Abstract:Rifapentine crystals with different habits were prepared by recrystallization from selected solvents, such as methanol, ethanol, chloroform, and acetic acid. Scanning electron microscopy, X-ray powder diffractometry, infrared spectrometry, and differential scanning calorimetry were used to investigate the physicochemical characteristics of the prepared crystals. The comparative dissolution behaviors of the newly developed crystals and of rifapentine without being treated were also studied. Results show that the newly developed crystals were different from each other with respect to physical properties but were identical chemically. Needle-shaped crystals were obtained from methanol, ethanol, and chloroform solvents, and the block-shaped crystals were obtained from acetic acid solvent. X-ray diffraction spectra and differential scanning calorimetry investigation on those developed crystals clearly indicate that rifapentine has different crystal structure modification. When the crystal was obtained from acetic acid, the change of crystal habit was originated from the crystal structure modification. The dissolution rate of newly developed crystals was found to be higher than that of rifapentine without being treated. However, the modified crystal obtained from acetic acid shows the lower dissolution rate than crystals obtained from other solvents.

Abstract:The solubility data of rifamycin S were measured in isopropanol, butyl acetate, and their mixed solvents across the temperature range of 283.15–323.15 K by the gravimetric method. The results demonstrate that the solubility of rifamycin S increases with the increasing temperature in the two pure solvents, and in the mixed solvents, it increases first and then decreases with increasing butyl acetate content. The experimental data of rifamycin S in the mixed solvents were better correlated using the modified Apelblat equation and ideal model equation. Furthermore, the relevant thermodynamic parameters of the dissolution process were determined based on the van't Hoff equation. The obtained dissolution enthalpy and Gibbs free energy are positive in all cases, which indicate that the dissolving process of rifamycin S is endothermic and nonspontaneous. The supersolubility data of rifamycin S were measured by the laser and thermal analytic method. The results demonstrate that the width of the metastable zone of rifamycin S becomes larger with decreasing cooling rate and increasing butyl acetate content. Furthermore, the crystallization process of rifamycin S was optimized on the basis of thermodynamic research. The results showed that when Vbutyl acetate:Vmixed solvent was 0.04, the cooling rate was 0.1 K/min, the stirring rate was 150 rpm, the final crystallization temperature was 283.15 K, and the aging time was 8 h, the purity of rifamycin S crystals could reach 98.5%, and the crystalline yield was 89.6%. After crystallization optimization, the size of rifamycin S crystals increased, and the dissolution in water was improved.This article has not yet been cited by other publications.

Abstract:From an ethanol/water solution containing (1R,3S)-camphoramic acid and a racemic mixture of mandelic acid, the title complex crystallizes in the space group P212121 with a = 8.015(2), b = 11.041(2), c = 21.781(4) Å. Both molecular complexes, the (1R,3S)-camphoramic acid (S)-mandelic acid complex and the (1R,3S)-camphoramic acid (R)-mandelic acid complex, randomly distribute in a disordered crystal structure with an approximate molar ratio of 3:1, thus a 3:1 separation of the mandelic acid enantiomers was achieved. Molecules of (S)-mandelic and (R)-mandelic acid form very similar intermolecular interactions with adjacent camphoramic acid and water molecules in the structure. This results in the formation of a cocrystal containing two diastereomers. The crystal structure consists of camphoramic acid layers and mandelic acid layers. An extensive H-bonding network occurs within and between the layers.

Abstract:PURPOSE: This article investigated the influence of novel rifampicin nanosuspension (RIF NS) for enhancing drug delivery properties.METHODS: RIF NS was fabricated using the antisolvent precipitation technique. The impact of solvent type and flow rate, stabilizer type and concentration, and stirring time and apparatus together with the solvent-antisolvent volume ratio on its controlled nanocrystallization has been evaluated. NSs were characterized by transmission electron microscopy, particle size and zeta potential analysis, solubility, and dissolution profiles. The compatibility between RIF and the stabilizer was investigated via Fourier transform infrared spectroscopy and the differential scanning calorimetry techniques. The shelf-life stability of the RIF NS was assessed within a period of 3 months at different storage temperatures. Cell cytotoxicity was evaluated using 3(4,5-dimethylthiazol-2-yl)-2,5-diphenyltetrazolium bromide (MTT) assay on lung epithelial cells.RESULTS: Polyvinyl alcohol at 0.4% w/v, 1:15 methanol to deionized water volume ratio and 30-minutes sonication were the optimal parameters for RIF NS preparation. Nanocrystals were obtained with a nanometeric particle size (101 nm) and a negative zeta potential (-26 mV). NS exhibited a 50-fold enhancement in RIF solubility and 97% of RIF was dissolved after 10 minutes. The RIF NS was stable at 4±0.5°C with no significant change in particle size or zeta potential. The MTT cytotoxicity assay of RIF NS demonstrated a good safety profile and reduction in cell cytotoxicity with half maximal inhibitory concentration values of 0.5 and 0.8 mg/mL for free RIF and RIF NS, respectively.CONCLUSION: A novel RIF NS could be followed as an approach for enhancing RIF physicochemical characteristics with a prominence of a safer and better drug delivery.

