Abstract:The increasing demand for rapid and sensitive roadside identification of trace illegal drugs drives the development of high-performance miniature mass spectrometric instrumentation and methodology. Here, we report a synchronized flash-thermal-desorption purging and ion injection (SFTDPI) method to increase the sensitive and rapid screening of volatile and nonvolatile illegal drugs for miniature ion trap mass spectrometry (ITMS). The flash-thermal desorption could reach 290 degrees C in 2.5 s, which could achieve efficient vaporization of nonvolatile noscapine with boiling point at 565 degrees C. ITMS using discontinuous atmospheric pressure interface (DAPI) has an ion utilization ratio of less than 1%, the synchronized purging for flash-thermal desorption and ion injection with DAPI could accurately control the time interval along with the desorption, gas purging, ionization and ion injection, and the sample utilization ratio increases more than five times. The miniature SFTDPI-ITMS presents good performance: (1) more than 60 times improvement in sensitivity was achieved compared to the previously reported thermal-desorption acetone-assisted photoionization ion trap mass spectrometer for nonvolatile drugs, and the minimum detectable quantity reaches 50 pg for fentanyl. (2) Ten kinds of mixing drugs with boiling point difference of 300 degrees C can be simultaneously identified within 3 s under a single analysis. SFTDPI-ITMS was deployed at the roadside checkpoint of Sino-Burmese border; fentanyl in the captured encapsulated powder and the suspected opiate had been successfully identified.

Abstract:Illegal drug smugglings and crimes have long been a global concern, and an apparatus which can identify drugs on-the-spot is urgently demanded by law enforcement. A thermal desorption acetone-assisted photoionization miniature ion trap mass spectrometer was developed for on-site and rapid identification of illegal drugs at checkpoints. Acetone was chosen for dopant-assisted photoionization, and the sensitivity of selected drugs was further enhanced with protonated analyte molecular ions [M + H]+. For example, the sensitivity of ephedrine was improved by as high as 22-fold. The mass discrimination effect, which was usually considered as a shortcoming of ion trap mass analyzer, was ingeniously utilized to eliminate the protonated acetone reagent ions and maximize the trapping efficiency of analyte ions in mass analyzer. Twenty-seven drugs were analyzed, and the limits of detection (LODs) of selected illegal drugs were at the nanogram level with analysis time of 2 s. Analyte/dopant ion peak intensity ratios in mass spectra could be used for quantitation to improve the quantitative analysis performance of miniature ion trap mass spectrometer equipped with a discontinuous atmospheric pressure interface (DAPI) with the prerequisite that dopant ions and analyte ions could be simultaneously and effectively trapped by the ion trap. The RSD of signal intensity was reduced from 25.3% to 8.5%, and the linear range was extended from 0.5-25 to 0.5-100 ng/μL for methamphetamine. A temperature-resolved thermal desorption sampling strategy was developed and used to distinguish illegal drug components in plant-based drug samples and drinks containing illegal drugs.

Abstract:The escalating need for prompt and highly sensitive on-site detection of trace-level drugs is fueling the advancement of miniature, high-performance mass spectrometers and analytical methodologies. In this study, a miniature continuous atmospheric pressure interfaced ion trap mass spectrometer integrated with thermal desorption acetone-assisted photoionization (TD-CAPI-ITMS) was developed for highly sensitive detection of nonvolatile drugs in saliva and blood. By strategically extending the sampling time of the TD-CAPI-ITMS to cover the entire desorption process, a remarkable two-order-of-magnitude enhancement in the signal intensity of individual drugs was observed. Moreover, the simultaneous detection of drug mixtures with widely varying boiling points and saturation vapor pressures was accomplished. Optimization of the parameters yielded a limit of detection (LOD) for ketamine and 5F-EMB-PICA of 1 pg/μL accompanied by a robust stability, as evidenced by a relative standard deviation (RSD) of 5.30 %. Combined with straightforward liquid-liquid extraction, the sensitivity of drugs in saliva as low as 10 pg/μL was achieved, which met the requirements of Chinese national standard GA1333-2017. Owing to its exceptional sensitivity, the matrix effect present in blood samples was significantly alleviated through dilution, allowing for accurate monitoring of antibiotic concentrations. The results underscore the substantial potential of the TD-CAPI-ITMS for lab-free applications in drug-related forensic analysis, therapeutic drug monitoring, and pharmacokinetic studies.
Parallel Coupling of Ion Mobility Spectrometry and Ion Trap Mass Spectrometry for the Real-Time Alarm Triggering and Identification of Hazardous Chemical Leakages

