Abstract:The intricate process of protein binding orchestrates crucial drug interactions within the bloodstream, facilitating the formation of soluble complexes. This research endeavours to improve the dissolution and oral bioavailability of Rifampicin (RMP) by strategically manipulating drug-protein binding dynamics and the hydrophobic characteristics of human serum albumin (HSA). Various precipitation techniques leveraging methanol, ammonium sulfate, and heat treatment were meticulously employed to tailor the properties of colloidal albumin (HSA NPs). The resultant complexes underwent comprehensive characterization encompassing evaluations of hydrophobicity, size distribution, surface charge, and structural analyses through FTIR, TG-DSC, XRD, and morphological examinations. The findings revealed a significant binding affinity of 78.07 ± 6.6% with native albumin, aligning with prior research. Notably, the complex RMP-HSA NPs-M13, synthesized via the methanolic precipitation method, exhibited the most substantial complexation, achieving a remarkable 3.5-fold increase, followed by the ammonium sulfate (twofold) and heat treatment (1.07-fold) methods in comparison to native albumin binding. The gastric simulated media exhibited accelerated drug release kinetics, with maximal dissolution achieved within two hours, contrasting with the prolonged release observed under intestinal pH conditions. These findings translated into significant improvements in drug permeation, as evidenced by pharmacokinetic profiles demonstrating elevated Cmax, AUC, t1/2, and MRT values for RMP-HSA NPs-M13 compared to free RMP. In summary, this innovative approach underscores the potential of precipitation methods in engineering stable colloidal carrier systems tailored to enhance the oral bioavailability of poorly soluble drugs, offering a pragmatic and scalable alternative to conventional surfactants, polymers, or high-energy methods for complex formation and production.

Study on Dissolution Thermodynamics and Cooling Crystallization of Rifamycin S

Abstract:The solubility data of rifamycin S were measured in isopropanol, butyl acetate, and their mixed solvents across the temperature range of 283.15–323.15 K by the gravimetric method. The results demonstrate that the solubility of rifamycin S increases with the increasing temperature in the two pure solvents, and in the mixed solvents, it increases first and then decreases with increasing butyl acetate content. The experimental data of rifamycin S in the mixed solvents were better correlated using the modified Apelblat equation and ideal model equation. Furthermore, the relevant thermodynamic parameters of the dissolution process were determined based on the van't Hoff equation. The obtained dissolution enthalpy and Gibbs free energy are positive in all cases, which indicate that the dissolving process of rifamycin S is endothermic and nonspontaneous. The supersolubility data of rifamycin S were measured by the laser and thermal analytic method. The results demonstrate that the width of the metastable zone of rifamycin S becomes larger with decreasing cooling rate and increasing butyl acetate content. Furthermore, the crystallization process of rifamycin S was optimized on the basis of thermodynamic research. The results showed that when Vbutyl acetate:Vmixed solvent was 0.04, the cooling rate was 0.1 K/min, the stirring rate was 150 rpm, the final crystallization temperature was 283.15 K, and the aging time was 8 h, the purity of rifamycin S crystals could reach 98.5%, and the crystalline yield was 89.6%. After crystallization optimization, the size of rifamycin S crystals increased, and the dissolution in water was improved.This article has not yet been cited by other publications.

Uniform and Amorphous Rifampicin Microspheres Obtained by Freezing Induced LLPS During Lyophilization

Abstract:By lyophilization of rifampicin (RIF) solution in TBA/water with various solvent compositions, uniform and amorphous rifampicin (RIF) microspheres were produced. Using 55% TBA solution, the obtained RIF microspheres have a mono-dispersive size distribution with diameters range from 1 to 3 μm. The RIF microspheres are found to be amorphous by X-ray diffraction, and are expected to dissolve much faster than the crystalline RIF upon inhalation. Mechanistic investigation revealed that the amorphous RIF microspheres were formed due to liquid-liquid phase separation (LLPS) occurred during the freezing of the TBA/water solution. We also observed that the RIF microspheres can be readily phagocytized by activated THP-1 cells within 15 min. The suitable size distribution, high solubility, and readiness for phagocytosis by macrophages, all suggest that the lyophilized amorphous RIF microspheres could be potentially used as an anti-tuberculosis inhalation therapy. In addition, similar process was used to lyophilize TBA/water solutions of several other drugs, including rifaximin, rifapentine, paclitaxel, and isoniazid. We found that for drugs with appropriate physiochemical properties, such as paclitaxel and rifaximin, mono-dispersive microspheres could be obtained as well, which demonstrated that freezing induced LLPS could be utilized as a novel particle engineering methodology to produce drug microspheres by lyophilization.
An aerosol nanocomposite microparticle formulation using rifampicin-cyclodextrin inclusion complexes for the treatment of pulmonary diseases

