Abstract:Transition metal catalyzed C-H activation/C-O coupling reaction of arenes serves as a powerful strategy for constructing phenols, aryl ethers and phenolic esters, and thus represents a hot topic in the field of organic chemistry. This strategy is effective and atom-economic as it allows people to construct C-O bond directly from arenes, avoiding pre-functionalization of arenes. Recent development on transition metal catalyzed C-H activation/C-O coupling reactions according to different species of transition metal catalysts is summarized. Possible mechanisms of typical protocols are explained and compared. Existing problems and limitations, and a brief outlook in this field is also presented.

Abstract:Homocoupling and cross-coupling of electron-deficient arenes, including nitrobenzene, (trifluoromethyl)benzene, ethyl benzoate, etc., have been developed to afford biaryls using a catalytic system of Pd(OAc)(2)/trifluoroacetic acid (TFA)/O-2 or K2S2O8. The crucial step is to tune the concentrations of arenes and the loading of TFA carefully to enhance the reaction rates and the selectivity of these couplings involving dual C-H activation.
Unactivated Pd-Catalyzed Oxidative Cross-Coupling Reactions of Arenes with Benzene: A Theoretical and Experimental study

Abstract:Oxidative C-H bond activated palladium-catalyzed aromatic cross-coupling reactions offer advantages of fewer required synthetic steps but disadvantages of loss of inherent regioselectivity and selectivity. In one proposed catalytic cycle, each arene adds to a palladium carboxylate catalyst according to a concerted metalation deprotonation (CMD) mechanism, followed by reductive elimination to couple the arenes and the replenishment of the catalyst by an oxidant. Using this proposed mechanism, molecular structures and free energy profiles were calculated for coupling of benzene with a range of heteroarenes containing various substituents and with several Pd-carboxylate catalysts. Reaction kinetics were analysed using the energetic span model, predicting TOFs of heterocoupling (HB & BH) and homocoupling (HH & BB) pathway and assessing degrees of turnover frequency control (XTOFs). More electron-withdrawing carboxylate ligands and more electron-donating heteroarene substituents decrease activation and reaction free energies of addition steps, with C-2 more favourable than C-3. When heteroarene and catalyst choice are particularly favourable, the first association can become exergonic, causing a significant change in catalytic kinetics. When not exergonic, free energy for addition of benzene is almost always higher, leading to large energetic spans and slower TOFs (BB < HB < BH < HH). Non-exergonic pathways are also faster with more electron-withdrawing ligands and more electron-donating heteroarene substituents because of a lower energy addition steps and smaller energetic span. For exergonic associations, the low energy arene-catalyst intermediate creates a larger energetic span, slowing down the HH and HB pathways. More electron-withdrawing ligands and more electron-donating substituents increase the relative TOF for BH and the reaction requires only small excesses of benzene to induce heterocoupling.

Up to date, many efforts have been made to activate the C—O bond of phenols with different strategies, for example, through in‐situ formed intermediates, through a catalytic reductive dearomatization‐condensation‐rearomatization sequence or catalytic deoxygenation. Abstract:Electrooxidation has emerged as an increasingly viable platform in molecular syntheses that can avoid stoichiometric chemical redox agents. Despite major progress in electrochemical C-H activations, these arene functionalizations generally require directing groups to enable the C-H activation. The installation and removal of these directing groups call for additional synthesis steps, which jeopardizes the inherent efficacy of the electrochemical C-H activation approach, leading to undesired waste with reduced step and atom economy. In sharp contrast, herein we present palladium-electrochemical C-H olefinations of simple arenes devoid of exogenous directing groups. The robust electrocatalysis protocol proved amenable to a wide range of both electron-rich and electron-deficient arenes under exceedingly mild reaction conditions, avoiding chemical oxidants. This study points to an interesting approach of two electrochemical transformations for the success of outstanding levels of position-selectivities in direct olefinations of electron-rich anisoles. A physical organic parameter-based machine learning model was developed to predict position-selectivity in electrochemical C-H olefinations. Furthermore, late-stage functionalizations set the stage for the direct C-H olefinations of structurally complex pharmaceutically relevant compounds, thereby avoiding protection and directing group manipulations.