Abstract:Hazardous chemical leakages involved in chemical terrorist attacks and chemical industrial accidents have been posing severe threats to human health and the environment. Vehicle-mounted mass spectrometry (MS) has been developed for continuous, on-road measurements to map the spatial and temporal distributions of hazardous chemicals. However, the detection of chemicals with small temporal scales and spatial scales is always challenging. In this study, a parallel coupling apparatus combining the techniques of ion mobility spectrometry and ion trap MS (p-IMS-ITMS) was developed to improve the detection rate and the time response capability of a stand-alone ITMS system for short time-span chemical tracking. A workflow was also proposed along with the apparatus, where the ITMS system can be triggered, as chemical suspects were discovered with the IMS system. The sampling positions of the ITMS system were investigated and optimized. In addition, a strategy was proposed to diminish the time span of samples from 1.5 to 0.5 s for evaluating the performances of the p-IMS-ITMS system. The detection rate of the stand-alone ITMS system was measured to be only 9.5, 32, and 87.5% for the time span of 0.5, 1, and 1.5 s, respectively. By comparison, the detection rates of the p-IMS-ITMS system were 99.5, 100, and 100%, where the detection rate was increased by a factor of 10 for 0.5 s time span. Moreover, the addition of an IMS system could provide temporal patterns of hazardous chemicals with a resolution of 33 ms. Finally, the potential of the p-IMS-ITMS system for environmental navigation monitoring and assessment was further demonstrated by detecting the leakages of dimethyl methyl phosphonate and dipropylene glycol monomethyl ether.

Abstract:A thermal desorption low temperature plasma (TD-LTP) ionization source was developed for the direct, rapid and sensitive detection of pesticides by mass spectrometry without complex sample pretreatment. A thermal desorption sampler was added in front of the plasma generator, and the analyte was collected on a polytetrafluoroethylene (PTFE) swab and desorbed into gas molecules, and then gas molecules were transported to the plasma generator by carrier gas to be ionized. The utilization of thermal desorption sampler helped to transform the interaction between gas phase plasma and sample from gas-solid or gas-liquid to gas-gas, which greatly increased the sensitivity and stability especially for non-volatile sample (e.g. pesticides) compared with conventional LTP ionization source. The TD-LTP ionization source was coupled to a homemade rectilinear ion trap mass spectrometry. The operation parameters of the TD-LTP ionization source were optimized and the characteristic ions of 12 different pesticides were successfully obtained and investigated. Then the TD-LTP ionization source was connected with commercial ACQUITY TQD mass spectrometer, and the pesticide residue level in broomcorn was evaluated.

Abstract:The detection of vapours of listed explosives and drugs by silicon chip based field asymmetric ion mobility spectrometry(FAIMS)technology was reported.The concentration of sample vapours was characterized by weighing mass loss from diffusion in 14-21days.Under the optimized conditions of detection,all 7explosives and 2drugs demonstrated good response in the FAIMS instrument at the concentration of 33-100ng/L.A novel method to extract characteristic signals was also developed to differentiate the fingerprint signals from presented samples.This simple process provided by FAIMSbased instrument demonstrated great potential in homeland security applications.

Abstract:The drug detection technology plays a pivotal role in the domains of pharmaceutical regulation and law enforcement. In this study, we introduce a method that combines thermal desorption corona discharge ionization (TD-CDI) with mass spectrometry for efficient drug detection. The TD-CDI module, characterized by its compact and simple design, enables the separation of analytes within seconds and real-time presentation of one or two analyte peaks on the mass spectrum most of the time, which reduces matrix interference and improves detection performance. Through experimental investigation, we studied the characteristics of TD-CDI for analyte separation and detection, even with the same mass number, and optimized the TD-CDI approach. TD-CDI-MS was employed for the rapid detection of drugs in various traditional medicine, food products, and human samples. Additionally, by utilizing TD-CDI for segmented hair direct analysis, it becomes possible to trace the drug usage cycle of individuals. This underscores the feasibility of the proposed analytical method within the realm of drug detection.

Abstract:In this study, we introduce the design, characterization, and application of an integrated thermal desorption corona discharge ionization (TD-CDI) device, which was developed for direct mass spectrometry (MS) analysis of drugs in solid or liquid samples. The structure and operational parameters of the TD module were optimized through simulation and experimental investigations. For the CDI module, a hollow needle was used as the discharge electrode and sample inlet, allowing self-aspiration and efficient ionization of the desorbed samples. The TD-CDI device was tested and characterized using different mass spectrometers, exhibiting satisfactory stability and quantitative performance. Owing to its compact structure, it can be coupled to a miniature MS instrument to implement rapid identification of various drugs in urine solutions.