Abstract:Rifampicin (RIF) is commonly used in the treatment of tuberculosis (TB), a bacterium that currently infects one fourth of the world's population. Despite the effectiveness of RIF in treating TB, current RIF treatment regimens require frequent and prolonged dosing, leading to decreased patient compliance and, ultimately, increased mortality rates. This project aims to provide an alternative to oral RIF by means of an inhalable spray-dried formulation. TB uses alveolar macrophages to hide and replicate until the cells rupture, further spreading the bacteria. Therefore, delivering RIF directly to the lungs can increase the drug concentration at the site of infection while reducing off-site side effects. Cyclodextrin (CD) was used to create a RIF-CD inclusion complex to increase RIF solubility and biodegradable RIF-loaded NP (RIF NP) were developed to provide sustained release of RIF. RIF NP and RIF-CD inclusion complex were spray dried to form a dry powder nanocomposite microparticles (nCmP) formulation (RIF-CD nCmP). RIF-CD nCmP displayed appropriate aerosol dispersion characteristics for effective deposition in the alveolar region of the lungs (4.0 µm) with a fine particle fraction of 89 %. The nCmP provided both a burst release of RIF due to the RIF-CD complex and pH-sensitive release of RIF due to the RIF NP incorporated into the formulation. RIF-CD nCmP did not adversely affect lung epithelial cell viability and RIF NP were able to effectively redisperse from the nCmP after spray drying. These results suggest that RIF-CD nCmP can successfully deliver RIF to the site of TB infection while providing both immediate and sustained release of RIF. Overall, the RIF-CD nCmP formulation has the potential to improve the efficacy for the treatment of TB.

Abstract:PURPOSE: This article investigated the influence of novel rifampicin nanosuspension (RIF NS) for enhancing drug delivery properties.METHODS: RIF NS was fabricated using the antisolvent precipitation technique. The impact of solvent type and flow rate, stabilizer type and concentration, and stirring time and apparatus together with the solvent-antisolvent volume ratio on its controlled nanocrystallization has been evaluated. NSs were characterized by transmission electron microscopy, particle size and zeta potential analysis, solubility, and dissolution profiles. The compatibility between RIF and the stabilizer was investigated via Fourier transform infrared spectroscopy and the differential scanning calorimetry techniques. The shelf-life stability of the RIF NS was assessed within a period of 3 months at different storage temperatures. Cell cytotoxicity was evaluated using 3(4,5-dimethylthiazol-2-yl)-2,5-diphenyltetrazolium bromide (MTT) assay on lung epithelial cells.RESULTS: Polyvinyl alcohol at 0.4% w/v, 1:15 methanol to deionized water volume ratio and 30-minutes sonication were the optimal parameters for RIF NS preparation. Nanocrystals were obtained with a nanometeric particle size (101 nm) and a negative zeta potential (-26 mV). NS exhibited a 50-fold enhancement in RIF solubility and 97% of RIF was dissolved after 10 minutes. The RIF NS was stable at 4±0.5°C with no significant change in particle size or zeta potential. The MTT cytotoxicity assay of RIF NS demonstrated a good safety profile and reduction in cell cytotoxicity with half maximal inhibitory concentration values of 0.5 and 0.8 mg/mL for free RIF and RIF NS, respectively.CONCLUSION: A novel RIF NS could be followed as an approach for enhancing RIF physicochemical characteristics with a prominence of a safer and better drug delivery.

