Abstract:Cellular packing of flexible nanofibers, including natural cytoskeletal microtubules, actin filaments, synthetic nanotubes and nanowires, is of fundamental interest to the understanding of a wide range of cell activities, including cell shape control, cell movement, cell division, and nano-cytotoxicity. Here, we perform molecular dynamics simulations and theoretical analysis to elucidate how the geometrical and mechanical properties of a flexible nanofiber influence its encapsulation within a lipid vesicle. Our analysis indicates that the packing morphology depends on the length and stiffness of the nanofiber, the initial configuration of the nanofiber–vesicle system and the pressure difference across the vesicle membrane. We establish a packing phase diagram based on three distinct vesicle morphologies in equilibrium, including a non-axisymmetric dumpling-shaped vesicle with a strongly curved nanofiber, a cherry-shaped vesicle with a tubular membrane protrusion enclosing a significant portion of the nanofiber, and an axisymmetric lemon-shaped vesicle with a pair of protruding tips induced by the encapsulated nanofiber.

Abstract:Recently, food grade nanofiber-based materials have received growing attentions in food packaging. In this work, novel active and intelligent packaging nanofibers based on gelatin/ chitosan with curcumin (GA/CS/ CUR) were developed via electrospinning technique. Effects of the incorporation of CUR content (0.1%- 0.3%, m/m) on the microstructure and functional properties of the electrospun nanofibers were investigated. Morphological studies using scanning electron microscopy indicated that loading CUR can affect the average diameter of nanofiber mats, which remained around 160-180 nm. The addition of an appropriate level CUR (0.2%, m/m) led to a stronger intermolecular interaction, and thus enhanced the thermal stability and tensile strength of the obtained nanofibers. Meanwhile, the incorporation of CUR significantly improved antioxidant activity and the antimicrobial activity of GA/CS/CUR nanofibers. Moreover, the sensitivity of nanofibers to ammonia results indicated that GA/CS nanofibers containing 0.2% CUR (GA/CS/CUR II) presented high sensitivity of colorimetric behavior to ammonia (within 3 min). These results suggest GA/CS/CUR II nanofibers has great potential as a multifunctional packaging to protect and monitor the freshness of protein -rich animal foods, such as meat and seafood. Packing of flexible 2D materials in vesicles

Abstract:To understand the mechanics of cellular packing of two-dimensional (2D) materials, we perform systematic molecular dynamics simulations and theoretical analysis to investigate the packing of a flexible circular sheet in a spherical vesicle and the 2D packing problem of a strip in a cylindrical vesicle. Depending on the system dimensions and the bending rigidity ratio between the confined sheet and the vesicle membrane, a variety of packing morphologies are observed, including a conical shape, a shape of three-fold symmetry, a cylindrically curved shape, an axisymmetrically buckled shape, as well as the initial circular shape. A set of buckling analyses lead to phase diagrams of the packing morphologies of the encapsulated sheets. These results may have important implications on the mechanism of intracellular packing and toxicity of 2D materials.
Compression-driven Migration of Nanoparticles in Semiflexible Polymer Brushes

Abstract:The conformations of binary nanoparticles (NPs) in semiflexible polymer brushes under compression are investigated by molecular dynamics simulations, and a migration phenomenon of large NPs in semiflexible polymer brushes is observed in our simulation. For flexible polymer brushes, small NPs disperse freely within polymer brushes and large NPs tend to be located at the brush interface during the compression process. However, for semiflexible polymer brushes, both small and large NPs enter polymer brushes at the beginning of compression process. Most of large NPs can penetrate into polymer brushes and migrate towards the grafting surface, while most of small NPs are always located within polymer brushes during the compression process. The micro-spatial distributions of binary NPs are controlled easily by compressing the impenetrable surfaces through the external forces. There is a free-energy barrier located in the middle region between two impenetrable surfaces, and a free-energy well near the grafting surface in the free-energy landscape of a large NP within semiflexible polymer brushes. The height of free-energy barrier and the depth of free-energy well depend mainly on the compression degree of two impenetrable surfaces and also rely on the size of NPs as well as the interaction between polymer brushes and NPs. However, for flexible polymer brushes, there only exists a free-energy well near the polymer brush surface during the compression process. This investigation may stimulate experimental interest in novel nano-materials.
Morphological Transformations of Vesicles with Confined Flexible Filaments.