Abstract:Illegal drugs and the explosives are harmful chemicals that present both social and national security issues. Existing analytical instruments and methods for identifying these substances are constrained by sensitivity, speed, and portability because of inherent technical defects. Miniaturized photoionization ion trap mass spectrometry is suitable for rapid and on-site identification of illegal drugs and the explosives; however, tandem mass spectrometry and the fragment ions have not been fully investigated. In this article, the abundant fragments of 22 illegal drugs and seven explosives were analyzed successfully using resonance excitation with a miniaturized ion trap mass spectrometer constructed in our laboratory and coupled with flash-thermal-desorption acetone-assisted photoionization. The mass range of the rectilinear ion trap was optimized through experimental mapping of stability diagrams to decrease the effect of the low mass cut-off. The secular frequencies for resonance excitation were simulated and validated using the experimental results. The fragments of illegal drugs and explosives were investigated and imported into a database. By comparing the tandem mass spectrometry fragments and the created database, a seized analyte could be identified within 3 s using a single analysis. This work indicates that ion resonance excitation is an effective approach for rapid analysis of illegal drugs and explosives. Furthermore, combination of a miniaturized ion trap mass spectrometer and compound database is useful for the identification of unknown and known compounds in forensic screening applications. Abstract:The thermal desorption (TD) technique is widely employed in modern mass spectrometry to facilitate the detection of non-volatile analytes. In this study, we developed a compact TD device based on a small resistance wire and coupled it with a self-aspirating corona discharge ionization (CDI) source to conduct direct MS analysis of various liquid and solid samples. Due to its small size and low heat capacity, the temperature of the TD module can be flexibly and rapidly modulated by controlling the power sequence. Multiple heating modes, including pulse heating (PH), isothermal heating, and step heating (SH), are realized and characterized, and then applied for the detection of different real samples. In particular, the PH mode is suitable for the simultaneous detection of multiple components in samples with relatively simple matrices, while the SH mode is capable of component separation. In addition, the sensitivity and quantitative capability of the TD-CDI system for DEP solutions were tested, showing acceptable stability with a relative standard deviation of about 6.7% and a detection limit of 0.088 ng. Overall, the developed TD-CDI system provides a simple, convenient, and versatile tool for direct mass spectrometry analysis of real samples.
Portable Digital Linear Ion Trap Mass Spectrometer Based on Separate-Region Corona Discharge Ionization Source for On-Site Rapid Detection of Illegal Drugs

Abstract:As narcotic control has become worse in the past decade and the death toll of drug abuse hits a record high, there is an increasing demand for on-site rapid detection of illegal drugs. This work developed a portable digital linear ion trap mass spectrometer based on separate-region corona discharge ionization source to meet this need. A separate design of discharge and reaction regions was adopted with filter air as both carrier gas for the analyte and protection of the corona discharge needle. The linear ion trap was driven by a digital waveform with a low voltage (±100 V) to cover a mass range of 50–500 Da with a unit resolution at a scan rate of 10,000 Da/s. Eighteen representative drugs were analyzed, demonstrating excellent qualitative analysis capability. Tandem mass spectrometry (MS/MS) was also performed by ion isolation and collision-induced dissociation (CID) with air as a buffer gas. With cocaine as an example, over two orders of magnitude dynamic range and 10 pg of detection limit were achieved. A single analysis time of less than 10 s was obtained by comparing the information of characteristic ions and product ions with the built-in database. Analysis of a real-world sample further validated the feasibility of the instrument, with the results benchmarked by GC-MS. The developed system has powerful analytical capability without using consumables including solvent and inert gas, meeting the requirements of on-site rapid detection applications.

