Abstract:3426Esophageal cancer is a common cancer with an increasing incidence worldwide. Squamous cancer, the most common form of this disease in non-Western countries, has a well-defined progression of pre-invasive dysplasia leading to invasive squamous cell carcinoma. We investigated the frequency of epigenetic changes in the promoter region of 14 genes epigenetically silenced in other forms of cancer and traced the most frequent changes in dysplastic lesions. Invasive squamous carcinomas, low to high grade dysplasia, and normal esophagus were examined for methylation changes in the promoter region of each gene using methylation specific PCR. Of these 42 carcinomas, methylation of at least one gene was present in 39 tumors (93%). The frequency of promoter hypermethylation varied among these genes: p16 (50%), O6-Methylguanine methyltransferase, MGMT (36%), MLH1 (33%), CDH1 (31 …
