Comparative Study of Different Coatings on Magnesium Alloys: The Effect on MSC Proliferation and Osteogenic Differentiation

Abstract:Magnesium (Mg) has been considered as a material with many advantages in medical use. Current study is aimed to investigate characterization of different modified surface coatings of Mg alloy and to compare the proliferation and differentiation of bone marrow-derived mesenchymal stem cell stromal cells (BMSCs) on different Mg alloy after surface modification. The adhesion and viability of BMSCs was analyzed by scanning electron microscopy (SEM). The proliferation and viability of the cells was measured by MU and calcein-AM/EthD-1 staining by immersion extracts co-culture system. Osteogenic differentiation was further assessed using real-time RT-PCR. CaP-AZ31B has rough surface with large amount of calcium phosphate deposition, while AZ31 B and F-AZ31B shows even and smooth surface morphology. Protein adsorption ability of CaP-AZ31B is higher than other groups at 24 h. BMSCs were more confluent and interconnective in CaP-AZ31B group than those on other groups. BMSCs on CaP-AZ31B group displayed the highest increase in viability at Day 5 and Day 7. Real-time RT-PCR analysis showed that CaP-AZ31B group significantly upregulated the expression of COL-1, ALP, OC and OPN in BMSCs. These results demonstrated that CaP coatings on AZ31B significantly promote BMSCs proliferation as well as osteogenic differentiation.
