Abstract:The diamondback moth, Plutella xylostella L. (Lepidoptera: Plutellidae), is a major pest of Brassica crops worldwide. This study explores the application of Brassica spp. allelochemicals in trapping diamondback moth adults. The effectiveness of eight commercially obtained Brassica spp. volatiles has been investigated, including - (Z)-3-hexenyl acetate, cis-3-hexene-1-ol, β-pinene, sabinene, n-heptanal, allyl isothiocyanate, and Brassica non-volatiles - sinigrin and 4-methylsulfinylbutyl glucosinolate. These allelochemicals were tested at varying concentrations (10 to 90 μg/μL) in laboratory and open-screen cage environment trapping experiments. Cis-3-hexene-1-ol, (Z)-3-hexenyl acetate, sabinene, and β-pinene significantly attracted and trapped more female moths than controls; attraction decreased at higher concentrations. A 1:1 blend of cis-3-hexene-1-ol and (Z)-3-hexenyl acetate displayed optimal attraction in lab and open environment trials for the tested blends. Among the colors tested, green-colored containers proved to be the most effective for trapping. While the plant allelochemicals attracted diamondback moths, signs of non-target cabbage loopers were observed on treated plants. Y-tube olfactometer assays revealed that (Z)-3-hexenyl acetate and cis-3-hexene-1-ol were attractive to both male and female moths. These findings demonstrate the potential of Brassica spp. allelochemicals, particularly cis-3-hexene-1-ol and (Z)-3-hexenyl acetate, for trapping diamondback moths. Abstract:The biological control function provided by natural enemies is regarded as a protection goal that should not be harmed by the application of any new pest management tool. Plants producing Cry proteins from the bacterium, Bacillus thuringiensis (Bt), have become a major tactic for controlling pest Lepidoptera on cotton and maize and risk assessment studies are needed to ensure they do not harm important natural enemies. However, using Cry protein susceptible hosts as prey often compromises such studies. To avoid this problem we utilized pest Lepidoptera, cabbage looper (Trichoplusia ni) and fall armyworm (Spodoptera frugiperda), that were resistant to Cry1Ac produced in Bt broccoli (T. ni), Cry1Ac/Cry2Ab produced in Bt cotton (T. ni), and Cry1F produced in Bt maize (S. frugiperda). Larvae of these species were fed Bt plants or non-Bt plants and then exposed to predaceous larvae of the green lacewing Chrysoperla rufilabris. Fitness parameters (larval survival, development time, fecundity and egg hatch) of C. rufilabris were assessed over two generations. There were no differences in any of the fitness parameters regardless if C. rufilabris consumed prey (T. ni or S. frugiperda) that had consumed Bt or non-Bt plants. Additional studies confirmed that the prey contained bioactive Cry proteins when they were consumed by the predator. These studies confirm that Cry1Ac, Cry2Ab and Cry1F do not pose a hazard to the important predator C. rufilabris. This study also demonstrates the power of using resistant hosts when assessing the risk of genetically modified plants on non-target organisms.
Effect of Bt Broccoli and Resistant Genotype of Plutella Xylostella (lepidoptera: Plutellidae) on Life History and Prey Acceptance of the Predator Coleomegilla Maculata (coleoptera: Coccinellidae)

