Abstract:Development is remarkably reproducible, producing organs with the same size, shape, and function repeatedly from individual to individual. For example, every flower on the Antirrhinum stalk has the same snapping dragon mouth. This reproducibility has allowed taxonomists to classify plants and animals according to their morphology. Yet these reproducible organs are composed of highly variable cells. For example, neighboring cells grow at different rates in Arabidopsis leaves, sepals, and shoot apical meristems. This cellular variability occurs in normal, wild-type organisms, indicating that cellular heterogeneity (or diversity in a characteristic such as growth rate) is either actively maintained or, at a minimum, not entirely suppressed. In fact, cellular heterogeneity can contribute to producing invariant organs. Here, we focus on how plant organs are reproducibly created during development from these highly variable cells.

Abstract:The shape and function of plant cells are often highly interdependent. The puzzle-shaped cells that appear in the epidermis of many plants are a striking example of a complex cell shape, however their functional benefit has remained elusive. We propose that these intricate forms provide an effective strategy to reduce mechanical stress in the cell wall of the epidermis. When tissue-level growth is isotropic, we hypothesize that lobes emerge at the cellular level to prevent formation of large isodiametric cells that would bulge under the stress produced by turgor pressure. Data from various plant organs and species support the relationship between lobes and growth isotropy, which we test with mutants where growth direction is perturbed. Using simulation models we show that a mechanism actively regulating cellular stress plausibly reproduces the development of epidermal cell shape. Together, our results suggest that mechanical stress is a key driver of cell-shape morphogenesis.

Abstract:Organ sizes and shapes are strikingly reproducible, despite the variable growth and division of individual cells within them. To reveal which mechanisms enable this precision, we designed a screen for disrupted sepal size and shape uniformity in Arabidopsis and identified mutations in the mitochondrial i-AAA protease FtsH4. Counterintuitively, through live imaging we observed that variability of neighboring cell growth was reduced in ftsh4 sepals. We found that regular organ shape results from spatiotemporal averaging of the cellular variability in wild-type sepals, which is disrupted in the less-variable cells of ftsh4 mutants. We also found that abnormal, increased accumulation of reactive oxygen species (ROS) in ftsh4 mutants disrupts organ size consistency. In wild-type sepals, ROS accumulate in maturing cells and limit organ growth, suggesting that ROS are endogenous signals promoting termination of growth. Our results demonstrate that spatiotemporal averaging of cellular variability is required for precision in organ size.

Abstract:How gene activities and biomechanics together direct organ shapes is poorly understood. Plant leaf and floral organs develop from highly similar initial structures and share similar gene expression patterns, yet they gain drastically different shapes later-flat and bilateral leaf primordia and radially symmetric floral primordia, respectively. We analyzed cellular growth patterns and gene expression in young leaves and flowers of Arabidopsis thaliana and found significant differences in cell growth rates, which correlate with convergence sites of phytohormone auxin that require polar auxin transport. In leaf primordia, the PRESSED-FLOWER-expressing middle domain grows faster than adjacent adaxial domain and coincides with auxin convergence. In contrast, in floral primordia, the LEAFY-expressing domain shows accelerated growth rates and pronounced auxin convergence. This distinct cell growth dynamics between leaf and flower requires changes in levels of cell-wall pectin de-methyl-esterification and mechanical properties of the cell wall. Data-driven computer model simulations at organ and cellular levels demonstrate that growth differences are central to obtaining distinct organ shape, corroborating in planta observations. Together, our study provides a mechanistic basis for the establishment of early aerial organ symmetries through local modulation of differential growth patterns with auxin and biomechanics.

Abstract:Significance Plant leaves and flower petals in nature exhibit a wide variety of complex 3D shapes. Formation of these shapes has largely been studied from genetic and biomolecular viewpoints, overlooking contributions from biophysical factors such as mechanical stress and deformation. By means of computational simulations and quantitative analyses of the growth strains in live plant organs, we develop fundamental mechanistic insights into how nature invokes mechanics in the evolution of four commonly found shapes in plant organs by differential growth. We also demonstrate how these common shapes can be synthetically reproduced in hydrogel using this mechanistic understanding. Our study provides a broad scientific framework for rationalizing plant organ morphogenesis, but also provides pathways for generating bioinspired 3D architectures in soft materials.

