Abstract:The present study reports the fabrication of medicated nanofibers for potential colon-targeted drug delivery using modified coaxial electrospinning, in which salt NaCl solutions were exploited as shell fluids to facilitate the preparation processes. A homemade concentric spinneret with an indented core capillary was developed to conduct the coaxial processes. Optical observations and scanning electron microscopic results demonstrated that the shell-to-core fluid flow rate ratio was a key parameter, which exerted a significant influence on the electrospinning processes and could be exploited to control the fibers’ morphology and diameters. A scaling law of D=0.173F-0.531R2=0.9976 was built, by which the nanofibers’ sizes can be predicted and manipulated easily. X-ray diffraction and attenuated total reflected FTIR tests verified that the medicated nanofibers were essentially a polymeric nanocomposite and the guest drug diclofenac sodium DS had fine compatibility with the host polymer. All the drug was encapsulated in the filament-forming carrier. In vitro dissolution experiments demonstrated that the medicated nanofibers could free the drug in a neutral condition, suggesting potential colon-targeted drug delivery applications. Ex vivo tests demonstrated that the medicated fiber mats could enhance the transmembrane of DS. Based on coaxial electrospinning, a new strategy is successfully developed for creating medicated nanomaterials.

Abstract:A major problem related to the fabrication of drug-loaded fibers by wet spinning is the significant loss of load when the drugs and spinning dopes are directly mixed together. We report a novel method to fabricate drug-loaded alginate fibers by spinning an aqueous mixture containing alginate and alginate nanocapsules incorporated with drugs. The alginate fibers exhibit spotty features of nanocapsules located close to the surface of the fibers. Adsorption experiments demonstrate the superabsorbent properties of the fibers, as well as the dependence of adsorption profiles on salt ionic strength. The adsorption processes of all the fibers are well-described by the pseudo-second-order kinetics model. Drug release tests show that the cumulative release amount increased with an increase in the proportion of nanocapsules present in the fiber. In addition, the burst release behaviors of the fibers incorporated with the nanocapsules were also diminished dramatically. This work suggests that adding nanocapsules containing drugs into the spinning dopes is a promising strategy to fabricate novel drug-loaded fibers for immediate drug delivery for wound dressing.
Aqueous-Based Coaxial Electrospinning of Genetically Engineered Silk Elastin Core-Shell Nanofibers

Abstract:A nanofabrication method for the production of flexible core-shell structured silk elastin nanofibers is presented, based on an all-aqueous coaxial electrospinning process. In this process, silk fibroin (SF) and silk-elastin-like protein polymer (SELP), both in aqueous solution, with high and low viscosity, respectively, were used as the inner (core) and outer (shell) layers of the nanofibers. The electrospinnable SF core solution served as a spinning aid for the nonelectrospinnable SELP shell solution. Uniform nanofibers with average diameter from 301 ± 108 nm to 408 ± 150 nm were obtained through adjusting the processing parameters. The core-shell structures of the nanofibers were confirmed by fluorescence and electron microscopy. In order to modulate the mechanical properties and provide stability in water, the as-spun SF-SELP nanofiber mats were treated with methanol vapor to induce β-sheet physical crosslinks. FTIR confirmed the conversion of the secondary structure from a random coil to β-sheets after the methanol treatment. Tensile tests of SF-SELP core-shell structured nanofibers showed good flexibility with elongation at break of 5.20% ± 0.57%, compared with SF nanofibers with an elongation at break of 1.38% ± 0.22%. The SF-SELP core-shell structured nanofibers should provide useful options to explore in the field of biomaterials due to the improved flexibility of the fibrous mats and the presence of a dynamic SELP layer on the outer surface.
Fabrication and Characterization of Absorbent and Antibacterial Alginate Fibers Loaded with Sulfanilamide

