Abstract:Extrusion-based three-dimensional (3D) printing has recently become a major field that provides significant benefits, as it is principally employed to fabricate 3D scaffolds, exploiting soft biomaterials. The 3D printing hydrogel-based ink requires crucial properties, such as printability and printing fidelity to fabricate the appropriate structure. However, it typically uses trial and error techniques to achieve a three-dimensional structure, which wastes material and time. This study employed multiphysics simulation to predicate the potential printability of chitosan hydrogel as a desirable biomaterial used in tissue engineering. The flow was presumed to be laminar and two-phased in the simulations. Furthermore, the impact of different velocities and viscosities in extrusion-based chitosan 3D printing was investigated. Moreover, the model validation of the printed chitosan hydrogel was investigated to confirm the simulation outcomes for high-quality printing. The effect of different printing settings was studied during the experimental test. The results obtained from the simulation and experiments provide information for deciding the optimum parameters for printing chitosan-based ink with high quality.

Abstract:Owing to flexible structural design and rapid prototyping manufacturing, 3D printable hydrogels with solid-like and fluid-like characters have emerged as a promising candidate for biomedical engineering, soft robotics, and flexible electronics. However, existing 3D printing of hydrogel place substantial constraints on hydrogel high-fidelity of the printed hydrogel complex structures due to the shape deformation and collapse caused by water evaporation. Herein, we propose a mixed solvent printing strategy followed with water dialysis to achieve vat photopolymerization 3D printing of high-fidelity hydrogel structure by restraining water evaporation. Concretely, dimethyl sulfoxide (DMSO) is introduced into the hydrogel ink as co-solvent to improve the anti-dehydration performance during 3D printing. The as-prepared gels are then transformed into hydrogels with good fidelity and excellent performance through solvent exchange and water dialysis. Enabled by these characteristics, various functional devices with complex structures are demonstrated with the present high-fidelity 3D printing of diverse hydrogels. The solvent assisted high-fidelity hydrogel 3D printing protocol combining with the compatibility to various high-performance hydrogels is believed to be promising for wearable devices, wound dressings, electronic skin, and intelligent robots.

Abstract:Chitosan (CS), as the only alkaline polysaccharide with inherent biocompatibility and biodegradability in nature, is one of the most studied biopolymers for hydrogel preparation. However, the inherent low mechanical strength and uncontrollable swelling of CS hydrogels limit their use as printing inks. Herein, we present an integrated rapid-prototyping and strength-increasing CS hydrogels 3D printing strategy using only saturated NaCl containing trace initiator as the ink-receiving solution. Moreover, the hydrogel also exhibits excellent anti-swelling and cytocompatibility, providing a customized, high-strength scaffold for tissue engineering applications.

Abstract:How to construct three-dimensional oriented chitosan scaffolds with improved mechanical property is essential to further design bone-regenerative scaffolds. Materials with multi-layer structures involve excellent mechanical properties. In this research, a facile approach was adopted to construct three-dimensional oriented chitosan scaffolds. The chitosan gel with multi-layer structure was first prepared with in-situ precipitation, and then lyophilization was applied to obtain porous scaffolds. SEM images indicated that the porous scaffolds had spoke-like framework in cross-section and multi-layer structure in vertical-section. Compared with the disordered scaffolds prepared with lyophilization method, the scaffolds prepared with in-situ precipitation method showed a significantly improved compressive strength. With chitosan concentration of 4–5% and drying time of 60min, the scaffold showed the best comprehensive property with a suitable porosity and a high compressive strength. This novel porous scaffold with three-dimensional oriented structure might have potential application in bone tissue engineering.