Abstract:Crystal morphology plays a pivotal role in the production process of pharmaceutical compounds and influences the properties of the final product. Achieving control over crystal morphology, especially for thin and needle-like crystals, presents significant challenges to the industry. In this study, we investigate the crystal morphology of rufinamide (RUF) as a model compound with a notorious thread-like morphology. RUF polymorphs crystallize with markedly poor aspect ratios, with the most stable form (form A) exhibiting the thinnest morphology among all. Our initial attempts to modulate RUF crystal morphologies using solvent screening and habit-modifying additives proved unsuccessful. We explored cocrystals as a novel approach to enhance crystal morphology, given their wide application in improving various physicochemical properties of pharmaceutical solids. We employed trimesic acid (TMA) as the auxiliary compound for cocrystallization, resulting in the synthesis of RUF-TMA. The cocrystal was then subjected to solvent screening in pursuit of crystals with isometric morphology. We successfully obtained RUF-TMA cocrystals with an improved morphology using 3-pentanone as the solvent. This study offers a detailed exploration of five crystal structures, encompassing three polymorphs of RUF and two polymorphs of RUF-TMA. The elucidation of crystal structures is accompanied by a comprehensive characterization, including SCXRD, powder X-ray diffraction, thermogravimetric analysis, and differential scanning calorimetry. This study provides valuable insights into controlling crystal morphology and highlights the potential of cocrystals in enhancing morphology when conventional techniques such as solvent and additive screening of pure active pharmaceutical ingredients fall short.

Abstract:The intricate process of protein binding orchestrates crucial drug interactions within the bloodstream, facilitating the formation of soluble complexes. This research endeavours to improve the dissolution and oral bioavailability of Rifampicin (RMP) by strategically manipulating drug-protein binding dynamics and the hydrophobic characteristics of human serum albumin (HSA). Various precipitation techniques leveraging methanol, ammonium sulfate, and heat treatment were meticulously employed to tailor the properties of colloidal albumin (HSA NPs). The resultant complexes underwent comprehensive characterization encompassing evaluations of hydrophobicity, size distribution, surface charge, and structural analyses through FTIR, TG-DSC, XRD, and morphological examinations. The findings revealed a significant binding affinity of 78.07 ± 6.6% with native albumin, aligning with prior research. Notably, the complex RMP-HSA NPs-M13, synthesized via the methanolic precipitation method, exhibited the most substantial complexation, achieving a remarkable 3.5-fold increase, followed by the ammonium sulfate (twofold) and heat treatment (1.07-fold) methods in comparison to native albumin binding. The gastric simulated media exhibited accelerated drug release kinetics, with maximal dissolution achieved within two hours, contrasting with the prolonged release observed under intestinal pH conditions. These findings translated into significant improvements in drug permeation, as evidenced by pharmacokinetic profiles demonstrating elevated Cmax, AUC, t1/2, and MRT values for RMP-HSA NPs-M13 compared to free RMP. In summary, this innovative approach underscores the potential of precipitation methods in engineering stable colloidal carrier systems tailored to enhance the oral bioavailability of poorly soluble drugs, offering a pragmatic and scalable alternative to conventional surfactants, polymers, or high-energy methods for complex formation and production.

Abstract:Rifamycins have been clinically utilized against mycobacterial infections for more than 50 years; however, their biosynthesis has not been fully elucidated. Here, on the basis of in vivo gene deletions, in vitro enzyme assays, isotope labeling, and site-directed mutations, we found that a flavin-dependent monooxygenase encoded by a rifamycin biosynthetic gene cluster, Rif-Orf17, not only converted the naphthoquinone chromophore of rifamycin S into benzo-γ-pyrone but also linearized rifamycin SV through phenolic hydroxylation. Abstract:The new compound was synthesized by the reaction of ZnCl2 and triadimefon (ratio 1: 2) in ethanol solution refluxing for 6 hours. The crystal used for X-ray analysis was obtained by slow evaporation of the solution at room temperature. It crystallizes in the monoclinic system, space group P2/ n. In the complex, the Zn (II) cation is tetrahedrally coordinated to two triadimefon ligands and two chloride in cis disposition. The title complex was also characterized by elemental analysis, molar conductance, IR and TG-DTA.