Abstract:Absorbent alginate fibers were fabricated using wet spinning technique and a broad-spectrum antibacterial and anti-inflammatory agent sulfanilamide was loaded from co-dissolving solution. The drug entrapment in the fibers during the processing was confirmed by optical microscope, SEM and FTIR. The water absorbency, in vitro drug release and antibacterial activities were performed to evaluate the potential application of the sulfanilamide-loaded alginate fibers for wound dressing. The results indicated that sulfanilamide was successfully encapsulated into the alginate fibers, and this system was stable in terms of high loading content and effectiveness in release. The in vitro release experiment showed a sustained and controlled release pattern of the drug from the fibers. And in vitro activity substantiated the fact that sulfanilamide was delivered in its active state and exhibited better antibacterial activities in comparison with the pure alginate fibers. Therefore the present investigation indicated that the sulfanilamide-loaded alginate fibers have potential application as ideal wound dressing.

Abstract:Coaxial electrospinning is a robust technique for one-step encapsulation of fragile, water-soluble bioactive agents, including growth factors, DNA and even living organisms, into core–shell nanofibers. The coaxial electrospinning process eliminates the damaging effects due to direct contact of the agents with organic solvents or harsh conditions during emulsification. The shell layer serves as a barrier to prevent the premature release of the water-soluble core contents. By varying the structure and composition of the nanofibers, it is possible to precisely modulate the release of the encapsulated agents. Promising work has been done with coaxially electrospun non-woven mats integrated with bioactive agents for use in tissue engineering, in local delivery and in wound healing, etc. This paper reviews the origins of the coaxial electrospinning method, its updated status and potential future developments for controlled release of the class of fragile, water-soluble bioactive agents.
Fabrication and characterization of zein-based nanofibrous scaffolds by an electrospinning method.

Abstract:Electrospun zein membranes were prepared using DMF as solvent. By changing the solution concentration, the electrospinning voltage and the distance between the spinneret and collector, nanofibrous meshes without bead defects could be obtained. In order to improve the mechanical strength of the hydrated zein meshes, core-shell-structured nanofibrous membranes with PCL as the core material and zein forming the shell were prepared by coaxial electrospinning. The core-shell structure of the composite fibers was confirmed by SEM characterization of the fibers, either extracted with chloroform to remove the inner PCL, or elongated to expose their cross-section. The composition and average diameter of the composite fibers could be modulated by the feed rate of the inner PCL solution. It was found that the core-shell fibrous membranes have similar wettability to the electrospun zein mesh. The presence of PCL in the fibers could significantly improve the mechanical properties of the zein membrane.

Abstract:Nanofibrous membranes of rigid polysaccharides including chitosan,alginate or hyaluronic acid(HA) were obtained by coaxial electrospinning of their corresponding aqueous solutions(the inner dope) and PEO(M_n=6.0×10~5) solution in a DMF/H_2O admixture(the outer dope) and subsequent removal of the outer PEO shell.The fiber structure was characterized by TEM and SEM.It is found that core-shell fibers with PEO forming the shell and polysaccharide as the core material can be conveniently prepared by coaxial electrospinning.The PEO shell can be removed by extraction of the core-shell fibers with chloroform.Compared with the polysaccharide fibers prepared by regular uni-axial electrospinning, the fibers(obtained) by coaxial electrospinning display integrated structure after PEO treatment.

Abstract:Electrospun nanofibers have shown great potential as drug vehicles and tissue engineering scaffolds. However, the successful encapsulation of multiple hydrophilic/hydrophobic therapeutic compounds is still challenging. Herein, sodium alginate/poly(ε-caprolactone) core/shell nanofibers were fabricated via water-in-oil emulsion electrospinning. The sodium alginate concentration, water-to-oil ratio, and surfactant concentration were optimized for the maximum stability of the emulsion. The results demonstrated that an increasing water-to-oil ratio results in more deviation from Newtonian fluid and leads to a broader distribution of the fibers' diameters. Moreover, increasing poly(ε-caprolactone) concentration increases loss and storage moduli and increases the diameter of the resulting fibers. The nanofibers' characteristics were investigated by scanning electron microscopy, transmission electron microscopy, confocal laser scanning microscopy, Fourier transform infrared spectroscopy, X-ray diffraction, and water contact angle measurements. It was observed that using an emulsion composition of 10% (w/v) PCL and a water-to-oil ratio of 0.1 results in smooth, cylindrical, and uniform core/shell nanofibers with PCL in the shell and ALG in the core. The in vitro cell culture study demonstrated the favorable biocompatibility of nanofibers. Overall, this study provides a promising and trustworthy material for biomedical applications.

