Abstract:Objective The purpose of this work was to investigate the distribution pattern of fibrinolytic factors and their inhibitors in rabbit tissues. Methods The components of the fibrinolytic system in extracts from a variety of rabbit tissues, including tissue plasminogen activator (tPA), plasminogen activator inhibitor-1 (PAI-1), plasminogen (Plg), plasmin (Pl) and α 2 plasmin inhibitor (α 2 PI), were determined by colorimetric assay. Results The tissue extracts in renal, small intestine, lung, brain and spleen demonstrated strong fibrinolytic function, in which high activity of tPA, Plg and Pl was manifested; whereas in skeletal muscle, tongue and stomach, higher activity of PAI-1 and α 2 PI showed obviously. Also excellent linear correlations were found between levels of tPA and PAI-1, Pl and α 2 PI, Plg and Pl. In related tissues, renal cortex and renal marrow showed distinctly higher activity of tPA and lower activity of PAI-1, with the levels of Plg and Pl in renal cortex being higher than those in renal marrow, where the α 2 PI level was higher than that in renal cortex. Similarly, the levels of tPA, Plg and Pl in small intestine were higher than those in large intestine, but with respect to PAI-1 and α 2 PI, the matter was reverse. In addition, the fibrinolytic activity in muscle tissue was lower, however, the levels of tPA, Plg, and Pl in cardiac muscle were obviously higher than those in skeletal muscles, and the levels of PAI-1 and α 2 PI were significantly lower than those in skeletal muscle. Conclusion Our data demonstrate that a remarkable difference of the fibrinolytic patterns exists in rabbit tissues, which has probable profound significance in understanding the relationship between the function of haemostasis or thrombosis and the physiologic function in tissues.

Abstract:Objective To evaluate the change of elasticity of rabbit liver following portal inflow vessels occlusion using acoustic radiation force impulse (ARFI). Methods Twelve Japanese white big ear rabbits were used to establish total hepatic vascular exclusion animal models by occluding the portal inflow vessels. The elasticity of rabbits' right liver was detected during the stages of baseline(T0), 5 min(T1),20min(T2) ,30min (T3)after occluding the portal inflow vessels using ARFI respectively. The elasticity of rabbit liver, levels of serum's aspartate aminotransferase (AST), alanine aminotransferase (ALT) and historical results of hepatic tissue during different stage were compared. Results Shear wave velocity(Vs)of rabbits' right livers in T2 was significantly higher than that in T0 ( P ＜0.05) and Vs of rabbits' right livers in T3 was significantly higher than that in T2 ( P ＜0.05). No significant differences of serum's AST,ALT levels between T2 and T0 were found,while the Vs of rabbit live in T2 was slightly higher than that in T0 (P＞0.05), serum's AST, ALT levels in T3 were significantly higher than that in T2 ( P ＜0.05).Historical changes of liver ischemia and injury were observed in T2 and T3 with light microscopy. Conclusions The liver ischemia and injury severity can be accurately and objectively evaluated using ARFI.
Protective effects of basic fibroblast growth factor against global cerebral ischemia/reperfuslon injury in rabbits