Abstract:The ecological implications on biological control of insecticidal transgenic plants, which produce crystal (Cry) proteins derived from the soil bacterium Bacillus thuringiensis (Bt), remains a contentious issue and affects risk assessment decisions. In this study, we used a unique system of resistant insects, Bt plants and a predator to critically evaluate this issue. The effects of broccoli type (normal or expressing Cry1Ac protein) and insect genotype (susceptible or Cry1Ac-resistant) of Plutella xylostella L. (Lepidoptera: Plutellidae) were examined for their effects on the life history of the predator, Coleomegilla maculata DeGeer (Coleoptera: Coccinellidae) over two generations. Additional behavioral studies were conducted on prey choice. C. maculata could not discriminate between Bt-resistant and susceptible genotypes of P. xylostella, nor between Bt and normal broccoli plants with resistant genotypes of P. xylostella feeding on them. The larval and pupal period, adult weight and fecundity of each female were not significantly different when C. maculata larvae fed on different genotypes (Bt-resistant or susceptible) of insect prey larvae reared on Bt or non-Bt broccoli plants. The life-history parameters of the subsequent generation of C. maculata fed on Bt broccoli-reared resistant P. xylostella were also not significantly different from those on non-Bt broccoli. These results indicated that Cry1Ac did not harm the life history or prey acceptance of an important predator after two generations of exposure. Plants expressing Cry1Ac are unlikely to affect this important predator in the field. (C) 2015 Elsevier Inc. All rights reserved.

Abstract:Cabbage looper, Trichoplusia ni (Hübner) is an important lepidopteran pest on many vegetable and greenhouse crops, and some field crops. Although there are no commercial transgenic Bt vegetable or greenhouse crops, T . ni is a target of Bollgard II cotton, which produces Cry1Ac and Cry2Ab. We expand on previous work that examined the effect of Bt crops on parasitoids using Bt-resistant lepidopteran populations as hosts. Cry1Ac/Cry2Ab-resistant T . ni larvae were used to eliminate host quality effects and to evaluate the direct effects of Bt cotton on the parasitoids Copidosoma floridanum (Ashmead) and Cotesia marginiventris (Cresson). These tri-trophic studies confirm that Bt cotton had no significant impact on development, success of parasitism, survival and adult longevity of C . marginiventris when using Bt-resistant T . ni fed on Bt cotton. Similarly, this Bt cotton had no significant impact on the development, mummy weight and the number of progeny produced by C . floridanum . Our studies verified that lyophilized Bt crop tissue maintained its insecticidal bioactivity when incorporated into an artificial diet, demonstrating that hosts and parasitoids were exposed to active Cry proteins. The egg-larval parasitoid C . floridanum , or similar species that consume their entire host, should be considered useful surrogates in risk assessment of Bt crops to non-target arthropods.

Abstract:This study introduced the protease inhibitor-encoding gene sporamin from sweet potato together with the wound-responsive promoter pMSPOA into the Chinese cabbage ( Brassica campestris ssp. chinensis var. parachinensis ) through Agrobacterium tumefaciens -mediated transformation. Transgenic lines were confirmed via PCR detection of both the sporamin and hygromycin gene fragments in the cloning vector. The expression level of sporamin in the different lines was determined using RT-PCR analysis. Leaves from four individual transgenic lines including T0-2, T0-3, T0-4 and T0-5, possessing higher levels of sporamin than control plants, were then used to conduct insect feeding studies. Second instar larvae of diamondback moth ( Plutella xylostella L.) were used in feeding assays. All four transgenic lines demonstrated high levels of resistance against diamondback based on average body weight of total incubated larvae following feeding assays and amount of leaf damage.

Abstract:The fall armyworm, Spodoptera frugiperda (J. E. Smith) (Lepidoptera: Noctuidae), is an important pest of maize in the United States and many tropical areas in the western hemisphere. In 2001, Herculex I ® (Cry1F) maize was commercially planted in the United States to control Lepidoptera, including S. frugiperda . In 2006, a population of S. frugiperda was discovered in Puerto Rico that had evolved resistance to Cry1F maize in the field, making it the first well-documented case of an insect with field resistance to a plant producing protein from Bacillus thuringiensis (Bt). Using this resistant population, we conducted tri-trophic studies with a natural enemy of S. frugiperda . By using resistant S. frugiperda , we were able to overcome possible prey-mediated effects and avoid concerns about potential differences in laboratory- or field-derived Bt resistance. We used the Cry1F-resistant S. frugiperda to evaluate effects of Cry1F on Cotesia marginiventris (Cresson) (Hymenoptera: Braconidae), a larval endoparasitoid of S. frugiperda, over five generations. Our results clearly demonstrate that Cry1F maize does not affect development, parasitism, survivorship, sex ratio, longevity or fecundity of C. marginiventris when they parasitize Cry1F maize-fed S. frugiperda. Furthermore, the level of Cry1F protein in the leaves was strongly diluted when transferred from Bt maize to S. frugiperda and was not detected in larvae, cocoons or adults of C. marginiventris. Our results refute previous reports of C. marginiventris being harmed by Bt proteins and suggest that such results were caused by prey-mediated effects due to using Bt-susceptible lepidopteran hosts.
Morphology and Glucosinolate Profiles of Chimeric Brassica and the Responses of Bemisia Tabaci in Host Selection, Oviposition and Development

