Abstract:How gene activities and biomechanics together direct organ shapes is poorly understood. Plant leaf and floral organs develop from highly similar initial structures and share similar gene expression patterns, yet they gain drastically different shapes later-flat and bilateral leaf primordia and radially symmetric floral primordia, respectively. We analyzed cellular growth patterns and gene expression in young leaves and flowers of Arabidopsis thaliana and found significant differences in cell growth rates, which correlate with convergence sites of phytohormone auxin that require polar auxin transport. In leaf primordia, the PRESSED-FLOWER-expressing middle domain grows faster than adjacent adaxial domain and coincides with auxin convergence. In contrast, in floral primordia, the LEAFY-expressing domain shows accelerated growth rates and pronounced auxin convergence. This distinct cell growth dynamics between leaf and flower requires changes in levels of cell-wall pectin de-methyl-esterification and mechanical properties of the cell wall. Data-driven computer model simulations at organ and cellular levels demonstrate that growth differences are central to obtaining distinct organ shape, corroborating in planta observations. Together, our study provides a mechanistic basis for the establishment of early aerial organ symmetries through local modulation of differential growth patterns with auxin and biomechanics.

Abstract:Although genetic control of morphogenesis is well established, elaboration of complex shapes requires changes in the mechanical properties of cells. In plants, the first visible sign of leaf formation is a bulge on the flank of the shoot apical meristem. Bulging results from local relaxation of cell walls, which causes them to yield to internal hydrostatic pressure. By manipulation of tissue tension in combination with quantitative live imaging and finite-element modeling, we found that the slow-growing area at the shoot tip is substantially strain-stiffened compared with surrounding fast-growing tissue. We propose that strain stiffening limits growth, restricts organ bulging, and contributes to the meristem's functional zonation. Thus, mechanical signals are not just passive readouts of gene action but feed back on morphogenesis.

Abstract:Significance Plant leaves and flower petals in nature exhibit a wide variety of complex 3D shapes. Formation of these shapes has largely been studied from genetic and biomolecular viewpoints, overlooking contributions from biophysical factors such as mechanical stress and deformation. By means of computational simulations and quantitative analyses of the growth strains in live plant organs, we develop fundamental mechanistic insights into how nature invokes mechanics in the evolution of four commonly found shapes in plant organs by differential growth. We also demonstrate how these common shapes can be synthetically reproduced in hydrogel using this mechanistic understanding. Our study provides a broad scientific framework for rationalizing plant organ morphogenesis, but also provides pathways for generating bioinspired 3D architectures in soft materials.

Abstract:Plant organ outgrowth superficially appears like the continuous mechanical deformation of a sheet of cells. Yet, how precisely cells as individual mechanical entities can act to morph a tissue reliably and efficiently into three dimensions during outgrowth is still puzzling especially when cells are tightly connected as in plant tissue. In plants, the mechanics of cells within a tissue is particularly well defined as individual cell growth is essentially the mechanical yielding of cell-wall in response to internal turgor pressure. Cell wall stiffness is controlled by biological signalling and, hence, cell growth is observed to respond to mechanical stresses building up within a tissue. What is the role of the mechanical feedback during morphing of tissue in three dimensions? Here, we develop a three dimensional vertex model to investigate tissue mechanics at the onset of organ outgrowth at the tip of a plant shoot. We find that organ height is primarily governed by the ratio of growth rates of faster growing cells initiating the organ to slower growing cells surrounding them. Remarkably, the outgrowth rate is higher when cells growth responds to the tissue-wide mechanical stresses. Our quantitative analysis of simulation data shows that tissue mechanical feedback on cell growth can act via twofold mechanism. First, the feedback guides patterns of cellular growth. Second, the feedback modifies the stress patterns on the cells, consequently amplifying and propagating growth anisotropies. This mechanism may allow plants to grow organs efficiently out of the meristem by reorganizing the cellular growth rather than inflating growth rates.

Abstract:ABSTRACT Rta, the gene product of Kaposi's sarcoma-associated herpesvirus (KSHV) encoded mainly in open reading frame 50 (ORF50), is capable of activating expression of viral lytic cycle genes. What was not demonstrated in previous studies was whether KSHV Rta was competent to initiate the entire viral lytic life cycle including lytic viral DNA replication, late-gene expression with appropriate kinetics, and virus release. In HH-B2, a newly established primary effusion lymphoma (PEL) cell line, KSHV ORF50 behaved as an immediate-early gene and autostimulated its own expression. Expression of late genes, ORF65, and K8.1 induced by KSHV Rta was eliminated by phosphonoacetic acid, an inhibitor of viral DNA polymerase. Transfection of KSHV Rta increased the production of encapsidated DNase-resistant viral DNA from HH-B2 cells. Thus, introduction of an ORF50 expression plasmid is sufficient to drive the lytic cycle to completion in cultured PEL cells.