Abstract:Photo-crosslinkable gelatin methacrylate (GelMA) has become an attractive ink in 3D printing due to its excellent biological performance. However, limited by low viscosity and long cross-linking time, it is still a challenge to directly print GelMA by extrusion-based 3D printing. Here, to balance the printability and biocompatibility, biomaterial ink composed of GelMA and nanoclay was specially designed. Using this ink, complex scaffolds with high shape fidelity can be easily printed based on the thixotropic property of nanoclay. In this study, we tried to answer some basic printing-required questions of this ink, including the printability window, general properties (porosity, mechanical strength, et al), and biocompatibility. We found that the GelMA/Nanoclay ink enabled printing complex 3D scaffolds, such as a bionic ear and a branched vessel. Furthermore, the addition of nanoclay improved the porosity, increased the mechanical strength, reduced the degradation ratio, and maintained a good biocompatibility of the printed scaffolds. Therefore, this method offers an easy way to print complex scaffolds with good shape fidelity and biological performance, and it might open up new potential applications for the customized therapy of tissue defects.

Abstract:Hydrogel bioprinting is a major area of focus in the field of tissue engineering. However, 3D printed hydrogel scaffolds often suffer from low printing accuracy and poor mechanical properties because of their soft nature and tendency to shrink. This makes it challenging to process them into structural materials. In this study, natural chitosan hydrogel scaffolds were, for the first time, reinforced with milled silk particles and fabricated by 3D printing. Compared with pure chitosan scaffolds, the addition of silk particles resulted in up to a 5-fold increase in compressive modulus as well as significantly better printing accuracy and improved scaffold stability. The chitosan/silk inks flowed well during printing; loading of up to 300% silk (w/w) resulted in only minor changes in the rheological properties of the ink. Particle loading also enabled tuning of the surface roughness of the scaffolds and improved scaffolds' biodegradability. The printed composite hydrogel scaffolds showed no cytotoxicity and supported adherence and growth of human fibroblast cells.

Abstract:Chitosan(CS) hydrogels prepare under novel alkaline conditions have attracted attention because they have more excellent mechanical properties than traditional acidic preparation conditions.We observe that the prepared CS sol still exhibits excellent printability even in the lower mass fraction range.It can be empolyed for fast printing and stable shape retention at room temperature.In order to meet the 3D printing of hydrogels with high mechanical properties, a solid cross-linked network is constructed based on polyvinyl alcohol (PVA) and CS .The hybrid hydrogel ink retains the excellent printability of CS hydrogel while also enhancing its mechanical properties (strain>400 %).Then graphene oxide (GO) is introduced into the system to give full play to the multi-functional characteristics of the composite hydrogels.This kind of hydrogel is used to print flexible sensors and shows very distinctive application value.Furthermore, it can be served as a flexible electrode material for the design of complex circuit systems, contributing to the development of flexible sensors.In short, this work will open a new research path for the application of fast 3D printing hydrogels with ultrastrenth properity.

Abstract:High-strength chitosan hydrogels (H-CS) were prepared without external crosslinker based on a LiOH/urea solvent system. The resultant H-CS hydrogels showed much better mechanical properties than conventional chitosan hydrogels (L-CS). Mechanical properties of H-CS can be tuned by adjusting the concentration of chitosan solution. SEM images of freeze-dried H-CS hydrogels displayed well-defined, interconnected, three-dimensional porous network structures that could greatly improve their mechanical properties. H-CS hydrogels fabricated in this study show great potential as a structural and functional material in the biomedical field.

Abstract:Three-dimensional (3D) printing is a promising method to prepare scaffolds for tissue regeneration. Collagen and chitosan composites are superior materials for tissue engineering scaffold but rarely printed due to their poor printability. Here, we prepared a series of tunable hybrid collagen/chitosan bioinks with significantly improved printability through hydrogen bond interaction and printed them into scaffolds by carefully controlling the temperature. Rheological tests proved the printable bioinks had sound shear thinning behavior, dramatical viscosity variation with temperature, and the gelation temperature from 7 to 10 °C. Chitosan could decrease the swelling ratio of the printed scaffolds, while their degradation rate increased with collagen proportion and the values of Young's modulus and tensile strength increased with chitosan proportion. Moreover, the scaffolds containing 2% (m/v) collagen and 2% (m/v) chitosan had a homogeneous and compact honeycomb-like structure, demonstrating the strengthening effect of chitosan. Cell viability assay presented vigorous cell growth on the surface of scaffolds, meanwhile, live cells were also found inside and at the bottom of the scaffolds, indicating the migration of cells. Therefore, chitosan can improve the printability of collagen and the hybrid collagen/chitosan bioinks can be printed into scaffolds with regulated properties, thus can fit different applications in tissue engineering.

