Abstract:A thermal desorption low temperature plasma (TD-LTP) ionization source was developed for the direct, rapid and sensitive detection of pesticides by mass spectrometry without complex sample pretreatment. A thermal desorption sampler was added in front of the plasma generator, and the analyte was collected on a polytetrafluoroethylene (PTFE) swab and desorbed into gas molecules, and then gas molecules were transported to the plasma generator by carrier gas to be ionized. The utilization of thermal desorption sampler helped to transform the interaction between gas phase plasma and sample from gas-solid or gas-liquid to gas-gas, which greatly increased the sensitivity and stability especially for non-volatile sample (e.g. pesticides) compared with conventional LTP ionization source. The TD-LTP ionization source was coupled to a homemade rectilinear ion trap mass spectrometry. The operation parameters of the TD-LTP ionization source were optimized and the characteristic ions of 12 different pesticides were successfully obtained and investigated. Then the TD-LTP ionization source was connected with commercial ACQUITY TQD mass spectrometer, and the pesticide residue level in broomcorn was evaluated.

Abstract:The application of thermal desorption-gas chromatography to determine the low boiling point components in tobacco was studied. By choosing the right materials and effective enrichment method to adsorb aroma components, the factors influencing the analysis was inspected, and a reasonable thermal desorption experiment program was established. Thus we can achieve the qualitative analysis of aroma components in tobacco. This method requires no sample manipulation, and is especially suitable for the determination of low volatile components in tobacco, also it can make up for the shortcomings of other methods of dealing with tobacco.

Abstract:The increasing demand for rapid and sensitive roadside identification of trace illegal drugs drives the development of high-performance miniature mass spectrometric instrumentation and methodology. Here, we report a synchronized flash-thermal-desorption purging and ion injection (SFTDPI) method to increase the sensitive and rapid screening of volatile and nonvolatile illegal drugs for miniature ion trap mass spectrometry (ITMS). The flash-thermal desorption could reach 290 degrees C in 2.5 s, which could achieve efficient vaporization of nonvolatile noscapine with boiling point at 565 degrees C. ITMS using discontinuous atmospheric pressure interface (DAPI) has an ion utilization ratio of less than 1%, the synchronized purging for flash-thermal desorption and ion injection with DAPI could accurately control the time interval along with the desorption, gas purging, ionization and ion injection, and the sample utilization ratio increases more than five times. The miniature SFTDPI-ITMS presents good performance: (1) more than 60 times improvement in sensitivity was achieved compared to the previously reported thermal-desorption acetone-assisted photoionization ion trap mass spectrometer for nonvolatile drugs, and the minimum detectable quantity reaches 50 pg for fentanyl. (2) Ten kinds of mixing drugs with boiling point difference of 300 degrees C can be simultaneously identified within 3 s under a single analysis. SFTDPI-ITMS was deployed at the roadside checkpoint of Sino-Burmese border; fentanyl in the captured encapsulated powder and the suspected opiate had been successfully identified.

Abstract:Microchip-based high-field asymmetric ion mobility spectrometry (FAIMS) technology was used for rapid identification of tobacco mildew.Sample gas was introduced by a dynamic headspace diffusion method.Using FAIMS technology,the sample was tested in positive and negative ion mode under atmospheric conditions with clean air (carrier gas:filtered by activated carbon and molecular sieve).To optimize detection conditions,carrier gas flow rate and sample flow rate were controlled at 2000mL/min and 100 mL/min,respectively.Headspace temperature was controlled at 25℃.Under optimal conditions,recognition rules of the degree of mildewy tobacco were established according to identification points.Identification points included three variables of the compensation voltage (CV),electric field (DF) strength and ionic current value (ICV).Characteristic substances of different mildewy tobacco were analyzed by TD-GC-MS.There was a significant difference among Ion current value of characteristic peaks for different mildewy tobacco.The recognition rate of this method was more than 90 percent with relative standard deviation (RSD) lower than 5％.Therefore,FAIMS detection technology has a great potential for application in rapid identification of tobacco mildew.
In Situ Analysis of Tobacco Leaves Based on Microwave Plasma Torch Desorption Ionization Mass Spectrometry