Abstract:The development of a hook-like structure at the apical part of the soil-emerging organs has fascinated botanists for centuries, but how it is initiated remains unclear. Here, we demonstrate with high-throughput infrared imaging and 2-D clinostat treatment that, when gravity-induced root bending is absent, apical hook formation still takes place. In such scenarios, hook formation begins with a de novo growth asymmetry at the apical part of a straightly elongating hypocotyl. Remarkably, such de novo asymmetric growth, but not the following hook enlargement, precedes the establishment of a detectable auxin response asymmetry, and is largely independent of auxin biosynthesis, transport and signaling. Moreover, we found that functional cortical microtubule array is essential for the following enlargement of hook curvature. When microtubule array was disrupted by oryzalin, the polar localization of PIN proteins and the formation of an auxin maximum became impaired at the to-be-hook region. Taken together, we propose a more comprehensive model for apical hook initiation, in which the microtubule-dependent polar localization of PINs may mediate the instruction of growth asymmetry that is either stochastically taking place, induced by gravitropic response, or both, to generate a significant auxin gradient that drives the full development of the apical hook.

Abstract:The growth of plant organs is a complex process powered by osmosis that attracts water inside the cells; this influx induces simultaneously an elastic extension of the walls and pressure in the cells, called turgor pressure; above a threshold, the walls yield and the cells grow. Based on Lockhart’s seminal work, various models of plant morphogenesis have been proposed, either for single cells, or focusing on the wall mechanical properties. However, the synergistic coupling of fluxes and wall mechanics has not yet been fully addressed in a multicellular model. This work lays the foundations of such a model, by simplifying as much as possible each process and putting emphasis on the coupling itself. Its emergent properties are rich and can help to understand plant morphogenesis. In particular, we show that the model can display a new type of lateral inhibitory mechanism that could contribute to the amplification of growth heterogeneities, essential for shape differentiation. Significance Statement Plant cells are surrounded by a rigid wall that prevents cell displacements and rearrangements as in animal tissues. Therefore, plant morphogenesis relies only on cell divisions, shape changes, and local modulation of growth rate. It has long been recognized that cell growth relies on the competition between osmosis that tends to attract water into the cells and wall mechanics that resists to it, but this interplay has never been fully explored in a multicellular model. The goal of this work is to analyze the theoretical consequences of this coupling. We show that the emergent behavior is rich and complex: among other findings, pressure and growth rate heterogeneities are predicted without any ad-hoc assumption; furthermore the model can display a new type of lateral inhibition based on fluxes that could complement and strengthen the efficiency of already known mechanisms.

Abstract:Significance Growth hormones are mobile chemicals that exert considerable influence over how multicellular organisms like animals and plants take on their shape and form. Of particular interest is the distribution of such hormones across cells and tissues. In plants, one of these hormones, gibberellin (GA), is known to regulate cell multiplication and cell expansion to increase the rate at which roots grow. In this work, biosensor measurements were combined with theoretical models to elucidate the biochemical mechanisms that direct GA distribution and how these patterns relate to root growth. Our detailed understanding of how GA distributions are controlled in roots should prove a valuable model for understanding the makings of the many other hormone distributions that influence how plants grow.

Fibrous Network Nature of Plant Cell Walls Enables Tunable Mechanics for Development

Abstract:During plant development, the mechanical properties of the cell walls must be tuned to regulate the growth of the cells. Cell growth involves significant stretching of the cell walls, yet little is known about the mechanical properties of cell walls under such substantial deformation, or how these mechanical properties change to regulate development. Here, we investigated the mechanical behavior of the Arabidopsis leaf epidermal cells being stretched. We found that the mechanical properties arise from the cell wall, which behaves as a fibrous network material. The epidermis exhibited a non-linear stiffening behavior that fell into three regimes. Each regime corresponded to distinct nonlinear behaviors in terms of transverse deformation (i.e., Poisson effect) and unrecoverable deformation (i.e., plasticity). Using a model, we demonstrated that the transition from reorientation and bending-dominated to stretch-dominated deformation modes of cellulose microfibrils cause these nonlinear behaviors. We found the stiffening behavior is more pronounced at later developmental stages. Finally, we show the spiral2-2 mutant has anisotropic mechanical properties, likely contributing to the spiraling of leaves. Our findings reveal the fibrous network nature of cell walls gives a high degree of tunability in mechanical properties, which allows cells to adjust these properties to support proper development.

