Abstract:We describe a new method for the stereoselective synthesis of (Z)-1,2-dithio-1-alkenes via copper-catalyzed cross-coupling of (Z)-2-bromovinyl sulfides and thiols. The desired products are obtained in good to excellent yields. The double bond geometry of the starting (Z)-2-bromovinyl sulfides is retained under these reaction conditions.

Abstract:We have developed a highly selective one-pot method for the synthesis of (E)-vinyl sulfones and sulfoxides from thiols with terminal alkynes. The sulfones and sulfoxides could be obtained with excellent selectivity in good isolated yields. It is simple, efficient and environmentally benign, and metal-free. The mechanism for the formation of the (E)-vinyl sulfones was also proposed.

Abstract:The addition reaction of sulfonyl chlorides with acetylenes in the presence of a copper salt is described. The reaction proceeds in toluene in the presence of CuCl with Me2S as the additive to afford only (Z)-beta-chlorovinyl sulfones in moderate to excellent yields.

Abstract:Vinyl sulfones are important structural motifs with a wide range of applications in the biomedical field. Although the hydrosulfonylation of alkynes have provided straightforward strategies for the synthesis of vinyl sulfones, these methods still exhibit several drawbacks, such as the use of large amounts of transition metal catalysts or/and additional additives. Herein, we report an efficient and mild multicomponent reaction (MCR) for the preparation of vinyl sulfones by hydrosulfonylation of alkynes with the corresponding aryl diazonium salts, K2S2O5 and thiophenols at room temperature without any metal catalysts or additives. This transformation is the first example of thiophenols as hydrogen source reacting with vinyl radical with the advantages of high chemoselectivity and step-economy. Notably, the method is also applicable to alkyl alkynes circumventing the limitations of existing methods, and the gram-scale experiment proceeded smoothly to prove the practicality of the methodology.

Abstract:Abstract Organic sulfides form the core scaffold of a wide range of pharmaceuticals, natural products, and materials, and serve as key intermediates in synthesis. Prior methods to organic sulfides require the use of transition metal (TM) catalysts, prefunctionalized or chelating group-containing substrates, and elevated reaction temperatures. A general TM-free C–H thiolation protocol using readily accessible sulfur source is highly desirable. Herein, we disclose a direct C(sp)–, C(sp2)–, and C(sp3)–H thiolation reaction using β-sulfinylesters as the versatile sulfur source. The key step of this protocol is chemoselective C–S bond cleavage of the sulfonium salts that is in situ formed from the corresponding (hetero)arenes, alkenes, alkynes, and 1,3-dicarboxyl compounds with β-sulfinylesters. The successful capture of acrylate byproduct supports a retro-Michael reaction mechanism. This method is expected to be used widely because of several advantageous aspects including TM-free, mild reaction conditions, and broad substrate scope including drug molecules.

Abstract:A sulfur stereogenic center is found in naturally occurring compounds and constitutes an important design element for pharmaceuticals and chiral auxiliaries. In view of the broad applications of S-stereogenic compounds, many methods were developed to enantioselectively construct the sulfur stereocenters. By contrast, the synthesis and isolation of S-stereogenic ylides was elusive although sulfonium ylides are important synthetic building blocks and such species have been proposed in the Doyle-Kirmse reaction. Using judicious substrate design, we have successfully developed the first catalytic enantioselective synthesis of sulfonium ylides with a chiral sulfur center. They are constructed from a broad array of sulfides in high yields (up to 98%) and enantioselectivities (up to 99%) through copper-catalyzed intramolecular carbene transfer reaction. The obtained ylides could be converted into alkylidenecyclopropanes with efficient sulfur-to-carbon chirality transfer.

Abstract:A new type of organocatalyzed 1,3-thiosulfonylation has been developed to straightforwardly access highly functionalized vinyl sulfones, which features mild conditions, atom- and step-economy, practicability, conciseness, and environmental friendliness. Moreover, these valuable products can be transformed to vinyl sulfides via a base-promoted isomerization. The versatile route can efficiently and rapidly introduce SCD3 groups with excellent levels of deuterium content (>99% D) by utilizing our newly developed SCD3 reagents. Gram-scale operations and further transformations are smoothly carried out, providing promising applications for drug discovery.

Abstract:A practical and straightforward approach for the highly stereoselective synthesis of β-amino sulfones was developed. With lithium bis(trimethylsilyl)amide as the base, the corresponding sulfone-stabilized carbanion derived from alkylphenyl sulfone can be transferred to N-(tert-butylsulfinyl) aldimines in excellent yields and with high diastereoselectivity.

Abstract:A palladium-catalyzed highly stereoselective sulfonylation of vinylethylene carbonates for the precise synthesis of structurally diverse (Z)-allylic sulfones was achieved with excellent regioselectivity and stereoselectivity (Z/E ratio, up to 99 : 1). This protocol used inexpensive sodium sulfinates as sulfonyl sources to construct valuable (Z)-allylic sulfones in good to excellent yields. The controlling experiment suggested that the hydroxyl proton came from the alpha-hydrogen of sulfone group through 1, 5-H shift.