Abstract:A major challenge in three-dimensional (3D) printing of hydrogels is the fabrication of stable constructs with high precision and good mechanical properties and biocompatibility. Existing methods typically feature complicated reinforcement steps or use potentially toxic components, such as photocuring polymers and crosslinking reagents. In this study, we used a thermally sensitive hydrogel, hydroxybutyl chitosan (HBC), for 3D-printing applications. For the first time, we demonstrated that this modified polysaccharide is affected by the specific ion effect. As the salt concentration was increased and stronger kosmotropic anions were used, the lower critical solution temperature of the HBC decreased and the storage modulus was improved, indicating a more hydrophobic structure and stronger molecular chain interactions. On the basis of the thermosensitivity and the ion effects of HBC, a 25-layered hydrogel scaffold with strong mechanical properties and an elaborate structure was prepared via a 3D-printing method and one-step ionic post-treatment. In particular, the scaffold treated with 10% NaCl solution exhibited a tunable elastic modulus of 73.2 kPa to 40 MPa and excellent elastic recovery, as well as biodegradability and cytocompatibility, suggesting the potential for its applications to cartilage tissue repair. By simply controlling the temperature and salt concentrations, this novel approach provides a convenient and green route to improving the structural accuracy and regulating the properties of 3D-printed hydrogel constructs.
Loose Pre-Cross-Linking Mediating Cellulose Self-Assembly for 3D Printing Strong and Tough Biomimetic Scaffolds

Abstract:The lack of an effective printable ink preparation method and the usual mechanically weak performance obstruct the functional 3D printing hydrogel exploitation and application. Herein, we propose a gentle pre-cross-linking strategy to enable a loosely cross-linked cellulose network for simultaneously achieving favorable printability and a strong hydrogel network via mediating the cellulose self-assembly. A small amount of epichlorohydrin is applied to (i) slightly pre-cross-link the cellulose chains for forming the percolating network to regulate the theological properties and (ii) form the loosely cross-linked points to mediate the cellulose chains' self-assembly for achieving superior mechanical properties. The fabrication of the complex 3D structures verifies the design flexibility. The printed cellulose hydrogels exhibit a biomimetic nanofibrous topology, remarkable tensile and compressive strength (5.22 and 11.80 MPa), as well as toughness (1.81 and 2.16 MJ/m(3)). As a demonstration, a bilayer scaffold (mimicking the osteochondral structure) consisting of a top pristine cellulose and a bottom cellulose/bioactive glass hydrogel is printed and exhibits superior osteochondral defect repair performance, showing a potential in tissue engineering. We anticipate that our loose pre-cross-linking 3D printing ink preparation concept can inspire the development of other polymeric inks and strong 3D printing functional hydrogels, eventually spreading the applications in diverse fields.
Direct 3D Printed Biomimetic Scaffolds Based on Hydrogel Microparticles for Cell Spheroid Growth