Abstract:Plant structures and chemicals, which are developed from the shoot apical meristem (SAM), form the main barriers to insect feeding. A plant chimera containing cells of different genetic origins in the SAM will be morphologically and chemically different compared with the parents and thus may result in differential resistance to herbivores. In this study, we explore if particular elements of plant resistance are localized in one of the layers of SAM; the replacement of one cell layer in a chimera may be linked to change of a single resistance trait to herbivores. The morphology and glucosinolate profiles of two periclinal chimeras (labeled as TTC and TCC, respectively) and grafted parents tuber mustard (labeled as TTT) and red cabbage (labeled as CCC) were compared and the performance of whitefly (Bemisia tabaci) in host selection, oviposition preference and development were assessed under controlled conditions. Both chimeras possessed leaf trichomes as parent tuber mustard TTT, however, TTC had significantly more trichomes than TCC and parent TTT. Leaf wax content of both chimeras was intermediate between the two parents. Five aliphatic and two indole glucosinolates were detected in both chimeras, whereas three aliphatic glucosinolates (3-methyl-sulfinylpropyl, 4-methyl-sulfinylbutyl and 2-hydroxy-3-butenyl) were not detected in tuber mustard, and one aliphatic glucosinolate (3-butenyl) was not detected in red cabbage. Unexpectedly for a chimera, the quantities of two aliphatic glucosinolates (3-methyl-sulfinylpropyl and 4-methyl-sulfinylbutyl) in both TTC and TCC were 3- to 5-fold higher than parents. In olfactory preference assays, B. tabaci showed preference to CCC, followed by TCC, TTC and TTT, and number of eggs laid showed the same pattern: CCC>TCC>TTC>TTT. Interestingly, more whiteflies landed on TTT plants than the other three types in a free choice experiment and the developmental duration from egg to adult was the shortest on TTT and increased in the order TTT
Comprehensive Evaluation on Qualities of Chinese Flowering Cabbage Resources and Resistance to Diamondback Moth

Abstract:[Objective] In order to provide the theory basis for quality improvement and cultivating varieties of high resistance to diamondback moth,the relations between the resistance to diamondback moth and qualities of Chinese flowering cabbage resources were researched and evaluated comprehensively.[Method] The study analyzed rules for soluble protein,sucrose,reducing sugar and soluble starch with eight Chinese flowering cabbage strains for materials,meanwhile,analyzed the resistance to diamondback moth of Chinese flowering cabbage strains by the evaluation method in vitro.[Result] The strains of No.8 and No.5 were high resistance to diamondback moth,the soluble protein content of two strains high,but the sucrose of the two strains were lower.The susceptible strains were No.1,No.2 and No.4.The soluble protein content of No.1,No.2 and No.4 were lower,and the sucrose of No.2 and No.4 were higher.[Conclusion] There was significant positive correlation between the resistance to diamondback moth and soluble protein content of Chinese flowering cabbage,and there was significant negative correlation between the resistance to diamondback moth and sucrose content of Chinese flowering cabbage.

