Abstract:The generation of thioesters through thioesterification of carboxylic acid has significant synthetic value given the easy availability of various carboxylic acids and the extensive application of thioesters. An economical and rapid thioester synthesis approach is still needed. Herein, we achieved thioesterification of carboxylic acids with valuable glycosyl thiol, aromatic and aliphatic thiols mediated by sulfonyl fluoride (SO2F2). The compatibility of numerous carboxylic acid substrates was tested, including (hetero)aromatic and aliphatic carboxylic acids and the reaction proceeded smoothly under mild conditions at good yields. Late‐stage modification of drug molecules rac‐Naproxen and Loxoprofen proved feasible. Besides, the gram‐scale reaction demonstrated the utility of this protocol.

Abstract:A dehydrative cross-coupling of unactivated allylic alcohols with sulfinic acids was achieved under catalyst-free conditions. This reaction proceeded via allyl sulfination and concomitant allyl sulfinate-sulfone rearrangement. Various allylic sulfones could be obtained in good to excellent yields with water as the only byproduct. This study expands the synthetic toolbox for constructing allylic sulfone molecules.

Abstract:A palladium-catalyzed highly stereoselective sulfonylation of vinylethylene carbonates for the precise synthesis of structurally diverse (Z)-allylic sulfones was achieved with excellent regioselectivity and stereoselectivity (Z/E ratio, up to 99 : 1). This protocol used inexpensive sodium sulfinates as sulfonyl sources to construct valuable (Z)-allylic sulfones in good to excellent yields. The controlling experiment suggested that the hydroxyl proton came from the alpha-hydrogen of sulfone group through 1, 5-H shift.

Abstract:An efficient approach to sulfones through a reaction of aryldiazonium tetrafluoroborates, sulfur dioxide, and aryliodonium tetrafluoroborates under metal-free conditions is reported. Abstract:A one-pot nickel-catalyzed synthesis of (hetero)aryl alkyl sulfones from readily available boronic acids, sodium metabisulfite and benzylic ammonium salts as electrophiles is described. This general and efficient synthetic process is of broad scope, occurs in two steps, and delivers structural diverse sulfones, and the intermediate sulfinate is isolated. The transformation exhibits application of benzylic ammonium salts as novel electrophiles for direct insertion of SO2 as a novel area to be investigated.

Abstract:A palladium catalyzed multicomponent cross-electrophile coupling involving sulfur dioxide under a metal-reductant free condition is presented here. Both aryl-alkyl and alkyl-alkyl sulfones could be successfully achieved. This protocol features mild reaction conditions, wide substrate scope, good functional group tolerance and excellent regioselectivity. Preliminary mechanistic studies reveal that two Pd catalytic cycles are involved and SO2 exclusively coupled with aryl iodides rather than allylic acetates, which are consistent with the DFT studies.

Abstract:A novel and controllable synthesis of thioacetals/thioketals and β-sulfanyl ketones mediated by the reaction of aldehyde/acetone with thiols has been developed. In this protocol, β-sulfanyl ketones can be generated without the prior preparation of α, β-unsaturated carbonyl compounds. A variety of thiols reacted with aldehyde/acetone and provided the corresponding thioacetals/thioketals and β-sulfanyl ketones in good to excellent yields, respectively. This protocol is operationally simple, mild, and atom-economical, providing controllable access to thioacetals/thioketals and thia-Michael addition products under mild conditions.

Abstract:Enantioselective sulfonylation of α,β-unsaturated carbonyl compounds from sulfur dioxide is achieved via the synergistic merger photoredox and hydrogen bonding catalysis. This approach provides a facile route to enantioenriched sulfones in the absence of transition metals by using DABCO·(SO2)2 as the sulfur dioxide surrogate. With this method, a variety of sulfones bearing a chiral centre in the α-position are prepared in moderate to good yields.

Abstract:A cobalt-promoted coupling reaction of triethoxysilanes, sulfur dioxide, and electrophiles is developed. Benzylic, allylic or alkyl bromides are all efficient as electrophiles in this transformation. Moreover, the reactions with other electrophilic partners including iodonium salts and electron-poor (hetero) aryl chlorides proceed smoothly as well under the standard conditions. The sulfinate intermediates are generated in situ from the reaction of triethoxysilanes and sulfur dioxide under cobalt-promoted conditions, which subsequently combine with various electrophiles to produce the corresponding sulfones in moderate to good yields.

Abstract:A practical strategy for the construction of diverse phosphonyl and thiofunctionalized sulfoxonium ylides via controllable monofunctionalization of hybrid I-(III)/S-(VI) ylides is presented. This process allows efficient P-H insertion of I-(III)/S-(VI) ylides under Cu catalysis, enabling the synthesis of phosphonyl sulfoxonium ylides, whereas reaction with sulfur-containing reagents including AgSCF3, KSC(S)OR, and KSCN under mild conditions resulted in alpha-trifluoromethylthiolation, dithiocarbanation, and thiocyanation of sulfoxonium ylides accordingly. Of note, wide substrate compatibility (108 examples), excellent efficiency (up to 99% yield), gram-scale experiments, and various product derivatizations highlight the synthetic utility of this protocol.