Abstract:Biocompatible hydrogel inks with shear‐thinning, appropriate yield strength, and fast self‐healing are desired for 3D bioprinting. However, the lack of ideal 3D bioprinting inks with outstanding printability and high structural fidelity, as well as cell‐compatibility, has hindered the progress of extrusion‐based 3D bioprinting for tissue engineering. In this study, novel self‐healable pre‐cross‐linked hydrogel microparticles (pcHμPs) of chitosan methacrylate (CHMA) and polyvinyl alcohol (PVA) hybrid hydrogels are developed and used as bioinks for extrusion‐based 3D printing of scaffolds with high fidelity and biocompatibility. The pcHμPs display excellent shear thinning when injected through a syringe and subsequently self‐heal into gels as shear forces are removed. Numerical simulations indicate that the pcHμPs experience a plug flow in the nozzle with minimal disturbance, which favors a steady and continuous printing. Moreover, the pcHμPs show a self‐supportive yield strength (540 Pa), which is critical for the fidelity of printed constructs. A series of biomimetic constructs with very high aspect ratio and delicate fine structures are directly printed by using the pcHμP ink. The 3D printed scaffolds support the growth of bone‐marrow‐derived mesenchymal stem cells and formation of cell spheroids, which are most important for tissue engineering.

Abstract:A porous chitosan-gelatin scaffold with specific geometry and complex internal microarchitectures was fabricated via a novel indirect rapid prototyping technique. A stereolithography (SL) technique was employed to build the resin mold, based on which a high-resolution poly-dimethylsilicone (PDMS) mold was microreplicated. The chitosan-gelatin hybrid solution was cast on the PDMS mold for pre-freezing and the monolayer scaffold with predefined features and highly porous structures was produced upon lyophilization. These monolayer structures along with the special interface layers can be stacked together to form a three-dimensional (3D) scaffold. The morphologies of the predefined features maintain well during the assembly process. Hepatocyte culture experiment indicates that the novel microstructured chitosan-gelatin porous scaffold could facilitate cells to migrate and proliferate into the deep layers of the scaffold in comparison with the traditional porous only scaffold. This approach provides a new way to fabricate a complex 3D scaffold using various natural biomaterials for vital organ engineering.
A route toward fabrication of 3D printed bone scaffolds based on poly(vinyl alcohol)-chitosan/bioactive glass by sol-gel chemistry

Abstract:Among different methods for the fabrication of bone scaffolds, 3D printing has created great advances in tissue engineering and regenerative medicine owing to its ability to make objects mimicking native tissues. Thanks to its abundant availability, structural features, and favorable biological properties, chitosan (CS) hydrogel was selected to be used for preparation of the bone scaffolds. However, the 3D printing of CS-based hydrogels is still under early exploration. Knowing the fact that natural polymers are not so competent at holding large amounts of water, poly(vinyl alcohol) as the second polymer was employed. The novelty of the present research lies in the concept of employing sol-gel chemistry in order to attain proper viscosity and rheological behavior to give self-standing filaments of the polymer blends. Employing sol-gel reaction in the preparation of the hybrid hydrogels had the advantage of endowing shape fidelity to the polymer blend without any solidifying in the needle. The obtained organic-inorganic hybrids were directly printed and subsequently cross-linked. The best performance in terms of mechanical strength, cell viability, and bio-mineralization was observed for the 50:50 ratio. The in vitro cell culture and the bioactivity results showed that the printed scaffolds with this method have great potential in bone tissue engineering. Further, this method could be expandable to print other hydrogels with diverse applications such as implantable devices, soft robotics, etc.
Direct 3D Printing of High Strength Biohybrid Gradient Hydrogel Scaffolds for Efficient Repair of Osteochondral Defect

