Abstract:The fabrication and surface morphology of methylene blue (MB)/perfluorosulfonated ionomer (Nafion) modified microcylinder carbon fiber electrode were studied. The stability of the modified electrode is good. The extraction coefficient K of MB between Nafion film and solution is similar to 110, and the diffusion coefficient of MB in Nafion film is similar to 5.3 x 10(-9) cm(2)/s. The MB/Nafion modified microcylinder electrode shows significant electrocatalytic activity for the oxidation of hemoglobin in weak acid media; it can be used for the determination of hemoglobin in the range of 5.0-50 mu M with a relative standard deviation of 3.5% at 6.0 mu M HB. A satisfactory result has been obtained for the determination of hemoglobin in clinical blood samples.

Abstract:Electrodeposition of polyphenothiazine films onto carbon fibre electrode from substitued phenothiazine and unsubstitued phenothiazine were achieved by anodization in acidic or basic media.The modified electrodes had good durability and redox properties.The mechanisms of electropolymerization were investigated.The chemically modified microelectrode,which was constructed by electropolymerizing a new mediator methylene green onto the carbon fibre had electrocatalytic activity to the reduction of hemoglobin.Under the potential range +0.6～(-0.4 V),and in NaAc-HAc(pH=4.4) buffer solution,the mediator behaved as reversible redox process involving two protons.The modified electrode showed that it has good stability and current response to hemoglobin.It can be used as a sensor for determining hemoglobin in FIA or IC-EC method.

Abstract:The hemoglobin concentration in diluted whole blood has been amperometrically measured using a toluidine blue O covalently modified microcylinder carbon fiber electrode. The modified electrode was prepared by modifying the surface of carbon fiber by cyclic voltammetric oxidation to form a lot of -COOH groups, and then covalently linking toluidine blue O (TBO) by acylation. The surface morphology and stability of the TBO modified electrode are discussed. The electrochemical behaviour of the modified electrode was studied by cyclic voltammetry. The number of H+ participating in the surface electrode process and some kinetic parameters in various pH solutions were determined. The TBO modified microelectrode showed significant electrocatalytic activity for the oxidation of hemoglobin(HB) in pH 5.4 solution. The amperometric response to HE increased linearly with increasing concentration from 8.0x10(-6) to 8.0x10(4) M. The obtained HB concentration correlated well with that obtained using a reference procedure. Copyright (C) 1996 by Marcel Dekker, Inc.

Abstract:The direct electrochemistry of hemoglobin (Hb) immobilized on the siliceous mesostructured cellular foam (MCF) modified glassy carbon electrode (GC) was presented. Using a simple physical adsorption method, Hb was immobilized on MCF and the influences of concentration of Hb and the pH value of solution were investigated. UV–vis and FTIR spectra displayed that Hb maintained its natural structure inside the pores of MCF. The electrochemical property of Hb/MCF/GC modified electrode was studied by cyclic voltammetry. A couple of quasi-reversible redox peaks of Hb were observed on the Hb/MCF/GC electrode at about −0.396V and −0.321V in PBS (pH 7.0). The results indicated that MCF provided a favorable microenvironment for immobilization of Hb and facilitated the electron transfer between Hb and electrode surface.

Abstract:Bismuth phosphate (BiPO4), a very attractive candidate for organics electrodegradation, harbors tremendous potential on removing contaminants from water. Here, four carbon microtube electrodes were prepared from corncob, each coated with BiPO4 by a different method to study the electrodegradation of methylene blue (MB). A thorough insight into the composite features of four electrodes was characterized. Better reversibility and electrocatalytic activity of the fourth electrode (BCC4) prepared by digital signal generator was presented with a current density of 5.71 mA cm−2 at a potential of 1.6 V vs Ag/AgCl. The electrochemical impedances and actual lifetime of BCC4 were 125 Ω and 833 h, respectively. The effectiveness of each kind of BiPO4/carbon electrode was preliminarily evaluated by analyzing the actual conversion rate of the MB concentration, which confirms MB electrodegradation by the BiPO4/carbon electrode was mainly dominated by the hydroxyl radical oxidation. The mass transfer rate was increased by carbon microtube; thereby, electrocatalysis of BiPO4/carbon electrode increased as revealed by an increase in the MB degradation rate. The rate constants k obtained for the degradation of MB by BiPO4/carbon electrode at 20 ℃ was 0.0046 mM−1 s−1, which was 11 times than that of BiPO4. The diffusion layer was decreased by carbon microtube, resulting in MB electrodegradation rate increased. The BiPO4 coated on the surface of the carbon microtube electrodes strengthened their electrocatalytic performance, which shed new light on effective selection of suitable carbon electrode for degradation of organics. Therefore, BiPO4/carbon electrode could be potentially applied in the electrodegradation of organic pollutants.