Abstract:The development of an organism from the earliest stages of fertilization to the production of the next generation of gametes is one of nature's most miraculous and mysterious processes. During this process, the entire blueprint for the organism encoded in the genome is converted into a 3-dimensional body plan complete with a myriad of different cell types, each with their own highly specialized functions. All of this requires exquisitely controlled communication between cells that evolves over time and space as the organism is elaborated. In plants, this situation is more complex as cell migration does not occur, and the developmental programs are constantly adjusting to environmental cues that help sculpt the final plant form or coordinate with favorable seasonal conditions. Across evolutionary time, we see common themes in these programs, yet species-specific differences contribute to the startling diversity of plant forms in nature. Understanding how all this is achieved is the holy grail for plant science and will likely inform our ability to further shape domesticated crop traits to improve their resilience to the continuously changing environment. Central to this is the need to define the pathways that shape plant stem cell regulation and cell differentiation, which drive all aspects of plant development. The series of articles in this issue highlights only a small fraction of the fascinating research being done by plant developmental biologists in the area of stem cell regulation and cell differentiation. Several key themes emerge. First, Arabidopsis, which has been and still is a genetic workhorse for cracking plant biology for decades, continues to provide foundational insights into plant developmental mechanisms. This is largely due to the wealth of data in this species, the creativity of labs working on it, and its sustained and expanding advantages for dissecting core plant processes. Arabidopsis continues to prove informative for studies in other species, including crop species. As such, strong and continued funding for basic Arabidopsis research will be highly impactful and should be a critical priority in plant sciences. The second emerging theme is that technology is changing what we can do in plant development, what questions we can ask, and even which species we can work on. Technological advances have enabled the ability to start to untangle the extensive crosstalk that occurs across the signaling modules that shape development. The explosion of CRISPR-based technologies and the advent of more sophisticated omics technologies have allowed researchers to probe the genetic programs behind development at scale and on highly defined temporal and spatial scales. It has also allowed us to dissect developmental mechanisms across species, complementing the extensive traditional genetic resources in crop species and allowing us to create new plant models. The curiosity-driven research in newly emerging model plants, basal plant species, and crop models is contributing to our understanding of plant development and providing directions for improving crop performance or even engineering new products with commercial values. The third theme is that the plant's developmental programs are not their own. These developmental programs are often at the mercy of environmental stresses and damage, and plants have elaborated extensive mechanisms to deal with this, and even repair and reconstruct their body plans if needed. Additionally, other species can hijack plant developmental programs and induce new developmental outcomes. This is sometimes in collaboration with the plant and at other times to the plants' detriment. Below we provide an overview of the review topics in this issue.

Abstract:Differential growth is central to eukaryotic morphogenesis. We showed using cellular imaging, simulations, and perturbations that light-induced differential growth in a curved organ, the Arabidopsis thaliana apical hook, emerges from the longitudinal expansion of subepidermal cells, acting in parallel with a differential in the material properties of epidermal cell walls that resist expansion. The greater expansion of inner hook cells that results in apical hook opening is gated by wall alkalinity and auxin, both of which are depleted upon illumination. We further identified mechanochemical feedback from wall mechanics to light stimulated auxin depletion, which may contribute to gating hook opening under mechanical restraint. These results highlight how plant cells coordinate growth among tissue layers by linking mechanics and hormonal gradients with the cell wall remodeling required for differential growth.

Abstract:As the development of research tools in molecular biology,plant developmental biology has been changed its focus from the descriptive analysis of plant morphology to the cellular and gene regulation level.Plants have distinct postembryonic development patterns compare to the animal,which give the plant a flexible developmental plasticity in response to different growth environments.In the long-term of evolution,plants are adapted to the environment changes through the continuous adjustment of their development strategy,which makes the plant world highly diverse.The growth and development of multicellular organisms depend on the maintenance and constant differentiation of stem cells.In plants,most of the organs originate from stem cells where resided in the shoot apical meristem,root apical meristem and cambium.The functional conservation of stem cells in those diverse plants is the basis for ontogenesis.However,from the perspective of evolutionary development,the diversity of key regulatory genes expression in different stem cell populations fulfills the continuous adjustments of development strategy to adapt to environmental changes.The plasticity in stem cell regulations determines the flexibility of plant development,which is conserved in plant kingdom from moss to angiosperms.Of course,the mechanism of stem cell maintenance and differentiation are even more complicated in angiosperms.In model plant of Arabidopsis thanian,the molecular mechanism of stem cell regulations has been extensively studied over the past decades.Multiple signaling molecules and transcription factors are found to tightly control the stem cell fate in Arabidopsis thaliana.The homeodomain transcription factor WUSCHEL (WUS) where expressed in the organizing center (OC) is a key regulator for plant stem cell fate determination.While,stem cells expressed secreted peptide CLV3,negatively regulates the expression of WUS.They form a negative feedback loop that tightly control plant stem cell fate.In addition,the classic phytohormones cytokinin and auxin also play essential roles in the maintenance of stem cells in shoot apical meristem,root apical meristem and cambium,and exhibit complex functional interactions.The molecular organization of the RAM is quite similar to that of the shoot.WOX5 (WUSCHEL-RELATED HOMEOBOX 5),a homologue of WUS,is expressed in the QC,and induces root stem cell fate in the surrounding cells.The GRAS family transcription factor SHORT-ROOT (SHR) is expressed in the innermost tissue of the root,and SHR moves to the surrounding cell layer activating SCARECROW (SCR),together with PLT1 and PLT2,defines the stem cells fate.In this review,we focused on the functions of plant stem cells in plant postembryonic development,and covered recent findings on plant hormonal regulation in stem cells.We also discussed the integration of endogenous genetic information and external environmental factors in plant development,and how they affected the development of organs,morphology and yield of crops.

