Abstract:Detecting nitroreductase activity with fluorescent probes has advantages over conventional monitoring approaches owing to their high sensitivity, selectivity and real‐time monitoring ability. This article reviews the activity and status of nitroreductases in bacteria, plants and animal cells as well as the significance of fluorescence‐imaging‐related studies by employing recently designed probes. Nitroreductases are the enzymes that can convert nitroaromatic substances including explosives like TNT to their corresponding nitrite, hydroxylamine or amino derivatives with reduced NADH or NADPH as an electron donor. A wide range of nitroreductases are known to present in bacterial genomes that are explored for their potential in the reduction of nitroaromatic compounds and they are used in converting prodrugs to toxic hydroxylamine derivatives causing tumor cell killing. Likewise, in cancer cells, the low oxygen condition called hypoxia is a characteristic of solid tumors and during hypoxia nitroreductase gene expression occurs. Moreover, due to the ability of nitroreductases to reduce nitroaromatic compounds, phytoremediation potential of plants was inspected by expressing bacterial nitroreductase genes in plants through genetic engineering. Due to its diversity and widespread applications in different biosystems, its detection has gained momentum among the scientific community. Designing fluorescent probes for detecting nitroreductases has emerged as a beneficial tool compared to conventional monitoring approaches due to their high sensitivity, selectivity, and real‐time monitoring ability. This review aims to provide an understanding of the activity and the status of nitroreductases in bacteria, plants and animal cells as well as the significance of fluorescent imaging‐related studies by employing recently designed probes.

Abstract:Nitric oxide synthase like enzyme (NOS-like enzyme), which produces nitric oxide, participates in many biological processes. However it remains unidentified and highly controversial that plants do possess a NOS-like enzyme. In this paper, a novel arginine analogue NP1 was designed and developed for the direct identification and real time tracking of NOS-like enzymes in plant by fluorescence sensing. It could bind NOS-like enzyme efficiently and enter the cell successfully. In vivo fluorescence response results directly proved that NOS-like enzymes did exist in tobacco leaf and would be stimulated by pathogen infection, which also provided a useful chemical tool for the study of the function of NOS-like enzyme in plants.

Abstract:A fluorescent nanoprobe is reported for rapid detection of nitrites (NO2-) in plant cells. The probe is fabricated by linking neutral reds (NR) to the surface of upconversion fluorescent core/shell nanocrystalline with the bridging of polyethylene glycol (PEG) molecules. The fluorescence of upconversion nanoparticles (UCNPs) is stored by NR through fluorescence resonance energy transfer (FRET) under 980 nm excitation that can be released by further linking to NO2-. It is observed that the intensity rate of green to red emission of NR-modified UCNPs changes linearly with increasing the amount of NO2-. So that concentration of NO2- can be accordingly addressed. Worth mentioning is that, comparing with bare core upconversion nanoparticles (NPs), core/shell UCNPs can greatly reduce the surface quenching of the fluorescence induced by solvents instead of NR and thus leading to the enhancement of signal-to-noise ratios. Moreover, excitation of core/shell UCNPs requires only a much lower power (0.06 W cm(-2)) than bare cores which is beneficial to reducing the decomposition of NR to stabilize the FRET processes. Under the optimum conditions, the detection limit of nitrite in plant cells was 0.1 mu g mL(-1).

Abstract:Rhodamine dyes bearing aromatic nitro group has been synthesized for nitroreductase enzyme chemosensing applications. The probe is showing very selective turn-on fluorescent response towards nitroreductase enzymes and in hypoxic conditions. The sensor displays a remarkable fluorescent enhancement at 557 nm (λex = 500 nm) without the interference of other biologically relevant species under hypoxic conditions in a physiological medium. The nitro group in the sensor is reduced by the nitroreductase enzyme to the amino group, resulting in the hydrolysis of the probe and subsequent release of highly fluorescent rhodamine 6G dye is observed. This rhodamine based fluorescent probe has been utilized for the imaging of nitroreductase enzymes as well as hypoxia in live cells.

