Enhanced Osteogenesis of Polyvinylpyrrolidone-Iodine As a Sterilizing Agent for Preservation of Allografts

Abstract:Sterilization for human allografts is important because of the widespread use of transplants in prosthesis revision, spinal fusion and reconstructive surgery. Here, we investigated the potential of polyvinylpyrrolidone-iodine for sterilization and preservation of allografts. Extracts of contaminated demineralized bone matrix sterilized by polyvinylpyrrolidone-iodine were co-cultured with Staphylococcus aureus and Candida albicans. The growth of the tested microbes was inhibited. The alkaline phosphatase activity of MC-3T3 E1 cells co-cultured with polyvinylpyrrolidone-iodine sterilized demineralized bone matrix extracts was significantly enhanced. We then used the thigh muscle pouch model in nude mice to investigate the ectopic bone formation ability of polyvinylpyrrolidone-iodine sterilized demineralized bone matrix, with Co60 sterilized demineralized bone matrix as the control. Bilateral operation was performed for self-control and the right femoral muscle pouch was implanted with polyvinylpyrrolidone-iodine sterilized demineralized bone matrix. No mice showed signs of postoperative infection; high density bone could be detected in the right side. Histological analysis revealed that the ectopic formation ability of polyvinylpyrrolidone-iodine sterilized demineralized bone matrix was better than Co60 sterilized demineralized bone matrix. Based on the obtained results, we concluded that polyvinylpyrrolidone-iodine is a very promising sterilizing agent for the preservation of allografts.