Abstract:Abstract Morphogenesis requires the coordination of cellular behaviors along developmental axes. In plants, gradients of growth and differentiation are typically established along a single longitudinal primordium axis to control global organ shape. Yet, it remains unclear how these gradients are locally adjusted to regulate the formation of complex organs that consist of diverse tissue types. Here we combine quantitative live imaging at cellular resolution with genetics, and chemical treatments to understand the formation of Arabidopsis thaliana female reproductive organ (gynoecium). We show that, contrary to other aerial organs, gynoecium shape is determined by two orthogonal, time-shifted differentiation gradients. An early mediolateral gradient controls valve morphogenesis while a late, longitudinal gradient regulates style differentiation. Local, tissue-dependent action of these gradients serves to fine-tune the common developmental program governing organ morphogenesis to ensure the specialized function of the gynoecium.

Abstract:The morphogenesis of organs necessarily involves mechanical interactions and changes in mechanical properties of a tissue. A long standing question is how such changes are directed on a cellular scale while being coordinated at a tissular scale. Growing evidence suggests that mechanical cues are participating in the control of growth and morphogenesis during development. We introduce a mechanical model that represents the deposition of cellulose fibers in primary plant walls. In the model both the degree of material anisotropy and the anisotropy direction are regulated by stress anisotropy. We show that the finite element shell model and the simpler triangular biquadratic springs approach provide equally adequate descriptions of cell mechanics in tissue pressure simulations of the epidermis. In a growing organ, where circumferentially organized fibers act as a main controller of longitudinal growth, we show that the fiber direction can be correlated with both the maximal stress direction and the direction orthogonal to the maximal strain direction. However, when dynamic updates of the fiber direction are introduced, the mechanical stress provides a robust directional cue for the circumferential organization of the fibers, whereas the orthogonal to maximal strain model leads to an unstable situation where the fibers reorient longitudinally. Our investigation of the more complex shape and growth patterns in the shoot apical meristem where new organs are initiated shows that a stress based feedback on fiber directions is capable of reproducing the main features of in vivo cellulose fiber directions, deformations and material properties in different regions of the shoot. In particular, we show that this purely mechanical model can create radially distinct regions such that cells expand slowly and isotropically in the central zone while cells at the periphery expand more quickly and in the radial direction, which is a well established growth pattern in the meristem.

Abstract:Understanding the mechanism by which patterned gene activity leads to mechanical deformation of cells and tissues to create complex forms is a major challenge for developmental biology. Plants offer advantages for addressing this problem because their cells do not migrate or rearrange during morphogenesis, which simplifies analysis. We synthesize results from experimental analysis and computational modeling to show how mechanical interactions between cellulose fibers translate through wall, cell, and tissue levels to generate complex plant tissue shapes. Genes can modify mechanical properties and stresses at each level, though the values and pattern of stresses differ from one level to the next. The dynamic cellulose network provides elastic resistance to deformation while allowing growth through fiber sliding, which enables morphogenesis while maintaining mechanical strength.

Abstract:One of the central problems in animal and plant developmental biology is deciphering how chemical and mechanical signals interact within a tissue to produce organs of defined size, shape and function. Cell walls in plants impose a unique constraint on cell expansion since cells are under turgor pressure and do not move relative to one another. Cell wall extensibility and constantly changing distribution of stress on the wall are mechanical properties that vary between individual cells and contribute to rates of expansion and orientation of cell division. How exactly cell wall mechanical properties influence cell behavior is still largely unknown. To address this problem, a novel, subcellular element computational model of growth of stem cells within the multilayered shoot apical meristem (SAM) of Arabidopsis thaliana is developed and calibrated using experimental data. Novel features of the model include separate, detailed descriptions of cell wall extensibility and mechanical stiffness, deformation of the middle lamella and increase in cytoplasmic pressure generating internal turgor pressure. The model is used to test novel hypothesized mechanisms of formation of the shape and structure of the growing, multilayered SAM based on WUS concentration of individual cells controlling cell growth rates and layer dependent anisotropic mechanical properties of subcellular components of individual cells determining anisotropic cell expansion directions. Model simulations also provide a detailed prediction of distribution of stresses in the growing tissue which can be tested in future experiments.
Dynamic coordination of cytoskeletal and cell wall systems during plant cell morphogenesis

