Abstract:A new and efficient [5+1] annulation reaction for the first synthesis of 5,5-disubstituted 4,5-dihydropyrazolo[1,5-a]quinazolines is described. This transition-metal-free tandem cyclization was performed with 5-amino-1H-phenylpyrazole and readily available electron-deficient olefinic ester. The reaction proceeds via an aza-Michael addition/Truce-Smiles rearrangement/SNAr cyclization pathway, which was verified by DFT calculations. The participation of Truce-Smiles rearrangement is substrate dependent and could prompt the process of cascade reactions. It is also the first report of alkenes acting as one-carbon synthons for [5+1] hetero-annulations.

Abstract:The 1-arylpyrazolo[5,1-b]quinazolin-9(1H)-one derivatives were obtained via intramolecular alkyne hydroamination reaction of 2-amino-N′-arylbenzohydrazide and alkynal diethyl acetal in the presence of CuBr and Cs2CO3. Abstract:An efficient and rapid synthesis of multiply substituted quinolines is described. This method is enabled by a three-component cascade annulation of readily available aryl diazonium salts, nitriles, and alkynes. This reaction is catalyst- and additive-free. Various aryl diazonium salts, nitriles, and alkynes can participate in this transformation, and the yields are up to 83%.

Abstract:The development of an efficient strategy for facile access to quinoline-based bis-heterocycles holds paramount importance in medicinal chemistry. Here in, we describe a unified approach for accessing 8-(indol-3-yl)methyl-quinolines by integrating Cp*Rh(III)-catalyzed C(sp3)–H bond activation of 8-methylquinolines followed by nucleophilic cyclization with o-ethynylaniline derivatives. Remarkably, methoxybiaryl ynones under similar catalytic conditions delivered quinoline tethered spiro[5.5]enone scaffolds via a dearomative 6-endo-dig C-cyclization. Moreover, leveraging this method for C8(sp2)−H bond activation of quinoline-N-oxide furnished biologically relevant oxindolyl-quinolines. This reaction proceeds via C(sp2)–H bond activation, regioselective alkyne insertion, oxygen-atom-transfer (OAT) and intramolecular nucleophilic cyclization in a cascade manner. One C−C, one C−N and one C=O bonds were created with concomitant formation of a quaternary center.

Abstract:Via an imine-protection strategy, we herein present an unprecedented copper-catalyzed oxidative multicomponent annulation reaction for direct synthesis of quinazolinones. The construction of various products is achieved via formation of three C-N and one C-C bonds in conjunction with the benzylic functionalization. The merits of easily available feedstocks, naturally abundant catalyst, good functional group and substrate compatibility, and release of H2O as the byproduct make the developed chemistry a practical way to access quinazolinones.

Abstract:An efficient sequence based on the Ugi-azide reaction and rhodium(III)-catalyzed intermolecular annulation has been established for the preparation of tetrazole-isoquinolone/pyridone hybrids. Several N -acylaminomethyltetrazoles were reacted with arylacetylenes to form the hybrid products in moderate to very good yields. The method relies on the capacity of the rhodium catalyst to promote C(sp 2 )–H activation in the presence of a suitable directing group. The Ugi-azide reaction provides broad molecular diversity and enables the introduction of the tetrazole moiety, which may further assist the catalytic reaction by coordinating the metal center. The scope of the isoquinolones is very wide and may be extended to the preparation of complex compounds having heterocyclic moieties such as pyridone, furan, thiophene and pyrrole, as well as the corresponding benzo-fused derivatives. The developed procedure is simple, reproducible and does not require inert conditions.

Abstract:An efficient one-pot synthesis of isoquinolines and heterocycle-fused pyridines by three-component reaction of aryl ketones, hydroxylamine, and alkynes is developed. The reaction involves condensation of aryl ketones and hydroxylamine, rhodium(III)-catalyzed C-H bond activation of the in situ generated aryl ketone oximes, and cyclization with internal alkynes. This protocol enables rapid assembly of multisubstituted isoquinolines as well as γ-carbolines, furo[2,3-c]pyridines, thieno[2,3-c]pyridines, and benzofuro[2,3-c]pyridines from readily available substrates.

Abstract:An efficient RhIII-catalyzed ortho-C-H bond activation for the synthesis of substituted isoquinolines and heterocycle-fused pyridines in aqueous medium has been developed. This method involves the in situ generation of ketimines from ketones and ammonium acetate and subsequent oxidative C-H bond activation/annulation of ketimines with alkynes to form the C-C/C-N bonds spontaneously. Various aryl ketones and internal alkynes were smoothly transformed into the desired heterocycles in moderate to excellent yields. The reaction exploits molecular oxygen as the sole oxidant by producing water as the only byproduct. Moreover, easily available NH4OAc was utilized as the nitrogen source, which makes this protocol practical and environmentally friendly.

Abstract:The synthesis of N -naphthyl pyrazoles has been realized by the direct three-component reactions of enaminones, aryl hydrazine hydrochlorides and internal alkynes via Rh( III ) catalysis. The synthetic reactions employing simple substrates lead to simultaneous construction of dual cyclic moieties, including a pyrazole ring and a phenyl ring, via sequential formation of two C–N and three C–C bonds.

Abstract:A cobalt‐catalyzed annulation of o‐phenylenediamines and internal alkynes for the synthesis of quinoxalines is developed. This method provides a variety of quinoxalines in good to high yields under a simple and mild reaction conditions with molecular O2 as a terminal oxidant. Preliminary mechanistic studies show that a novel reaction mechanism is involved in the current reaction, which is different from the previous reported dicarbonyl compound mechanism via the oxidation of the alkyne.
