Abstract:A new "turn-on" fluorescent probe, composed of a protected phenol group with a p-nitrobenzyl moiety that functions as a latent donor and conjugated with two benzo[f] indolinium acceptors, was developed and applied for imaging nitroreductase (NTR) in hypoxic tumor cells. Nitrobenzyl moieties on the substrate can be conveniently converted into aminobenzyl groups with a nitroreductase-catalyzed reaction in the presence of reduced nicotinamide adenine dinucleotide (NADH). This is followed by a cleavage reaction and the release of the free phenol moiety, which is manifested in enhanced fluorescence intensity during the detection process. Our experimental results show that the NTR detection ability is over 100 equivalents of other biological reductants (1 mM), whereas the confocal fluorescence imaging of tumor cells indicates the possibility of its application in biomedical research fields for tumor hypoxia diagnosis.

Abstract:Nitroreductase (NTR) may be more active under the environment of hypoxic conditions, which are the distinctive features of the multiphase solid tumor. It is of great significance to effectively detect and monitor NTR in the living cells for the diagnosis of hypoxia in a tumor. Here, we synthesized a novel turn-on fluorescent probe NTR-NO2 based on a fused four-ring quinoxaline skeleton for NTR detection. The highly efficient probe can be easily synthesized. The probe NTR-NO2 showed satisfactory sensitivity and selectivity to NTR. Upon incubation with NTR, NTR-NO2 could successively undergo a nitro reduction reaction and then generate NTR-NH2 along with significant fluorescence enhancement (30 folds). Moreover, the fluorescent dye NTR-NH2 exhibits a large Stokes shift (Delta lambda = 111 nm) due to the intramolecular charge transfer (ICT) process. As a result, NTR-NO2 displayed a wide linear range (0-4.5 mu g mL(-1)) and low detection limit (LOD = 58 ng mL(-1)) after responding to NTR. In addition, this probe was adopted for the detection of endogenous NTR within hypoxic HeLa cells.

Abstract:Nitroreductase (NTR) is to be pivotal in the biodegradation of nitroaromatics. NTR is produced in tumor tissues under hypoxic conditions, which is one of the markers for early tumor diagnosis. In this study, a novel probe FD-NTR was developed for NTR detection. Probe FD-NTR can exhibit a specific reaction with NTR in the presence of NADH. The probe displayed satisfactory selectivity and sensitivity towards NTR with a calculated detection limit of 12 ng/mL. Under the conditions of low cytotoxic hypoxia, the FD-NTR probe has shown successful application in imaging both MCF-7 cells and tumor tissues, which indicated that the FD-NTR probe holds promising application prospects for detecting NTR in tumors.

Abstract:Nitroreductase as a potential biomarker for aggressive tumors has received extensive attention. In this work, a novel NIR fluorescent probe for nitroreductase detection was synthesized. The probe Py-SiRh-NTR displayed excellent sensitivity and selectivity. Most importantly, the confocal fluorescence imaging demonstrated that HepG-2 cells treated with Py-SiRh-NTR under hypoxic conditions showed obvious enhanced fluorescence, which means that the NTR was overexpressed under hypoxic conditions. Moreover, the probe showed great promise that could help us to study related anticancer mechanisms research.

Abstract:Nitroreductase (NTR) can be overexpressed in hypoxic tumors, thus the selective and efficient detection of NTR is of great importance. To date, although a few optical methods have been reported for the detection of NTR in solution, an effective optical probe for NTR monitoring in vivo is still lacking. Therefore, it is necessary to develop a near-infrared (NIR) fluorescent detection probe for NTR. In this study, five NIR cyanine dyes with fluorescence reporting structure decorated with different nitro aromatic groups, Cy7-1-5, have been designed and explored for possible rapid detection of NTR. Our experimental results presented that only a para-nitro benzoate group modified cyanine probe (Cy7-1) could serve as a rapid NIR fluorescence-enhanced probe for monitoring and bioimaging of NTR. The structure function relationship has been revealed by theoretical study. The linker connecting the detecting and fluorescence reporting groups and the nitro group position is a key factor for the formation of hydrogen bonds and spatial structure match, inducing the NTR catalytic ability enhancement. The in vitro response and mechanism of the enzyme-catalyzed reduction of Cy7-1 have been investigated through kinetic optical studies and other methods. The results have indicated that an electro-withdrawing group induced electron-transfer process becomes blocked when Cy7-1 is catalytically reduced to Cy7-NH2 by NTR, which is manifested in enhanced fluorescence intensity during the detection process. Confocal fluorescence imaging of hypoxic A549 cells has confirmed the NTR detection ability of Cy7-1 at the cellular level. Importantly, Cy7-1 can detect tumor hypoxia in a murine hypoxic tumor model, showing a rapid and significant enhancement of its NIR fluorescence characteristics suitable for fluorescence bioimaging. This method may potentially be used for tumor hypoxia diagnosis.