Abstract:Nitrate (NO 3 − ) uptake and distribution are critical to plant life. Although the upstream regulation of NO 3 − uptake and downstream responses to NO 3 − in a variety of cells have been well studied, it is still not possible to directly visualize the spatial and temporal distribution of NO 3 − with high resolution at the cellular level. Here, we report a nuclear-localized, genetically encoded fluorescent biosensor, which we named NitraMeter3.0, for the quantitative visualization of NO 3 − distribution in Arabidopsis thaliana . This biosensor tracked the spatiotemporal distribution of NO 3 − along the primary root axis and disruptions by genetic mutation of transport (low NO 3 − uptake) and assimilation (high NO 3 − accumulation). The developed biosensor effectively monitors NO 3 − concentrations at the cellular level in real time and spatiotemporal changes during the plant life cycle.

Abstract:Among pathogenic bacteria, Escherichia coli, Enterococcus faecium, Staphylococcus aureus, Klebsiella pneumoniae, Acinetobacter baumannii, and Pseudomonas aeruginosa were the six leading causes for the deaths associated with antibiotic resistance in 2019. Although new treatment options are urgently needed, the precise identification of the bacterial species remains pivotal for an accurate diagnosis and effective treatment. Clinically, mass spectrometry is used to distinguish these bacteria based on their protein mass pattern at the genus and species level. Herein, we report an alternative approach to identify these bacteria using the nitroreductase-based "turn-on" fluorescent probes (ETH1-NO2 and ETH2-NO2), with potential visual indicators for the six individual bacteria species. The limits of detection (LODs) of the probes for NTRs are 0.562 (ETH1-NO2) and 0.153 μg/mL (ETH2-NO2), respectively. They respond effectively to both Gram-positive and Gram-negative bacteria, with the lowest LOD at 1.2 × 106 CFU/mL for E. coli. In particular, different bacteria show noticeable difference in the apparent color of ETH1-NO2 samples, allowing possible identification of these bacteria visually. In addition, ETH1-NO2 also has potential applications in bacterial fluorescence imaging. Thus, our study provides an alternative approach for bacteria identification and new reagents for bacteria imaging.

Abstract:Nitroxyl (HNO), a reactive nitrogen species (RNS), is essential for plant growth. However, the action of HNO in plants has been difficult to understand due to the lack of highly sensitive and real-time in-situ monitoring tools. Herein, we presented a near-infrared fluorescent probe, DCI-HNO , based on dicyanoisophorone fluorophore, for real-time mapping HNO in plants. The introduction of a phosphine moiety as a specific HNO recognition unit can inhibit the intramolecular charge transfer (ICT) of probe DCI-HNO . However, in the presence of HNO, the ICT process occurred, leading to the emission at 665 nm. Probe DCI-HNO exhibited high sensitivity (97 nM), rapid response time (8 min), large Stokes shift (135 nm) for detection of HNO in plants. The novel developed probe has successfully imaged endogenous HNO produced during NO/H 2 S cross-talk in plant tissues. Additionally, the up-regulated in HNO levels during tobacco aging and in response to stress has been confirmed. Therefore, probe DCI-HNO has provided a reliable method for monitoring the NO/H 2 S cross-talk and revealing the role of HNO in plants.

Abstract:Nitroreductase (NTR) is a type of cytoplasmic enzyme that depends on flavin mononucleotide or flavin adenine dinucleotide, and that is widely present in tumor cells where it increases during hypoxia. NTR can effectively reduce aromatic nitro compounds to corresponding amino compounds, with the assistance of reduced nicotinamide adenine dinucleotide (NADH). In this study, a nitro-containing fluorescent turn-on probe, TP-NO, was synthesized and characterized. TP-NO exhibited a fast and selective response towards NTR (LOD = 1.96 ng/mL), and which reaction mechanism was proved through molecular docking. In addition, TP-NO has been successfully applied in imaging hypoxia levels in tumor cells, and zebrafish. This work provided a probe that can be applied to investigate NTR in biological systems.

