Abstract:We report a novel design, based on a combination of lanthanide-doped upconversion nanoparticles and manganese dioxide nanosheets, for rapid, selective detection of glutathione in aqueous solutions and living cells. In this approach, manganese dioxide (MnO(2)) nanosheets formed on the surface of nanoparticles serve as an efficient quencher for upconverted luminescence. The luminescence can be turned on by introducing glutathione that reduces MnO(2) into Mn(2+). The ability to monitor the glutathione concentration intracellularly may enable rational design of a convenient platform for targeted drug and gene delivery.

Abstract:Schematic diagram of GSH fluorescence sensing and E. coli O157:H7 imaging based on SQDs‐MnO2 nanoprobe system. Glutathione (GSH) is one of the most abundant bioethanol antioxidants in living cells. Here, a fluorescent probe based on MnO2 nanosheets and sulfur quantum dots (SQDs) was fabricated. Because of the synergistic effect of IFE and FRET, the fluorescence from SQDs was quenched by MnO2 nanosheets. In the presence of GSH, the fluorescence of SQDs could be recovered because of the reduction of MnO2 nanosheets by GSH. The method can detect GSH in the concentration range of 5 ~ 1000 μM with the detection limit as low as 1.26 μM. This quick, easy, and cost‐effective sensor could be used for the quantification of GSH in serum samples and the imaging of GSH in Escherichia coli O157:H7.

Abstract:A ratiometric fluorescence assay for glutathione (GSH) was developed. The novel assay is based on a nanoprobe composed of manganese dioxide nanosheets (MnO2 NS) and dual-emission carbon dots (de-CDs) with intrinsic GSH-response property. After construction of the nanoprobe, two emission peaks of de-CDs were suppressed to varying degrees by MnO2 NS. The suppression was relieved and the two emission peaks recovered proportionally when MnO2 NS was decomposed by GSH, thus realizing the ratiometric assay for micromolar GSH. The intrinsic responsiveness of de-CDs to millimolar GSH broadens the analytical range of the nanoprobe. An appropriate precursor, calcon-carboxylic acid, was screened out to synthesize de-CDs via one-step hydrothermal treatment. The de-CD@MnO2 NS nanoprobe can measure GSH concentrations through the fluorescence intensity ratio between 435 and 516 nm excited at 365 nm. The range of response was from 1 μM to 10 mM and the detection limit reached 0.6 μM (3σ criterion). Benefiting from its good biocompatibility, the proposed nanoprobe has excellent applicability for intracellular GSH imaging.

Abstract:Near infrared (NIR) emitting semiconductor quantum dots can be excellent fluorescent nanoprobes, but the poor biodegradability and potential toxicity limits their application. The authors describe a fluorescent system composed of graphene quantum dots (GQDs) as NIR emitters, and novel MnO2 nanoflowers as the fluorescence quenchers. The system is shown to be an activatable and biodegradable fluorescent nanoprobe for the “turn-on” detection of intracellular glutathione (GSH). The MnO2-GQDs nanoprobe is obtained by adsorbing GQDs onto the surface of MnO2 nanoflowers through electrostatic interaction. This results in the quenching of the NIR fluorescence of the GQDs. In the presence of GSH, the MnO2-GQDs nanoprobe is degraded and releases Mn2+ and free GQDs, respectively. This gives rise to increased fluorescence. The nanoprobe displays high sensitivity to GSH and with a 2.8 μM detection limit. It integrates the advantages of NIR fluorescence and biodegradability, selectivity, biocompatibility and membrane permeability. All this makes it a promising fluorescent nanoprobe for GSH and for cellular imaging of GSH as shown here for the case of MCF-7 cancer cells.Graphical abstractA biodegradable NIR fluorescence nanoprobe (MnO2-GQDs) for the “turn-on” detection of GSH in living cell was established, with the NIR GQD as the fluorescence reporter and the MnO2 nanoflower as the fluorescence quencher.

Abstract:Persistent luminescence nanoparticles (PLNPs) hold great promise for the detection and imaging of biomolecules. Herein, we have demonstrated a novel nanoprobe, based on the manganese dioxide (MnO2)-modified PLNPs, that can detect and image glutathione in living cells and in vivo. The persistent luminescence of the PLNPs can be efficiently quenched by the MnO2 nanosheets. In the presence of glutathione (GSH), MnO2 was reduced to Mn2+ and the luminescence of PLNPs can be restored. The persistent luminescence property can allow detection and imaging without external excitation and avoid the background noise originating from the in situ excitation. This strategy can offer a promising platform for detection and imaging of reactive species in living cells or in vivo.
A Fluorescence Sensing Platform with the MnO2 Nanosheets As an Effective Oxidant for Glutathione Detection