Abstract:Tobacco is an important economic crop worldwide. Tobacco leaves contain a large number of natural products. Therefore, the analysis of tobacco composition is crucial for the control of tobacco safety and quality assessment. Microwave plasma torch desorption ionization mass spectrometry (MPT-MS) is an emerging ambient mass spectrometry (AMS) technique that can be used for rapid real-time analysis of samples without pretreatment. Our work focuses on the chemical composition analysis of tobacco using MPT-MS, and 14 compounds were detected in green flue-cured tobacco leaves, while 30 compounds were detected in cured flue-cured tobacco leaves, which confirms that curing can enhance the flavor of cigarettes. Furthermore, we successfully in situ identified characteristic fragment ions of α/β-CBD (α/β-cembranoids), an important aroma compound in tobacco by MPT-MS. The results demonstrate the capability of MPT-MS for in situ analysis of tobacco components and its potential for evaluating tobacco quality.

Abstract:A miniature ion trap mass spectrometer with capillary direct sampling and vacuum ultraviolet photoionization source was developed to conduct trace analysis of organic compounds in liquids. Self-aspiration sampling is available where the samples are drawn into the vacuum chamber through a capillary with an extremely low flow rate (less than 1 μL/min), which minimizes sample consumption in each analysis to tens of micrograms. A pulsed gas-assisted inlet was designed and optimized to promote sample transmission in the tube and facilitate the cooling of ions, thereby improving instrument sensitivity. A limit of detection of 2 ppb could be achieved for 2,4-dimethylaniline in a methanol solution. The sampling system described in the present study is specifically suitable for a miniature photoionization ion trap mass spectrometer that can perform rapid and online analysis for liquid samples. Abstract:A method incorporating headspace liquid-phase microextraction (HS-LPME) coupled to matrix-assisted laser desorption/ionization (MALDI) with Fourier transform mass spectrometry (FTMS) was established to analyze volatile basic components in tobacco. The sample preparation volume for MALDI-MS was compatible with the volume of the solvent microdrop in the HS-LPME procedure. The pH and the polarity of the solvent for HS-LPME were adjusted by choice of the MALDI matrix and matrix additive. Based on the elemental composition and tandem mass spectrometry information, 25 volatile nitrogenous compounds in tobacco were detected and identified. The approach is fast and sensitive, and has the potential for automation for high-throughput analysis. Abstract:A novel infrared-assisted extraction coupled to headspace solid-phase microextraction followed by gas chromatography with mass spectrometry method has been developed for the rapid determination of the volatile components in tobacco. The optimal extraction conditions for maximizing the extraction efficiency were as follows: 65 μm polydimethylsiloxane-divinylbenzene fiber, extraction time of 20 min, infrared power of 175 W, and distance between the infrared lamp and the headspace vial of 2 cm. Under the optimum conditions, 50 components were found to exist in all ten tobacco samples from different geographical origins. Compared with conventional water-bath heating and nonheating extraction methods, the extraction efficiency of infrared-assisted extraction was greatly improved. Furthermore, multivariate analysis including principal component analysis, hierarchical cluster analysis, and similarity analysis were performed to evaluate the chemical information of these samples and divided them into three classifications, including rich, moderate, and fresh flavors. The above-mentioned classification results were consistent with the sensory evaluation, which was pivotal and meaningful for tobacco discrimination. As a simple, fast, cost-effective, and highly efficient method, the infrared-assisted extraction coupled to headspace solid-phase microextraction technique is powerful and promising for distinguishing the geographical origins of the tobacco samples coupled to suitable chemometrics.