Abstract:An amperometric microsensor for hemoglobin (HB) has been constructed using a carbon fiber electrode modified with poly-(pAAQ). Scanning electron microscopic studies showed that the potential range and time of electrochemical polymerization had strong effects on the morphology of the polymer film at the microelectrode. The stability and the electrochemical properties of the modified electrode were studied using cyclic voltammetry. The pAAQ modified microelectrode showed an excellent electrocatalytic activity for the reduction of HB in a weakly acidic solution. The catalytic current in cyclic voltammetry increased linearly with the HB concentration between 1.0–400 μM. The amperometric response to HB had a linear range of 0.5 μM to 340 μM with a correlation coefficient of 0.990(n = 18). The biosensor was stable for at least one month when it was stored in a phosphate buffer (pH 7.4) below 20 °C. The obtained HB concentration in the whole blood agreed well with the one obtained with the reference instrument.

Abstract:Heteroatom doping is an effective way to elevate the fluorescent performance of carbon dots. In this study, a microwave one-pot approach for the synthesis of SiBCDs was proposed by using citric acid (C6H8O7), boric acid (H3BO3) and (3-aminopropyl) triethoxy sliane (APTES) as source materials. PAAS- SiBCDs were prepared with the assistance of microwave when sodium polyacrylate (PAAS) was added in the precursor of SiBCDs. The products were characterized by X-ray diffraction (XRD), X-ray photoelectron spectroscopy (XPS) and Fourier transform infrared (FT-IR) spectra, respectively, confirming that the product was amorphous CDs, with small amount of Si and B. The synthesized SiBCDs had good monodispersity with size of 4-8 nm, the average size of PAAS-SiBCDs was 5. 2 nm, and the excitation/emission maximum was 350 nm/445 nm. The quantum yield of SiBCDs was 20. 1%, and that of PAAS. SiBCDs was 34. 6%. Based on the quenching effect of hemoglobin (Hb) on the fluorescence PAAS. SiBCDs, a sensitive Hb detection method was developed. A linear range of 0. 21-5. 22 mu mol/L was obtained together with a detection limit of 0. 06 mu mol/L. The method was applied in the determination of Hb in human blood samples.

Abstract:The electrochemical behavior of methylene blue (MB) at a carbon fiber microcylinder electrode, was studied by cyclic voltammetry. The charge transfer coefficient cu of the electrode reaction with two-electron transfer is 0.5, and the number of Hf participating in the electrode process is 3 at pH 2.2-5.4, 2 at pH 5.4-6.0 and 1 at pH 6.0-10.7, respectively. The standard electron transfer rate constant k degrees', and standard formal potential E degrees' of MB at various pH were determined by using the carbon fiber microcylinder electrode. The electrode reaction mechanism of methylene blue at various pH is proposed. The adsorbability of MB at the electrode is discussed and explored by cyclic voltammetry and chronocoulometric technique.

Abstract:We developed a novel mesoporous carbon/whisker-like carbon (MCWC) composite which can promote the direct electron transfer of hemoglobin (Hb) immobilized on its surface. The cyclic voltammetric results showed that Hb immobilized on the surface of the MCWC composite could undergo a direct quasi-reversible electrochemical reaction. Its formal redox potential, E0′ is −0.313V in the phosphate buffer solution (pH 6.9) at a scan rate of 200mV/s and is almost independent of the scan rate in the range of 100–600mV/s. The dependence of E0′ on the pH of phosphate buffer solution indicated that the redox reaction of Hb includes a one-electron-transfer reaction process coupled with one-proton-transfer. The experiment obtained larger value of electron transfer rate constant, ks, than that of Hb immobilized on other carriers reported previously due to its special structure of loosely packed nanometer-scale carbon whiskers and thus formed “V-type” nano-pores. Furthermore, Hb immobilized on the surface of the MCWC composite can retain the stable bioelectrocatalytic activity for the reduction of H2O2.

