Abstract:Antibacterial materials have always suffered from high toxicity. In this study, we fabricated injectable and biodegradable hydrogels utilizing poly(hexamethylene guanidine) hydrochloride and poly (ethylene glycol) as the backbone through a thiol-ene "click" reaction under physiological conditions, and they showed remarkably low toxicity and high and broad-spectrum antibacterial activity.
Shear-Thinning Catechol-Modified Chitosan Hydrogel Loaded with Silver Nanoparticles for Endoscopic Submucosal Dissection

Abstract:Shear-thinning hydrogels have emerged for endoscopic submucosal dissection, while wound intervention after surgery has rarely been mentioned. Herein, a catechol-modified chitosan hydrogel with shear-thinning property was developed for simultaneously facilitating endoscopic submucosal dissection and postoperative wound healing. Benefiting from the shear-thinning and self-healing characteristics, the asprepared hydrogel showed easily endoscopic injectability. It also performed very well as submucosal cushion, which could remain above 70% after injection for 120 min in ex vivo porcine large intestine model. In fact, the cushion height of normal saline dramatically decreased to 46% of the initial height at 30 min. Ag nanoparticles encapsulated into the network endowed the hydrogel with almost reached 100% antibacterial effect against E. coli and S. aureus. The hemolysis ratio of the hydrogel was calculated to be as low as 0.8%. Combined with good hemocompatibility and cytocompatibility, the as-prepared hydrogel displayed much higher in vivo wound closure and healing efficacy than normal saline. These results demonstrated the superiority of the shear-thinning chitosan hydrogel in facilitating clinical endoscopic submucosal dissection surgery.
Ultrastretchable, Adhesive, and Antibacterial Hydrogel with Robust Spinnability for Manufacturing Strong Hydrogel Micro/Nanofibers

Abstract:The ultrastretchable (over 12 400%) hydrogel with long-lasting adhesion, strong antibacterial activity, and robust spinnability is developed based on the oxidative decarboxylation and quinone-catechol reversible redox reaction induced by Ag-lignin nanoparticles in a precursor solution containing citric acid (CA), acrylic acid (AA), and poly (acrylamide-co-acrylic acid) (P(AAm-co-AA)). With massive reversible interactions including hydrogen bonds and electrostatic forces, such hydrogel exhibits promising injectability and is facilely spun via manual drawing, draw-spinning, and electrospinning for manufacturing strong hydrogel micro/nanofibers. The resulting fibers exhibit excellent mechanical properties, including tensile stress of 422.0 MPa, strain of 86.5%, Young's modulus of 8.7 GPa, and toughness of 281.6 MJ m<sup>-3</sup> . The hydrogel microfibers obtained from a house-built spinner are scaled-up fabricated while retaining promising mechanical properties, as evidenced by lifting a load (317.2 g) using the spun fibers of ≈33 000 times lighter weight (9.5 mg), indicating their great potentials in the applications such as net and safety cord which require robust mechanical properties. Moreover, assisted by a commercial electrospinning machine, nanosized hydrogel fibers are facilely spun on personal protective equipment such as a mask to offer an antiseptic coating with near 100% killing efficiency against airborne bacteria aerosols, demonstrating the capability of spun hydrogel fibers on disinfection-related applications.

Shear-Thinning Extrudable Hydrogels Based on Star Polypeptides with Antimicrobial Properties

Abstract:Hydrogels with low toxicity, antimicrobial potency and shear-thinning behavior are promising materials to combat the modern challenges of increased infections. Here, we report on 8-arm star block copolypeptides based on poly(L-lysine), poly(L-tyrosine) and poly(S-benzyl-L-cysteine) blocks. Three star block copolypeptides were synthesized with poly(S-benzyl-L-cysteine) always forming the outer block. The inner block comprised either two individual blocks of poly(L-lysine) and poly(L-tyrosine) or a statistical block copolypeptide from both amino acids. The star block copolypeptides were synthesized by the Ring Opening Polymerization (ROP) of the protected amino acid N-carboxyanhydrides (NCAs), keeping the overall ratio of monomers constant. All star block copolypeptides formed hydrogels and Scanning Electron Microscopy (SEM) confirmed a porous morphology. The investigation of their viscoelastic characteristics, water uptake and syringe extrudability revealed superior properties of the star polypeptide with a statistical inner block of L-lysine and L-tyrosine. Further testing of this sample confirmed no cytotoxicity and demonstrated antimicrobial activity of 1.5-log and 2.6-log reduction in colony-forming units, CFU/mL, against colony-forming reference laboratory strains of Gram-negative Escherichia coli and Gram-positive Staphylococcus aureus, respectively. The results underline the importance of controlling structural arrangements in polypeptides to optimize their physical and biological properties.