Abstract:Underlying the architectural complexity of plants are diverse cell types that, under the microscope, easily reveal relationships between cell structure and specialized functions. Much less obvious are the mechanisms by which the cellular growth machinery and mechanical properties of the cell interact to dictate cell shape. The recent combined use of mutants, genomic analyses, sophisticated spectroscopies, and live cell imaging is providing new insight into how cytoskeletal systems and cell wall biosynthetic activities are integrated during morphogenesis. The purpose of this review is to discuss the unique geometric properties and physical processes that regulate plant cell expansion, then to overlay on this mechanical system some of the recent discoveries about the protein machines and cellular polymers that regulate cell shape. In the end, we hope to make clear that there are many interesting opportunities to develop testable mathematical models that improve our understanding of how subcellular structures, protein motors, and extracellular polymers can exert effects at spatial scales that span cells, tissues, and organs.
Assessing the hydromechanical control of plant growth

Abstract:Multicellular organisms grow and acquire their shapes through the differential expansion and deformation of their cells. Recent research has addressed the role of cell and tissue mechanical properties in these processes. In plants, it is believed that growth rate is a function of the mechanical stress exerted on the cell wall, the thin polymeric layer surrounding cells, involving an effective viscosity. Nevertheless, recent studies have questioned this view, suggesting that cell wall elasticity sets growth rate or that uptake of water is limiting for plant growth. To assess these issues, we developed a microfluidic device to quantify growth rates, elastic properties, and hydraulic conductivity of individual plants in a controlled environment with a high throughput. We characterized the effect of osmotic treatment and of abscisic acid on growth and hydromechanical properties. Overall, the instantaneous growth rate of individuals is correlated to both bulk elastic modulus and hydraulic conductivity. Our results are consistent with a framework in which growth rate is determined primarily by elasticity of the wall and its remodelling, and secondarily by hydraulic conductivity. Accordingly, the coupling between chemistry of the cell wall and hydromechanics of the cell appears as key to set growth patterns during morphogenesis.

Abstract:Petals and leaves are usually curled and exhibit intriguing morphology evolution upon growth, which contributes to their important biological functions. To understand the underlying morphoelastic mechanism and to determine the crucial factors that govern the growth-induced instability patterning in curved petals and leaves, we develop an active thin shell model that can describe variable curvatures and spontaneous growth, within the framework of general differential geometry based on curvilinear coordinates and hyperelastic deformation theory. Analytical solutions of distinguished growing shapes such as saddle surface and cylindrical mode are then derived. We reveal distinct morphological evolutions of doubly curved leaves/petals with different curvatures κx (along the main vein) and κy (perpendicular to the main vein) upon differential growth. Compared to the flat (zero curvature) configuration, leaves/petals with longitudinal curvature κx experiences a global bending deformation. With the increase of growth strain, the leaf/petal undergoes a coupling behavior of edge wrinkling and global bending deformation, associated with a pitchfork bifurcation. Conversely, the transverse curvature κy does not lead to significant bending behavior, but results in delayed critical buckling threshold and reduced wrinkling amplitude. Physical insights into curvature effects on morphology evolutions are further provided by the analysis of nonlinear competition between bending and membrane energies. Moreover, we explore the effect of vein constraint on pattern formation, showing that different from edge wrinkling observed in leaves with strong vein constraint, the ones with weak vein constraint are prone to grow into a saddle shape, which is consistent with analytical solutions. The results uncover the intricate interplay between configurational curvature and vein confinement on plant morphogenesis, providing fundamental insights into a variety of growing shapes of curled petals and leaves.

Abstract:The principles underlying plant development are extended to allow a more molecular mechanism to elaborate the schema by which ground cells differentiate into vascular cells. Biophysical considerations dictate that linear dynamics are not sufficent to capture facilitated auxin transport (e.g., through PIN). We group these transport facilitators into a non-linear model under the assumption that they attempt to minimize certain {\em differences} of auxin concentration. This Constant Gradient Hypothesis greatly increases the descriptive power of our model to include complex dynamical behaviour. Specifically, we show how the early pattern of PIN1 expression appears in the embryo, how the leaf primordium emerges, how convergence points arise on the leaf margin, how the first loop is formed, and how the intricate pattern of PIN shifts during the early establishment of vein patterns in incipient leaves of Arabidopsis. Given our results, we submit that the model provides evidence that many of the salient structural characteristics that have been described at various stages of plant development can arise from the uniform application of a small number of abstract principles.