Abstract:Rifampicin is characterized by a conjugated system with a weak native fluorescence that was enhanced after incorporation into β‐cyclodextrin micelles. Rifampicin is a frontline antibiotic in the management of tuberculosis. Since no spectrofluorimetric methods are reported for this drug, this approach was challenged to craft a sensitive, reliable, valid, fast, and green methodology. In recent years, fluorescence spectroscopy has received a lot of interest. Its benefits include ecological greenness and analytical performance. The pharmaceutical industries greatly like this approach because of its low energy and decreased solvent usage, which make it both economical and environmentally friendly. This methodology was based on utilizing the enhanced native fluorescence of the rifampicin at 341 nm after excitation at 241 nm in a beta‐cyclodextrin micellar system. Modern developments in analytical chemistry have been applied to reduce risks to the workplace and environment by using distilled water as a dilution solvent for method application and optimization. The method was found excellent green with 97 eco‐scale and 0.86 AGREE scores besides an 89.6 overall whiteness score. The range of linearity for rifampicin raw material was 0.2–1.5 μg·mL−1, and the average recoveries for raw material and spiked plasma were 100.15% and 99.64%, respectively. The suggested technique worked well for the commercial oral syrup of Rimactane® and did not conflict with any common additives.

Abstract:Acinetobacter baumannii is a gram-negative bacterium causing severe hospital-acquired infections such as bloodstream infections or pneumonia. Moreover, multidrug resistant A. baumannii becomes prevalent in many hospitals. Consequently, the World Health Organization made this bacterium a critical priority for the research and development of new antibiotics. Rifabutin, a semisynthetic product from the rifamycin class, was recently found to be very active in nutrient-limited eukaryotic cell culture medium against various A. baumannii strains, including extremely drug-resistant strains, with minimal inhibitory concentrations as low as 0.008 μg/mL. Moreover, this in vitro potency translates into in vivo efficacy. Thus, rifabutin appears to be an attractive novel antibiotic against A. baumannii. In this work, our objective was to design and synthetize rifabutin prodrugs with increased aqueous solubility to allow intraveneous use. Synthetic methodologies were developed to selectively functionalize the hydroxyl group in position 21 and to afford 17 prodrugs. We measured the water solubility of the prodrugs, the stability in human and mouse plasma and their antimicrobial activity against A. baumannii after incubation in human serum. Finally, a pharmacokinetic release study of rifabutin was performed in CD1 mice with three selected prodrugs as a proof of concept.
Crystal Modification of Rifapentine Using Different Solvents

Abstract:Rifapentine crystals with different habits were prepared by recrystallization from selected solvents, such as methanol, ethanol, chloroform, and acetic acid. Scanning electron microscopy, X-ray powder diffractometry, infrared spectrometry, and differential scanning calorimetry were used to investigate the physicochemical characteristics of the prepared crystals. The comparative dissolution behaviors of the newly developed crystals and of rifapentine without being treated were also studied. Results show that the newly developed crystals were different from each other with respect to physical properties but were identical chemically. Needle-shaped crystals were obtained from methanol, ethanol, and chloroform solvents, and the block-shaped crystals were obtained from acetic acid solvent. X-ray diffraction spectra and differential scanning calorimetry investigation on those developed crystals clearly indicate that rifapentine has different crystal structure modification. When the crystal was obtained from acetic acid, the change of crystal habit was originated from the crystal structure modification. The dissolution rate of newly developed crystals was found to be higher than that of rifapentine without being treated. However, the modified crystal obtained from acetic acid shows the lower dissolution rate than crystals obtained from other solvents.

Abstract:This study aimed to investigate the effect of different polymers (polyethylene glycol 4000 and 6000 and Soluplus®) on the enhancement of solubility, dissolution, and stability of cefixime trihydrate as a selected class II model drug. Different solid dispersions have been prepared using conventional methods and supercritical fluid technology. The effect of co-solvent incorporation in supercritical fluid technology was also studied. Physicochemical properties for solid dispersions were investigated using Fourier transform infrared analysis, differential scanning calorimetry, thermogravimetric analysis, powder X-ray diffraction, and scanning electron microscopy. The solubility of the prepared solid dispersions increased except for those prepared with Soluplus® using supercritical fluid technology without co-solvent. The best enhancement in the release profile was recorded by Soluplus®-based solid dispersions prepared using a conventional method. The conventional methods of preparation and the presence of co-solvent in supercritical fluid technology converted cefixime into its amorphous form.

