Abstract:Objective:To investigate the effect of surface modifications on the peroxidase-like activity of iron oxide nanoparticles.Methods:Magnetic iron oxide nanoparticles were prepared by co-precipitation method.The morphology of particles was observed by using TEM.The obtained γ-Fe2O3 nanoparticles were modified with meso-2,3-Dimercaptosuccinic acid(DMSA),citric acid,tartaric acid and 3-aminopropyltriethoxysilanes(APTS),respectively,and the resultant products were characterized by zeta potential measurements.Like horseradish peroxidase(HRP),γ-Fe2O3 nanoparticles were found to be able to catalyze a colour reaction of the substrate 3,3′,5,5′-tetramethylbenzidine(TMB) in the presence of H2O2.The catalysis was evaluated by detecting the colour reaction using UV-visible spectrophotometer and microplat reader.Results:(1) TEM measurements showed that the mean diameter of γ-Fe2O3 nanoparticles was 13 nm.(2) The peroxidase-like activity of γ-Fe2O3 nanoparticles was found to be related to their surface charge.TMB carries two amine groups,likely yielding stronger affinity toward a negatively charged nanoparticle surfaces.(3)γ-Fe2O3/DMSA nanoparticles with the strongest negative surface charge exhibited the highest peroxidase-like activity than others when TMB acts as a substrate.The kinetic parameter of γ-Fe2O3/DMSA nanoparticles were determined to be Km(TMB)=0.3 mmol·L-1,exhibiting a similar enzyme activity to the natural HRP.Conclusions:The peroxidase-like activity of γ-Fe2O3 nanoparticles can be readily regulated by surface modifications,thus providing a potential advantage for application as a HRP mimic enzyme.

Abstract:We found that the peroxidase-like activity of gold nanoparticles (GNPs) followed the MichaelisMenten kinetic model and was dependent on environmental pH and temperature, which was very similar to natural Horseradish Peroxidase (HRP). However, unlike HRP, which needs a lower H2O2 concentration with a very narrow range to reach a maximum reaction rate and avoid enzyme poisoning, GNPs have very high activity, even at an H2O2 concentration two orders of magnitude higher than HRP. It was demonstrated that H2O2 treatment could enhance the peroxidase-like activity of GNPs, resulting thus in the activity increase in a circular catalytic reaction by the reduplicative use of GNPs. It was also found that the peroxidase-like activity of GNPs responded sensitively to nanoparticle size and surface modifications. When used in an immunoassay, GNPs were generally conjugated with antibody and blocked with hydrophilic macromolecules to construct a nanoprobe. This strongly reduced the peroxidase-like activity and detection sensitivity of GNPs, therefore, restricting their use as peroxidase mimetics. We presented a novel strategy that combined the nanoprobes with gold staining to expose fresh catalytic gold surfaces and obtained a great increase in detection sensitivity.
2D PdCu Alloy Nanodendrites Manifest Effective Peroxidase-Like Activity Against Biofilms

Abstract:Noble metal nanomaterials with peroxidase-like catalytic activity have received great interest lately for their potential applications in biomedicine and environmental protection; however, it is still challenging to achieve high catalytic efficiency despite enormous efforts. In this work, a novel but simple route was developed to synthesize 2D PdCu alloy nanodendrites (PdCu NDs) as a high-performance peroxidase mimic for biofilm elimination. Catalytic kinetics shows that the composition-dependent synergy between Pd and Cu in the PdCu NDs can strongly enhance the peroxidase-like activity. Density functional theory calculations further provide the underlying mechanisms at both atomic and electronic levels for the effective adsorption and dissociation of H2O2 molecules on PdCu NDs surfaces. Owing to their superior peroxidase-like activity, the PdCu NDs exhibit striking biofilm inhibition properties, which suggests that the controlled synthesis of 2D noble metal alloy may open up new opportunities for enhancing enzyme-like activities of noble metal nanomaterials.
Surface gold atoms determine peroxidase mimic activity in gold alloy nanoparticles