Abstract:In this paper, a simple method of headspace analysis (HSA) was applied to determine aroma components in Chinese southwest tobacco. Ninety-seven aroma compounds were extracted from tobacco under optimized experimental conditions. Meanwhile, it took 4 h for traditional method namely simultaneous distillation and extraction (SDE) for analysis, which is time-consuming and a waste of solvent. Compared with simultaneous distillation and extraction, headspace analysis is faster and more convenient in extracting tobacco aroma compounds. Moreover, six aroma compounds were first proposed in the research. The main aromatic components in tobacco detected by headspace can evaluate the tobacco quality and distinguish a specific variety from others. Headspace analysis method was proposed as an easy, rapid and environment-friendly method for the determination of aroma components in tobacco. The proposed method can not only enhance the extracting rate of aromas but meet the demand of qualitative analysis.

Abstract:Taste and odor (T&O) compounds are widespread in water environments and have attracted considerable public attention. Nowadays, the standard detections of these chemicals rely mainly on off-line methods such as GC-MS or evaluation by trained analysts' senses. In this study, we report a method for the rapid detection of T&O compounds in water by exploiting a newly invented chemi-ionization source, in combination with headspace vapor measurement at room temperature. The calibrated limits of detection (LODs) of 2-methylbutyraldehyde, methyl tert-butyl ether (MTBE), methyl methacrylate (MMA), 2-isobutyl-3-methyoxypyrazine (IBMP), and 2-isopropyl-3-methoxypyrazine (IPMP) are in the range of 3.5-50.2 ng L-1, and the estimated LODs of 2-methylisoborneol (2-MIB) and geosmin (GSM) are 0.25 and 0.77 ng L-1, respectively. The calibration results reveal that the instrumental LODs for 2-methylbutyraldehyde, MTBE, MMA, beta-cyclocitral, 2-MIB, and GSM are 1-2 orders of magnitude better than the odor thresholds of humans. The accuracy, precision, recovery, and linearity (R-2) of the method are tested. Water samples from city tap water and three rivers in Beijing are assessed using this technique, and the typical T&O compositions are observed with concentrations ranging from 0.2 to 297 ng L-1. The new ultra-sensitive rapid detection method shows comparable sensitivities to the existing off-line technique and displays great potential for real-time detection of T&O pollution in water environments. Real-time characterization and quantification of aerosol components for open- and Closed-Ended heated tobacco products

Abstract:To characterize fast and dynamic thermophysical processes of aerosol formation from heated tobacco products (HTPs), a real-time chemical detection and quantification at sub-second time resolution is of great significance. Herein a novel vacuum ultraviolet (VUV) photoionization time-of-flight mass spectrometer (PI-TOFMS) was adapted for on-line chemical analysis of gaseous and particulate aerosol fractions formed from two HTP systems of contrasting designs. The two HTP systems used a battery-powered heating mechanism to heat tobacco rods containing granulated tobacco materials, one puffing air to flow through the tobacco bed and the other with no puffing air going through the tobacco section (termed as open-ended and close-ended, respectively). The on-line PIMS analyzer was able to detect a great number of mass peaks, including nicotine (Nic), glycerin (VG) and propylene glycol (PG) etc in the particulate phase, and flavor components such as 2,3-Butanedione and triethyl citrate in both gaseous and particulate phases. In particular, some short-lived intermediates such as ethenol and propen-2-ol from pyrolysis of the tobacco granules were observed and identified on-line. Nic, VG and PG were quantified with a high time resolution of 0.5 s using their standard calibration curves established with PI-TOFMS. The time-resolved evolving mass profiles of Nic, VG and PG were obtained in near real-time, as well as Nic release rate within a single puff for the first time. In addition, the present results clearly revealed that the change in puffing air flow pathway along the tobacco rod from the open-ended to the close-ended configurations significantly altered the Nic delivery pattern, in which a stable Nic delivery profile in the open-ended HTP at prolonged puff durations was reduced in the close-ended HTP. These findings can be utilized to develop suitable HTPs that could mimic the Nic delivery and hence help smokers to quit smoking.