Abstract:Diazo compounds had been widely employed as a class of environment friendly and highly efficient coupling reagents in transition-metal-catalyzed C-H bonds insertion reactions.Recently, C-H alkylation of various arenes and heteroarenes has been achieved enormous successes using diazo compounds as the alkylation reagents under a new catalytic model.The recent progress in diazo compounds mediated transition-metal-catalyzed C-H alkylation and tandem C-H alkylation/cyclization is reviewed, including reaction mechanism and synthetic applications.Furthermore, the prospects of this reaction are also discussed.

Abstract:The asymmetric cross-coupling reaction is developed as a straightforward strategy toward 1,1-diaryl alkanes, which are a key skeleton in a series of natural products and bioactive molecules in recent years. Here we report an enantioselective benzylic C(sp(3))-H bond arylation via photoredox/nickel dual catalysis. Sterically hindered chiral biimidazoline ligands are designed for this asymmetric cross-coupling reaction. Readily available alkyl benzenes and aryl bromides with various functional groups tolerance can be easily and directly transferred to useful chiral 1,1-diaryl alkanes including pharmaceutical intermediates and bioactive molecules. This reaction proceeds smoothly under mild conditions without the use of external redox reagents.

Abstract:A tandem coupling-ketooxygenation reaction of readily accessible enediyne-carboxylic compounds with inner alkynes has been developed that utilizes the PdCl2/CuBr2 catalytic system under an O2 atmosphere and assembles a class of isoindolinones and o-acylbenzoic acids. The two oxygen atoms are regioselectively incorporated into enediyne units at the 1- and 6-positions from atmospheric molecular oxygen and H2O, respectively, during the present process. This study uncovered a formal [4C + 2C] benzannulation-diketonization of enediynes and alkynes via a coupling and decoupling strategy.

Abstract:Transition-metal catalyzed cross-coupling reactions have emerged as a powerful tool for constructing biaryl compounds. Aryl halides and aryl metallic reagents (typically prepared from aryl halides) are used as coupling partners. It would be desirable to replace either aryl halide or aryl metallic reagents used in cross-couplings reactions with more readily available surrogates. Oxidative dehydrogenative cross-coupling between two different "inert" aryl C-H bonds represents an ideal system that would revolutionize cross-coupling chemistry. Furthermore, cross-coupling reactions might be improved by developing new catalytic protocols based on cheap transition-metal catalysts or even transition-metal-free systems to decrease costs and avoid the use of heavy metal and noble transition metals. It would be desirable to promote both catalytic systems and replace either or both coupling partners. We have used different strategies to improve cross-coupling reactions for constructing biaryls, which we categorized into four groups as follows. First, we focused on developing methodologies to be applied to easily produced and naturally abundant arenol-based electrophiles in cross-coupling via C-O activation. We have extended coupling partners to aryl carboxylates and arenols. Direct application of arenes as surrogates for organohalides and organometallic reagents avoids the tedious preparation of these reagents from arenes and considerably reduces the cost of starting materials. We have also explored cross-coupling reactions of arenes with various organometallic reagents, such as arylboronic acids, arylsilanes, and aryl Grignard reagents. Second, we summarize oxidative cross-coupling reactions based on C-H activation with aryl metallic reagents. On the basis of the reactivity patterns of different organometallic reagents, we adapted different catalytic systems to achieve effective cross-coupling reactions. Third, we improved a well-developed cross-coupling between arenes and organohalides through a strategy of replacing one coupling partner and using a new catalytic system. We have applied earth-abundant transition metals, such as Fe, and Co, and even developed transition-metal-free catalytic systems. Finally, our ultimate goal is to construct biaryls by cross dehydrogenative arylation between two different arenes. Owing to the structural similarity of both arenes, in particular two substituted benzenes, the greatest challenges are not only achieving regio- and chemo-selective C-H activation reactions but also matching both the reactivities and selectivities of both substrates to avoid homocouplings of either arene. Through our efforts, we have developed and applied four different strategies by introducing directing groups, controlling electronic and steric properties, and using dual directing strategies. We hope our studies will stimulate interest and new thinking on cross-couplings reactions for building carbon-carbon bonds from readily available and inexpensive chemicals from basic petroleum chemistry and nature.