Abstract:Cultivating insect pest-resistant varieties is one of the most effective ways to prevent or mitigate pest infestation in Chinese cabbage ( Brassica campestris ssp. chinensis ). Via the agrobacterium tumefaciens-mediated transformation method, this study introduced the protease inhibitor encoding gene sporamin into two widely cultured cultivars ‘Youdonger’ and ‘Shanghaiqing’, of the common variety of Chinese cabbages ( B. campestriss ssp. chinensis var. communis ), getting transgenic plants with high sporamin expression. In vitro insect bioassays indicated that, compared with the wild type plants, the transgenic plants exhibited improved resistance to diamondback moth ( Plutella xylostella L.) The analysis of inheritance pattern of exogenous sporamin in the progenies of single copy insertion transgenic lines demonstrated that sporamin could be inherited and expressed stably in transgenic progenies. Field survey of the insect resistance under the normal culture condition confirmed the enhanced resistance in transgenic progenies to diamondback moth. Our results strongly suggest that sporamin is an efficient candidate gene for insect-resistant genetic engineering in Chinese cabbage.

Abstract:Chinese cabbage (Brassica rapa L. ssp. pekinensis) ranks among the most cultivated and consumed vegetables in China. A major threat to its production is Plasmodiophora brassicae, which causes large root tumors, obstructing nutrient and water absorption and resulting in plant withering. This study used a widely targeted metabolome technique to identify resistance-related metabolites in resistant (DH40R) and susceptible (DH199S) Chinese cabbage varieties after inoculation with P. brassicae. This study analyzed disease-related metabolites during different periods, identifying 257 metabolites linked to resistance, enriched in the phenylpropanoid biosynthesis pathway, and 248 metabolites linked to susceptibility, enriched in the arachidonic acid metabolism pathway. Key metabolites and genes in the phenylpropanoid pathway were upregulated at 5 days post-inoculation (DPI), suggesting their role in disease resistance. In the arachidonic acid pathway, linoleic acid and gamma-linolenic acid were upregulated at 5 and 22 DPI in resistant plants, while arachidonic acid was upregulated at 22 DPI in susceptible plants, leading to the conclusion that arachidonic acid may be a response substance in susceptible plants after inoculation. Many genes enriched in these pathways were differentially expressed in DH40R and DH199S. The research provided insights into the defense mechanisms of Chinese cabbage against P. brassicae through combined metabolome and transcriptome analysis.

Effects of Host Plant on the Oviposition Preference of Plutella Xylostella (L.) and Host-Selection Behavior of Cotesia Plutellae (Kurdjmov)

Abstract:The ovipositional preference of the diamondback moth, Plutella xylostella (L.) between different Brassica vegetable plants and the host selection by Cotesia plutellae (Kurdjmov) between host larvae on different Brassica plants were studied. With the presence of both the Chinese cabbage, B. campestris L. ssp. pekinensis, the common cabbage, Brassica oleracea L. var. capitata, P. xylostella, moths laid three times as many eggs on the former as that on the latter plants. When C. plutellae wasps were provided with equal numbers of larvae of P. xylostella on both species of plants in one area, parasitism rate of hosts on B. campestris was almost five times that on B. oleracea. The possible effects of infochemicals from the Brassica plants on the oviposition preference of P. xylostella and on the host selection by C. plutellae were discussed.

Abstract:[Objective] The changes of carbohydrate metabolism and expression of proteins were analyzed to provide the basic theory information for exploring the interactions between the diamondback moth feeding and defense response of Chinese flowering cabbage.[Method] Mter 24 h the diamondback moth feeding on ‘ Sijiu'Chinese flowering cabbage,the study analyzed rules for soluble starch,soluble protein,sugar and reducing sugar,meanwhile,analyzed expression of proteins by Tandem mass tag (TMT) coupled to two-dimensional liquid chromatography tandem mass spectrometry (2D LC-MS/MS).[Result] The soluble starch accumulation decreased,and soluble protein and soluble sugar (including sugar,reducing sugar) content increased.430 discrepant proteins were detected by bioinformatics analysis using the SEQUEST search engine,211 proteins were up-regulated with a more than 1.2 folds change,and 119 proteins were down-regulated with a less than 0.83 folds.Among them,the ones of photosynthesis were down regulated,while the others related to stress response,some of hydrolytic enzymes and energy metabolism were up regulated.[Conclusion] The TMT was a powerful method for isolating and identifying differentially expressed proteins between the diamondback moth feeding and defense response of Chinese flowering cabbage.Most of them were involved in stress resistance,glycometabolism,photosynthesis,respiration,energy and substance metabolism.
Eco-Geographical and Botanical Patterns of Resistance to Lepidoptera Insects in Brassica rapa L.