Abstract:A novel method of microwave-assisted deep eutectic solvent extraction coupled with solid-phase microextraction was developed to determine the volatile compounds in tobacco by gas chromatography-mass spectrometry. During the process, the deep eutectic solvent played the roles of absorbing medium for microwave radiation, destroyer of cell walls, and solvent for dissolving the compounds released from the cells. Combined with solid-phase microextraction, the volatile components were extracted and concentrated in one step. In this method, several experimental parameters, such as fibre type, deep eutectic solvent type, microwave power, set-up temperature and irradiation time were studied. Compared with microwave-assisted water extraction coupled with solid-phase microextraction, and conventional solid-phase microextraction, more volatile components were obtained with higher responses. It was demonstrated that this method has improved extraction efficiency. Moreover, the proposed method has been applied to analyzing tobacco volatiles from different regions and nine different types of components have been detected. The results indicate that microwave-assisted deep eutectic solvent extraction coupled with solid-phase microextraction is an efficient, environmentally-friendly and fast miniaturization pretreatment technology for the extraction of volatiles from tobacco samples. This is a potentially promising technique for other complex and valuable matrices with low volatile content.
Analysis of Pesticide Residues in Tobacco Using Heart-cutting Two Dimensional Liquid Chromatography-tandem Mass Spectrometry

Abstract:A new concept for sample cleanup, based on size exclusion of macro molecular compounds in matrix, has been developed by coupling of gel permation chromatography (GPC) with LC-MS/MS. Heart-cutting two-dimensional liquid chromatography tandem mass spectrometry system was used by transferring the target elution through a loop. In the two dimensional liquid chromatography -tandem mass spectrometry system, a GPC column was employed in the first dimension to separate the macromolecule interference compound weight. After split, the target fluent were heart-cutting to a loop connected to a 6-way valve. Then the valve switched, so the second dimensional mobile phase took the target compounds for further reversed phase liquid chromatographic separation. Finally, about 80 different species of pesticide residues were determined qualitatively and quantitatively under the multi reaction monitoring (MRM) mode. The recoveries of most pesticides were in the range of 72%-136% and the limits of quantitation (LODs) for this method were from 0.022 to 0.152 mg/kg. Compared to conventional approaches for pesticide residue analysis in tobacco, the new technique involves less contamination of ion source in MS without sacrificing method LODs. In addition, the 2DLC-MS/MS method was fully automatic and the results were also accurate when utilized to detection of tobacco samples.
Development of Dual-Photoionization Ion Trap Mass Spectrometry and Its Application for Direct Analysis of VOCs in Fruit Aroma

Abstract:Photoionization-ion trap mass spectrometry (PI-ITMS) is one of the major directions of mass spectrometer miniaturization because of its great potential for rapid on-site VOCs detection in many cases. Traditionally, PI has always been investigated separately and is restrained by ion transmission structure, so a new structure needs to be designed and investigated for simplifying and improving the ion transmission efficiency. Interestingly, our preliminary experiments found that the signal intensity and mass range can be effectively improved by combing atmospheric pressure photoionization (APPI) and low-pressure photoionization (LPPI). Therefore, in this paper, a new dual photoionization - ion trap mass spectrometry (DPI-ITMS) was developed, explored and used to directly analyze complex VOCs. Compared with traditional single PI configuration, it presents two obvious merits: (1) simplified ion transmission structure, eliminating the need to use deflection electrode to repel ions and avoiding breakdown risk. (2) some missing/weak low m/z ion mass spectral peaks in APPI and some high m/z ion mass spectral peaks in LPPI were improved in DPI detection mode. In addition, by combining multivariate statistical analysis, we preliminary achieved in differentiating fruit types and maturity level. In summary, we concluded that the developed DPI-ITMS has moderate detection sensitivity (limited by the homemade ITMS, 0.1-1 ppmv with RSD of 6.36 %), and the DPI-ITMS configuration can be referenced by future PI-MS, and this study also provides a high-throughput, simple, noninvasive and no chemical contamination solution for analyzing main VOCs in fruit aroma.
Rapid Determination of Flavorings in e-Cigarette Liquid by Direct Analysis in Real Time (DART) Mass Spectrometry (MS) in the Quick Strip Mode