Abstract:Pd-catalyzed oxidative C-H/C-H coupling reaction is an emerging type of C-H activation reaction, which attracts great interests because both reaction partners do not require pre-functionalization. In the present study, we employed DFT methods to investigate the mechanism of Pd(OAc)(2)-catalyzed oxidative C-H/C-H coupling of pentafluorobenzene with benzene. Four possible pathways were examined in the C-H activation part: path A benzene-pentafluorobenzene mechanism (C-H activation of benzene occurs before the C-H activation of pentafluorobenzene), path B pentafluorobenzene-benzene mechanism (C-H activation of benzene occurs after the C-H activation of pentafluorobenzene), path C benzene-pentafluorophenyl silver mechanism (C-H activation of benzene and subsequent transmetalation with pentafluorophenyl silver complex), path D pentafluorophenylsilver-benzene mechanism (transmetalation with pentafluorophenyl silver complex and subsequent C-H activation of benzene). Based on the calculations, the sequences of two C-H activation steps are found to be different in the oxidative couplings of same substrates (i.e. pentafluorobenzene and benzene) in different catalytic systems, where the additive Ag salts played a determinant role. In the absence of Ag salts, the energetically favored pathway is path B (i.e. the C-H activation of pentafluorobenzene takes place before the C-H cleavage of benzene). In contrast, with the aid of Ag salts, the coordination of pentafluorophenylsilver to Pd center could occur easily with a subsequent C-H activation of benzene in the second step, and the second step significantly raises the whole reaction barrier. Alternatively, in the presence of Ag salts, the kinetically preferred mechanism is path C (i.e. the C-H activation of benzene takes place in the first step followed by transmetalation with pentafluorophenylsilver complex), which is similar to path A. The calculations are consistent with the H/D exchange experiment and kinetic isotope effects. Thus the present study not only offers a deeper understanding of oxidative C-H/C-H coupling reaction, but also provides helpful insights to further development of more efficient and selective oxidative C-H/C-H coupling reactions.

Abstract:Substantial attention has been given to modem organocopper chemistry in recent years since copper salts are naturally abundant, cheap, and less toxic in comparison to precious metals. Copper salts also exhibit versatility in catalyzing and mediating carbon-carbon and carbon-heteroatom bond forming reactions. Despite the wide applications of copper salts in catalysis, reaction mechanisms have remained elusive. Using azacalix[1]arene[3]pyridine, an arene-embedded macrocycle, and its isolated and structurally well-defined ArCu(II) and ArCu(III) compounds as molecular tools, we now report an in-depth experimental and computational study on the mechanism of a Cu(II)-catalyzed oxidative cross-coupling reaction between arenes and boronic acids with air as the oxidant. Stoichiometric reaction of organocopper compounds with p-tolylboronic acid validated arylcopper(II) rather than arylcopper(III) as a reactive organometallic intermediate. XPS, EPR, H-1 NMR, HRMS, and UV-vis spectroscopic evidence along with the isolation and quantification of all products and copper speciation, combined with computational analysis of the electronic structure and energetics of the transient intermediates, suggested a reaction sequence involving electrophilic metalation of arene by Cu(II), transmetalation of arylboronate to ArCu(II), the redox reaction between the resulting ArCu(II)Ar' and ArCu(II) to form respectively ArCu(III)Ar' and ArCu(I), and finally reductive elimination of ArCu(III)Ar'. Under aerobic catalytic conditions, all Cu(I) ions released from reductive elimination of ArCu(III)Ar' and from protolysis of ArCu(I) were oxidized by oxygen to regenerate Cu(II) species that enters into the next catalytic cycle. The unraveled reactivity of arylcopper(II) compounds and the catalytic cycle would enrich our knowledge of modern organocopper chemistry and provide useful information in the design of copper -catalyzed reactions.

Abstract:A protocol for the synthesis of bibenzyls via nickel acetylacetonate-catalyzed coupling of arenes was devloped using di-tort-butperoxide as the oxidant with the asistance of 1,4-diazabicyclo[2.2.2]octane (DABCO). The process tolerated to different substrates, among of which, isopropylbenzene provided the most satisfactory yield of bibenzyls in 73% yield. The cross-coupling of arenes with cycloalkanes was also realized under the optimal reaction conditions but accompanied with the self-coupling of substrates. The method could effectively restrain the formation of alcohol, aldehyde, acid.