Abstract:In the context of the widespread expansion of damage by herbivorous pests of Brassica crops, taking into account the requirements for minimizing pesticide pollution of the environment, it is important to have fundamental knowledge of the geographical features of the distribution of pests and about the botanical confinement of plant resistance in order to develop a strategy for creating new Brassica cultivars with complex resistance to insects. The relevance of our work is related to the study of the variability in the degree of resistance of the extensive genetic diversity of Brassica rapa accessions to the main herbivorous pests of Brassica crops in contrasting ecological and geographical zones of the Russian Federation (Arctic, northwestern, and southern zones). We have studied the distribution and food preferences of Lepidoptera insects (diamondback moth Plutella xylostella and cabbage moth Mamestra brassicae) on a set of 100 accessions from the VIR B. rapa collection (Chinese cabbage, pakchoi, wutacai, zicaitai, mizuna, and leaf and root turnips) in the field in three zones of the Russian Federation. We have found that the diamondback moth and cabbage moth are largely harmful in three zones of the European part of the Russian Federation, although the degree of damage to plants by these insects varies by year of cultivation. On average, for the set studied during the two years of the experiment, the degree of plant damage by both pests in the Arctic zone was low and almost low, and in the northwestern and southern zones, it was medium. It was noted that diamondback moth damage was greater in the northwestern zone in both years and in the southern and Arctic zones in 2021, while in 2022, the degree of cabbage moth damage was slightly higher in the southern and Arctic zones. Under the conditions of field diamondback moth damage, the accessions of Chinese cabbage, wutacai, and mizuna turned out to be the most resistant (the damage score was 1.92–1.99), whereas the accessions of wutacai and pakchoi were the most resistant to the cabbage moth (the damage score was 1.62–1.78). A high variability in the degree of resistance of Brassica crops to Lepidoptera insects from complete resistance to susceptibility was revealed. We have identified sources of resistance to insects, including complex resistance in all study areas, among landraces and some modern cultivars of Chinese cabbage, pakchoi, wutacai, and mizuna from Japan and China, as well as European turnips. The highest susceptibility to pests in the studied set was noted in the accession of root turnip "Hinona" (k-1422, USA) (average damage score of 3.24–3.53 points). We were not able to establish the morphological features of resistant plants or the geographical confinement of the origin of resistance of B. rapa crop accessions.

Abstract:The term 'dead-end trap cropping' has recently been proposed to identify a plant that is highly attractive for oviposition by an insect pest, but on which offspring of the pest cannot survive. The potential of the wild crucifer Barbarea vulgaris R. Br. to allure and serve as a dead-end trap crop for the diamondback moth Plutella xylostella (L.), an important pest of cruciferous crops worldwide, was examined in laboratory experiments. When P. xylostella adults were provided with a dual-choice of plants of B. vulgaris, and Chinese cabbage Brassica campestris (L.), in one arena, adult moths laid 2.5-6.8 times more eggs on the former than on the latter. When P. xylostella adults were provided with a dual-choice of plants of B. vulgaris and common cabbage Brassica oleracea L., adult moths laid virtually all their eggs on the former and ignored the latter. Nearly all P. xylostella eggs laid on the three species of plants hatched successfully, but nearly all individuals on plants of B. vulgaris died as neonates or early instar larvae, while 87-100% of the larvae on Chinese cabbage and common cabbage survived to pupation. Dual choice tests with a Y-tube olfactometer showed that volatiles from B. vulgaris were much more attractive to P. xylostella adults than those from common cabbage. The results demonstrate that B. vulgaris has a great potential as a dead-end trap crop for improving management of P. xylostella. Factors that may influence the feasibility of using B. vulgaris as a trap crop in the field are discussed, and ways to utilize this plant are proposed.
Expression of cry1Aa gene in cabbage imparts resistance against diamondback moth (Plutella xylostella)