Abstract:Electronic cigarettes (E-cigarettes) have become popular around the world. An important reason is the variety of available flavors. However, many countries have imposed restrictions on flavor substances in E-cigarettes due to their potential health risks. Therefore, it is necessary to develop an effective way to identify flavorings in E-cigarette liquids. In this work, direct analysis in real time-mass spectrometry (DART-MS) in the quick strip (QS) mode was developed and applied to determine flavorings in the E-cigarettes. Vanillin, ethyl vanillin, maltol, and ethyl maltol were quantified. Meanwhile, the ionization efficiency of flavorings in the different solvents was compared in the QS mode, which revealed that the solvent boiling point was related to the analyte ionization efficiency. The effect of endogenous matrix (propylene glycol and glycerol) on flavoring ionization was also investigated. Calibration curves with satisfactory linearity ( R 2 > 0.99) were established from 1 to 1000 μg L −1 . Good sensitivity (limits of detection [LODs] < 0.3 μg L −1 ), recoveries (92.6% to 104.7%), and acceptable repeatability (relative standard deviations < 14.6%, n = 5) were obtained. The limits of quantification (LOQs; 10–50 μg g −1 ) for the E-cigarette liquid sample were much lower than the regulatory limits. The developed method provided a simple way for rapidly determining flavorings in E-cigarettes.

Abstract:Ambient ionization mass spectrometry (AIMS) is simple to operate for analytes adsorbed on the surface of various shaped probes. However, gaseous substances or liquids that are easy to evaporate, diffuse, and escape in the atmosphere are harder to capture. In this work, a Tee-shaped sample introduction device coupled with direct analysis in real time mass spectrometry (DART-MS) is developed. The Tee-shaped device is placed between the DART ion source and the MS inlet with a heated sample transfer tube. Gaseous samples from either a Tedlar sampling bag or liquids evaporated from a graduated syringe were tested. The Tee-shaped device was used for several volatile organic compounds with a wide range of boiling points, and detection limits of ng/mL to fg/mL were obtained. To test the device for real-life samples, puff-by-puff analysis of a complex gaseous mainstream smoke was performed. Individual puffs can be analyzed rapidly, and there is no cross contamination between consecutive puffs. The dynamic changes of chemical components among different puffs for different types of cigarettes can be observed. This work provides a universal Tee-shaped sampling device to enhance AIMS for the analysis of volatile compounds and gases, which is adapted to different sampling modules applicable for various forms of samples. The device enables direct exploration of chemical components in complex gaseous samples without tedious sample preparation and time-consuming LC or GC separation.

Abstract:With the development of instrumental miniaturization, the portable mass spectrometer is becoming a new tool for on-site rapid analysis of environmental samples. Membrane inlet (MI) and photoionization (PI) are two commonly used sampling and ionization techniques, respectively, as they both exhibit detection selectivity for volatile organic compounds (VOCs). In this paper, a membrane inlet photoionization ion trap mass spectrometer was developed for the direct analysis of VOCs in gaseous samples. With the new structure and timing design, various operation modes were proposed and tested. In particular, the use of pulse carrier gas can integrate the appropriate pressure conditions required by each module, thus improving the efficiency of analyte transport, ionization, and mass analysis. The detection limit of sub-ppb was obtained, and the response time can be greatly reduced by increasing the sample flow rate. Furthermore, the capability of selective enrichment for organic analytes was also realized by using a special accumulation mode with a modified sequence, which is easy to operate because no additional devices are needed.

Abstract:The thermal desorption (TD) technique is widely employed in modern mass spectrometry to facilitate the detection of non-volatile analytes. In this study, we developed a compact TD device based on a small resistance wire and coupled it with a self-aspirating corona discharge ionization (CDI) source to conduct direct MS analysis of various liquid and solid samples. Due to its small size and low heat capacity, the temperature of the TD module can be flexibly and rapidly modulated by controlling the power sequence. Multiple heating modes, including pulse heating (PH), isothermal heating, and step heating (SH), are realized and characterized, and then applied for the detection of different real samples. In particular, the PH mode is suitable for the simultaneous detection of multiple components in samples with relatively simple matrices, while the SH mode is capable of component separation. In addition, the sensitivity and quantitative capability of the TD-CDI system for DEP solutions were tested, showing acceptable stability with a relative standard deviation of about 6.7% and a detection limit of 0.088 ng. Overall, the developed TD-CDI system provides a simple, convenient, and versatile tool for direct mass spectrometry analysis of real samples.
Rapid On-Site Detection of Illegal Drugs in Complex Matrix by Thermal Desorption Acetone-Assisted Photoionization Miniature Ion Trap Mass Spectrometer.