Abstract:This study reports the successful incorporation of rifampicin into a stearic acid‐based solid lipid nanoparticle formulation, to be used for drug delivery. Using the microwave microemulsion technique, monodisperse nanosized particles are produced. Characterization through various methods, including cryo‐TEM, visually confirms drug encapsulation. Antimicrobial testing shows a dose‐dependent increase in the activity with higher rifampicin loading, highlighting the potential of this system for targeted therapeutic applications. Rifampicin (RF) is a potent antibiotic, but its clinical effectiveness is limited by poor water solubility and stability. To overcome these limitations, RF is encapsulated in solid‐lipid nanoparticles (SLNs) using a one‐pot microwave microemulsion method. This study aimed to synthesize and characterize RF‐loaded SLNs, evaluating their physicochemical characteristics and antimicrobial efficacy. The synthesized SLNs exhibit particle sizes ranging from 150 to 200 nm, with a high encapsulation efficiency of ≈55% and high loading capacity. The particles are imaged by cryogenic‐transmission electron microscopy and found to be circular and elongated, with RF homogenously distributed throughout the particles. The RF‐loaded SLNs demonstrate potent antimicrobial activity, as evidenced by disc diffusion assays. The SLNs exhibits a sustained drug release profile, highlighting their potential as an effective drug delivery system for antimicrobial treatment. These findings suggest that SLNs can enhance the stability, bioavailability, and therapeutic efficacy of hydrophobic drugs like RF.

Uniform and Amorphous Rifampicin Microspheres Obtained by Freezing Induced LLPS During Lyophilization

Abstract:By lyophilization of rifampicin (RIF) solution in TBA/water with various solvent compositions, uniform and amorphous rifampicin (RIF) microspheres were produced. Using 55% TBA solution, the obtained RIF microspheres have a mono-dispersive size distribution with diameters range from 1 to 3 μm. The RIF microspheres are found to be amorphous by X-ray diffraction, and are expected to dissolve much faster than the crystalline RIF upon inhalation. Mechanistic investigation revealed that the amorphous RIF microspheres were formed due to liquid-liquid phase separation (LLPS) occurred during the freezing of the TBA/water solution. We also observed that the RIF microspheres can be readily phagocytized by activated THP-1 cells within 15 min. The suitable size distribution, high solubility, and readiness for phagocytosis by macrophages, all suggest that the lyophilized amorphous RIF microspheres could be potentially used as an anti-tuberculosis inhalation therapy. In addition, similar process was used to lyophilize TBA/water solutions of several other drugs, including rifaximin, rifapentine, paclitaxel, and isoniazid. We found that for drugs with appropriate physiochemical properties, such as paclitaxel and rifaximin, mono-dispersive microspheres could be obtained as well, which demonstrated that freezing induced LLPS could be utilized as a novel particle engineering methodology to produce drug microspheres by lyophilization.
Variations in the capacity of the steroid binding beta globulin (SBbetaG) in plasma from normal young twins.

Abstract:The capacity of human plasma to bind specifically dihydrotestosterone (DHTBC) has been determined in normal monozygotic and dizygotic twins less than 20 years old. The total variation in DHTBC was analysed by stepwise multiple regression, combined with variance analysis, applied to the pair sums in DHTBC, body height and body weight, as well as to age. A significant part of the variation in DHTBC was explained. Due to the strong interwining between body weight, body height and age in growing children it was impossible to make out which one of the latter variable contributed most to the negative correlation which was observed. Previous studies in adults did, however, show that only body weight and not body height or age correlated negatively with DHTBC. By doing the same calculations for the within-pair differences in DHTBC, body height and body weight it was hoped to delineate the within-family part of the environmental variation in DHTBC. Body weight and body height did, however, not offer any significant explanation as to this part of the variation in DHTBC. The upper limit of heritability (h2) for DHTBC was estimated by comparing intraclass-correlation coefficients in monozygotic and dizygotic twins. According to the method used to calculate the latter intraclass-correlation coefficients h2 estimates for DHTBC of 0.774 and 0.960 were arrived at. However, due to the rather small number of dizygotic twins available the confidence limits of these estimates were rather large.

Abstract:The specific crystalline form of a compound remarkably affects its physicochemical properties. Therefore, a detailed analysis of the structural features and intermolecular interactions of a multi-component crystal is feasible to understand the relationships among the structure, physicochemical properties and the formation mechanism. In the present study, three novel cocrystal salt solvates of rhein and berberine were reported for the first time. Various solid characterizations and theoretical computations based on density functional theory (DFT) were carried out to demonstrate the intermolecular interactions. The theoretical computation shows that the strongest interaction existed between berberine cation and rhein anion, and the electrostatic interaction play a dominant role. However, no salt bond was observed between them. Further intrinsic dissolution rate analysis in water shows that the monohydrate exhibits 17 times enhancement in comparison with rhein. The rhein and berberine combined in ionic state in cocrystal salt is the main reason for the solubility improvement. This paper suggests that the interactions between the different components can be visualized and qualitatively and quantitatively analyzed by theoretical computation, which is helpful to understand the relationship between stereochemical structure and physicochemical properties of multi-component complex.