Abstract:Herein, a sensitive, selective, rapid and cost-effective fluorescence sensing platform has been designed for the detection of glutathione (GSH). In this platform, the manganese dioxide (Mn0(2))nanosheets served as both oxidant, which could effectively oxidize o-phenylenediamine (OPDA) to produce fluorescence for highly sensitive detection, and recognition element. When GSH was introduced, the Mn02 nanosheets were reduced to Mn2+, which inhibited the formation of fluorescent oxidation products of OPDA. Under optimal conditions, the Mn0(2)-OPDA sensing platform showed very sensitive response to GSH in the range of 10 nM-40 mu M. The detection limit of GSH reached as low as 10 nM. The high selectivity of inhibition of fluorescent product formation by GSH allows this platform to be used for detecting GSH in practical biological samples (such as cell extracts). Abstract:We synthesized six 1-oxo-1H-phenalene-2,3-dicarbonitrile (OPD)-based probes with various leaving groups using an arylthioether linker and for the first time identified the probe O-NH2 capable of highly selective detection of glutathione over cysteine/homocysteine in vitro and in vivo based on an aromatic nuclear substitution reaction (SNAr) mechanism. The fluorescence of the probe O-NH2 was quenched because of the photoinduced electron transfer (PET) process, but switched on by a glutathione-triggered specific recognition reaction between the probe O-NH2 and glutathione. The recognition mechanism for glutathione was explored and verified by theoretical calculations and ESI-MS analysis. Using O-NH2 as the probe, the GSH fluorescence images were demonstrated in HeLa cells and the intracellular GSH levels in different imatinib-resistant K562 tumor cells were firstly determined. Further, O-NH2 was utilized to detect glutathione in D. magna and zebrafish embryos. The combined results indicate that O-NH2 can be applied as an effective tool for detecting glutathione in biological investigations.

Abstract:Glutathione (GSH) plays a crucial role in several physiological processes, including anti-oxidation and heavy metal detoxification. GSH is produced endogenously in the human body and can also be obtained through diet. The development of fast, highly sensitive, and multi-application fluorescent probes remains a challenging task. In this study, we have designed and synthesized a coumarin-based fluorescent probe (NFRF) for the sensitive and rapid detection of GSH in 100 % aqueous solution. By loading probe NFRF on the filter paper, the real-time visual detection of GSH is achieved in both daylight and fluorescence modes, providing a convenient, economical and rapid on-site detection tool. Probe NFRF could be used for the detection of GSH in real samples, with recoveries rates of 81.74 %-115.12 %. Notably, the probe imaged changes in GSH concentrations in oxidative stress environments and during ferroptosis. This work provides a prospective method for GSH detection in food and complex biological systems.

Abstract:Glutathione (GSH) serves vital cellular biological functions, and its abnormal levels are associated with many diseases. To better understand its physiological and pathological functions, efficient methods for monitoring of GSH in living systems are desired. Although quite a few small molecule-based and nanomaterial-based one photon fluorescence probes have been reported for GSH, two-photon (TP) probes, especially nanoprobes with good membrane permeability, are more favorable for bioimaging applications, since TP fluorescence imaging can provide improved spatial localization and increased imaging depth. In this work, we for the first time reported a "turn-on" TP fluorescence nanoprobe for efficient detection of GSH in aqueous solutions and TP excited fluorescence imaging of GSH in living cells and tissues. The nanoprobe consists of two-photon mesoporous silica nanoparticles (TP-MSNs) with a large TP excitation action cross-section (Φδ) value of 103 GM and MnO2 nanosheets, which show intense and broad optical absorption and could act as efficient quenchers for TP fluorescence. In the sensing system, the negatively charged MnO2 nanosheets are adsorbed on the positively charged MSNs through electrostatic interaction, resulting in efficient quenching of their fluorescence, with very low background fluorescence observed. The addition of GSH could reduce MnO2 into Mn(2+), lead to the decomposition of the MnO2 nanosheets, and thereby result in remarkable enhancement of both one photon and TP excited fluorescence of the nanosystem. The nanoprobe shows a highly sensitive response to GSH in aqueous solutions, with a detection limit of 200 nM achieved. It also exhibits a high selectivity toward GSH relative to other biomolecules and electrolytes, with good membrane permeability and excellent biocompatibility. The nanoprobe was successfully applied in monitoring the change of the intracellular GSH in living cells and tissues via TP fluorescence imaging, demonstrating its value of practical application in biological systems.