Abstract:A miniature ion trap mass spectrometer with capillary direct sampling and vacuum ultraviolet photoionization source was developed to conduct trace analysis of organic compounds in liquids. Self-aspiration sampling is available where the samples are drawn into the vacuum chamber through a capillary with an extremely low flow rate (less than 1 μL/min), which minimizes sample consumption in each analysis to tens of micrograms. A pulsed gas-assisted inlet was designed and optimized to promote sample transmission in the tube and facilitate the cooling of ions, thereby improving instrument sensitivity. A limit of detection of 2 ppb could be achieved for 2,4-dimethylaniline in a methanol solution. The sampling system described in the present study is specifically suitable for a miniature photoionization ion trap mass spectrometer that can perform rapid and online analysis for liquid samples. Abstract:The sensitivity of mass analysis is highly dependent on the efficiencies of the sample introduction, analyte ionization, and subsequent ion transfer to the mass analyzer. For miniature mass spectrometry (MS) systems, it is highly desirable to maximize the amount of the samples that can be introduced at the pumping capability highly compromised and to have the MS analysis performed with simple electronics and robust in-vacuum components. In this study, a discharge ionization source synchronized with the sample introduction has been developed for hand-held mass spectrometers with discontinuous atmospheric pressure interfaces (DAPI). The flow rate of the sample introduced with a DAPI can be much higher than that allowed with a conventional continuous atmospheric pressure interface. The pressure variation associated with the DAPI operation was used to turn on and off the synchronized discharge ionization (SDI). For analysis of volatile organic compounds (VOCs) in air samples, the SDI has been shown to be soft and molecular ions of the VOCs have been observed as the major species. Since the ions could be transferred directly to the ion trap mass analyzer without a barrier for maintaining pressure differences, high sensitivity in chemical analysis was also achieved, with limits of detection (LODs) better than 1 ppbv obtained for PAHs (polycyclic aromatic hydrocarbons) in air.
Miniaturized Ion Mobility Spectrometer with a Dual-Compression Tristate Ion Shutter for On-Site Rapid Screening of Fentanyl Drug Mixtures.

Abstract:The hidden presence of fentanyl and new psychoactive substances in established illicit drugs has led to a great number of unintentional fatal overdoses, justifying the urgent need for portable tools for the rapid screening of various drugs. Ion mobility spectrometry, as a rapid detection technique, has been widely used in detecting illicit drugs, whereas it is insufficiently sensitive for bigger ions due to the mobility discrimination of an ion shutter. In this work, a miniaturized ion mobility spectrometer with an inner diameter of 14 mm and a drift length of 38.9 mm and equipped with a novel dual-compression tristate ion shutter has been developed, which could greatly reduce the mobility discrimination by compressing the ion packet twice during the injection process. The best gating performance of the dual-compression tristate ion shutter was about three times as high as that of the tristate ion shutter and twice as high as that of the Tyndall-Powell shutter. For example, the peak height was increased by 150 or 40% when the resolving power was the same, whereas the resolution of two neighboring peaks was improved by 46 or 19% when the peak heights were the same. In screening of fentanyl drug mixtures, the new shutter enhanced the identification accuracy of constituents by making peaks of slow dimer ions and complex ions observable. Besides, the new shutter helped to achieve high sensitivity for drug ions spreading a wide ion mobility range from 0.644 to 2.032 cm2 s-1 V-1, demonstrating its potential use in the analysis of other mixtures and the detection of large ions.

Spray-inlet microwave plasma torch ionization tandem mass spectrometry for the direct detection of drug samples in liquid solutions

Abstract:Rationale: Drug abuse or dependence results in a series of social problems, including crime and traffic accidents. Spray-inlet microwave plasma torch tandem mass spectrometry (MPT-MS/MS) was developed and used for the direct detection of such drugs in liquid solutions. Methods: Drug sample solutions were directly sprayed into the flame of an MPT by a sampling pump and the ions produced by Penning ionization and ion-molecule reactions were guided into a quadrupole time-of-flight (QTOF) tandem mass spectrometer for mass analysis. The MPT was operated at 40 W and 2.45 GHz in a 700 mL/min argon flow both for the inner and middle plasma. Results: Intact quasi-molecular and molecular ions of various drugs were successfully characterized by spray-inlet MPT-MS/MS. The analysis of one sample was finished within 30 s. Furthermore, the method exhibited excellent efficiency, precision and sensitivity, and the limits of detection and limits of quantification of the samples in methanol were in the range of 5.25-60.0 and 17.5-200 ng g-1 , respectively. Excellent linearities with coefficients of determination (R2 ) of 0.9627-0.9980 were verified in the range 0.05-50 μg g-1 . Four different beverages purchased locally were also analyzed with spray-inlet MPT-MS/MS, and caffeine was directly determined in two of the beverages. By adding six standard drug samples to sport drinks (each drug was 1 μg g-1 ) and Chinese spirit (each drug was 0.1 μg g-1 ), all the drugs except for caffeine were detected successfully. Conclusions: This study indicates that spay-inlet MPT-MS/MS is an effective method for direct and rapid identification of drug solutions, and it has substantial potential for fast and sensitive drug residue detection.