Abstract:Extensins are abundant proteins presumed to determine physical characteristics of the plant cell wall. We have cloned a cDNA encoding LeExt1 from a tomato (Lycopersicon esculentum Mill.) root hair cDNA library. The deduced sequence of the LeExt1 polypeptide defined a novel type of extensin-like proteins in tomato. Patterns of mRNA distribution indicated that expression of the LeExt1 gene was initiated in the root hair differentiation zone of the tomato rhizodermis. Cloning of the corresponding promoter and fusion to the -glucuronidase (GUS) reporter gene allowed detailed examination of LeExt1 expression in transgenic tomato plants. Evidence is presented for a direct correlation between LeExt1 expression and cellular tip growth. LeExt1/GUS expression was detectable in trichoblasts (=root hair-bearing cells), but not in atrichoblasts of the tomato rhizodermis. Both hair formation and LeExt1 expression was inducible by the plant hormone ethylene. Comparative analysis of the LeExt1/GUS expression was performed in transgenic tomato, potato (Solanum tuberosum), tobacco (Nicotiana tabacum), and Arabidopsis plants. In the apical/basal dimension, GUS staining was absent from the root cap and undifferentiated cells at the root tip in all species investigated. It was induced at the distal end of the differentiation zone and remained high proximally to the root/hypocotyl boundary. In the radial dimension, GUS expression was root hair specific in the solanaceous species. Whereas LeExt1 mRNA was exclusively detectable in the rhizodermis, root hair-specific expression correlated with GUS expression in germinating pollen tubes. This is correlative evidence for a role of LeExt1 in root hair tip growth [corrected].

Abstract:A novel chemical structure named pharmaceutical cocrystal was synthesized with the purpose to enhance the solubility of levofloxacin (LVFX). Nicotinic acid (NA) was selected as cocrystal coformer (CCF). The cocrystal was successfully obtained by solvent evaporation method combined with ultrasonic technology. The crystal structure of LVFX-NA is triclinic with a P1 space group, unit cell parameters a = 7.1392 Å, b = 10.0188 Å, c = 17.5394 Å, α=86.109 (7), β=86.082 (6), γ=73.809 (7), z = 2, R1=0.0518, ωR2=0.1425. The supramolecular structure of LVFX-NA is mainly controlled by OH···N and CH···O intermolecular hydrogen bond. Moreover, intramolecular hydrogen bonds CH···F have also been found in the cocrystal structural units. The obtained cocrystal exhibits unique properties of thermoanalysis, Fourier transform infrared (FT-IR) spectrum and X-ray powder diffraction (PXRD) spectrum, demonstrated the new formed crystal material. Equilibrium solubility measurement revealed that LVFX-NA exhibited improved aqueous solubility, that is 5.02 times higher than the original LVFX.

Abstract:The solubility of rifapentine in acetic acid and n-octanol solvent mixtures was measured at temperatures ranging from (278.15 to 323.15) K. The mole fraction of n-octanol in the solvent mixtures (x(3)) range from 0.0 to 0.5993. For the seven group data studied, the experimental solubility data were well-correlated with the data, calculated by means of a semiempirical equation.

Abstract:Riccardin D is a new compound extracted from liverwort Marchantia polymorpha L. It has been proved to be useful in antifungal therapy and reversing the resistance of Candida albicans against fluconazole. However, the poor solubility leads to the poor bioavailability and limits its development. In this study, nanocrystals were prepared in the evaporative precipitation into aqueous solution (EPAS) and the microfluidisation process. The characterizations of nanocrystals were compared by transmission electron microscope, size distribution, and zeta potential. In the EPAS method, the drug was dissolved in the organic phase and F68, HPMC, PVP K30 were dissolved in water with the mass ratio of 2:1:2:1. In the microfluidisation process, two key factors – pressure and number of cycles were screened and 8 cycles at 2000bar was the most efficient parameter. The nanocrystals made in EPAS process were smaller, more uniform and had a narrower distribution than the microfluidisation nanocrystals. Differential scanning calorimetry (DSC) and X-ray diffraction confirmed the crystalline states that were both reserved. The solubility was greatly improved by the two methods and the EPAS nanocrystals were more soluble due to the smaller size. An enhanced dissolution was obvious in vitro. And the stable nanocrystals were successfully achieved by the two methods.