Abstract:A one-step, mild procedure based on coaxial electrospinning was developed for incorporation and controlled release of two model proteins, BSA and lysozyme, from biodegradable core-shell nanofibers with PCL as shell and protein-containing PEG as core. The thickness of the core and shell could be adjusted by the feed rate of the inner dope, which in turn affected the release profiles of the incorporated proteins. It was revealed that the released lysozyme maintained its structure and bioactivity. The current method may find wide applications for controlled release of proteins and tissue engineering.

Abstract:Within the past few years electrospinning has been paid much attention due to the increasing interests of the researchers in the nanotechnology and the unique properties of the nanomaterials. This review focused on the electrospinning behaviors of biomacromolecules including synthetic biomedical polymers, natural-occurring biopolymer and the composite nanofibrous membranes. The electrospinning parameters of the biomacromolecules and the properties of the electrospun membranes such as the morphological stability, mechanical properties and degradation behavior were discussed in details. In addition, the application of the electrospun membranes for controlled drug release was also included. The incorporation method and the release behavior of the drugs were discussed and summarized, according to the solubility of the drugs.

Abstract:Colon-targeted drug delivery is gradually attracting attention because it can effectively treat colon diseases. Furthermore, electrospun fibers have great potential application value in the field of drug delivery because of their unique external shape and internal structure. In this study, a core layer of hydrophilic polyethylene oxide (PEO) and the anti-colon-cancer drug curcumin (CUR), a middle layer of ethanol, and a sheath layer of the natural pH-sensitive biomaterial shellac were used in a modified triaxial electrospinning process to prepare beads-on-the-string (BOTS) microfibers. A series of characterizations were carried out on the obtained fibers to verify the process–shape/structure–application relationship. The results of scanning electron microscopy and transmission electron microscopy indicated a BOTS shape and core–sheath structure. X-ray diffraction results indicated that the drug in the fibers was in an amorphous form. Infrared spectroscopy revealed the good compatibility of the components in the fibers. In vitro drug release revealed that the BOTS microfibers provide colon-targeted drug delivery and zero-order drug release. Compared to linear cylindrical microfibers, the obtained BOTS microfibers can prevent the leakage of drugs in simulated gastric fluid, and they provide zero-order release in simulated intestinal fluid because the beads in BOTS microfibers can act as drug reservoirs.

Abstract:<p>It is of great importance and a major challenge to achieve a controlled delivery of specific types of active ingredients for tissue regeneration. Herein, we report a system comprised of electrospun quad-axial nanofibers fabricated by quadriaxial electrospinning, with the material components in the different layers can be well regulated. The quad-axial nanofibers allow the regulation of the type of materials in different layers as well as the manipulation of the location of drug in the nanofibers, representing a promising controlled drug release system. In one typical example, we apply polycaprolactone to construct both the outermost and second innermost layers while gelatin to construct both the second outermost and innermost layers. The nanofibers with a cleared four-layered nanostructure are confirmed by morphological evaluations. Moxifloxacin, a type of antibacterial drug serving as a model of therapeutic payload, is encapsulated in the different layers of the nanofibers, realizing an effective control of the drug delivery. In addition, the efficacy for drug delivery with the use of quad-axial nanofibers is superior to that of core-sheath and blended nanofibers. This quadriaxial electrospinning technique can be widely used for the co-delivery of factor cocktails in a designed sequence, which will show great potential for tissue engineering.</p>