Abstract:Most hydrogels are chemically crosslinked hydrogels, the use of excessive chemical crosslinkers limits their application in biomedicine and other fields. Herein, a completely physically crosslinked double-network hydrogel of sodium alginate/chitosan/zinc ion (SA/CS/Zn2+ PDH) which does not contain toxic chemical reagents was prepared. Compared with the sodium alginate/chitosan single-network hydrogel crosslinked by electrostatic interactions, the mechanical properties of the PDH is significantly improved. Gelation time, swelling behavior, mechanical performance, and antibacterial ability were evaluated with zinc concentration as a variable. It was found that with the increase of zinc content, the gelation rate increased, the swelling rate decreased, and the mechanical properties improved. The PDH exhibited outstanding antibacterial activity against both Staphylococcus aureus and Escherichia coli. Our work provides a new idea for the fabrication of environment-friendly physically crosslinked double-network hydrogels based on natural polysaccharides.

Abstract:Antibacterial hydrogels have attracted significant attention due to their diverse applications, efficient antimicrobial properties, and adaptability to various environments and requirements. However, their relatively fragile structure, coupled with the potential for environmental toxicity when exposed to their surroundings for extended periods, may significantly limit their practical application potential. In this work, a composite hydrogel was synthesized with outstanding mechanical features and antibacterial capability. The hydrogel was developed through the combination of the eco-friendly and enduring antibacterial agent, lignin silver nanoparticles (Lig-Ag NPs), with polyvinyl alcohol (PVA) and sodium alginate (SA), in varying proportions. The successful synthesis of the hydrogel and the dispersed distribution of Lig-Ag NPs within the hydrogel were confirmed by various analytical techniques, including field emission scanning electron microscopy (SEM), energy-dispersive spectroscopy (EDS), mercury intrusion porosimetry (MIP), Fourier transform infrared spectroscopy (FT-IR), X-ray diffraction (XRD), and X-ray photoelectron spectroscopy (XPS). The formation of multiple hydrogen bonds between Lig-Ag NPs and the composites contributed to a more stable and dense network structure of the hydrogel, consequently enhancing its mechanical properties. Rheological tests revealed that the hydrogel exhibited an elastic response and demonstrated outstanding self-recovery properties. Significantly, the antibacterial hydrogel demonstrated effectiveness against Escherichia coli (E. coli) and Staphylococcus aureus (S. aureus), achieving a <5% survival of bacteria within 12 h. This study presented a green and straightforward synthetic strategy for the application of antibacterial composite hydrogels in various fields.

Abstract:Hydrogel materials are the preferred candidates for antimicrobial materials due to their high water absorption, superior biocompatibility and structural diversity. In this work, we present for the first time that diamine-alkyl and 1,3-dibromo-2-propanol can be rapidly cross-linked to prepare a series of new cationic polymer gels by polycondensation reactions under a strongly alkaline environment. The presence of C−N+ bonds are demonstrated in the formation of dendritic structured polymers by XPS analysis. In addition, the physicochemical properties of b-PHIs hydrogels were evaluated and it was found that the variation of diamine alkyl chain length could modulate the storage modulus, water uptake properties, thermal stability and zeta-potential of the gels. The increase in hydroxyl content and the C−N+/C-C ratio is evidenced by the elevated water absorption properties of the gels and the variations in zeta potential, respectively. In addition, the antibacterial performance evaluation showed that gels had good antibacterial activity against S. aureus and E. coli, with antimicrobial rates of b-PHOI as high as 96% and 97%, respectively. This work pioneers a brand-new idea for the simple and rapid synthesis of branched cationic polymers with low cytotoxicity in the field of antimicrobial biomaterials and proceeds a new attempt in the directional modulation of the physical properties of the materials.