Abstract:Abstract The emerging 3D printing technique allows for tailoring hydrogel‐based soft structure tissue scaffolds for individualized therapy of osteochondral defects. However, the weak mechanical strength and uncontrollable swelling intrinsic to conventional hydrogels restrain their use as bioinks. Here, a high‐strength thermoresponsive supramolecular copolymer hydrogel is synthesized by one‐step copolymerization of dual hydrogen bonding monomers, N ‐acryloyl glycinamide, and N ‐[tris(hydroxymethyl)methyl] acrylamide. The obtained copolymer hydrogels demonstrate excellent mechanical properties—robust tensile strength (up to 0.41 MPa), large stretchability (up to 860%), and high compressive strength (up to 8.4 MPa). The rapid thermoreversible gel ⇔ sol transition behavior makes this copolymer hydrogel suitable for direct 3D printing. Successful preparation of 3D‐printed biohybrid gradient hydrogel scaffolds is demonstrated with controllable 3D architecture, owing to shear thinning property which allows continuous extrusion through a needle and also immediate gelation of fluid upon deposition on the cooled substrate. Furthermore, this biohybrid gradient hydrogel scaffold printed with transforming growth factor beta 1 and β‐tricalciumphosphate on distinct layers facilitates the attachment, spreading, and chondrogenic and osteogenic differentiation of human bone marrow stem cells (hBMSCs) in vitro. The in vivo experiments reveal that the 3D‐printed biohybrid gradient hydrogel scaffolds significantly accelerate simultaneous regeneration of cartilage and subchondral bone in a rat model.

Abstract:In this work, a 3D printed double-network (DN) hydrogel scaffold is designed with chitosan (CS) and polyvinyl alcohol (PVA) as the framework matrix. The addition of PVA into the CS-based hydrogel clearly enhances the mechanical properties and lowers the swelling behaviors of the hydrogels. The crosslinking of CS with genipin can perform the pre-crosslinking to improve the viscosity and 3D printability of the hydrogel precursor, while increasing the PVA content results in lowering the viscosity and 3D printability of the pre-crosslinked hydrogel. The antibacterial property results of the DN hydrogel display that the hydrogel have favorable long-lasting antibacterial ability. The appropriate pre-crosslinked hydrogel with the CS/PVA mass ratio of 3:10 and pre-crosslinking time of 7 h is used for 3D printing to prepare the 3D printed porous DN hydrogels. Moreover, the anti-tumor drug doxorubicin (DOX) is loaded into the 3D printed porous DN hydrogels and the in vitro release study displays the sustainable drug release behavior. And the DOX release from hydrogel scaffold can be adjusted by the pH value of release environment. All of the results indicate that the porous DN CS/PVA hydrogel scaffolds have great application potential for tissue regeneration.

Abstract:To mimic biological tissues with high toughness such as cartilage, it is highly desired to fabricate stable and tough hydrogels with intricate shapes to act as a structural support. Extrusion-based 3D printing is a promising method to fabricate 3D scaffolds with various architectures; however, printing tough hydrogel structures with high fidelity and resolution is still a challenge. In this work, we adopt the fast sol-to-gel transition of κ-carrageenan in the solution of acrylamide upon cooling to fix the printed scaffolds and polymerize the precursor solution to form the second network. The printed constructs of κ-carrageenan/polyacrylamide double-network gels are toughened by soaking in an aqueous solution of zirconyl chloride to form coordination complexes between the Zr4+ ions and sulfate groups of κ-carrageenan. The obtained hydrogels are stable in water and possess good mechanical properties, with a tensile breaking stress of 1-2 MPa, breaking strain of 100-150%, and Young's modulus of 4-10 MPa. The printed grid can hold 150 times its own weight. 3D printed constructs with a high aspect ratio and shape fidelity are obtained by optimizing the printing parameters. Furthermore, a biomimetic strategy is applied to construct a hydrogel composite by filling the printed tough hydrogel scaffold with a cell-laden fibrin hydrogel as the soft substance. Chondrocytes in the hydrogel composite maintain high viability after cyclic compression, demonstrating the load-bearing capacity of the tough scaffold and favorable microenvironment for cells provided by the embedded soft fibrin gel. We envision that this printing strategy for hydrogel constructs with high toughness and good stability, as well as the method to form tough-soft hydrogel composites, can be extended to other systems to develop structural elements and scaffolds towards applications in biomedical devices and tissue engineering.