Abstract:Background and Aims In the roots of most vascular plants, the growth zone is small, the meristem and the elongation zone are sharply separated, and only meristematic cells divide. This statement is based almost entirely on studies with soil-rooted plants. Whether aerial roots of structurally dependent (=epiphytic/hemiepiphytic) species differ is virtually unexplored. Methods Growth of aerial roots in 20 structurally dependent plant species from eight families was studied ex situ . In 12 species, we studied the anatomical structure and distribution of cortex cell lengths and rhizoderm in the growth zone. Key Results All the studied aerial roots had an open apical meristem, and mitoses were not restricted to the meristem. In contrast to belowground roots, relative growth rate did not strongly increase upon transition to the elongation zone, while elongating growth was often prolonged. Still, the relative growth rate was lower than in belowground roots in soil, and in different species, it did not change considerably compared to each other. Conclusions A distinct elongation zone with rapid cell growth was missing in the studied aerial roots. Rather, there was a growth zone in which division, growth, and differentiation co-occurred. We observed a generally low relative growth rate in aerial roots and a surprisingly similar initial growth pattern in spite of the diversity in taxonomy and ecology, which resembled initial cellular growth in leaves, stems, and fleshy dicotyledonous fruit.

One role is to separate different cell groups, for example the differentiating cells of a leaf primordium from the pluripotent cells of the apical meristem. Boundary zones are also established during compound leaf development, to separate young leaflets from each other, and in many other positions of the plant body. Recent studies have demonstrated that different boundary zones share similar properties. They are characterized by a low rate of cell divisions and specific patterns of gene expression. In addition, the levels of the plant hormones auxin and brassinosteroids are down-regulated in boundary zones, resulting in a low differentiation level of boundary cells. This feature seems to be crucial for a second important role of boundary zones, the formation of new meristems. The primary shoot meristem, as well as secondary and ectopic shoot meristems, initiate from boundary cells that exhibit competence for meristem formation.

Abstract:Plant organ morphogenesis spans several orders of magnitude in time and space. Because of limitations in live-imaging, analysing whole organ growth from initiation to mature stages typically rely on static data sampled from different timepoints and individuals. We introduce a new model-based strategy for dating organs and for reconstructing morphogenetic trajectories over unlimited time windows based on static data. Using this approach, we show that Arabidopsis thaliana leaves are initiated at regular 1-day intervals. Despite contrasted adult morphologies, leaves of different ranks exhibited shared growth dynamics, with linear gradations of growth parameters according to leaf rank. At the sub-organ scale, successive serrations from same or different leaves also followed shared growth dynamics, suggesting that global and local leaf growth patterns are decoupled. Analysing mutants leaves with altered morphology highlighted the decorrelation between adult shapes and morphogenetic trajectories, thus stressing the benefits of our approach in identifying determinants and critical timepoints during organ morphogenesis.

Abstract:Abstract Morphogenesis requires the coordination of cellular behaviors along developmental axes. In plants, gradients of growth and differentiation are typically established along a single longitudinal primordium axis to control global organ shape. Yet, it remains unclear how these gradients are locally adjusted to regulate the formation of complex organs that consist of diverse tissue types. Here we combine quantitative live imaging at cellular resolution with genetics, and chemical treatments to understand the formation of Arabidopsis thaliana female reproductive organ (gynoecium). We show that, contrary to other aerial organs, gynoecium shape is determined by two orthogonal, time-shifted differentiation gradients. An early mediolateral gradient controls valve morphogenesis while a late, longitudinal gradient regulates style differentiation. Local, tissue-dependent action of these gradients serves to fine-tune the common developmental program governing organ morphogenesis to ensure the specialized function of the gynoecium.