Abstract:The development of peroxidase mimic nanocatalysts is relevant for oxidation reactions in biosensing, environmental monitoring and green chemical processes. Several nanomaterials have been proposed as peroxidase mimic, the majority of which consists of noble metals and oxide nanoparticles (NPs). Yet, there is still limited information about how the change in the composition influences their catalytic activity. Here, the peroxidase mimic behaviour of gold NPs is compared to a traditional nanoalloy as Au‐Ag and to the Au‐Fe and the Au‐Co nanoalloys, which were not tested before as oxidation catalysis. Since the alloys of gold with iron and cobalt are thermodynamically unstable, laser ablation in liquid (LAL) is exploited for the synthesis of these NPs. Using LAL, no chemical stabilizers or capping agents are present on the NPs surface, allowing the evaluation of the oxidation behaviour as a function of the alloy composition. The results point to the importance of surface gold atoms in the catalytic process, but also indicate the possibility of obtaining active nanocatalysts with a lower content of Au by alloying it with iron, which is earth‐abundant, non‐toxic and low cost. Overall, Au nanoalloys are worth consideration as a more sustainable alternative to pure Au nanocatalysts for oxidation reactions.

Ph-Dependent Growth of Atomic Pd Layers on Trisoctahedral Gold Nanoparticles to Realize Enhanced Performance in Electrocatalysis and Chemical Catalysis

Abstract:In this work, the controlled epitaxial growth of ultrathin Pd shells of a few atomic layers (denoted as nL) on the surfaces of gold nanoparticle (Au NP) cores of different morphologies (trisoctahedral, cubic, and spherical shapes) in the presence of cetyltrimethylammonium chloride (CTAC) was achieved by regulating the pH value of the aqueous CTAC solution and finely tuning the amount of the Pd precursor. It was found that the critical shell thickness for epitaxial Pd growth at the optimal pH value was 4 atomic layers, taking {331}-faceted trisoctahedral (TOH) Au@Pd-nL NPs as an example, on the basis of the results of atomic-resolution high-angle annular dark-field scanning transmission electron microscopy (HAADF-STEM) images. Moreover, the resulting TOH Au@Pd-1L NPs (100.9 m(2) g(-1), 13.2 A mg(Pd)(-1) and 13.1 mA cm(-2)) exhibited excellent electrocatalytic performance and long-term electrocatalytic activity for ethanol oxidation, around 4.8-fold, 66-fold, and 21.8-fold better than commercial Pd/C catalysts (31 m(2) g(-1), 0.2 A mg(Pd)(-1), and 0.6 mA cm(-2)). Furthermore, the resulting TOH Au@Pd-1L NPs not only markedly enhance the chemical catalytic activity for the reduction of 4-nitrophenol (4-NP), but also allow the in situ surface-enhanced Raman spectroscopy (SERS) monitoring of the reaction process of the Pd-catalyzed reduction of 4-NTP. Thus, our work may provide a new way to fabricate core-shell (CS) bimetallic NPs with the merits of both metal outer shells (excellent catalytic performance in electrocatalysis and chemical catalysis) and Au NP cores (reaction process by in situ SERS monitoring).
A simple route to CoFe2O4 nanoparticles with shape and size control and their tunable peroxidase-like activity

Abstract:Recent studies have suggested that the physical and chemical properties of nanoparticles (NPs) strongly depend on local chemical composition, size, and shape. Here, we report a new precursor-mediated growth of monodisperse magnetic cobalt ferrite (CoFe2O4) NPs with controlled size and shape. CoFe2O4 NPs with near corner-grown cubic, near cubic and polyhedron shape can be successfully prepared by simply tuning the amount of iron and cobalt acetylacetonates in oleic acid. Interestingly, the product shape varies from near corner-grown cubic to starlike by only changing the reaction temperature from 320 degrees C to 330 degrees C. These CoFe2O4 NPs exhibit size and shape-dependent peroxidase-like activity towards 3,30,5,50-tetramethylbenzdine (TMB) in the presence of H2O2, and thus exhibited different levels of peroxidase-like activities, in the order of spherical > near corner-grown cubic > starlike > near cubic > polyhedron; this order was closely related to their particle size and crystal morphology. CoFe(2)O(4)NPs exhibited high stability in HAc-NaAc buffer (pH = 4.0) and high activity over a broad pH (2.5-6.0). Furthermore, the Michaelis constants K-m value for the CoFe2O4 NPs (0.006 mM) with TMB as the substrate was lower than HRP (0.062 mM) and Fe3O4 NPs (0.010 mM). After further surface functionalization with folic acid (FA), the folate-conjugated CoFe2O4 nanoparticles allow discrimination of HeLa cells (folate receptor overexpression) from NIH-3T3 cells (without folate receptor expression). Such investigation is of great significance for peroxidase nanomimetics with enhanced activity and utilization.