Abstract:The fabrication process for a novel three-dimensional (3D) chitosan/gelatin scaffold with predefined multilevel internal architectures and highly porous structures is presented combining solid freeform fabrication (SFF), microreplication and lyophilization techniques. The computer model of the scaffold is designed with biological data such as branching angle in liver vascular cast incorporated. Stereolithography (SL), known as a SFF technique, is utilized to build the resin mould, based on which poly-dimethylsilicone (PDMS) mould is produced by microreplication. The chitosan/gelatin hybrid solution is then cast onto the PDMS mould for pre-freeze and the monolayer porous structures with organized internal morphology are produced upon lyophilization. The 3D scaffold can be constructed via stacking these monolayer structures. The properties of porous structure, such as porosity, pore size and micromorphology as well as wall thickness, were investigated. Scanning electron microscopy (SEM) demonstrated that the scaffold possesses multilevel organized internal morphologies including vascular systems (portal vein, artery and hepatic vein) and parenchymal component (hepatocyte chamber). These organized structures enable orderly arrangement of hepatocyte and hepatic nonparenchymal cells and co-culture in the same 3D scaffold to guide liver regeneration in a controlled manner. Cell culture experiment in vitro showed that hepatocytes perform better in the well-defined chitosan/gelatin scaffold than in porous scaffold. This approach makes it flexible to investigate the relationship between internal scaffold microstructure and hepatocyte behavior in vitro. It also provides a new way to fabricate complex 3D scaffold using various natural biomaterials for vital organ engineering.

Abstract:Three-dimensional (3D) printing holds great potential for preparing sophisticated scaffolds for tissue engineering. As a result of the shear thinning properties of an alginate solution, it is often used as 3D printing ink. However, it is difficult to prepare scaffolds with complexity structure and high fidelity, because the alginate solution has a low viscosity and alginate hydrogels prepared with Ca2+ crosslinking are mechanically weak. In this work, chitosan powders were dispersed and swelled in an alginate solution, which could effectively improve the viscosity of an alginate solution by 1.5-4 times. With the increase of chitosan content, the shape fidelity of the 3D printed alginate-chitosan polyion complex (AlCh PIC) hydrogels were improved. Scanning electron microscope (SEM) photographs showed that the lateral pore structure of 3D printed hydrogels was becoming more obvious. As a result of the increased reaction ion pairs in comparison to the alginate hydrogels that were prepared with Ca2+ crosslinking, AlCh PIC hydrogels were mechanically strong, and the compression stress of hydrogels at a 90% strain could achieve 1.4 MPa without breaking. In addition, human adipose derived stem cells (hASCs) adhered to the 3D printed AlCh PIC hydrogels and proliferated with time, which indicated that the obtained hydrogels were biocompatible and could potentially be used as scaffolds for tissue engineering.

Abstract:Chitosan-based hydrogel has been widely used in the field of tissue engineering due to its favorable biocompatibility and good biodegradability. However, this kind of hydrogel generally exhibits poor mechanical stability, which greatly limits its application in the field of 3D bioprinting. In this study, we provided a hybrid bioink created from photocurable chitosan and acrylamide (AM) for digital light processing (DLP) based 3D bioprinting in tissue engineering applications. This hybrid bioink was facilely prepared by combining AM and chitosan modified with methacryloyl groups (CHIMA). The gelling point of the hybrid pre-hydrogel bioink was greatly dependent on the photoinitiators. Both the mechanical properties and cytocompatibility of the hydrogel formed by the bioink can be modulated by varying the AM contents. The hybridization of natural CHIMA and synthetic AM enables the hybrid hydrogels with desirable biological activity and mechanical properties. Utilizing the DLP based 3D printing, this hybrid bioink can be processed into complex 3D hydrogel constructs with high-strength and good biocompatibility. Therefore, the photocurable hybrid bioink composed of CHIMA and AM in proper proportion is well suitable for use in DLP based 3D bioprinting, which would play a promising role in constructing tissues and organs in the future.