Abstract:Nitroreductase (NTR) is a highly expressed endogenous enzyme in hypoxic tumor cells, which is associated with the hypoxic degree in tumors. Therefore, it is particularly important to develop low-toxicity and high-sensitivity fluorescent probes for NTR. Herein, we report an NTR-activated near-infrared probe with good selectivity and sensitivity. More importantly, the background fluorescence signal is effectively reduced due to the quenching effect of nitro group as well as the spironolactone (ring-closed) form of rhodamine moiety. Upon reaction with NTR, the probe exhibits a remarkable fluorescence off-on response at 740 nm (about 32-fold intensity enhancement), with an extremely low detection limit of 1.09 ng/mL. Moreover, the probe is able to target mitochondria. With this probe, we have achieved the visualization of NTR overexpression in hypoxic cancer cells, as well as in hypoxic tumors in vivo as revealed in the tumor-bearing mouse model. By using this probe, some essential information may be revealed about hypoxia-relevant diseases.

Abstract:Nitroreductase (NTR) acts as a detective enzyme that can be used to judge the hypoxic level of solid tumors. To date, only a few optical probes can effectively and selectively monitor NTR in vivo. Therefore, it is important to develop an ideal probe with near-infrared (NIR) excitation and emission for NTR detection and imaging. In this study, we report a NIR frequency upconversion luminescence (FUCL) probe (Cy7-NO2) based on a cyanine structure decorated with an aromatic nitro group to detect NTR in vivo. Cy7-NO2 not only was excited at 850 nm and emitted at 790 nm, but also showed a fast and selective response to NTR. Docking calculations revealed that NTR was able to form hydrogen bonds with Cy7-NO2 and underwent an enzyme-catalyzed reduction reaction. Compared with absorption and the Stokes emission methods, FUCL method exhibited a lower detection limit (3.4 ng mL(-1)). In addition, Cy7-NO2 was used for FUCL bioimaging of NTR in living cells and mice. All of these results proved that Cy7-NO2 as a FUCL probe supplies a new method for biodetection and bioimaging of NTR in vitro and in vivo.

Abstract:Nitroreductase (NTR) is a type of cytoplasmic enzyme that depends on flavin mononucleotide or flavin adenine dinucleotide, and that is widely present in tumor cells where it increases during hypoxia. NTR can effectively reduce aromatic nitro compounds to corresponding amino compounds, with the assistance of reduced nicotinamide adenine dinucleotide (NADH). In this study, a nitro-containing fluorescent turn-on probe, TP-NO, was synthesized and characterized. TP-NO exhibited a fast and selective response towards NTR (LOD = 1.96 ng/mL), and which reaction mechanism was proved through molecular docking. In addition, TP-NO has been successfully applied in imaging hypoxia levels in tumor cells, and zebrafish. This work provided a probe that can be applied to investigate NTR in biological systems.

Abstract:Hypoxia is a common characteristic of solid tumors, which is caused by the imbalance of oxygen supply and consumption. As the expression level of nitroreductase (NTR) increases in hypoxic solid tumors, NTR is one of the common biomarkers of hypoxia and widely used to evaluate the degree of tumor hypoxia. In this study, we designed and synthesized a highly water-soluble chemiluminescent probe, CL-NTR, for the detection of NTR activity in hypoxic tumors. It was found that the probe could be used to detect NTR with high sensitivity, and the total light photons increased tremendously with 6000-fold after the probe was treated with NTR. The chemiluminescence total light photons emission was directly proportional to the concentration of nitroreductase in the range of 3-55 ng/mL, with a detection limit of 0.947 ng/mL. Finally, the probe was successfully used to evaluate NTR activity in living mice by chemiluminescent imaging. In general, this probe has a remarkable response to NTR, which provides a promising method for the determination of NTR activity in vivo.