Abstract:Redox changes of pyridine nucleotides in cellular compartments are highly dynamic and their equilibria are under the influence of various reducing and oxidizing reactions. To obtain spatiotemporal data on pyridine nucleotides in living plant cells, typical biochemical approaches require cell destruction. To date, genetically encoded fluorescent biosensors are considered to be the best option to bridge the existing technology gap, as they provide a fast, accurate, and real-time readout. However, the existing pyridine nucleotides genetically encoded fluorescent biosensors are either sensitive to pH change or slow in dissociation rate. Herein, we employed the biosensors which generate readouts that are pH stable for in planta measurement of NADH/NAD(+) ratio and NADPH level. We generated transgenic Arabidopsis lines that express these biosensors in plastid stroma and cytosol of whole plants and pollen tubes under the control of CaMV 35S and LAT52 promoters, respectively. These transgenic biosensor lines allow us to monitor real-time dynamic changes in NADH/NAD(+) ratio and NADPH level in the plastids and cytosol of various plant tissues, including pollen tubes, root hairs, and mesophyll cells, using a variety of fluorescent instruments. We anticipate that these valuable transgenic lines may allow improvements in plant redox biology studies.

Abstract:A novel near-infrared (NIR) fluorescent probe based on cyanine dyes was designed and synthesized to detect nitroreductase (NTR) activity. Upon reaction with NTR, the nitro-group of p-nitrobenzyl moiety was reduced to amino-group and therefore renews the π-electron conjugation of the dye, resulting in the intense NIR fluorescent emission (708 nm). This redox-induced NIR-fluorescence probe has been successfully applied to the detection of NTR activity under physiological conditions with high selectivity.

Abstract:Nitroreductase (NTR) has long been a target of interest for its important role involved in the nitro compounds metabolism. Various probes have been reported for NTR analysis, but rarely able to distinguish the extracellular NTR from intracellular ones. Herein we reported a new NTR sensor, HCyS‐NO2, which was a hemicyanine molecule with one nitro and two sulfo groups attached. The nitro group acted as the reporting group to respond NTR reduction. Direct linkage of nitro group into the hemicyanine π conjugate system facilitated the intramolecular electron transfer (IET) process and thus quenched the fluorescence of hemicyanine core. Upon reduction with NTR, the nitro group was rapidly converted into the hydroxylamino and then the amino group, eliminating IET process and thus restoring the fluorescence. The sulfo groups installed significantly increased the hydrophilicity of the molecule, and introduced negative charges at physiological pH, preventing the diffusion into bacteria. Both gram‐negative and gram‐positive bacteria were able to turn on the fluorescence of HCyS‐NO2, without detectable diffusion into cells, providing a useful tool to probe the extracellular reduction process.

Abstract:Cytochrome P450 55B1(CYP55B1) from Chlamydomonas reinhardtii reduces nitric oxide (NO) to dinitrogen oxide (N2O) with the electron supply from NAD(P)H in vivo. Here a novel nitric oxide biosensor was developed by immobilized CYP55B1 on the surface of pyrolytic graphite electrode (PGE) by cross-linking with glutaraldehyde (GA) and bovine serum albumin (BSA). The direct electrochemistry of CYP55B1 was realized with the redox peak potential of -0.355 V and -0.385 V and the catalytic reduction peak of NO by CYP55B1 is at -0.85 V at the scan rate of 0.5 V S-1 in pH 7.0 phosphate buffer. The apparent coverage (Γ = 1.43 × 10-11 mol cm-2), the electron transfer rate constant (ks = 17.39 s-1) and apparent affinity to NO (Kmapp = 11.64 nM) of CYP55B1 in GA/BSA film were obtained. The catalytic mechanism of CYP55B1 towards NO with NADH was examined by the biosensor. The linear range of NO detection was investigated by differential pulse voltammetry with the results of 5-50 nM and the detection limit of 0.5 nM (S/N = 3). The selectivity and stability of the electrochemical biosensor were investigated. Furthermore, the CYP55B1electrochemical biosensor was applied to monitor NO release from Arabidopsis protoplasts with the average content of 0.848 fmol per cell under anaerobic condition.

Abstract:As an endogenously generated signal-inducing agent in the immune, cardiovascular and nervous systems, nitric oxide (NO) is involved in various biological processes and a large number of pathologies. Hence, sensitive and reliable detection of NO in biological samples and inside living cells has been of great interest. Herein, we reported a carbon-dot-based fluorescent ratiometric nanosensor for NO detection in water and imaging in living cells. Ratiometric sensing is realized through the fluorescence resonance energy transfer (FRET) process, and the carbon dots (CDs) serve as both the energy donor and the anchoring site for the NO probe moiety, which turns into the energy acceptor in the presence of NO. This nanosensor is highly selective for NO, and is operable in totally aqueous media with a very low detection limit of 3 nM. Due to the biocompatible nature and small size of the carbon dots, the nanosensor exhibits very low cytotoxicity and can easily permeate through the cell membrane. The nanosensor is capable of tracking the exogenous NO levels in several cell lines such as Hela, L929 and Raw 264.7 cells, and it can be also used to visualize the endogenously produced NO in the Raw 264.7 macrophage cell line.
An efficient ratiometric fluorescence and colorimetric dual-mode probe for convenient determination of nitrite in real samples and E. coli

