Abstract:1-Iodoalkynes are one type of the most reactive and the most practical intermediates in organic synthesis. Here, some facile and efficient methods for the 1-iodination of terminal alkynes are developed using N-iodosuccinimide (NIS) as an iodination reagent and some inexpensive mild bases as catalysts. K2CO3 and 4-dimethylaminopyridine (DMAP) have been proven to be the most suitable inorganic and organic bases, succeeding in 17 examples with excellent yields (up to 99%).

Abstract:A novel iodine-mediated intermolecular iodosulfonylation reaction of alkenes for dual C-S and C-I bond formation is described. A series of alkenes could be selectively converted into the corresponding beta-iodo sulfones, which are versatile building blocks in organic synthesis and medicinal chemistry. Molecular iodine was the iodine source in this reaction.

Abstract:We report the first catalyst-free and trans-selective iodoalkylation reaction of alkynes with a series of α-carbonyl compounds. This unprecedented three-component iodoalkylation reaction is enabled by using (iodoethynyl)trimethylsilane as a radical initiator and iodide source. The 1,2-difunctionalization affords alkenyl iodides, which are versatile building blocks for the construction of tri-substituted alkene derivatives.

Abstract:A highly regioselective iodosulfonylation of allenes in the presence of CuI and 1,10-phenanthroline has been developed for the synthesis of various useful ( E)-α-iodomethyl vinylsulfones in moderate to excellent yields. This practical reaction is fast, operationally simple, and in particular, proceeds under very mild conditions to afford the target products with high regio- and stereoselectivity. The selectivity was illustrated by a conceptual DFT analysis.

Abstract:An efficient one-pot method for the preparation of alpha-iodo-substituted alpha,beta-unsaturated aldehydes (alpha-iodoenals) from propargylic alcohol is developed. The reaction proceeds via an iodoallene intermediate, which is generated in situ by the reaction of propargylic alcohol with aqueous HI. The iodoallene intermediate is further transformed to an alpha-iodoenal derivative in good overall yield by oxidation with molecular O(2).

Abstract:A Rh(iii)-catalyzed C-H alkynylation of substituted N-phenoxyacetamides has been developed with the aid of hypervalent iodine-alkyne reagents. Complementary to the Sonogashira coupling reaction, this protocol provides an efficient and straightforward method to access aryl alkynes at room temperature. The multifunctional directing group is preserved which can be further employed for ortho-directed functionalizations to obtain additional new complex products.

Abstract:An efficient synthesis toward 2-alkyl substituted oxazoles has been achieved through Suzuki cross-coupling reaction. Treatment of various alkyl iodides with 9-MeO-BBN, followed by in situ palladium-catalyzed carbon–carbon formation with 2-iodooxazoles obtained the 2-alkyl substituted oxazoles in good to excellent yields. Regioselective C2-alkylation on 2,4-di-iodooxazole under the same reaction conditions was firstly disclosed.

Abstract:Among all functional groups, alkynes occupy a privileged position in synthetic and medicinal chemistry, chemical biology, and materials science. Thioalkynes, in particular, are highly useful, as they combine the enhanced reactivity of the triple bond with a sulfur atom frequently encountered in bioactive compounds and materials. Nevertheless, general methods to access these compounds are lacking. In this article, we describe the mechanism and full scope of the alkynylation of thiols using ethynyl benziodoxolone (EBX) hypervalent iodine reagents. Computations led to the discovery of a new, three-atom concerted transition state with a very low energy barrier, which rationalizes the high reaction rate. On the basis of this result, the scope of the reaction was extended to the synthesis of aryl- and alkyl-substituted alkynes containing a broad range of functional groups. New sulfur nucleophiles such as thioglycosides, thioacids, and sodium hydrogen sulfide were also alkynylated successfully to lead to the most general and practical method yet reported for the synthesis of thioalkynes.

Abstract:In this work, we have developed a successful Sonogashira coupling reaction utilizing IPrCl-Cl as the activation reagent to couple carboxylic acids with terminal alkynes. The reaction employs CuI/Pd(PPh3)2Cl2 as the co-catalyst and Et3N as the solvent, resulting in excellent yields (up to 88%) of ynones. The use of carboxylic acids as a starting material makes this method a valuable contribution to organic synthesis.