Abstract:Structural polymeric nanohybrids is presently a popular topic and can be conceived for numerous functional applications, including the pH-sensitive oral colon-targeted drug-delivery system. In this paper, a brand-new Janus core@shell (JCS) nanostructure was fabricated using a trifluid electrospinning, in which three polymers and a model drug 5-fluorouracil (5-FU) were elaborately and intentionally positioned. In the structural hybrids, the pH-sensitive polymer hydroxypropyl methyl cellulose acetate succinate was located in the common shell layer, and the 5-FU-loaded ethyl cellulose (EC) and polyethylene oxide (PEO) were organized in a side-by-side manner in the core sections. The JCS fiber had a fine linear morphology with a multiple-chamber structure and a shell thickness of about 24 nm. The drug presented in the fibers in an amorphous state, owing to the secondary intermolecular interactions between EC and 5-FU. The ex vivo adhesion experiments suggested that the JCS fibers could stick firmly to the intestine membranes. In vitro dissolution tests showed the JCS fibers released only 7.8% ± 3.5% of the loaded 5-FU in an acid condition. In vivo gavage administration verified that the JCS fibers effectively promoted the absorbance of 5-FU in a synergistic manner, better than the double-layer core–shell and Janus nanofibers, and near fourfold than the drug solutions as a control. The present protocol opens a new way for developing novel multifunctional nanomaterials with the JCS nanostructure as a powerful supporting platform.Graphical abstract

Abstract:Introduction: As an interdisciplinary field, drug delivery relies on the developments of modern science and technology. Correspondingly, how to upgrade the traditional dosage forms for a more efficacious, safer, and convenient drug delivery poses a continuous challenge to researchers. Methods, results and discussion: In this study, a proof-of-concept demonstration was conducted to convert a popular traditional liquid dosage form (a commercial oral compound solution prepared from an intermediate licorice fluidextract) into a solid dosage form. The oral commercial solution was successfully encapsulated into the core–shell nanohybrids, and the ethanol in the oral solution was removed. The SEM and TEM evaluations showed that the prepared nanofibers had linear morphologies without any discerned spindles or beads and an obvious core–shell nanostructure. The FTIR and XRD results verified that the active ingredients in the commercial solution were compatible with the polymeric matrices and were presented in the core section in an amorphous state. Three different types of methods were developed, and the fast dissolution of the electrospun core–shell nanofibers was verified. Conclusion: Coaxial electrospinning can act as a nano pharmaceutical technique to upgrade the traditional oral solution into fast-dissolving solid drug delivery films to retain the advantages of the liquid dosage forms and the solid dosage forms.

Tri-Layer Core–Shell Fibers from Coaxial Electrospinning for a Modified Release of Metronidazole

Abstract:Polymers are the backbone of drug delivery. Electrospinning has greatly enriched the strategies that have been explored for developing novel drug delivery systems using polymers during the past two decades. In this study, four different kinds of polymers, i.e., the water-soluble polymer poly (vinyl alcohol) (PVA), the insoluble polymer poly(ε-caprolactone) (PCL), the insoluble polymer Eudragit RL100 (ERL100) and the pH-sensitive polymer Eudragit S100 (ES100) were successfully converted into types of tri-layer tri-polymer core–shell fibers through bi-fluid coaxial electrospinning. During the coaxial process, the model drug metronidazole (MTD) was loaded into the shell working fluid, which was an emulsion. The micro-formation mechanism of the tri-layer core–shell fibers from the coaxial emulsion electrospinning was proposed. Scanning electron microscope and transmission electron microscope evaluations verified the linear morphology of the resultant fibers and their obvious tri-layer multiple-chamber structures. X-ray diffraction and Fourier transform infrared spectroscopy measurements demonstrated that the drug MTD presented in the fibers in an amorphous state and was compatible with the three polymeric matrices. In vitro dissolution tests verified that the three kinds of polymer could act in a synergistic manner for a prolonged sustained-release profile of MTD in the gut. The drug controlled-release mechanisms were suggested in detail. The protocols reported here pioneer a new route for creating a tri-layer core–shell structure from both aqueous and organic solvents, and a new strategy for developing advanced drug delivery systems with sophisticated drug controlled-release profiles.