Abstract:Developing an effective and convenient nitrite detection method is indispensable in food safety, environmental monitoring, clinical diagnosis of diseases, and many other areas. Herein, a dicyanoisophorone derivative, TMN-NH2 with large Stokes shift and near-infrared (NIR) emission, was proposed as a ratiometric fluorescence and colorimetric dual-mode probe for the rapid determination of NO2- in acidic media, showing excellent selectivity and high sensitivity. The sensing mechanism is based on the diazotization of TMN-NH2 with NO2- and subsequent diazonium salt hydrolysis to form a hydroxyl-substituted product (TMN-OH). Under the optimized conditions of reaction and detection, a new quantitative analysis method based on TMN-NH2 was established for NO2- detection, exhibiting good linear relationships to NO2- in the range of 0.5 to 15 μM with practical detection limits of 26.6 nM and 17.6 nM for the colorimetric and fluorescent readout, respectively. The quantitative detection of NO2- in real samples demonstrated satisfactory recoveries and repeatability. Moreover, TMN-NH2 was successfully applied for monitoring NO2- in Escherichia coli by confocal fluorescence imaging.

Abstract:To set up effective analysis method is important for environmental monitoring. In this work, the 4-aminophenol functionalized nitrogen and sulfur co-doped carbon quantum dots (AP-NSCDs) were synthesized based on the functionalization of NSCDs obtained by one-step hydrothermal method and used as a new fluorescent sensor for the determination of nitroalkene pesticide and medicine, such as nitenpyram, nizatidine and ranitidine. Nitroalkenes could quench the fluorescence of the functionalized carbon dots due to the fluorescence resonance energy transfer (FRET) principle. The sensor showed high sensitivity, selectivity, and stability, with a wide linear range of 0.05 ∼ 10 mg/L and a low detection limit of 0.02 mg/L. This sensor was successfully employed to detect nitenpyram in rice, orange and cabbage, and to determine nizatidine and ranitidine in serum, urine and river water as well. The developed method was rapid and sensitive, and had potential for monitoring the nitroalkene compounds in the environment and food.

Abstract:Crop diseases caused by pathogenic microorganisms pose severe threats to the global food supply. Effective diagnostic tools for timely determination of plant diseases become essential to the assurance of agricultural sustainability and global food security. Nucleic acid- and antibody-based molecular assays are gold-standard methodologies for the diagnosis of plant diseases, but the analyzing procedures are complex and laborious. The prominent physical or chemical properties of nanomaterials have enabled their use as innovative and high-performance diagnostic tools for numerous plant pathogens and other important disease biomarkers. Engineered nanomaterials have been incorporated into traditional laboratory molecular assays or sequencing technologies that offer notable enhancement in sensitivity and selectivity. Meanwhile, nanostructure-supported noninvasive detection tools combined with portable imaging devices (e.g., smartphones) have paved the way for fast and on-site diagnosis of plant diseases and long-term monitoring of plant health conditions, especially in resource-poor settings.

Abstract:A new "turn-on" fluorescent probe, composed of a protected phenol group with a p-nitrobenzyl moiety that functions as a latent donor and conjugated with two benzo[f] indolinium acceptors, was developed and applied for imaging nitroreductase (NTR) in hypoxic tumor cells. Nitrobenzyl moieties on the substrate can be conveniently converted into aminobenzyl groups with a nitroreductase-catalyzed reaction in the presence of reduced nicotinamide adenine dinucleotide (NADH). This is followed by a cleavage reaction and the release of the free phenol moiety, which is manifested in enhanced fluorescence intensity during the detection process. Our experimental results show that the NTR detection ability is over 100 equivalents of other biological reductants (1 mM), whereas the confocal fluorescence imaging of tumor cells indicates the possibility of its application in biomedical research fields for tumor hypoxia diagnosis.