Abstract:β-Chiral sulfones are structural motifs widely found in natural products and bioactive molecules, while also serving as important intermediates for the synthesis of valuable chiral scaffolds. In contrast to the rapid growth of sulfur dioxide insertion chemistry over the last decade, enantioselective catalytic variants for accessing β-chiral sulfones, especially those bearing a quaternary carbon stereocenter, remain rare. Herein, we report an enantioselective copper/bisoxazoline catalyzed oxysulfonylation of alkenes to yield isoxazolinyl-containing β-chiral sulfones bearing quaternary carbon stereocenters with moderate to excellent yields and up to 96:4 er. The advantage and irreplaceability of this sulfur dioxide insertion catalytic system have been demonstrated by achieving transformations clearly unavailable with the previously reported sulfonyl chloride system. Additionally, the recoverability of the new-developed bisoxazoline ligand L9 and its reusability without any erosion of the catalytic activity and enantioselectivity further demonstrate the usefulness of the protocol.

Abstract:An efficient method for the synthesis of alkynyl sulfides via C(sp3)−S bond cleavage of α-bromostyrene sulfonium salts has been developed. This base-promoted nucleophilic ring-opening pathway that allows for the preparation of diverse alkynyl sulfide compounds using tetramethylene sulfoxide as the sulfur source. The reaction proceeds with good functional group tolerance and could be applied to late-stage functionalization of bioactive molecules and drugs. Abstract:Recent advances in the sulfonylation of alkenes via the insertion of sulfur dioxide are summarized. The sulfur dioxide surrogate of DABCO<bold></bold>(SO2)(2) or inorganic sulfites is used in the transformation through a radical process. Two strategies for the sulfonylation of alkenes with the insertion of sulfur dioxide have been developed. In most cases, the vicinal difunctionalization of alkenes is initiated by the arylsulfonyl radical generated in situ. Although the sulfonylation of alkenes with the insertion of sulfur dioxide is efficient, the asymmetric versions are still challenging and less to be explored. It is expected that the asymmetric vicinal difunctionalization of alkenes with the insertion of sulfur dioxide will be developed rapidly in the near future.

Abstract:Abstract Organic sulfides form the core scaffold of a wide range of pharmaceuticals, natural products, and materials, and serve as key intermediates in synthesis. Prior methods to organic sulfides require the use of transition metal (TM) catalysts, prefunctionalized or chelating group-containing substrates, and elevated reaction temperatures. A general TM-free C–H thiolation protocol using readily accessible sulfur source is highly desirable. Herein, we disclose a direct C(sp)–, C(sp2)–, and C(sp3)–H thiolation reaction using β-sulfinylesters as the versatile sulfur source. The key step of this protocol is chemoselective C–S bond cleavage of the sulfonium salts that is in situ formed from the corresponding (hetero)arenes, alkenes, alkynes, and 1,3-dicarboxyl compounds with β-sulfinylesters. The successful capture of acrylate byproduct supports a retro-Michael reaction mechanism. This method is expected to be used widely because of several advantageous aspects including TM-free, mild reaction conditions, and broad substrate scope including drug molecules.

Abstract:Trisulfide unit is widely found in natural products and has garnered attention due to the unique pharmacological and physiochemical properties. However, despite limited progress, widely applicable approaches for constructing unsymmetrical trisulfides have so far remain scarce. In this work, an easy-to-prepare, solid-state and scalable reagent, S-substituted sulphenylthiosulphate, has been developed for the divergent synthesis of unsymmetrical trisulfides. Alkyl electrophile substrates, including bromides, chlorides, iodides and tosylates, with diverse substituents are smoothly converted to the corresponding trisulfides with S-substituted sulphenylthiosulphates and thiourea as another sulfur source. Furthermore, the late-stage modification of drug molecules was successfully achieved through this method.

Abstract:A simple method to synthesize diverse sulfonyl chlorides through NaClO2-mediated oxidative chlorosulfonation of S-alkyl isothiourea salts is presented. The approach features safe operation, environmental friendliness, convenient purification procedures, and delivers high yields of up to 96%. The procedure is also applicable to substrates such as thiols, disulfides, thioacetates, and xanthates. It is a versatile and convenient method for the synthesis of various sulfonyl chlorides from different thiol precursors and derivatives.

Abstract:Comprehensive Summary The selective oxidative sulfonylation of alkenes with selenium sulfonate depended on the reaction conditions. The electrochemical C—H sulfonylation proceeded smoothly to afford ( E )‐vinyl sulfones with good selectivity in an undivided cell without external oxidant. While aerobic trifunctionalization of alkenes occurred in the presence of KI in the air, which provides β ‐keto selenosulfones via the formation of C—O, C—S, and C—Se bonds in one‐pot. Following control experiments, a plausible mechanism is proposed to rationalize the experimental results.