Abstract:Surfactant-based microemulsions have emerged as an advanced material for universal drug carriers for poorly soluble drugs. These microemulsions (MEs) are based on ionic liquid as oil in presence of traditional surfactants which have been receiving ample attention from research communities all over the world. In this study, we have reported oil-in-water MEs containing a hydrophobic ionic liquid (IL), 1-butyl-3-methylimidazolium hexafluorophosphate (BmimPF6) as oil phase, nonionic surfactants Brij35/Triton X100 and water. The microstructure of these BmimPF6-in-water (IL/w) MEs were identified by using UV-Vis and fluorescence spectroscopy. The breaks observed in the absorption spectra of both probe (p-nitroanaline) and drug (rifampicin) versus wt% of IL can be used to confirm the formation of MEs. Similar behavior has been observed in the emission spectra of fluorescence probe (pyrene) and drug in presence of varied wt% of IL which supported the results of the absorption spectroscopy. The results from dynamic light scattering (DLS) and scanning electron microscopy (SEM) measurements showed that the MEs had a nearly monodisperse droplet size distribution (PDI < 0.4) which indicates the homogeneity in distribution of the ME formulations. The incorporation of rifampicin-drug in the MEs shows notable changes which suggests the accumulation of drug in the palisade layers of the MEs. Further, the surface free energy properties of the MEs are explored from surface tension (γ) measurements. The changes in γ in these different MEs may be due to the changes in microstructures of the ME systems which can be helpful to recognize the transitions occurring in the ME structures. This study offers a better understanding of MEs features and paves path towards the potential applications of MEs in pharmaceutical industry.

Abstract:RECENT reports from most parts of the world show a definite progressive increase in the incidence of early syphilitic infection. 1 Moore et al 2 state that there were 124,000 cases of syphilis reported in the USA in 1962 and also conclude that this figure represents but ⅕ of the total number. The most disturbing factor in the recent upsurge is the large number of young people involved with the possibility of transmission of the disease to their progeny. Abstract:Based on the morphologic characteristics of oocysts of the genus Isospora, we have demonstrated that I. ohioensis is a relatively common Isospora species in dogs which are resident in Chonbuk province, Korea. The prepatent period of I. ohioensis was four days. The size of the unsporulated oocysts in fresh stool specimens was 22.9 x 19.8 microns (R = 1.16). The size of the contracted sporonts was 17.4 x 16.3 microns (R = 1.06). By 96 hours, sporulation is complete and the ratio of length/width was constant relatively. And the sizes of oocysts and sporocysts were 22.8 x 20.5 microns (R = 1.11) and 15.0 x 10.8 microns (R = 1.39), respectively. At this time it is most reliable for the measurements of oocyst sizes of I. ohioensis to provide with the identifiable clues against the others. It is therefore recommended that the clinical fecal specimens suspected of isosporosis should first be incubated and aerated for 96 hours before a definitive parasitological diagnosis can be reached. These and other observations contribute to the understanding of the biological characteristics and laboratory and clinical diagnosis of isosporosis.
Quantification of rifampicin loaded into inhaled polymeric nanoparticles by reversed phase high-performance liquid chromatography in pulmonary nonphagocytic cellular uptake

Abstract:Rifampicin is an antibiotic effective against both Gram-negative and Gram-positive bacteria and is commonly used as a first-line treatment for tuberculosis caused by Mycobacterium tuberculosis. In this study, a reversed-phase high- performance liquid chromatography method was developed and validated to assess rifampicin, either free or combined with ascorbic acid, loaded into chitosan/tween 80-coated alginate nanoparticles. The method utilized a reversed-phase C18 Restek column with specific chromatographic conditions: a mobile phase of 60:40 ratios of methanol/buffer phosphate (pH 7.0), at a flow rate of 0.8 mL/min, and an injection volume of 15 μL. rifampicin and the internal standard (rifamycin) had retention times of 4.0 and 2.5 min, respectively, and were detected at 334 nm. The method demonstrated the stability of stored samples after freezing-thawing cycles and specificity for rifampicin, even in the presence of degradation products from stress conditions. The high-performance liquid chromatography method was found to be specific, precise, robust, and sensitive. Results indicated that rifampicin accumulation and uptake kinetics varied based on cell type, formulation (free or loaded in nanoparticles), rifampicin concentration, and incubation time. Confocal fluorescence microscopy images supported these findings, showing isothiocyanate fluorescein nanoparticles distribution in different intracellular regions depending on the cell type used.