Abstract:Fentanyl and fentanyl analogues represent a current and emerging threat in the United States as pure illicit narcotics and in mixtures with heroin. Because of their extreme potency, methods to safely and rapidly detect these compounds are of high interest. This work investigates the use of thermal desorption direct analysis in real time mass spectrometry (TD-DART-MS) and ion mobility spectrometry (IMS) as tools for the rapid and sensitive (nanogram to picograms) detection of fentanyl, 16 fentanyl analogues, and five additional opioids. Competitive ionization studies highlight that detection of these compounds in the presence of heroin is readily achievable, down to 0.1% fentanyl by mass with TD-DART-MS. With IMS, detection of nanogram levels of fentanyl in a binary fentanyl and heroin mixture is possible but can be complicated by decreased resolution in certain commercial instrument models. Modifications to the alarm windows can be used to ensure detection of fentanyl in binary mixtures. Additionally, three complex background matrices (fingerprint residue, dirt, and plasticizers) are shown to have a minimal effect of the detection of these compounds. Wipe sampling of the exterior of bags of questioned powders is shown to be a safe alternative method for field screening and identification, removing the need to handle potentially lethal amounts of material.

Abstract:Synchronized discharge ionization (SDI) was previously developed for hand-held mass spectrometers with discontinuous atmospheric pressure interfaces. The function of SDI has been demonstrated for analysis of volatile organic compounds in air at high sensitivity, which is attributed to the fact that ions were produced next to the ion trap mass analyzer inside the vacuum manifold. In this study, a simple sampling device was designed and fitted to a hand-held mass spectrometer to characterize its potential in direct analysis of low-volatility chemicals on surfaces. Nine chemicals of vapor pressures ranging from 10(-4) to 10(-8) Torr (at room temperature), including pesticides, illicit drugs, and explosives, were selected to evaluate and demonstrate the analytical capability of the designed system. Compounds of vapor pressures below 10(-7) Torr, such as tetryl, cocaine, and tetrahydrocannabinol (THC), have been successfully detected. Direct analysis of pesticides from fruit and explosives from a large surface area has also been demonstrated. Tandem mass analysis was performed, which helped to confirm the analyte identity as well as to improve the signal-to-noise ratio (S/N).

Abstract:Increasing in cases involving drugs of abuse leads to heavy burden for law enforcement agencies, exacerbating demand for rapid screening technique. In this study, atmospheric pressure ionization technologies including direct analysis in real time (DART) ion source coupled to a time-of-flight mass spectrometer (DART-TOF-MS)as well asdopant-assisted positive photoionization ion mobility spectrometry (DAPP-IMS) without radioactivity were utilized together as the powerful analytical tool for the rapid screening and identification of 53 abused drugs.The limits of detection (LOD) were 0.05-2μg/mL when using DART-TOF-MS and 0.02-2μg when using DAPP-IMS which could satisfy the actual requirement in forensic science laboratory. The advantages of this method included fast response, high-throughput potential, high specificity, and minimal sample preparation. A screening library of reduced mobility (K0), accurate mass of informative precursor ion ([M+H]+) and fragment ions was established respectively by employing a bench-top DAPP-IMS and TOF-MS in-source collision induced dissociation (CID) mode. Then the standardized screening procedure was developed with criteria for the confirmation of positive result. A total of 50 seized drug samples provided by local forensic laboratory we reanalyzed to testify the utility of the method. This study suggests that a method combing DART-TOF-MS and DAPP-IMS is promising for the rapid screening and identification of abused drugs with minimal sample preparation and absence of chromatography.
In-line Thermal Desorption and Dielectric Barrier Discharge Ionization for Rapid Mass Spectrometry Detection of Explosives

Abstract:Thermal desorption (TD) of wipe-based samples was coupled with an in-line dielectric barrier discharge ionization (DBDI) source and rugged compact time-of-flight mass spectrometer (MS) for the detection of explosives, propellants, and post-blast debris. The chromatography-free TD-DBDI-MS platform enabled rapid and sensitive detection of organic nitramine, nitrate ester, and nitroaromatic explosives, as well as black powder and black powder substitute propellants. Parametric investigations characterized the response to TD temperature, and optimized DBDI voltage, aerodynamically assisted entrainment, and fragmentation through in-source collision induced dissociation (isCID). Excess nitrate generated by the DBDI source yielded predominantly nitrate-adduct formation. Sub-nanogram sensitivities were demonstrated for all explosives investigated, except for nitroglycerin, specifically due to its volatility. Further, most analytes/explosives exhibited tens of picogram sensitivities. The platform also demonstrated the detection of propellant and military explosives from post-blast debris. The TD-DBDI-MS system performed well without the need for aerodynamically assisted entrainment (and the associated rough pump), which along with requiring no additional gasses (i.e., N2 ¬or He) or solvents, aid in potential field deployment. The ease of TD-DBDI attachment and removal added trace solid or liquid residue detection to the rugged mass spectrometer, designed primarily for analysis of volatile organic and inorganic compounds.
