Epigenetics changes of HIN 1 during esophageal squamous cancer progression

Abstract:1849HIN1( high in normal-1) is a putative cytokine that was initially identified by using serial analysis of gene expression (SAGE). HIN-1 was identified as one of the most abundant transcripts in normal luminal mammary epithelial cells and was found to be absent (silenced) in non-invasive breast cancers such as ductal carcinoma in-situ (DCIS). HIN-1 expression is significantly down regulated in 94% of human breast carcinomas and in 95% of preinvasive lesions, such as ductal and lobular carcinoma in-situ. This decrease in HIN-1 expression is accompanied by hypermethylation of its promoter in the majority of breast cancer cell lines (>90%) and primary tumors (74%). To confirm the report, we evaluated methylation status of HIN-1 in samples of DCIS and primary breast tumors using methylation specific PCR (MSP).
