and 3-O-β-D-glucopyranosyl-28-O-[α-L-rhamnopyranosyl(1 → 2)]-β-D-glucopyranosyl-21β,22α-dihydroxyl-olean-12-ene (3), on the basis of the spectral analysis of NMR and chemical methods. Cytotoxic assay indicated that none of them showed obvious inhibitory effect on the proliferation of two human tumor cell lines.

Abstract:Three new iridoid glycosides, epiasperuloside (1), epipaederosidic acid (2), and epipaederoside (3), together with 7 known compounds, were isolated from the 85% ethanol extract of the leaves of Saprosma ternatum Hook. f. Their structures were determined by application of spectroscopic (NMR, MS) and chemical methodologies. The antioxidant activity was evaluated for all isolates in terms of both DPPH and ABTS bioassays. Epiasperuloside (1) showed some antioxidant activity.

Abstract:Two new oleanane-type triterpenoid saponins, kakkasaponin II (1) and kakkasaponin III (2), were isolated from the methanol extract of the flowers of Pueraria thomsonii (Leguminosae), together with seven known oleanane-type triterpenoid saponins, phaseoside IV (3), sophoradiol monoglucuronide (4), kakkasaponin I (5), kaikasaponin III (6), soyasaponin I (7), soyasaponin III (8), and soyasaponin IV (9). The structures of 1 and 2 were elucidated by spectroscopic methods including IR, ESI-TOF-MS, and 1D and 2D NMR experiments.

Abstract:Two new oleanane-triterpenoid saponins, clinograsaponins A (1) and B (2), together with twelve known ones (3-14), were isolated from the whole herb of Clinopodium gracile (Bentham) Matsumura. Their structures were determined by spectroscopic analysis and chemical method. All the isolated compounds were evaluated for their activities against ATP-citrate lyase (ACLY) and nuclear factor kappa B (NF-κB).

Abstract:A new triterpenoid saponin containing five sugars was isolated from the ethanolic root extract of Dipsacus japonicus Miq., and its structure was established as 3-O-[beta-D-glucopyranosyl (1-->4)] [alpha-L-rhamnopyransoyl (1 --> 3)]-beta-D-glucopyranosyl (1-->3)-alpha-L-rhamnopyranosyl (1-->2)-alpha-L-arabinopyranosyl-oleanoic acid. The sites of glycosylation and the sequence of sugars in the glycoside can be determined unambiguously and total assignment of severely overlapping proton resonance of sugar residues were achieved by a combined use of the 1D-SEMDY and NOE difference spectroscopy in rotating frame techniques, without having recourse to chemical degration or modification. The results showed that these new NMR techniques are very effective and convenient for the structure determination and spectral assignment of this class of compounds.
Oxepine and Bibenzyl Compounds from Bulbophyllum Kwangtungense

Abstract:Phytohemical investigation of the EtOAc extract of leaves and stems of Bulbophyllum kwangtungense Schlecht., belonging to the orchid family was carried out. Two new stilbenods were isolated by repeat column chromatography and characterized: one is bibenzyl compound, 5-(2, 3-dimethoxyphenethyl)-6-methylbenzo [d] [1, 3] dioxole (1), and the other dibenz [b, f] oxepine, 10, 11-dihydro-2, 7-dimethoxy-3, 4-methylenedioxy dibenzo b, f] oxepine (2). Their structures were established on the basis of analyses of spectroscopic evidence, particularly by two-dimensional NMR spectroscopic analysis. Bibenzyl compound with a methyl group in the benzene ring is rarely found from natural sources. Natural oxepines are very rare, and most of them were reported to have good anti-tumor activities. In addition, anti-tumor activities of the new compounds were also evaluated. Among the compounds isolated, compound 2 showed anti-tumor activities with IC50 value of 47.2 mu g/mL in vitro against cultures of Hela cells.

Abstract:A new oleanane-type triterpenoid saponin with a carbonyl group at C-15, named polybosaponin A (1), together with a known triterpenoid saponin, dehydrosoyasaponin I (2), were isolated from Radix Hedysari. On the basis of one- and two-dimensional NMR and high resolution electrospray ionization mass spectrometry, the structure of 1 were elucidated and the 1H and 13C NMR spectral signals were assigned totally. In addition, the complete assignment of 1H NMR signals and revision of incorrect assignment of 13C NMR signals for 2 were also achieved based on one- and two-dimensional NMR spectrometry.

Abstract:Four new lanostane-type triterpenes, 24-ethyl-3beta-methoxylanost-9(11)-en-25-ol (1), 3beta-methoxy-24-methylenelanost-9(11)-en-25-ol (2), 3beta-methoxy-25-methyl-24-methylenelanost-9(11)-en-21-ol (3) and 3beta-methoxy-24-methyllanosta-9(11),25-dien-24-ol (4) together with three known triterpenes, betulinaldehyde, betulinic acid methyl ester, and ursaldehyde have been isolated from the methanol extract of the twigs of Diospyros discolor. The structures of those new compounds were elucidated by spectroscopic methods.

Abstract:Three previously undescribed biflavonoids, oxytrodiflavanone A (1), and oxytrochalcoflavanones A,B (2,3), were isolated from the aerial part of Oxytropis chiliophylla, together with their putative biosynthetic monomers, i.e., (2S)-5,7-dihydroxyflavanone (4), (2S)-7-hydroxyflavanone (5), and 2',4'-dihydroxychalcone (6). The structures of these compounds were elucidated by a combination analysis of spectroscopic data. The cytotoxic activities of all the isolated compounds against PC-3 human prostate cancer cell line are also presented.