Abstract:A fundamental understanding of cell shaping with confined flexible filaments, including microtubules, actin filaments, and engineered nanotubes, has been limited by the complex interplay between the cell membrane and encapsulated filaments. Here, combining theoretical modeling and molecular dynamics simulations, we investigate the packing of an open or closed filament inside a vesicle. Depending on the relative stiffness and size of the filament to the vesicle as well as the osmotic pressure, the vesicle could evolve from an axisymmetric configuration to a general configuration with a maximum of three reflection planes, and the filament could bend in or out of plane or even coil up. A plethora of system morphologies are determined. Morphological phase diagrams predicting conditions of shape and symmetry transitions are established. Organization of actin filaments or bundles, microtubules, and nanotube rings inside vesicles, liposomes, or cells are discussed. Our results provide a theoretical basis to understand cell shaping and cellular stability and to help guide the development and design of artificial cells and biohybrid microrobots.

Abstract:Employing the discrete element method with a multi-sphere model, this paper presents a numerical analysis of the packing structure of fibers during the preparation of the fiber porous blank, in which effects of such shape parameters of fibers as the aspect ratio and the curl index have been taken into account. For ten kinds of straight fibers and eight kinds of curved fibers with different aspect ratios and curl indexes, the microstructure of fiber packing formed by gravity settling is quantified in terms of the packing porosity, coordination number, contact orientation and fiber orientation. The results show that a larger packing porosity can be realized by increasing the aspect ratio or decreasing the curl index. Similarly, an individual fiber with a large aspect ratio in the packing structure can have more lap joints by increasing the curl index. Meanwhile, the phenomenon that curved fiber can fit better in an interstice than straight fibers is the main cause for the sharp increase of the mean coordination number when the shape of fiber changes from straight to curved. With regard to straight fibers, a preferable contact angle of 15° is confirmed almost for all cases while the intersection angle is in the vicinity of 50° on the whole. Correspondingly, the curvature has a significant influence on the heterogeneity of the contact orientation, and a strong tendency of horizontal alignment can be found for both straight and curved fibers. It is noted that the predictions of the packing porosity for straight fibers in this work are comparable to available experimental data and simulations. Those finds would be useful for a better understanding of the packing structure of fibers at the particle scale.
Determination of the Packing Model of a Supramolecular Nanofiber Via Mass-Per-length Measurement and De Novo Simulation

Abstract:Herein, we report an example of using scanning transmission electron microscopy to determine the mass-per-length of supramolecular nanofibers. Together with the measurement of the diameter of nanofibers via transmission electron microscopy, we could estimate the packing density of assembling molecules along the nanofibers. A parallel unbiased de novo simulation screens and reveals the most plausible molecular packing pattern of small molecules along the supramolecular nanofibers. Remarkably, the simulated packing patterns and density correlate well with the experimental measurements. Unexpectedly, the naphthalene groups are likely facing outward, creating a hydrophobic surface, which is driven by the geometry of the hydrogelator molecule. Overall, this study establishes a complementary method to determine molecular arrangement along the supramolecular nanofibers, which is potentially useful for the guidance of rational design of biomaterials based on self-assembly.

Abstract:This article examines the mechanical response of flexible fiber packings subject to triaxial compression. Short fibers yield in a manner similar to typical granular materials in which the deviatoric stress remains nearly constant with increasing strain after reaching a peak value. Interestingly, long fibers exhibit a hardening behavior, where the stress increases rapidly with increasing strain at large strains and the packing density continuously increases. Phase diagrams for classifying the bulk mechanical response as yielding, hardening, or a transition regime are generated as a function of the fiber aspect ratio and fiber-fiber friction coefficient. Large fiber aspect ratio and large fiber-fiber friction coefficient promote hardening behavior. The positions of boundaries between different regimes depend on the confining pressure and fiber flexibility. The hardening packings can support much larger loads than the yielding packings, but larger internal axial forces within fibers and larger fiber-fiber contact forces occur.
Modification of carbon nanotubes using poly(vinylidene fluoride) with assistance of supercritical carbon dioxide: the impact of solvent

Abstract:We report herein a typical piezoelectric polymer, poly(vinylidene fluoride) (PVDF) to be successfully wrapped on single-walled carbon nanotubes (SWCNTs) using a simple supercritical carbon dioxide (SC CO(2)) antisolvent-induced polymer epitaxy method. Our study focused on the effect of different solvents on the morphology of PVDF wrapping on SWCNTs. Three organic solvents, dimethyl sulfoxide (DMSO), N,N-dimethylformamide (DMF), and N,N-dimethylacetamide (DMAc) were chosen for PVDF. When DMSO was used as solvent, the decorating degree of PVDF on the surface of SWCNTs increases significantly with the increase of SC CO(2) pressure, and nanocrystals wrapping on SWCNTs can be observed at high pressure. FTIR and Raman spectra indicated that there exist interactions between SWCNTs and PVDF chains. What's more, FTIR results also show that there exists a transformation from the beta-phase to the alpha-phase of PVDF in DMSO with the assistance of SC CO(2), which is similar to the action of elongation/shear flow field. It indicated that the alpha-phase is the predominant form occurring on the surface of SWCNTs after treatment with SC CO(2). And the helical structure on SWCNTs observed from the TEM image reflected the alternate trans- and gauche-bond conformation of the alpha-form. When DMF or DMAc was used as the solvent, although nanocrystal wrapping and helical structure was not visible, the samples had more excellent dispersion than that in DMSO. Particularly, for DMF, a typical network structure was observed, which is similar to a spider web. Therefore, this work supplies a clue that the various morphologies of nanohybrid structure can be obtained just by changing the solvent during the treatment process of SC CO(2), and accordingly, the tailored nanohybrid structure are promising and important for functional design as a basic component in microfabrication and other fields.