Abstract:Illegal drug smugglings and crimes have long been a global concern, and an apparatus which can identify drugs on-the-spot is urgently demanded by law enforcement. A thermal desorption acetone-assisted photoionization miniature ion trap mass spectrometer was developed for on-site and rapid identification of illegal drugs at checkpoints. Acetone was chosen for dopant-assisted photoionization, and the sensitivity of selected drugs was further enhanced with protonated analyte molecular ions [M + H]+. For example, the sensitivity of ephedrine was improved by as high as 22-fold. The mass discrimination effect, which was usually considered as a shortcoming of ion trap mass analyzer, was ingeniously utilized to eliminate the protonated acetone reagent ions and maximize the trapping efficiency of analyte ions in mass analyzer. Twenty-seven drugs were analyzed, and the limits of detection (LODs) of selected illegal drugs were at the nanogram level with analysis time of 2 s. Analyte/dopant ion peak intensity ratios in mass spectra could be used for quantitation to improve the quantitative analysis performance of miniature ion trap mass spectrometer equipped with a discontinuous atmospheric pressure interface (DAPI) with the prerequisite that dopant ions and analyte ions could be simultaneously and effectively trapped by the ion trap. The RSD of signal intensity was reduced from 25.3% to 8.5%, and the linear range was extended from 0.5-25 to 0.5-100 ng/μL for methamphetamine. A temperature-resolved thermal desorption sampling strategy was developed and used to distinguish illegal drug components in plant-based drug samples and drinks containing illegal drugs.

Simultaneous Enrichment and Analysis of Tobacco Alkaloids by Microextraction Coupled with Mass Spectrometry Using a Poly (N-Isopropyl-acrylamide-co-divinyl-benzene-co-n, N'-methylene Diacrylamide) Monolithic Column.

Abstract:Herein, we realized the simultaneous online detection of six tobacco alkaloids (TM) by in-tube solid-phase microextraction (In-tube SPME) coupled with mass spectrometry by a rapid, sensitive, and matrix effect-free method requiring no chromatographic separation and only minimal sample pre-treatment. A poly (N-isopropylacrylamide-co-divinylbenzene-co-N, N'-methylenediacrylamide) [Poly (NIPAAm-co-DVB-co-MBAA)] monolithic column was designed according to the chemical structures of selected TM and used as an extraction medium engaging in hydrophobic, pi-pi, and hydrogen bonding interactions with analytes, allowing them to be effectively extracted. A number of important parameters were systematically optimized to achieve maximal extraction efficiency. The ion intensity of the TM signals obtained by in-tube SPME-MS were higher than the direct MS mode by about 400 folds with the signal-to-noise ratio improved by 2-7 folds. The detection limits of the six TM were determined as 1.99-4.06 ng g(-1), with good linearity with correlation coefficients exceeding 0.99 obtained under optimal extraction conditions. Besides, TA recoveries in cigarette tobacco spiked at three concentration levels were in the range of 76.4-100.2%, and the corresponding RSDs (n = 5) were obtained as 4.32-7.16%. The extraction performance of the poly (NIPAAm-co-DVB-co-MBAA) monolithic column was well reproducible, with intra- or inter-day precision RSDs determined not to exceed 7.38%. Finally, no marked matrix effects were observed when the developed method was applied to the analysis of both high-abundance and trace level TM in practical samples, and the above technique was therefore concluded to be well suited for the detection of TAs in cigarette tobacco or other products.