Abstract:The search for higher-quality nanomaterials for medicinal applications continues. There are similarities between electrospun fibers and natural tissues. This property has enabled electrospun fibers to make significant progress in medical applications. However, electrospun fibers are limited to tissue scaffolding applications. When nanoparticles and nanofibers are combined, the composite material can perform more functions, such as photothermal, magnetic response, biosensing, antibacterial, drug delivery and biosensing. To prepare nanofiber and nanoparticle hybrids (NNHs), there are two primary ways. The electrospinning technology was used to produce NNHs in a single step. An alternate way is to use a self-assembly technique to create nanoparticles in fibers. This paper describes the creation of NNHs from routinely used biocompatible polymer composites. Single-step procedures and self-assembly methodologies are used to discuss the preparation of NNHs. It combines recent research discoveries to focus on the application of NNHs in drug release, antibacterial, and tissue engineering in the last two years.

Abstract:BSTRACT Purpose To design and fabricate multicomponent amorphous electrospun nanofibers for synergistically improving the dissolution rate and permeation profiles of poorly water-soluble drugs. Methods Nanofibers were designed to be composed of a poorly water soluble drug, helicid, a hydrophilic polymer polyvinylpyrrolidone as filament-forming matrix, sodium dodecyl sulfate as transmembrane enhancer and mannitol as taste masking agent, and were prepared from hot aqueous co-dissolving solutions of them. An elevated temperature electrospinning process was developed to fabricate the composite nanofibers, which were characterized using FESEM, DSC, XRD, ATR-FTIR, in vitro dissolution and permeation tests. Results The composite nanofibers were homogeneous with smooth surfaces and uniform structure, and the components were combined together in an amorphous state because of the favorable interactions such as hydrogen bonding, electrostatic interaction and hydrophobic interactions among them. In vitro dissolution and permeation tests demonstrated that the composite nanofibers had a dissolution rate over 26-fold faster than that of crude helicid particles and a 10-fold higher permeation rate across sublingual mucosa. Conclusions A new type of amorphous material in the form of nanofibers was prepared from hot aqueous solutions of multiple ingredients using an electrospinning process. Abstract:Nanomaterials, particularly nanofibers are demonstrating exceptional potential for medical applications considered as promising systems for the integration of drug molecules, providing an enhancement of drug effectiveness and safety, while reducing drug side effects. Nanofibers based on two polymeric matrices poly(vinyl alcohol) (PVA) and chitosan (CS) have been demonstrated to be safe and promising materials for the drug integration and targeted drug delivery. This article studied the factors influencing on the fabrication and properties of PVA–CS nanofibers, including the impact of solution components on solution structure, rheological properties, electrospinning technology parameters, as well as the structure and thermomechanical properties of the obtained nanofibers have been investigated in details. The increasing of acetic acid concentration has the effect of pH lowering as well as electrical conductivity while heightening its viscosity. The pH, conductivity, and viscosity values of the solution rises as the CS concentration increases. Based on the Hildebrand‐Scatchard solubility theory, calculated Teas diagrams that account intermolecular interactions allow to predict low‐defect nanofibers based on PVA–CS in binary solvent systems with a greater concentration of hydrogen bonds. The optimal solution content and electrospinning parameters were determined. Infrared spectroscopy confirmed that the two polymer materials have a good interaction and the acetic acid was completely separated from the PVA–CS nanofibers. The XRD spectrum reveals that the crystal lattice parameters of the nanofibers change according to the polymer ratio. The polymers ratio also influence on the thermal and tensile properties. Such biosafe polymer materials may be considered as the promising drug delivery systems with the targeted mode of action.