Abstract:In this work, we report an approach to prepare segmented polymer nanofibers (SPNFs) composed of rodlike subunits by kinetically controlled self-assembly of polystyrene-b-poly(4-vinylpyridine)-based supramolecules in microfluidic chips. The length and morphology of the SPNFs could be effectively adjusted by changing the total flow rate (Vtotal) and the molar ratio (x) of 4-vinylpyridine (4VP) unit to a hydrogen-bonding molecule, 3-n-pentadecyphenol. Moreover, the subunits of SPNFs could transform from short rods to spheres when the interfacial tension between PS core and solvent increased. On the contrary, the SPNFs elongated along the major axis when the interfacial tension decreased. Abstract:To understand the mechanics of cellular/intracellular packing of one-dimensional nanomaterials, we performed theoretical analysis and molecular dynamics simulations to investigate how the morphology and mechanical behaviors of a lipid vesicle are regulated by encapsulated rigid nanorods of finite and non-uniform diameters, including a cylindrical rod, a rod with widened ends, a cone-shaped rod, and a screw-driver-shaped rod. As the rod length increases, the vesicle evolves from a sphere into different shapes, such as a lemon, a conga drum, a cherry, a bowling pin, or a tubular shape for long and thick rods. The contact between the vesicle protrusion and the rod plays an important role in regulating the vesicle tubulation, membrane tension, and axial contact force on the rod. Our analysis provides a theoretical basis to understand a wide range of experiments on morphological transitions that occur in cellular packing of actin or microtubule bundles, mitotic cell division, and intracellular packing of carbon nanotubes.

Abstract:We investigated the co-assembly of nanoparticles P and amphiphilic diblock copolymers AB in selective solvents using a dissipative particle dynamics (DPD) method. By controlling the nanoparticle concentration and the interaction parameter between the hydrophobic blocks and the solvents, we found that the aggregation morphology can be changed from rod-like micelles to disk-like micelles and further to vesicles. The ratio of the hydrophobic/hydrophilic block and the nanoparticle concentration largely affects the structural characteristics of vesicles and the dispersion of nanoparticles. Copolymers with a longer hydrophobic block length are more likely to form vesicles with a smaller aqueous cavity size and vesicle size as well as a thicker wall. At the same time, the nanoparticles in the hydrophobic membrane tend to locate closer to the center of the vesicle and they become more compactly organized. A significant discovery has found that the larger the nanoparticle concentration, the smaller the aqueous cavity and the larger the vesicle size. We can also locate the nanoparticles at the center of spherical micelles or the hydrophobic membranes of vesicles by varying the nanoparticle concentration. This provides an effective and simple method to prepare size-controlled vesicles containing nanoparticles, project the localization of nanoparticles within the vesicles, and even tune the distance between the nanoparticles.

Abstract:A fundamental understanding of cell-nanomaterial interaction is of essential importance to nanomedicine and safe applications of nanotechnology. Here we investigate the adhesive wrapping of a soft elastic vesicle by a lipid membrane. We show that there exist a maximum of five distinct wrapping phases based on the stability of full wrapping, partial wrapping, and no wrapping states. The wrapping phases depend on the vesicle size, adhesion energy, surface tension of membrane, and bending rigidity ratio between vesicle and membrane. These results are of immediate interest to the study of vesicular transport and endocytosis or phagocytosis of elastic particles into cells.
Tailoring the Morphologies of Pvdf Nanofibers by Interfacial Diffusion During Coaxial Electrospinning

Abstract:The morphology regulation of nanofiber (NF) plays an important role in its applications. In the present study, a simple method to tailor the cross-section morphology of polyvinylidene fluoride (PVDF) NFs by controlling the interfacial diffusion during coaxial electrospinning is presented. By choosing a soft bad-solvent of PVDF to prepare a partially miscible inner solution, the regulation of the interfacial diffusion can be simplified into the adjustment of the viscosity difference between the inner and outer solutions. When the viscosity of the inner solution surpassed that of the outer solution, the diffusion direction changed from outer-to-inner to inner-to-outer, which leaded to the change in the cross-section morphology of resultant NFs from a net-like to a hollow structure. Water contact angle also showed the effect caused by the interfacial diffusion. BET surface area of the as-prepared porous PVDF NFs can reach up to 42m2/g, which is almost the highest data ever reported in the literature for polymer nanofiber.