Abstract:Protein-CMC films were made by casting a solution of myoglobin (Mb) or hemoglobin (Hb) and carboxymethyl cellulose (CMC) on pyrolytic graphite electrodes. In pH 7.0 buffers, Mb and Hb incorporated in CMC films gave a pair of well-defined and quasi-reversible cyclic voltammetric peaks at about −0.34 V vs. SCE, respectively, characteristic of heme FeIII/FeII redox couples of the proteins. The electrochemical parameters such as apparent standard heterogeneous electron transfer rate constants (ks) and formal potentials (E°′) were estimated by square wave voltammetry with nonlinear regression analysis. In aqueous solution, stable CMC films absorbed large amounts of water and formed hydrogel. Scanning electron microscopy of the films showed that interaction between Mb or Hb and CMC would make the morphology of dry protein-CMC films different from the CMC films alone. Positions of Soret absorbance band suggest that Mb and Hb in CMC films retain their secondary structure similar to the native states in the medium pH range. Trichloroacetic acid, nitrite, oxygen, and hydrogen peroxide were catalytically reduced at protein-CMC film electrodes.

Abstract:Hemoglobin (Hb) can take direct electron-transfer reactions after being entrapped in poly-3-hydroxybutyrate (PHB) film. A pair of well-defined, quasi-reversible cyclic voltammetric peaks is thus obtained at an Hb-PHB modified pyrolytic graphite electrode. The anodic and cathodic peaks are located at -224 and -284 mV for a pH 5.0 acetate buffer solution. Meanwhile, the peroxidase activity of the protein in the membrane has been greatly enhanced, with the apparent Michaelis-Menten constant calculated to be 1076 microM. According to the direct electron transfer property and enhanced peroxidase activity of Hb in the membrane, a Hb-PHB based hydrogen peroxide biosensor is prepared, with a linear range 6.0 x 10(-7) to 8.0 x 10(-4) M. The pathway of reductive dehalogenation of trichloroacetic acid is also discussed in detail. The highly reduced form of Hb produced in PHB film can be used to dechlorinate di- and monochloroacetic acid. The catalytic ability of Hb toward the reduction of nitric oxide has been investigated as well. Due to its biodegradability, low cost, chemical inertness, and especially its biocompatibility and non-toxicity, PHB would be a desirable film in the sensor field.

Abstract:Using chitosan as an effective linker between CMK-3 and glassy carbon electrode surface, {Hb/CMK-3}n multilayer film-modified electrodes were constructed through layer-by-layer assembly. The morphology of thus-formed {Hb/CMK-3}n film was characterized by scanning electron micrographs, and the interaction of hemoglobin (Hb) with CMK-3 was studied by UV–vis spectroscopy and electrochemical methods. Under optimal conditions, {Hb/CMK-3}6 film showed a couple of stable and well-defined redox peaks at about −377 and −296mV in pH 7.0 buffers. Furthermore, the {Hb/CMK-3}6 film displayed excellent electrocatalysis to the reduction of both H2O2 and O2. Based on thus-formed film and its direct electron transfer behavior, a novel biosensor was presented for the determination of H2O2 ranging from 1.2 to 57μM with the detection limit of 0.6μM at S/N=3. CMK-3 provided a desirable matrix for protein immobilization and biosensor preparation.

Abstract:The functional group -COOH on carbon fiber microcylinder electrode(CFMCE) was formed by electrochemical oxidation with cyclic voltammetry, and the Nile blue modified CFMCE was manufactured by means of electrostatic-attraction. The electrochemical behaviour of the modified electrode was studied, and its stability was discussed. The apparent standard electron transfer rate constant and charge transfer coefficient of fixed Nile blue A at various pH, and the number of H+ participating in electrode reaction were determined. The experiments indicated the modified electrode can catalyze the electrode reaction of hemoglobin at carbon fiber microcylinder electrode.

Abstract:Hemoglobin voltammetrically responds at a bare silver microelectrode. By the introduction of a proper amount of sodium dodecyl sulfate, the peak current of the protein greatly increased and its voltammogram became symmetrical, so the method could be directly used for the determination of protein. Experimental results revealed that the dependence of peak current on hemoglobin concentration was linear in the range of 5. 0��10 -7�� 5. 0��10 -6 mol/L. The determination for a clinical human blood sample was satisfactory.

Abstract:Hemoglobin modified electrode was successfully fabricated to realize direct electrochemistry by immobilizing of Hemoglobin (Hb) in bimodal mesoporous silica (BMS) and chitosan (CS) inorganic–organic hybrid film. Here, BMS acted as a support to immobilize Hb due to its large pores and CS acted as a binder to increase film adherence and stabilizer to prevent the leakage of Hb. The resulting electrode (Hb/BMS/CS) gave a well-defined, reversible redox couple for HbFe(III)/Fe(II) with a formal potential of about −0.32V (vs. Ag/AgCl) in pH 7.0 phosphate buffer solution. Hb/BMS/CS electrode showed a better electrocatalytial performance to H2O2 with wider linear detection range, lower detection limit, and higher sensitivity than that at electrode without BMS. The improved electrocatalytic performance for Hb/BMS/CS electrode was possibly contributed to BMS bimodal structure, whose large pores with 10–40nm provide favorable conditions for protein immobilization and small pores with 2–3nm avoid the mass-transfer limitations. In addition, UV–Vis and FTIR spectra indicated that Hb well maintained its native structure in the hybrid film.