Design and implementation of fuzzy P^2ID controller for handlebar control of a bicycle robot

Abstract:This paper presents the design and implementation of a new fuzzy proportional-integration plus fuzzy proportional-derivative control scheme, named the fuzzy P$^2$ID controller hereafter, for controlling the handlebar of a bicycle robot with time-varying load (disturbance). The fuzzy P$^2$ID controller combines some advantages of the fuzzy PI controller and the ones of the fuzzy PD controller, which can compensate their individual shortcomings. Detailed design is described, along with the implementation of the integrated computer-aided designed bicycle control system. Functionally, the controller is a nonlinear controller with variable gains, providing the expected fast response and robustness. Structurally, it is an embedded version of a conventional PI controller and a PD controller, which has an additional fuzzy control block, where a switching rule of control signal is also implemented. Experimental results show the effectiveness of the new control scheme for controlling the handlebar of a bicycle robot, built from "off-the-shelf" parts with completely unknown hardware parameters. Comparison tests among several existing fuzzy controllers as well as the conventional PID controller have been conducted, so as to evaluate the superior performance of the new scheme. This new control scheme can be directly applied to general robotic systems and other industrial processes under similar physical conditions.
Assessing Polymorphic Purity of Rifampicin in Double and Triple-Drug Fixed-Dose Combination Products

Abstract:First-line tuberculostatic agents, Rifampicin (RIF), Isoniazid (ISH), Ethambutol (ETB), and Pyrazinamide (PZA) are generally administered as a fixed-dose combination (FDC) for improving patient adherence. The major quality challenge of these FDC products is their variable bioavailability, where RIF and its solid state are key factors. In this work, the analysis of the impact of the polymorphism in the performance of RIF in RIF-ISH and PZA-RIF-ISH combined products was carried out by an overall approach that included the development and validation of two methodologies combining near-infrared (NIR) spectroscopy and partial least squares (PLS) to the further evaluation of commercial products. For NIR-PLS methods, training and validation sets were prepared with mixtures of Form I/Form II of RIF, and the appropriate amount of ISH (for double associations) or ISH-PZA (for triple associations). The corresponding matrix of the excipients was added to the mixture of APIs to simulate the environment of each FDC product. Four PLS factors, reduced spectral range, and the combination of standard normal variate and Savitzky-Golay 1st derivative (SNV-D') were selected as optimum data pre-treatment for both methods, yielding satisfactory recoveries during the analysis of validation sets (98.5±2.0%, and 98.7±1.8% for double- and triple-FDC products, respectively). The NIR-PLS model for RIF-ISH successfully estimated the polymorphic purity of Form II in double-FDC capsules (1.02 ± 0.02w/w). On the other hand, the NIR-PLS model for RIF-ISH-PZA detected a low purity of Form II in triple FDC tablets (0.800 ± 0.021w/w), these results were confirmed by X-ray powder diffraction. Nevertheless, the triple-FDC tablets showed good performance in the dissolution test (Q=99-102%), implying a Form II purity about of 80% is not low enough to affect the safety and efficacy of the product.

Abstract:Despite the large number of works on the synthesis of polylactide-co-glycolide (PLGA) nanoparticles (NP) loaded with antituberculosis drugs, the data on the influence of various factors on the final characteristics of the complexes are quite contradictory. In the present study, a comprehensive analysis of the effect of multiple factors, including the molecular weight of PLGA, on the size and stability of nanoparticles, as well as the loading efficiency and release of the antituberculosis drug rifampicin (RIF), was carried out. Emulsification was carried out using different surfactants (polyvinyl alcohol, Tween 80 and Pluronic F127), different aqueous-to-organic phase ratios, and different solvents (dichloromethane, dimethyl sulfoxide, ethyl acetate). In this research, the PLGA nanoemulsion formation process was accompanied by ultrasonic dispersion, at different frequencies and durations of homogenization. The use of the central composite design method made it possible to select optimal conditions for the preparation of PLGA-RIF NPs (particle size 223 ± 2 nm, loading efficiency 67 ± 1%, nanoparticles yield 47 ± 2%). The release of rifampicin from PLGA NPs was studied for the first time using the flow cell method and vertical diffusion method on Franz cells at different pH levels, simulating the gastrointestinal tract. For the purpose of the possible inhalation administration of rifampicin immobilized in PLGA NPs, their mucoadhesion to mucin was studied, and a high degree of adhesion of polymeric nanoparticles to the mucosa was shown (more than 40% within 4 h). In the example of strain H37Rv in vitro, the sensitivity of Mycobacterium tuberculosis to PLGA-RIF NPs was proven by the complete inhibition of their growth.