Abstract:Abstract Owing to their remarkable properties, gold nanoparticles are applied in diverse fields, including catalysis, electronics, energy conversion and sensors. However, for catalytic applications of colloidal gold nanoparticles, the trade-off between their reactivity and stability is a significant concern. Here we report a universal approach for preparing stable and reactive colloidal small (~3 nm) gold nanoparticles by using multi-dentate polyoxometalates as protecting agents in non-polar solvents. These nanoparticles exhibit exceptional stability even under conditions of high concentration, long-term storage, heating and addition of bases. Moreover, they display excellent catalytic performance in various oxidation reactions of organic substrates using molecular oxygen as the sole oxidant. Our findings highlight the ability of inorganic multi-dentate ligands with structural stability and robust steric and electronic effects to confer stability and reactivity upon gold nanoparticles. This approach can be extended to prepare metal nanoparticles other than gold, enabling the design of novel nanomaterials with promising applications.

Evaluation of Biochemical Behavior and Stability of Gold Nanoparticles with High Intrinsic Peroxidase-Like Activity

Abstract:In this study, the biochemical behavior and stability of gold nanoparticles with intrinsic peroxidase-like activity were evaluated as potential native enzyme alternatives. The gold nanoparticles were synthesized at physiological temperature using bovine serum albumin as the stabilizer and then characterized by the TEM imaging method. Afterward, their peroxidase-like activity was checked upon irreversible oxidation of 3,3’-diaminobenzidine to produce a brown-colored indamine polymer, and the specific enzyme-like activity of the as-prepared nanoparticles was also calculated. The results showed a specific activity as high as 0.4212 UI μM-1 for the as-prepared gold nanoparticles. Thereafter, their stability and biochemical performances were evaluated considering their enzyme-like activity as a reliable index. The as-prepared nanoparticles showed their maximal activity at pH=5.0 and 20.0±1.0 °C according to the results of pH and thermal stability studies, in order. Besides, the nanoparticles saved above 80.0% of their maximal activity over pH=3.0-4.0. As a significant advantage compared to the natural enzymes, the as-synthesized gold nanoparticles revealed a pHindependent enzyme-like activity over a wide pH range of pH=7.0-10.0 along with a temperature-independent activity over t=23-28°C. The salt stability studies showed that their activity was not affected by variations in the ionic strength of the reaction media. The kinetics results showed a Vmax of 83.3 μM min-1 and a Km as very low as 0.005 M for the gold nanoparticles. Considering the above results, the as-prepared gold nanoparticles can be considered high stable nanozymes with high intrinsic peroxidase-like activity and excellent catalytic efficiency.
Enhanced peroxidase-like activity of Fe@PCN-224 nanoparticles and their applications for detection of H2O2and glucose

Abstract:Inspired by natural hemin structure, the fabrication of metal organic framework (MOF) nanomaterials constructed by metal ions and iron porphyrin for peroxidase mimicking has attracted extensive attention recently due to their high concentration of active sites and great channeling for substrate diffusion. Herein, by using benzoic acid as capping agent, we fabricated uniform PCN-224 nanoparticles (NPs), which were composed of Zr6 cluster and tetrakis(4-carboxyphenyl)porphyrin (H2TCPP). Then Fe(Ⅲ) ions were incorporated into the center of H2TCPP producing iron porphyrin unit similar to hemin serving as catalytic active site. The procedure of iron incorporation didn't change the size and morphology of PCN-224 NPs. The newly formed Fe@PCN-224 NPs possessed peroxidase-like activity with much lower Km values and higher Kcat values than most of peroxidase-like nanozymes as well as natural horseradish peroxidase (HRP), indicating their enhanced catalytic activity. Taking advantage of the excellent peroxidase-like activity of Fe@PCN-224 NPs, we successfully developed a colorimetric method for simple and sensitive detection of hydrogen peroxide and glucose in combination with glucose oxidase.