Abstract:A silver electrode modified by silver nanoparticles was prepared and its application to the electrochemical determination of hemoglobin (Hb) was studied. Silver nanoparticles were prepared by reduction of AgNO3 with NaBH4. They can be efficientyl immobilized on the surface of a silver electrode. It showed that the silver nanoparticles could clearly enhance the electron transfer process between hemoglobin and the electrode compared with bulk silver. Linear sweep voltammetric measurement of Hb at the chemical modified electrode indicated that the oxidative peak currents of Hb were linear to its concentration ranging from 2.0 x 10(-7) to 1.0 x 10(-5) mol/L with the detection limits of 7.4 x 10(-8) mol/L. The direct electrochemistry of Hb was also studied by cyclic voltammetry. The method has been used to detect Hb in artificial mixture sample.

Abstract:A well-defined amphiphilic diblock copolymer, poly [N-(2-methacryloyloxyethyl) pyrrolidone]-block-poly [glycidyl methacrylate] (PNMP-b-PGMA), was applied to immobilize hemoglobin (Hb) in a simple procedure. The characterization of PNMP-b-PGMA/Hb film was demonstrated by ultraviolet–visible (UV–vis) spectra, atomic force microscopy (AFM), and electrochemical impedance spectroscopy (EIS). The electrochemical behaviors of Hb in PNMP-b-PGMA film have been investigated and a pair of well-defined and nearly reversible cyclic voltammetric peaks for the protein heme Fe(III)/Fe(II) redox couples were observed at about −0.409V (vs. SCE) in pH 7.0 phosphate buffer solution (PBS). The results showed that the PNMP-b-PGMA film improved the protein loading with the retention of bioactivity and greatly promoted the direct electron transfer between Hb and electrode surface. This PNMP-b-PGMA/Hb film modified electrode also exhibited high electrocatalytic effects towards reduction of nitric oxide (NO). These results indicated the potential applicability of the films as new types of third-generation biosensors or bioreactors based on direct electrochemistry of the proteins.

Abstract:Hemoglobin (Hb) was incorporated in dimethylaminoethyl methacrylate (DMAEMA) to form a film and the film was modified on pyrolytic graphite (PG) electrode. UV-Vis spectra suggested that Hb in the film could keep its secondary structure. Consequently, a pair of stable, well-defined, and quasi-reversible cyclic voltammetric peaks could be observed with the formal potential at -206 mV ( vs. saturated calomel electrode), characteristic of heme Fe(III)/Fe(II) redox couple of the protein. The apparent heterogeneous electron transfer rate constant and other electrochemical parameters were presented. The catalytic activity of Hb in DMAEMA films toward hydrogen peroxide was also investigated.

Abstract:Several aspects of the application of silver electrode to the electrochemical studies of hemoglobin have been discussed in this paper . The silver electrode could not only be used directly as the electrode for the electrochemical studies of hemoglobin, but also react with phenothiazine and benzimidazole to give stable and useful mediator-coated electrodes. In addition, the silver electrode could help sodium dodecyl sulfate to give full play to its promoting effect on the protein.

Abstract:Effects of pore structure of mesoporous silicas (MPS) on the direct electron transfer of hemoglobin (Hb) were investigated. MCM-41 and SBA-15 were used to supply 1D channel and hexagonal mesoporous structure with the average pore diameters of 3.2 and 6.9nm, respectively, while MCF was used to supply 3D and caged mesoporous network structure with the average pore diameter of 15.7nm. Hb was immobilized by the physical adsorption method without significant denaturation and have the saturated binding amounts of 15.8, 240.0 and 325.7mgg−1 for MCM-41, SBA-15 and MCF, respectively. The electrochemical properties of Hb/MPS/GC modified electrode were studied by cyclic voltammetry (CV). A couple of redox peaks were observed at Hb/SBA-15/GC and Hb/MCF/GC, while no obvious redox peak was found at Hb/MCM-41/GC modified electrode because Hb molecule could not enter into the channels of MCM-41. The CV signal of Hb/MCF/GC is about 2× as large as that of Hb/SBA-15/GC electrode when the same amount of Hb was immobilized in the pores of MCF and SBA-15, which suggests that the 3D porous structure may be the key factor for promoting the electron transfer of Hb.