Abstract:The unique benefit of electrostatic self-assembly of microscale components in solution is demonstrated for the first time. In particular, positive and negative treatment of poly(ethylene glycol) (PEG) facilitates a novel bottom-up assembly approach using electrostatic interaction from microgels with opposite charges. Fundamental investigations of electrostatic interaction of microgels reveal that the contact area of microgels determines the total energy of construct and thus the final patterns. The electrostatic self-assembly approach enables the fabrication of large and complex biological related structures (e.g., multi-layer spheroid) with accurate control. By the design of the microgels, the thickness and number of microgels in each layer can be controlled. Biological investigations of positive and negative treatments of PEG further prove the possibility of using this approach in tissue engineering, regenerative medicine and drug delivery.

Abstract:Viscoelastic microfluidics becomes an efficient and label-free hydrodynamic technology to enrich and separate micrometer-scale particles, including blood cells, circulating tumor cells, and bacteria. However, the manipulation of nanoscale particles by viscoelastic microfluidics remains a major challenge, because the viscoelastic force acting on the smaller particle decreases dramatically. In contrast to the commonly used polymer solutions of high molecular weight, herein we utilize the aqueous solutions of poly(ethylene oxide) (PEO) of low molecular weight with minimized shear thinning but sufficient elastic force for high-quality focusing and separation of various nanoparticles. The focusing efficiencies of 100 nm polystyrene (PS) nanoparticles and λ-DNA molecules are 84% and 85%, respectively, in a double spiral microchannel, without the aid of sheath flows. Furthermore, we demonstrate the size-based viscoelastic separation of two sets of binary mixtures-100/2000 nm PS particles and λ-DNA molecules/blood platelets-all achieving separation efficiencies of >95% in the same device. Our proposal technique would be a promising approach for enrichment/separation of the nanoparticles encountered in applications of analytical chemistry and nanotechnology.

Abstract:Hydrogels usually are fabricated by using monomers or preexisting polymers in precursor solutions. Here, we report a polyelectrolyte complex biohydrogel (Bio‐PEC hydrogel) made from a precursor dough, by kneading, annealing, and crosslinking the dough of two oppositely charged polysaccharides, cationic chitosan quaternary ammonium salt (HACC) and anionic sodium hyaluronate (HA), photoinitiator (α‐ketoglutaric acid), crosslinker glycidyl methacrylate (GMA), and water of very small quantity. Controlled kneading and annealing homogenized the dough with respect to transforming randomly distributed, individual polymer chains into tightly wound double‐stranded structures, which, upon UV irradiation, covalently sparsely crosslinked into a highly entangled network and subsequently, upon fully swollen in water, resulted in Bio‐PEC hydrogel, HACC/HA, exhibiting near‐perfect elasticity, high tensile strength, and high swelling resistance. via the same kneading and annealing, tetracarboxyphenylporphyrin iron (Fe‐TCPP) metal nanoclusters were incorporated into HACC/HA to obtain photocatalytic, antibacterial, and biocompatible Bio‐PEC hydrogel composite, Fe‐TCPP@HACC/HA. Using SD rat models, we demonstrated the efficacy of Fe‐TCPP@HACC/HA in inhibiting E. coli growth in vitro and the ability to promote wound healing and scar‐free skin regeneration in vivo, or its high potential as a wound dressing material for biomedical applications. Abstract:We report a simple and facile self-assembly approach to fabricate polyelectrolyte complex (PEC) hydrogel films with positively charged chitosan (CS) and negatively charged heparin sodium (HS) by combining hydrogen bonding and electrostatic interactions. The CS/HS hydrogel films exhibited excellent tensile strength and toughness, good self-recovery ability, superior water absorbency, and pH-dependent surface charge characteristics. The gelation mechanism was investigated by zeta potential measurements. The CS/HS hydrogel films exhibited high antibacterial efficacy against E. coli at selected pHs or when coordinated with various metal ions and a significant effect on accelerating wound healing. The self-assembly approach presented in this work may serve as a generic strategy for the fabrication of novel multi-functional PEC hydrogels for broad biomedical applications.

