Abstract:Rh(III)-catalyzed C(sp(3)) - H bond aminocarbonylation of 8-methylquinolines and isocyanates has been realized under mild conditions. This approach is applicable to different aryl and alkyl isocyanates, leading to the synthesis of various alpha-quinolinyl amide compounds in moderate to excellent yields. A plausible mechanism for this transformation is proposed according to the experimental results obtained.

Abstract:A Rh(III)-catalyzed oxidative annulation of pyridin-2(1H)-ones with alkynes via double C–H activation to produce highly functionalized 4H-quinolizin-4-ones is disclosed. This reaction features easily available starting materials, simple manipulation, a relatively wide substrate scope, and good functional group tolerance. The application of this protocol is demonstrated by the synthesis of a known fluorescent quinolizino[3,4,5,6-ija]quinolinium salt.

Abstract:One-pot syntheses of 1-benzyl-, 1-alkoxyl-, and 1-alkylamino- isoquinolines through automatic directing group (DGauto)-assisted, rhodium(III)-catalyzed aryl C-H activation and annulation with internal alkynes were developed. The reactions affording 1-benzylisoquinolines involve a cascade oximation of diarylacetylenes with hydroxylamine, forming aryl benzyl ketone oxime, and oxime-assisted rhodium(III)-catalyzed aryl C-H activation and followed annulation with another molecule of diarylacetylene in a one-pot manner. The formation of 1-alkoxyl/amino isoquinolines includes the addition of nucleophilic alcohols or amines to aryl nitriles, imine-assisted rhodium-catalyzed aryl C-H activation, and subsequent alkyne annulation.

Abstract:The cascade oxidative annulation reactions of aryl imidazoles with two molecules of alkynes via multiple C-H activation proceed efficiently in the presence of [Cp*RhCl2]2 and Cu(OAc)2·H2O to give substituted benzo[ij]imidazo[2,1,5-de]quinolizine-based polyheteroaromatic compounds. This method is compatible with various functional groups, which are very useful for further synthetic transformations.

Abstract:A Rh(III)-catalyzed C-H activation/cyclization cascade was developed for the first divergent synthesis of pyrazolo[1,5-a]quinazolines through a [5 + 1] annulation reaction exclusively. The one-pot procedure is recognized for its broad substrate scope, functional group tolerance, and high atom economy. Mechanistic studies reveal the reaction pathway, addressing current limitations. Notably, this catalytic transition metal-assisted tandem annulation smoothly proceeds through the reaction of substituted phenyl-1H-pyrazol-5-amine with an alkyne ester or amide, where a one ring carbon is provided by the alkynoate building block. This transformation highlights the potential of transition metal-catalyzed methods for synthesizing diverse pyrazolo[1,5-a]quinazoline frameworks.

Abstract:Rhodium (Rh)-catalysed [4+2] annulation of aromatic oximes with common terminal alkenes for the synthesis of 3,4-dihydroisoquinolines is presented. Through the cooperation of a Rh(III) catalyst and a catalytic amount of K2HPO4 base, the reaction enables the formation of two new bonds, a C(sp(2))-C(sp(3)) bond and a C(sp(3))-N bond, in a single reaction via functionalization of both the C-H and N-O bonds and provides a practical method to produce 3,4-dihydroisoquinolines with exquisite selectivity and excellent compatibility of functional groups.

Abstract:Here we report a novel Rh-catalyzed C-H/C-H alkenylation of N-arylmethanimines with vinylene carbonate acting as a vinylene unit. Forty examples of C3,C4-nonsubstituted quinolines were achieved from commercially available starting materials. This identified process features an exceedingly simple system, a lower loading of catalyst, and the capacity for postfunctionalization with bioactive molecules.

Abstract:A highly enantioselective rhodium(I)-catalyzed dearomative arylation or alkenylation of easily available N-alkylquinolinium salts is reported, thus providing an effective and practical approach to the synthesis of dihydroquinolines in up to 99 % ee. This reaction tolerates a wide range of functional groups with respect to both the organic boronic acids and the quinoline starting materials. Moreover, the synthetic utility of this protocol is demonstrated in the formal asymmetric synthesis of bioactive tetrahydroquinoline and the total syntheses of (-)-angustureine and (+)-cuspareine.