Abstract:We report a facile, green, and high yielding approach for the synthesis and stabilization of relatively monodisperse gold nanoparticles using typical nonionic biosurfactant ethoxylated sterol (BPS-30). The obtained gold nanoparticles capped by BPS-30 not only showed remarkable stability after several months of storage under ambient conditions, but also exhibited exciting stability in high concentrations of electrolyte aqueous solutions such as acid, base, salt, and even physiological phosphate buffered saline (PBS). Derjaguin-Landau-Verwey-Overbeek (DLVO) theory was introduced to interpret the relationship between the capping mechanism and the excellent stability of the nanoparticles. UV-vis absorption spectroscopy was used to determine the yield of the gold nanoparticles. Moreover, the catalytic efficiency of the gold nanoparticles was evaluated by using the reduction of 4-nitroaniline by potassium borohydride in aqueous solution.

Abstract:Au nanoparticles half-encapsulated in nano iron oxide are prepared and loaded on alumina as a support. The donation of electrons from nano iron oxide to Au nanoparticles is detected and both the properties of gold and iron oxide are adjusted by the donation. The properties are different from the bulk iron oxide supported gold catalysts, in which the iron oxide is little influenced by the electronic interaction between the two components. The catalyst shows noticeably promoted activity for the aerobic oxidation of 1-phenyl-ethanol over Au-Al2O3 and Au-bulk FeOx. The enhanced catalytic behavior may result from the cooperative effect between the Au nanoparticles and nano iron oxide.

Abstract:The introduction of functional molecules to the surface of magnetic iron oxide nanoparticles (NPs) is of critical importance. Most previously reported methods were focused on surface ligand attachment either by physisorption or covalent conjugation, resulting in limited ligand loading capacity. In this work, we report the seeded growth of a nucleotide coordinated polymer shell, which can be considered as a special form of adsorption by forming a complete shell. Among all of the tested metal ions, Fe(3+) is the most efficient for this seeded growth. A diverse range of guest molecules, including small organic dyes, proteins, DNA, and gold NPs, can be encapsulated in the shell. All of these molecules were loaded at a much higher capacity compared to that on the naked iron oxide NP core, confirming the advantage of the coordination polymer (CP) shell. In addition, the CP shell provides better guest protein stability compared to that of simple physisorption while retaining guest activity as confirmed by the entrapped glucose oxidase assay. Use of this system as a peroxidase nanozyme and glucose biosensor was demonstrated, detecting glucose as low as 1.4 μM with excellent stability. This work describes a new way to functionalize inorganic materials with a biocompatible shell.
Surface coating–modulated peroxidase-like activity of maghemite nanoparticles for a chromogenic analysis of cholesterol

Abstract:Maghemite (γ-Fe2O3) nanoparticles (NPs) emerging as an artificial enzymes have demonstrated an excellent peroxidase-like activity and thus gained much attention in various biological and medical applications. But naked γ-Fe2O3 NPs are aqueously instable and prone to aggregation in biological solutions such as blood plasma. Surface coating for γ-Fe2O3 NPs is thus necessitated to achieve better stability and biocompatibility. In this work, three typical coating layers including poly(lactic-co-glycolic acid) (PLGA), carboxymethyl chitosan (CMCS), and human serum albumin (HSA) were utilized as modifiers to decorate γ-Fe2O3 NPs and fabricate compound NPs including NPPLGA, NPCMCS, and NPHSA, respectively, and subsequently, the peroxidase-like activity of these NPs was evaluated with colorimetric analysis of cholesterol detection. The results showed that the surface coating barely affected peroxidase-like activity of NPs but could remarkably amend stability in the determined pH and temperature ranges. As evidenced with kinetic parameters, the enzymatic catalysis of NPs accorded well with Michaelis–Menten kinetics. Moreover, the catalytic assay demonstrated that the fabricated NPPLGA, NPCMCS, and NPHSA showed a capable catalytic activity using cholesterol as substrate, and especially, the NPPLGA showed a higher peroxidase-like activity compared with the NPCMCS and NPHSA. In conclusion, herein we obtained a coating layer-modulated peroxidase-like activity of γ-Fe2O3 NPs for a visualized analysis of cholesterol, which could be extended for cholesterol detection in biomedical analyses in the future.

