Abstract:An efficient and highly regioselective synthesis of 2-amino-3-arylindoles via the BF3-promoted reaction of 3-diazoindolin-2-imines with aryl aldehydes is described. This reaction could be carried out under mild reaction conditions with a broad scope of substrates. Moreover, the operation is of practical usage and could be scaled up.

Abstract:An efficient [bis-(trifluoroacetoxy)iodo]benzene (PIFA)-mediated oxidative C-N bond formation is developed for the synthesis of 1H-indazoles from readily available arylhydrazones. The reaction tolerates a wide range of functional groups and has broad scope of substrates. Moreover, this method is a relative green and reliable method for rapid preparation of substituted 1H-indazoles under mild conditions.

Abstract:A novel [3+3] annulation reaction of 2‐alkenylindoles with hydrazonyl chlorides in the presence of base for the formation of pyridazino[4,5‐b]indoles was developed. This approach provided a convenient and efficient way to synthesize a series of functionalized pyridazino[4,5‐b]indoles derivatives. The method exhibited a broad substrate scope and provided the corresponding pyridazino[4,5‐b]indoles products with excellent yields.

Abstract:A novel method has been developed to synthesize a unique class of highly functionalized isochromeno[4,3-c]pyridazines. This reaction features an intermolecular functionalization of terminal nitrogen atom of diazo group of 4-diazoisochoman-3-imine with two dimethylsulfonium ylide components, followed by a base promoted 6-exo-trig cyclization step. Readily available starting materials, a broad substrate scope, and operationally simple, mild, and catalyst-free reaction conditions are the prominent features of this method.

Abstract:An unprecedented silver-promoted regioselective (4 + 1) annulation of isocyanoacetates with pyridinium salts is reported. The established protocol provides controlled, facile, and modular access to a range of synthetically useful N-fused heterocyclic scaffolds containing indolizines, pyrrolo[1,2-a]quinolines, pyrrolo[2,1-a]isoquinolines, and 1H-imidazo[4,5-a]indolizin-2(3H)-ones. A mechanistic pathway involving nucleophilic addition/protonation/elimination/cycloisomerization is proposed.

Abstract:A protocol based on a newly developed N-bromosuccinimide (NBS)-induced cycloisomerization was described to prepare tricyclic azepino[4,5-b]indoles from simple β-enaminoesters or β-enaminones containing an indole unit. A mechanism involving a Pictet-Spengler cyclization, an aziridine ring formation, and a regioselective C-N bond cleavage was proposed to account for the medium-sized ring formation and the migration of electron-withdrawing group (ester, ketone).

Abstract:An efficient, direct, and novel Lewis acid-catalyzed regioselective C-H carboxamidation of indolizines with dioxazolones via an acyl nitrene type rearrangement under metal-free conditions has been documented. A diverse array of indolizine-3-carboxamides were achieved in moderate to good yields with wide substrate scope. In these transformations, isocyanatobenzene was formed by the ring opening of dioxazolones and a subsequent Curtius-type rearrangement, which could be harnessed in C-H carboxamidation of N-heterocycles. The present protocol is satisfactorily complementary to nitrene transfer chemistry and C-H functionalization of N-heterocycles. Furthermore, photophysical experiments revealed that a few compounds exhibited high fluorescence absorption and emission intensity.

Abstract:The chiral pyrrolo[1,2-a]indole represents a privileged stuctural motif in many natual products and pharmaceutical agents. Herein we developed an efficient [3+2] annulation of 2-nitrovinylindoles with azlactones via the cascade Michael addition and intramolecular acylation under the catalysis of bifunctioanl tertiary amine in combination with DABCO, delivering a wide range of pyrrolo[1,2-a]indoles in good yields with high diastereo- and enantioselectivities. Alternatively, 7-nitrovinylindoles are served as 4C-synthons to perform the enantioselective [4+2] annulation under the identical condition, smoothly giving the pyrrolo[3,2,1-ij]quinoline skeleton with good to high stereoselectivities. Furthermore, a novel enantio-enriched indole-based tetracyclic skeleton was facilely derived from the obtained pyrrolo[1,2-a]indoles.

Abstract:An unexpected isomerization of azomethine ylides generated in situ from isatin with indoline-2-carboxylic acid has been disclosed, providing direct access to N -functionalized indole scaffolds. This protocol has good functional group tolerance and provides various 3-(1 H -indol-1-yl)indolin-2-one derivatives in moderate to high yields simply by using alcohol as the solvent, with no additional additive being required.

Abstract:A green and efficient protocol for the preparation of benzo[a]carbazoles via visible-light-promoted and electron donor-acceptor (EDA) complex-driven intermolecular cyclization of 2-arylindoles with Z-α-bromocinnamaldehydes in the absence of external photocatalysts, transition metals, and oxidants was reported. This new approach demonstrates an intermolecular cyclization model using indole derivatives as electron donors under visible light. Mechanistic investigations have showed that 2-arylindoles with Z-α-bromocinnamaldehydes form EDA complexes, which undergo sequential single-electron transfer, radical coupling, 6π-electrocyclization, and dehydroaromatization to generate benzo[a]carbazoles under visible light irradiation. The current photochemical method features readily accessible starting materials, mild conditions, simple operation, and a broad substrate scope.

Abstract:A novel copper-catalyzed tandem cyclization/direct C(sp2)-H annulation of phenyl azide-ynamides via α-imino copper carbenes has been developed, which provides a concise and flexible approach for the construction of a range of valuable azepino[2,3-b:4,5-b']diindoles in mostly good to excellent yields with high chemoselectivities. This protocol enables the one-pot construction of N-fused benzimidazo-β-carbolines in good yields.