Abstract:Hypoxia plays an important role in tumor progression, and the development of efficient methods for monitoring hypoxic degree in living systems is of great biomedical importance. In the solid tumors, the nitroreductase level is directly corresponded with the hypoxic status. Many one-photon excited fluorescent probes have been developed for hypoxia imaging in tumor cells via the detection of nitroreductase level. However, two-photon excited probes are more suitable for bioimaging. In this work, a two-photon probe 1 for nitroreductase detection and hypoxic status monitoring in living tumor cells and tissues was reported for the first time. The detection is based on the fact that the nitro-group of probe 1 could be selectively reduced to an amino-group by nitroreductase in the presence of reduced NADH, following by a 1,6-rearrangement-elimination to release the fluorophore, resulting in the enhancement of fluorescence. The probe exhibited both one-photon and two-photon excited remarkable fluorescence enhancement (∼70-fold) for nitroreductase, which afforded a high sensitivity for nitroreductase, with a detection limit of 20 ng/mL observed. Moreover, the applications of the probe for fluorescent bioimaging of hypoxia in living cells and two-photon bioimaging in tissues were carried out, with tissue-imaging depths of 70-160 μm observed, which demonstrates its practical application in complex biosystems.

Abstract:Nitroreductase (NTR), one of the flavin-dependent enzymes and an upregulated enzyme under tumor hypoxia, has been studied for decades. Many fluorescent probes were developed to detect NTR activity; however, these probes tend to diffuse away from their reaction site (NTR) inevitably, leading to inappropriate sample fixation, lower accuracy of NTR localization, and weaker signal-to-noise ratio. Herein, we present the design, synthesis, in vitro evaluation, and biological applications of an NTR-activatable fluorogenic and labeling probe FY. By integrating with quinone methide (QM) proximity-based protein labeling, the additional fluoromethyl group on FY serves as a potential origin of QM. Compared with conventional fluorescent probes, this new NTR probe not only offers mitochondrial localizable and fluorogenic response but also achieves permanent retention on the site of activation with an enhanced spatial resolution to improve the detection sensitivity even after cell fixation. We believe our work could offer an expandable synthetic approach to develop these permanent labeling and imaging fluorescence probes for deciphering complex biological events.

Abstract:Peroxynitrite (ONOO-) has been proven to participate in various physiological and pathological processes, and may also be a contributing factor in many diseases. In view of this, there is a need to develop detection tools for unambiguously tracking a small amount of endogenous ONOO- to reveal its exact mechanisms. In this paper, a colorimetric and red-emitting fluorescent probe Red-PN, based on a rhodamine-type fluorophore and hydrazide reactive site is described. The probe Red-PN possesses the advantages of rapid response (within 5 s), visual color change (from colorless to pink), preeminent sensitivity (detection limit = 4.3 nM) and selectivity. Because of these outstanding performances, it was possible to accurately detect endogenous ONOO-. It was encouraging that the probe Red-PN could be used effectively for tracking the relatively low levels of endogenous and exogenous ONOO-in living cells and zebrafish. Thus, it is envisioned that the probe Red-PN would have promising prospects in applications for imaging ONOO- in a variety of biological settings.

Abstract:Rhodamine dyes bearing aromatic nitro group has been synthesized for nitroreductase enzyme chemosensing applications. The probe is showing very selective turn-on fluorescent response towards nitroreductase enzymes and in hypoxic conditions. The sensor displays a remarkable fluorescent enhancement at 557 nm (λex = 500 nm) without the interference of other biologically relevant species under hypoxic conditions in a physiological medium. The nitro group in the sensor is reduced by the nitroreductase enzyme to the amino group, resulting in the hydrolysis of the probe and subsequent release of highly fluorescent rhodamine 6G dye is observed. This rhodamine based fluorescent probe has been utilized for the imaging of nitroreductase enzymes as well as hypoxia in live cells.

Abstract:Hypoxia is a typical feature of solid tumors. To detect hypoxic tumor cells, we designed new selective turnon probes by conjugating (1-methyl-2-nitro-1H-imidazol-5-yl)methanol to the near-infrared fluorescence probe DCPO through an ether linkage and bis-carbamate linkage specifically. The proposed activation mechanism was demonstrated through the nitroreductase and cytochrome P450 assays in vitro. However, two probes revealed different hypoxia selectivities when evaluated on H460, HeLa and A549 cell lines. The hypoxia selectivity of IOD was higher than that of IND. The probe IOD (containing the ether linkage) was considered to be a promising hypoxia-selective fluorescent probe.

Abstract:A new cresyl violet-based fluorescent off-on probe has been developed through a one-step synthesis for the detection of nitroreductase (NTR) and hypoxia. The detection mechanism is based on the NTR-catalyzed reduction of the probe to cresyl violet, accompanied with a large fluorescence enhancement at a long wavelength of 625 nm. The probe can detect NTR in aqueous solution with high selectivity and sensitivity, and the detection limit is 1 ngmL(-1) NTR. Most importantly, the probe has been successfully used to image not only NTR and hypoxia in living cells, but also the distribution of NTR in zebrafish in vivo.