Abstract:Meristems are three-dimensional generative structures that maintain a population of stem cells whilst producing new organs and tissues. Meristems develop in all land plants, however we know relatively little about the spatial and temporal regulation of meristem structure in lineages such as the bryophytes. Here we describe the three-dimensional anatomy of the meristem during the development of the liverwort, Using optical reconstructions of the frontal, sagittal and transverse planes through the mature meristem, we show that the apical stem cell is sub-apical, ventral, and located in the outer cell layer. The anatomy of the mature meristem is therefore asymmetrical in the dorsoventral axis, which is reflected by the domain specific protein localisation of Marchantia Class III and Class IV Homeodomain-Leucine-Zippers (MpC3HDZ and MpC4HDZ) and promoter activity of Mp . The dorsoventral asymmetry that defines the mature meristem is absent in the juvenile meristems of the asexual propagules known as gemmae. We discovered that anatomical dorsoventral asymmetry of the meristem forms after two days of gemmaling growth and is accompanied by expression of the dorsal identity reporter, MpC3HDZ. We conclude that the gemma meristem is in a state of arrested development and undergoes anatomical rearrangement to develop the three-dimensional meristem structure of the mature plant.

Epidermal Restriction Confers Robustness to Organ Shapes.

Abstract:The shape of comparable tissues and organs is consistent among individuals of a given species, but how this consistency or robustness is achieved remains an open question. The interaction between morphogenetic factors determines organ formation and subsequent shaping, which is ultimately a mechanical process. Using a computational approach, we show that the epidermal layer is essential for the robustness of organ geometry control. Specifically, proper epidermal restriction allows organ asymmetry maintenance, and the tensile epidermal layer is sufficient to suppress local variability in growth, leading to shape robustness. The model explains the enhanced organ shape variations in epidermal mutant plants. In addition, differences in the patterns of epidermal restriction may underlie the initial establishment of organ asymmetry. Our results show that epidermal restriction can answer the longstanding question of how cellular growth noise is averaged to produce precise organ shapes, and the findings also shed light on organ asymmetry establishment.

Abstract:In animals and plants, organ shape is primarily determined during primordium development by carefully coordinated growth and cell division . Rare examples of post-primordial change in morphology (reshaping) exist that offer tractable systems to study mechanisms required for organ-shape determination and diversification. One such example is the heart-shape formation of fruits that occurs by reshaping the ovate spheroid gynoecium upon fertilization . Here we use whole-organ live-imaging to show that dynamic changes in growth and cell division coupled with local maintenance of meristematic identity drives fruit-shape formation. At the molecular level, we reveal an auxin-induced mechanism ultimately descending on a single regulatory element to mediate morphological alteration. This element resides in the promoter of the ( ) gene. The CrSTM meristem identity factor positively regulates its own expression through binding to this element thereby providing a feed-forward loop at the position and time when protrusions emerge to form the heart. Independent evolution of the STM-binding element in promoters across Brassicaceae species correlates with those undergoing a gynoecium-to-fruit metamorphosis. Accordingly, genetic and phenotypic studies showed that the STM-binding element is required to facilitate the shape transition and reveals a conserved molecular mechanism for organ morphogenesis.

Abstract:Abstract One of the most transformative events in the history of life on earth was the transition of plants from water to land approximately 470 million years ago. Within the Charophyte green algae, the closest living relatives of land plants, body plans have evolved from those that comprise simple unicells to those that are morphologically complex, large and multicellular. The Charophytes developed these broad ranging body plans by exploiting a range of one-dimensional and two-dimensional growth strategies to produce filaments, mats and branches. When plants were confronted with harsh conditions on land, they were required to make significant changes to the way they shaped their body plans. One of the fundamental developmental transitions that occurred was the evolution of three-dimensional growth and the acquisition of apical cells with three or more cutting faces. Plants subsequently developed a range of morphological adaptations (e.g. vasculature, roots, flowers, seeds) that enabled them to colonise progressively drier environments. 3D apical growth also evolved convergently in the brown algae, completely independently of the green lineage. This review summarises the evolving developmental complexities observed in the early divergent Charophytes all the way through to the earliest conquerors of land, and investigates 3D apical growth in the brown algae.

Abstract:Abstract Growth extent and direction determine cell and whole-organ architecture. How they are spatio-temporally modulated to control size and shape is not well known. Here we tackled this question by studying the effect of brassinosteroid (BR) signalling on the structure of the root meristem. Quantification of the three-dimensional geometry of thousands of individual meristematic cells across different tissue types showed that the modulation of BR signalling yields distinct changes in growth rate and anisotropy, which affects the time that cells spend in the meristem and has a strong impact on the final root form. By contrast, the hormone effect on cell volume was minor, establishing cell volume as invariant to the effect of BR. Thus, BR has the highest effect on cell shape and growth anisotropy, regulating the overall longitudinal and radial growth of the meristem, while maintaining a coherent distribution of cell sizes. Moving from single-cell quantification to the whole organ, we developed a computational model of radial growth. The simulation demonstrates how differential BR-regulated growth between the inner and outer tissues shapes the meristem and thus explains the non-intuitive outcomes of tissue-specific perturbation of BR signalling. The combined experimental data and simulation suggest that the inner and outer tissues have distinct but coordinated roles in growth regulation.