Abstract:Brassica oleracea cv. Pride of India is one of the most promising vegetable cultivars commercially grown as cash crop in Himachal Pradesh, India. However, its overall production is severely hampered by diamondback moth (Plutella xylostella), a notorious pest. To avoid yield losses caused by this pest, cryIAa gene was introduced into cabbage cv. Pride of India using Agrobacterium-mediated transformation method. In an attempt to maximize the transformation frequency, critical determinant factors such as explant type, pre-incubation and co-cultivation period, and acetosyringone effect were successfully optimized. The highest transformation frequency (4.67% and 14.50%) in cotyledon and hypocotyl explant was achieved with a pre-incubation period of 72 h and co-cultivation period of 48 h. Furthermore, transformation frequency was enhanced in cotyledon (18.66%) and hypocotyl (32.00%) explants, when selective regeneration medium was fortified with 100 µM acetosyringone, respectively. The transgene (cryIAa) integration and copy number were confirmed using PCR and Southern blotting. Reverse transcriptase PCR and quantitative real-time PCR analyses were performed that proved transcriptional expression of cryIAa gene in PCR-positive transgenic events. Transgenic cabbage-fed diamondback moth larvae showed significantly higher mortality, thereby proving transgene effectiveness against insect pest control.

Abstract:Clubroot is a serious soil-borne disease of crucifers caused by the obligate parasite Plasmodiophora brassicae . The genetic basis and histopathology of clubroot resistance in two Chinese cabbage ( Brassica rapa ssp. pekinensis ) inbred lines Bap055 and Bap246, challenged with pathotype 4 of P. brassicae , was evaluated. The Chinese cabbage cultivar “Juxin” served as a susceptible check. The resistance in Bap055 was found to be controlled by the CRa gene, while resistance in Bap246 fit a model of control by unknown recessive gene. Infection of the roots by P. brassicae was examined by inverted microscopy. Despite their resistance, primary and secondary infection were observed to occur in Bap055 and Bap246. Primary infection was detected at 2 days post-inoculation (DPI) in “Juxin,” at 4 DPI in Bap055, and at 6 DPI in Bap246. Infection occurred most quickly on “Juxin,” with 60% of the root hairs infected at 10 DPI, followed by Bap055 (31% of the root hairs infected at 12 DPI) and Bap246 (20% of the root hairs infected at 14 DPI). Secondary infection of “Juxin” was first observed at 8 DPI, while in Bap055 and Bap246, secondary infection was first observed at 10 DPI. At 14 DPI, the percentage of cortical infection in “Juxin,” Bap055 and Bap246 was 93.3, 20.0, and 11.1%, respectively. Although cortical infection was more widespread in Bap055 than in Bap246, secondary infection in both of these hosts was restricted relative to the susceptible check, and the vascular system remained intact. A large number of binucleate secondary plasmodia were observed in “Juxin” and the vascular system was disrupted at 16 DPI; in Bap055 and Bap246, only a few secondary plasmodia were visible, with no binucleate secondary plasmodia. The defense mechanisms and expression of resistance appears to differ between Chinese cabbage cultivars carrying different sources of resistance.