Abstract:Electro-responsive dynamic hydrogels, which possess robust mechanical properties and precise spatiotemporal resolution, have a wide range of applications in biomedicine and energy science. However, it is still challenging to design and prepare electro-responsive hydrogels (ERHs) which have all of these properties. Here, we report one such class of ERHs with these features, based on the direct current voltage (DCV)-induced rearrangement of sodium dodecyl sulfate (SDS) micelles, where the rearrangement can tune the hydrogel networks that are originally maintained by the SDS micelle-assisted hydrophobic interactions. An enlarged mesh size is demonstrated for these ERHs after DCV treatment. Given the unique structure and properties of these ERHs, hydrophobic cargo (thiostrepton) has been incorporated into the hydrogels and is released upon DCV loading. Additionally, these hydrogels are highly stretchable (>6000%) and tough (507 J/m2), showing robust mechanical properties. Moreover, these hydrogels have a high spatiotemporal resolution. As the cross-links within our ERHs are enabled by the non-covalent (i.e., hydrophobic) interactions, these hydrogels are self-healing and malleable. Considering the robust mechanical properties, precise spatiotemporal resolution, dynamic nature (e.g., injectable and self-healing), and on-demand drug delivery ability, this class of ERHs will be of great interest in the fields of wearable bioelectronics and smart drug delivery systems.

Abstract:In this work, fully biodegradable antimicrobial hydrogels were prepared facilely via a thiol-ene "click" reaction under human physiological conditions using multifunctional poly(ethylene glycol) (PEG) derivatives as precursors. Water soluble and degradable PEG derivatives with multi-enes and multi-thiols, respectively, were synthesized by polycondensation of oligo(ethylene glycol) (OEG) with "clickable" monomers. Ammonium groups with long alkyl chains were incorporated into one of the precursors covalently, using dodecyl bis(2-hydroxyethyl) methylammonium chloride as a comonomer. Proton nuclear magnetic resonance (H-1-NMR) spectroscopy, gel permeation chromatography (GPC) and Fourier transform infrared spectroscopy (FT-IR) were used to characterize the precursors and hydrogels. These types of cationic PEG-type hydrogels showed strong antibacterial abilities against both Gram-negative and Gram-positive bacteria due to the ammonium moieties. Moreover, the hydrogel with fewer ammonium moieties still possessed significant antibacterial abilities, but low toxicity, and has the potential to be used as a medical material.

Facile Synthesis of Thermo-Sensitive Composite Hydrogel with Well Dispersed Ag Nanoparticles for Application in Superior Antibacterial Infections

Abstract:In this study, we have described a facile process for fabrication of multifunctional composite hydrogel, in which sodium alginate was subjected to cross-linking using Ca2+ derived from ZnO/CaCO₃/Ag composite nanospheres. The ZnO/CaCO₃/Ag composite nanospheres were prepared based on our previously reported AA-[Zn(OH)₄]2- composite nanosphere reaction conducted with silver and calcium salt following hydrothermal method, that led to the disintegration and release of Ca2+ under acidic conditions for application as a cross-linking agentto catalyze reaction with sodium alginate. Ag nanoparticles were well-dispersed in the multifunctional composite hydrogel, exhibiting excellent antibacterial activity. Additionally, polydopamine (PDA) with photothermal effect was also added to obtain a multifunctional composite hydrogel, and this hydrogel showed photothermal conversion performance and facilitated the release of Ag+ to achieve the rapid antibacterial effect. Simultaneously, PDA NPs could scavenge free radicals and improve cell adhesion. All such features would promote wound healing. The potent antimicrobial activity of the prepared composite hydrogel was demonstrated in the mouse model of S. aureus infection, and biosafety of the hydrogel was confirmed by conducting histopathological examination in the mouse model. This type of multifunctional hydrogel wound dressing with photosensitive and antibacterial properties presents with broad applications and prospects in antibacterial treatment.