Iron oxide coated gold nanorods: synthesis, characterization, and magnetic manipulation

Abstract:We report a simple process to generate iron oxide coated gold nanorods. Gold nanorods, synthesized by our three-step seed mediated protocol, were coated with a layer of polymer, poly(sodium 4-styrenesulfonate). The negatively charged polymer on the nanorod surface electrostatically attracted a mixture of aqueous iron(II) and iron(III) ions. Base-mediated coprecipitation of iron salts was used to form uniform coatings of iron oxide nanoparticles onto the surface of gold nanorods. The magnetic properties were studied using a superconducting quantum interference device (SQUID) magnetometer, which indicated superparamagnetic behavior of the composites. These iron oxide coated gold nanorods were studied for macroscopic magnetic manipulation and were found to be weakly magnetic. For comparison, premade iron oxide nanoparticles, attached to gold nanorods by electrostatic interactions, were also studied. Although control over uniform coating of the nanorods was difficult to achieve, magnetic manipulation was improved in the latter case. The products of both synthetic methods were monitored by UV-vis spectroscopy, zeta potential measurements, and transmission electron microscopy. X-ray photoelectron spectroscopy was used to determine the oxidation state of iron in the gold nanorod-iron oxide composites, which is consistent with Fe2O3 rather than Fe3O4. The simple method of iron oxide coating is general and applicable to different nanoparticles, and it enables magnetic field-assisted ordering of assemblies of nanoparticles for different applications.

Abstract:Pt modified Fe3O4 magnetic nanoparticles (Fe3O4@Pt NPs) were synthesized by a simple method. The morphology and crystal structure of the as-prepared nanocomposite were characterized by TEM, XRD and EDS. The peroxidase-like catalytic activity of the synthesized nanoparticles was also investigated. Catalysis was found to follow Michaelis–Menten kinetics. The calculated kinetic parameters of the Fe3O4@Pt NPs show that they exhibited strong affinity with substrates and enhanced catalytic activity than that of Fe3O4 nanoparticles, suggesting a higher peroxidase-like activity.

Abstract:Iron oxides are directly coated on the surface of cetyl-trimethylammonium bromide (CTAB)-capped gold nanorods (AuNRs) in aqueous solutions at room temperature, which results in AuNR@Fe2O3, AuNR@Fe3O4, and AuNR@Fe2O3@Fe3O4 core-shell heterostructures. The iron oxide shells are uniform, smooth, with characteristic porous structure, and their thickness can be readily tuned. The shell formation is highly dependent on the reaction parameters including pH and CTAB concentration. The Fe2O3 shell is amorphous and exhibits nearly zero remanence and coercivity, while the Fe3O4 shell is ferromagnetic with a low saturation magnetization of about 0.5 emu g-1 due to its low crystallinity and the porous structure. At elevated temperatures achieved by plasmonic heating of the Au core, the Fe2O3 shell transforms from amorphous to γ-Fe2O3 and α-Fe2O3 phases, while the Fe3O4 phase disappears because of the oxidation of Fe2+. A 1.4-fold increase of photocatalytic performance is observed due to the plasmonic resonance provided by the Au core. The photocatalytic efficiency of Fe3O4 is about 1.7-fold higher than Fe2O3 as more surface defects are present on the Fe3O4 shell, promoting the adsorption and activation of reagents on the surface during the catalytic reactions. This approach can be readily extended to other nanostructures including Au spherical nanoparticles and nanostars. These highly uniform and multifunctional core-shell heterostructures can be of great potential in a variety of energy, magnetic, and environment applications.