Abstract:A novel turn-on red fluorescent BODIPY-based probe (Probe 1) for the detection of glutathione was developed. Such a probe carries a para-dinitrophenoxy benzyl pyridinium moiety at the meso position of a BODIPY dye as self-immolative linker. Probe 1 responds selectively to glutathione with the detection limit of 109nM over other amino acids, common metal ions, reactive oxygen species, reactive nitrogen species, and reactive sulfur species. A novel electrostatic interaction to modulate the SNAr attack of glutathione was believed to play significant role for the observed selective response to glutathione. The cleavage of dinitrophenyl ether by glutathione leads to the production of para-hydroxybenzyl moiety which is able to self-immolate through an intramolecular 1,4-elimination reaction to release the fluorescent BODIPY dye. The low toxic probe has been successfully used to detect mitochondrial glutathione in living cells.
MnO 2 Nanospheres Assisted by Cysteine Combined with MnO 2 Nanosheets as a Fluorescence Resonance Energy Transfer System for “Switch-on” Detection of Glutathione

Abstract:Manganese dioxide nanosheets combined with cysteine-assisted emitting manganese dioxide nanospheres (Cys-MnO2 nanospheres) is fabricated for the first time as an "off–on" fluorescence detection platform for glutathione (GSH). In this sensing system, Cys-MnO2 nanospheres served as energy donors, while MnO2 nanosheets were used as both energy acceptors and recognition units. MnO2 nanosheets can effectively quench the fluorescence of Cys-MnO2 nanospheres through the fluorescence resonance energy transfer (FRET). The addition of GSH could reduce MnO2 nanosheets into Mn2+, disrupting the FRET process and restoring the fluorescence of Cys-MnO2 nanospheres. Under the optimum conditions, the "switch-on" platform we established has a wide response to GSH with a range of 5–50 μM and 150–800 μM, as well as a superior specificity. Importantly, all components of the sensor are nontoxic, biocompatible, easily prepared, and have a high utilization of raw materials. Abstract:A novel magnetic/fluorometric bimodal sensor was built from carbon dots (CDs) and MnO2. The resulting sensor was sensitive to glutathione (GSH), leading to apparent enhancement of magnetic resonance (MR) and fluorescence signals along with visual changes. The bimodal detection strategy is based on the decomposition of the CDs-MnO2 through a redox reaction between GSH and MnO2. This process causes the transformation from non-MR-active MnO2 to MR-active Mn(2+), and is accompanied by fluorescence restoration of CDs. Compared with a range of other CDs, the polyethylenimine (PEI) passivated CDs (denoted as pCDs) were suitable for detection due to their positive surface potential. Cross-validation between MR and fluorescence provided detailed information regarding the MnO2 reduction process, and revealed the three distinct stages of the redox process. Thus, the design of a CD-based sensor for the magnetic/fluorometric bimodal detection of GSH was emphasized for the first time. This platform showed a detection limit of 0.6 μM with a linear range of 1-200 μM in the fluorescence mode, while the MR mode exhibited a linear range of 5-200 μM and a GSH detection limit of 2.8 μM with a visible change being observed rapidly at 1 μM in the MR images. Furthermore, the introduction of the MR mode allowed the biothiols to be easily identified. The integration of CD fluorescence with an MR response was demonstrated to be promising for providing detailed information and discriminating power, and therefore extend the application of CDs in sensing and imaging.

Abstract:Glutathione is a major endogenous antioxidant in biological systems. As a result, highly selective fluorescent probes are significant components of studies aimed at gaining a better understanding of the role(s) glutathione plays in biological functions. In this study, we developed a dual-channel turn-on fluorescent probe for glutathione, comprising naphthalimide and cyanine, bridged by a thiol-reactive sulfonamide moiety. The probe selectively responds to glutathione and is not affected by cysteine and homocysteine. Important information has been gained about the chemical mechanism for operation of the probe, which responds to glutathione in visible and near-infrared channels synchronously and independently. Moreover, this probe exhibits the remarkable capability of targeting mitochondria and dual-channel tracking of intracellular glutathione in a spatiotemporal and synchronous manner. We believe that the strategy used to design the new dual-channel fluorescence-on probe will be applicable to the design of fluorescent probes that display specific responses in multiple channels.
MnO2 nanoparticles decorated with Ag/Au nanotags for label-based SERS determination of cellular glutathione

Abstract:A novel stimulus-responsive surface-enhanced Raman scattering (SERS) nanoprobe has been developed for sensitive glutathione (GSH) detection based on manganese dioxide (MnO2) core and silver/gold nanoparticles (Ag/Au NPs). The MnO2 core is not only capable to act as a scaffold to amplify the SERS signal via producing "hot spots", but also can be degraded in the presence of the target and thus greatly enhance the nanoprobe sensitivity for sensing of GSH. This approach enables a wide linear range from 1 to 100 µM with a 2.95 µM (3σ/m) detection limit. Moreover, the developed SERS nanoprobe represents great possibility in both sensitive detection of intracellular GSH and even can monitor the change of intracellular GSH level when the stimulant occurs. This sensing system not merely offers a novel strategy for sensitive sensing of GSH, but also provides a new avenue for other biomolecules detection.