Abstract:Polymeric conductive hydrogels have been highlighted for use in flexible electronics, and there is still a challenge to design and fabricate a high-performance hydrogel electrolyte with integrated and multiple properties, such as sustainability, stretchability and compressibility, ionic conductivity, antibacterial property, strain sensitivity, and low-temperature tolerance. Herein, chitosan (CS) derived poly(aprotic/protic ionic liquids) (CS-PAPILs) are facilely prepared by taking the structural and sustainable features of chitosan and betaine hydrochloride, which are used as a functional component for the fabrication of CS-PAPILs/polyacrylamide (PAM)/LiCl (CS-PAPILs/PAM/LiCl) double-network (DN) hydrogel electrolyte via an in situ polymerization of AM and then a soaking strategy in LiCl aqueous solution. The achieved DN hydrogels exhibit tunable mechanical performance (tensile strength of 70-900 kPa, elastic modulus of 31.5-484 kPa, high compressibility (4450 kPa at 80 % strain), superior low-temperature tolerance (freezing point < -85 degrees C), anti-dehydration performance, high ionic conductivity at 25/-50 degrees C (94.8/4.6 mS cm(-1)), and excellent antibacterial activity. As a proof of concept, an assembled flexible and anti-freezing supercapacitor by the as-prepared DN hydrogel electrolyte demonstrated a high specific capacitance of 108.4F/g (at 2 A/g) and impressive cycling stability over 50,000 cycles at temperature as low as -50 degrees C. Furthermore, a wearable strain sensor based on the DN hydrogel was also demonstrated, and the sensor can be successfully attached to the human joints to monitor various human motions.

Abstract:Polymer hydrogels find extensive application in biomedicine, serving specific purposes such as drug delivery, biosensing, bioimaging, cancer therapy, tissue engineering, and others. In response to the growing threat of bacterial infections and the escalating resistance to conventional antibiotics, this research introduces a novel injectable, self‐healing antimicrobial hydrogel comprising bioactive aldolized hyaluronic acid (AHA) and quaternized chitosan (QCS). This designed QCS/AHA hydrogel incorporates self‐assembling peptide nanofibers (PNFs) and small‐sized silver nanoparticles (AgNPs) for tailored functionality. The resulting hybrid QCS/AHA/PNF/AgNPs hydrogel demonstrates impressive rheological characteristics, broad‐spectrum antimicrobial efficacy, and high biocompatibility. Notably, its antimicrobial effectiveness against E. coli and S. aureus surpasses 99.9%, underscoring its potential for treating infectious wounds. Moreover, the rheological analysis confirms its excellent shear‐thinning and self‐healing properties, enabling it to conform closely to irregular wound surfaces. Furthermore, the cytotoxicity assessment reveals its compatibility with human umbilical vein endothelial cells, exhibiting no significant adverse effects. The combined attributes of this bioactive QCS/AHA/PNF/AgNPs hydrogel position it as a promising candidate for antimicrobial applications and wound healing. This article is protected by copyright. Abstract:<p>As a smart wearable sensor device, the mildew of the biocompatible hydrogel limits its application. In this paper, silver nanoparticles were prepared by solid-state reduction of hydroxyethyl cellulose and compounded into a chemically cross-linked hydrogel as an antibacterial, flexible strain sensor. Because the high surface energy of silver nanoparticles can quench free radicals, we designed three initiators to synthesize hydrogels: ammonium persulfate (APS), 2,2'-Azobis(2-methylpropionitrile) (AIBN) and 2,2'-azobis(2-methylpropionamidine) dihydrochloride (AIBA). Impressively, silver nanoparticles composite hydrogel could only be successfully fabricated and triggered by the AIBN. The mechanical property of the composite hydrogel (0.12 MPa at 704.33 % strain) was significantly improved because of dynamic crosslinking point by HEC. Finally, the composite hydrogels are applied to the field of antibacterial strain sensor and the highest Gauge Factor (GF) reached 4.07. This article proposes a novel, green and simple strategy for preparing silver nanoparticles and compounding them into a hydrogel system for antibacterial strain sensor.</p>