Cerec3D endocrowns--two-year clinical examination of CAD/CAM crowns for restoring endodontically treated molars.

Abstract:The present clinical study evaluates the clinical prognosis of Cerec3D endocrowns over an observation period of two years. Twenty Cerec endocrowns (VITABLOCS Mark II, Vita Zahnfabirk, Bad Säckingen, Germany) were placed with PanaviaTM F 2.0, a dual-curing bonding composite. The control examinations took place semi-annually and the restorations were evaluated based on modified US Public Health Service criteria (USPHS). The two-year survival rate of the Cerec endocrowns was 90%. Two out of 20 endocrowns were assessed as failure because of fractures. The first fracture was observed after 12 months and the second fracture after 18 months. No recurrent caries was diagnosed during the entire examination period. Slight percussion symptoms were found on three restorations in the first weeks after placement. The results of the present study show that good esthetic and functional results similar to those of other restoration types can be achieved with endocrowns and that CAD/CAM-fabricated crowns represent a very promising treatment alternative for endodontically treated molars.

Abstract:This study reports the successful incorporation of rifampicin into a stearic acid‐based solid lipid nanoparticle formulation, to be used for drug delivery. Using the microwave microemulsion technique, monodisperse nanosized particles are produced. Characterization through various methods, including cryo‐TEM, visually confirms drug encapsulation. Antimicrobial testing shows a dose‐dependent increase in the activity with higher rifampicin loading, highlighting the potential of this system for targeted therapeutic applications. Rifampicin (RF) is a potent antibiotic, but its clinical effectiveness is limited by poor water solubility and stability. To overcome these limitations, RF is encapsulated in solid‐lipid nanoparticles (SLNs) using a one‐pot microwave microemulsion method. This study aimed to synthesize and characterize RF‐loaded SLNs, evaluating their physicochemical characteristics and antimicrobial efficacy. The synthesized SLNs exhibit particle sizes ranging from 150 to 200 nm, with a high encapsulation efficiency of ≈55% and high loading capacity. The particles are imaged by cryogenic‐transmission electron microscopy and found to be circular and elongated, with RF homogenously distributed throughout the particles. The RF‐loaded SLNs demonstrate potent antimicrobial activity, as evidenced by disc diffusion assays. The SLNs exhibits a sustained drug release profile, highlighting their potential as an effective drug delivery system for antimicrobial treatment. These findings suggest that SLNs can enhance the stability, bioavailability, and therapeutic efficacy of hydrophobic drugs like RF.

Abstract:Background: Natural isoflavones are recognized for their diverse pharmacological activities; however, their low aqueous solubility presents a significant challenge for further development. Here, we aimed to develop a cocrystal of formononetin (FMN) to improve its solubility. Methods: The formononetin-imidazole (FMN-IMD) cocrystal was prepared using liquid-assisted grinding method. The prepared cocrystal was identified through a thermal analysis of physical mixtures with various coformers. FTIR and solid-state NMR confirmed the presence of hydrogen bonds and π-π interactions in the FMN-IMD cocrystal. Results: The solubility of FMN-IMD was two to three times higher than that of crystalline FMN. The FMN-IMD cocrystal showed a 4.93-fold increase in the Cmax value and a 3.58-fold increase in the AUC compared to FMN after oral administration in rats. There were no changes in the PXRD of the FMN-IMD cocrystal after six months of storage at 40 °C. Conclusions: Thus, the FMN-IMD cocrystal is proposed as an effective solid form for oral delivery, offering enhanced solubility and physical stability.