Abstract:Submicron-sized poly(N-isopropyl acrylamide)/polyethyleneimine core-shell microgels were prepared in aqueous media by using tert-butyl hydroperoxide (TBHP) as an initiator, and then the gold nanoparticles (similar to 8 nm) were formed on the surface of the microgels. The amino groups on the polyethyleneimine (PEI) chains act as the binder for the assembly of the gold nanoparticles/microgel complex. In aqueous media the microgels are highly stable with the gold nanoparticles on their extended PEI chains, and this multi-scale nanoparticle complex can be recovered from water and redispersed in water. The nanogold/microgel particles were conjugated with the enzymes horseradish peroxidase (HRP) and urease. It is found that under identical assay conditions the enzyme/nanogold/microgel systems exhibit enhanced biocatalytic activity over free enzymes in solution, especially at lower enzyme concentrations. In addition, compared to free HRP, the HRP/nanogold/microgel systems show higher activity at varied pHs and temperatures, as well as higher storage stability. Thus the novel nanogold/microgel particles can serve as an excellent support for enzymes.
Peroxidase-like activity of gold nanoparticles stabilized by hyperbranched polyglycidol derivatives over a wide pH range

Abstract:The aim of this work was to carry out comparative studies on the peroxidase-like activity of gold nanoparticles (AuNPs) stabilized with low molecular weight hyperbranched polyglycidol (HBPG-OH) and its derivative modified with maleic acid residues (HBPG-COOH). The influence of the stabilizer to gold precursor ratio on the size and morphology of nanoparticles obtained was checked, and prepared nanoparticles were characterized by means of transmission electron microscopy and UV-Vis spectroscopy. The results indicated the divergent effect of increasing the concentration of stabilizers (HBPG-OH or HBPG-COOH) on the size of the nanostructures obtained. The gold nanoparticles obtained were characterized as having intrinsic peroxidase-like activity and the mechanism of catalysis in acidic and alkaline mediums was consistent with the standard Michaelis-Menten kinetics, revealing a strong affinity of AuNPs with 2, 2'-azino-bis(3-ethylbenzothiazoline-6-sulfonic acid) (ABTS) and 3, 3', 5, 5'-tetramethylbenzidine (TMB), and significantly lower affinity towards phenol. By comparing the kinetic parameters, a negligible effect of polymeric ligand charge on activity against various types of substrates (anionic or cationic) was indicated. The superiority of steric stabilization via the application of tested low-weight hyperbranched polymers over typical stabilizers in preventing salt-induced aggregation and maintaining high catalytic activity in time was proved. The applied hyperbranched stabilizers provide a good tool for manufacturing gold-based nanozymes, which are highly stable and active over a wide pH range.

Abstract:Chitosan stabilized iron oxide magnetic nanoparticles (Chit-MNPs) were prepared with the theoretical Chit/Fe3O4 weight ratios equal to 0.5, 1, 2, 3 w/w and characterized by different techniques. The effect of various Chit/Fe3O4 weight ratios on the samples’ hydrodynamic diameter, zeta potential, and isoelectric point was studied by the dynamic light-scattering method. Using TEM analysis, the average magnetic core size was found to be ranging from 6 nm to 9 nm. The obtained results are comparable to those from magnetic measurements. The magnetic measurements confirmed successful chitosan coating on MNPs by means of the decrease in saturation magnetizations from 69.2 emu·g−1 for unmodified MNPs to 66.6, 65.9, 65.5 and 65.2 emu·g−1 for Chit-MNPs at different ratios of Chit/MNPs 0.5, 1, 2, and 3. TGA was used to quantify the amount of chitosan adsorbed on MNPs surface – the max. amount was 0.045 mg chitosan/mg MNPs. The peroxidase-like catalytic activity of unmodified MNPs and chitosan Chit-MNPs in present of a substrate such as N,N-diethyl-p-phenylenediamine sulfate salt (DPD) and hydrogen peroxide was studied. The results confirmed the peroxidase-like activity of samples, – they catalytically oxidise substrate DPD by H2O2 to produce a typical colour reaction. The chitosan coating was found to slightly decrease catalytic activity compared to MNPs at pH 7.4. Despite the fact, these findings can be useful for creating sets of MNPs and Chit-MNPs samples possessing predefined peroxidase-like activity and could be applied for chromogenic reactions in the future.