Abstract:Herein, a bovine serum albumin-stabilized gold clusters-MnO2 nanocomposite (BSA@AuNCs-MnO2) was constructed. Manganese dioxide (MnO2) was generated in situ on the gold clusters (BSA@AuNCs) based on the redox reaction between bovine serum albumin (BSA) and potassium permanganate (KMnO4). The fluorescence of BSA@AuNCs can be quenched by the in-situ grown MnO2, which has strong light absorption ability. It is worth noting that the quenched fluorescence of the BSA@AuNCs can be restored in the presence of glutathione (GSH), and MnO2 was reduced to Mn2+ in return. Encouragingly, 1 mu M GSH can cause a detectable fluorescence change. This sensitivity is comparable to other nanomaterials based fluorescent probes. Furthermore, this nanocomposite has obvious superiorities, such as good uniformity, simple preparation and mild reaction. The nanocomposite also has good stability and specificity, which can be further used for visualizing of intracellular GSH.
A Turn-on MOF-based Luminescent Sensor for Highly Selective Detection of Glutathione

Abstract:Glutathione (GSH), as a key regulator for the redox balance of biological processes, was detected with a metal-organic framework (MOF) (UiO-67-sbdc) fluorescence sensor. The fluorescence of UiO-67-sbdc could be effectively enhanced by GSH via an electronic effect between GSH and the ligand of MOF, thus achieving the recognition of GSH. It is the first attempt to design a MOF-based fluorescent sensor to distinguish GSH from cysteine (Cys). This sensor exhibited short response time (< 120 s), suitable linear range, and high selectivity towards GSH. Further research conducted in serum also showed a quantitative and selective detection of GSH. Thus, the reported UiO-67-sbdc in this work has its application prospect in clinically judging antioxidant capacity of the body.

Abstract:Glutathione (GSH) is a crucial non-protein thiol and an indispensable endogenous antioxidant. The aberrant expression of GSH in plasma and cytosol is closely related to numerous diseases, including cancer. Therefore, establishing a sensitive method for analyzing GSH has important application value for biomedical research and clinical medical detection. Herein, A method for the rapid and simple detection of GSH was proposed, which is based on an anti-etching mechanism by utilizing gold nanotetrapods (Au NTPs) and manganese dioxide nanosheets (MnO 2 NSs). In the absence of GSH, Au NTPs solution can cause a distinct color change from gray-green to red through the etching effect of MnO 2 NSs. However, in the presence of GSH, the redox reaction between GSH and MnO 2 NSs inhibits the etching of Au NTPs by MnO 2 NSs, and Au NTPs solution maintains persistent gray-green color. The colorimetric probe exhibited excellent selectivity for GSH. The limits of detection for GSH were 43.5 nM (UV–vis spectrum) and 0.25 μM (naked eyes). The sensing technique exhibited excellent linearity between wavelength shift and GSH concentration within the range of 0.25 μM–1.5 μM. The outcomes of GSH detection in actual biological samples demonstrate that this probe has the potential to be applied to GSH detection in intricate biological samples.

Dual sensing of glutathione and acidic pH values by using MnO 2 nanosheets and 3-acetyl-7-hydroxy-2H-chromen-2-one as a fluorescent pH probe

Abstract:A multifunctional nanoprobe is described for dual sensing of acidic pH values and glutathione (GSH) by combining the pH-responsive fluorescent probe 3-acetyl-7-hydroxy-2H-chromen-2-one (AHC) and MnO 2 nanosheets. The fluorescence of the MnO 2 /AHC composite is weak due to an inner filter effect. If, however, the MnO 2 nanosheets are reductively decomposed by GSH, the blue fluorescence of the pH probe AHC (with excitation/emission maximum at 417/456 nm) will be restored. The MnO 2 nanosheets also are decomposed by acidic pH values, and the fluorescence of AHC is decreased. According to absorbance and fluorescence signal changes, the pH and GSH induced responses can be easily distinguished. Thus, the nanoprobe can be used for logical analysis of acidic pH values and GSH. The nanoprobe works in the pH range from 4 to 7, and GSH can be determined in the concentration range from 0.5 to 200 μM.