Abstract:Potential trap crops for the diamondback moth, Plutella xylostella (L.) (Lepidoptera: Plutellidae), were evaluated through a series of ovipositional preference and larval survival experiments in outdoor screenhouses in 2002 and 2003. Hosts examined as trap crops were glossy and waxy collards, Brassica oleracea L. variety acephala; Indian mustard, Brassica juncea (L.) Czern; and yellow rocket, Barbarea vulgaris (R. Br.) variety arcuata. More eggs were laid on the potential trap crops, with the exception of waxy collards, than on cabbage. When P. xylostella was offered multiple hosts at the same time, numbers of eggs laid on glossy collards, Indian mustard, and yellow rocket were 3, 18, and 12 times greater than on cabbage, respectively. Similarly, when P. xylostella was offered a single trap crop host and cabbage, numbers of eggs laid on glossy collards, Indian mustard, and yellow rocket were 300, 19, and 110 times greater than on cabbage, respectively. Our studies suggest differences in oviposition between the potential trap crops and cabbage were likely due to host volatiles, leaf morphology and color, or a combination of these factors, rather than to total leaf areas, leaf shape, or plant architecture. Two-choice tests with a Y-tube olfactometer indicated that plant volatiles were major factors in P. xylostella host preference. The percentage larval survival from egg to pupation was 22.2% on cabbage, 18.9% on waxy collards, and 24.4% on Indian mustard, whereas survival was significantly lower on glossy collards (6.7%) and yellow rocket (0%). Based on our tests, it seems that yellow rocket may be the best candidate for use as a trap crop for P. xylostella because it is highly attractive for oviposition, but larvae do not survive on it.

Abstract:Chinese cabbage is the most consumed leafy crop in East Asian countries. However, premature bolting induced by continuous low temperatures severely decreases the yield and quality of the Chinese cabbage, and therefore restricts its planting season and geographic distribution. In the past 40 years, spring Chinese cabbage with strong winterness has been selected to meet the market demand. Here, we report a genome variation map of Chinese cabbage generated from the resequencing data of 194 geographically diverse accessions of three ecotypes. In-depth analyses of the selection sweeps and genome-wide patterns revealed that spring Chinese cabbage was selected from a specific population of autumn Chinese cabbage around the area of Shandong peninsula in northern China. We identified 23 genomic loci that underwent intensive selection, and further demonstrated by gene expression and haplotype analyses that the incorporation of elite alleles of VERNALISATION INSENTIVE 3.1 (BrVIN3.1) and FLOWER LOCUS C 1 (BrFLC1) is a determinant genetic source of variation during selection. Moreover, we showed that the quantitative response of BrVIN3.1 to cold due to the sequence variations in the cis elements of the BrVIN3.1 promoter significantly contributes to bolting-time variation in Chinese cabbage. Collectively, our study provides valuable insights into the genetic basis of spring Chinese cabbage selection and will facilitate the breeding of bolting-resistant varieties by molecular-marker-assisted selection, transgenic or gene editing approaches.
Biology of diamond back moth, Plutella xylostella Linn.

Abstract:Accepted : June, 2009 Diamond back moth, Plutella xylostella Linn. is the most noxious pest of cabbage crop which occurs almost all over the world and causes severe damage to the crop. The pest was first recorded from South America in 1890 and then from Venezuala in 1939 (Saravaiya and Patel, 2005). The larvae mine the cabbage leaves on their undersurfaces in the earlier stages while in the later stages they feed on leaves. The larvae feed on the leaves to the extent of 62 to 78 per cent, causing irregular patches on the leaves (Harcourt, 1957). All the leaf tissues are consumed by the larvae except the veins. The last stage larvae are voracious feeders which cause more injury than the first three larval instars creating a ‘windows’ to the leaves. Thus, the larvae skeletonise the plants and growth of plants remains stunted in cases of severe attack. The pest causes the damage right from seedling stage to till harvest. As a result there is a reduction in quality and yield of the produce. Abraham and Padmanaban (1968) reported 31 per cent yield loss to cabbage crop due to P. xylostella attack while Krishnakumar et al. (1986) estimated 52 per cent losses in the marketable yield due to the attack of this pest.