Light-Gated Nano-Porous Capsules from Stereoisomer-Directed Self-Assemblies

Abstract:Structuring pores into stable membrane and controlling their opening is extremely useful for applications that require nanopores as channels for material exchange and transportation. In this work, nanoporous vesicles with aggregation-induced emission (AIE) properties were developed from the amphiphilic polymer PEG550-TPE-Chol, in which the hydrophobic part is composed of a tetraphenylethene (TPE) group and a cholesterol moiety and the hydrophilic block is a poly(ethylene glycol) (PEG, Mn = 550 Da). Two stereoisomers, trans-PEG550-TPE-Chol and cis-PEG550-TPE-Chol, were successfully synthesized. These thermally stable stereoisomers showed distinct self-assembly behavior in water: trans-PEG550-TPE-Chol formed classical vesicles, while cis-PEG550-TPE-Chol self-assembled into cylindrical micelles. Interestingly, trans/cis mixtures of PEG550-TPE-Chol (trans/cis = 60/40), either naturally synthesized without isomers' separation during the synthesis or intentionally mixed using trans- and cis-isomers, constructed perforated vesicles with nanopores. Moreover, under the illumination of high intensity UV light (365 nm, 15 mW/cm2), the classical vesicles of trans-PEG550-TPE-Chol were perforated by its cis counterparts generated from the trans–cis photoisomerization, while the cylindrical micelles of cis-PEG550-TPE-Chol interweaved to form meshes and nanoporous membranes due to the trans-isomers produced by cis–trans photoisomerization. All of these assemblies in water emitted bright cyan fluorescence under UV light, while their constituent molecules were not fluorescent when solubilized in organic solvent. Characterization of glycosylated gelatin/pullulan nanofibers fabricated by multi-fluid mixing solution blow spinning

Abstract:In this work, multi-fluid mixing solution blow spinning was applied to develop gelatin/pullulan composite nanofibers, and then the nanofibers were glycated to enhance the physical properties. The results show that the grafting degree of the nanofibers increased significantly from 17.5 % to 36.0 % as the glycation time increased, and the morphology results indicated that 72 h of glycation did not destroy the structure of the nanofibers. FTIR results show that the glycation consumed the the–NH2 groups, cleaved sugar units of polysaccharide, and affected the secondary structure of the protein. The glycation enhanced the thermal stability and improved the rigidity of the nanofibers. Besides, after 120 h of glycation, the water contact angle of nanofibers increased from 0° to 79.1°, and the water vapor transmission rates decreased from 12.49 to 8.97 g mm/m2 h kPa, indicating the enhanced hydrophobicity and barrier properties. In addition, the glycation improved the water stability of the nanofibers, which increased the applicability of the gelatin/pullulan nanofibers in food packaging. The present work provides a green and efficient method for improving the physical properties of gelatin/pullulan nanofibers.

Abstract:In this study, zanthoxylum bungeanum essential oil/β-cyclodextrin inclusion complexes (ZBEO/β-CD-ICs) were first prepared by precipitation method. When the addition of ZBEO was 1 g, the reaction time was 4 h and the reaction temperature was 55 °C, the recovery (73.88%) and loading content (9.53%) reached the highest value. The characterization results showed inclusion complexation changed the crystalline structure, enhanced interaction among molecules and increased the thermal stability. Then, nanofiber films containing ZBEO/β-CD-ICs were prepared by electrospinning. When the total polymer concentration was constant at 20%, with the increase of ZBEO/β-CD-IC content, the diameter of nanofiber and mechanical strength decreased, but the temperature corresponding to the maximum rate of weight loss increased. X-ray diffraction analysis proved that the addition of ZBEO/β-CD-IC increased the crystallinity degree of film. The Fourier transform infrared spectra indicated hydrogen bond interactions among molecules. Releasing behavior of ZBEO indicated that increase of temperature and relative humidity accelerated the releasing speed. Antibacterial and antioxidant activity results demonstrated the increase of ZBEO content enhanced antibacterial and antioxidant efficiency, Z40P10 nanofibers had the maximum antibacterial rate of 62.02% against S. aureus and the maximum antioxidant activity of 60.18%.