Abstract:Objective To investigate the effects of basic fibroblast growth factor (bFGF) on global cerebral ischemia/reperfusion (I/R) injury.Methods Twenty-four rabbits of both sexes aged 6 months weighing 2.2-3.0 kg were randomized into 3 groups ( n = 8 each) : group I sham operation; group Ⅱ I/R and group Ⅲ I/R + bFGF. The animals were anesthetized with diazepam, haloperidol and fentanyl. The right femoral artery was cannulated for BP monitoring and blood-letting. The animals were tracheotomized and mechanically ventilated (VT = 8 ml ·kg -1 , RR = 40 bpm, I: E = 1:2). PET CO2 was maintained at 40 mm Hg. Global cerebral I/R was induced by "occlusion of four vessels + hypotension" . Bilateral common carotid arteries and vertebral arteries were occluded for 30 min combined with hypotension, which was produced by blood-letting and maintained at 50%-60% of the baseline level. The four vessels were then released for reperfusion and BP was returned to baseline level by retrieval of the removed blood. In sham operation group ( Ⅰ ) the four blood vessels were exposed but not occluded. In group Ⅲ a loading dose of 30 μg·kg-1 bFGF was given Ⅳ at the end of 30 min ischemia followed by bFGF infusion at 10 μg· kg-1· h-1 for 6 h. In I/R group ( Ⅱ ) normal saline was given IV instead of bFGF. Blood samples were taken before I/R (T0 , baseline) and at 0, 0.5, 1, 3, 6 h (T1-5) of reperfusion for determination of serum neuron specific enolase (NSE) and S-100B concentrations. At the end of the experiment the animals were killed and brains removed for determination of brain water content and microscopic examination. Results Serum NSE and S-100B concentrations in group Ⅱ and Ⅲ were significantly increased at T4 5 as compared to the baseline (T0) and were significantly higher at T4 5 than those in sham operation group ( Ⅰ ) . Serum NSE and S-100B concentrations were significantly lower in group Ⅲ (I/R + bFGF) than in groupⅡ (I/R). Serum NSE and S-100B were positively correlated ( r = 0.736, P < 0.01) . Brain water content was significantly higher in group Ⅱ and Ⅲ than in sham operation group ( Ⅰ ) ( P < 0.01). The histo-pathological changes were milder in I/R + bFGF group (Ⅲ) than in I/R group (Ⅱ ). Conclusion Basic fibroblast growth factor has protective effects against global cerebral I/R injury.
[Recombinant Expression of Human Fibrogenic Factors and Preparation of Polyclonal Antibodies].

Abstract:AIM:To express the recombinant proteins of human fibrogenic factors and prepare the polyclonal antibodies of rabbit-anti-human fibrogenic factors.METHODS:The human fibrogenic factors were variant splicing forms of one gene. Their consensus sequence (ChS) was cloned into plasmid pQE30 to construct recombinant prokaryotic expression system. The recombinant expression vectors were transformed into E.coli M15, and then the expression vectors were induced by IPTG and many insoluble inclusion body proteins was obtained. The obtained proteins were used to immunize rabbits to obtain polyclonal antiserum with high titer and specificity. The eukaryotic expression of the recombinants of human fibrogenic factors was detected by Western blot.RESULTS:The insoluble inclusion body proteins were also used to prepare polyclonal antibodies with high titer and specificity.CONCLUSION:The expression of the recombinant proteins of human fibrogenic factors and the preparation of polyclonal antiserum provide a useful tool for further study of the expression and function of human fibrogenic factors.
Effect of Simultaneous Treatment of Phlegm and Stasis on the Balance of Blood Coagulant and Fibrinolytic Activity in Atherosclerotic Rabbits

Abstract:Objective To study the effects of simultaneous treatment of phlegm and stasis on the changes of blood fat,tissue plasminogen activator(t-PA)and plasminogen activator inhibitor(PAI),platelet agglutinate rate and hemorheology in atherosclrotic rabbits.Methods The 50 white rabbits were divided randomly into five groups with 10 in each:blank control group,model group,Di'ao Xinxuekang(0.02g/kg)group,phlegm and stasis simultaneous treatment small and large dose(7.5g/kg,15g/kg)groups.After four weeks' high-fat feedstuff feed,the rabbits were given medicine gavage per day for 4 weeks and then killed.The blood fat,t-PA and PAI,platelet agglutinate rate and hemorheology were measured.Results The phlegm and stasis simultaneous treatment in the treated group could significantly improve the level of blood fat,increase t-PA and decrease PAI,significantly inhibit the platelet one minute agglutinate rate and maximum agglutinate rate,significantly decrease the whole blood high cut rate and low cut rate viscosity,whole blood revert viscosity,platelet adhesion,and erythrocyte sedimentation rate(ESR)in atherosclerosis(AS)rabbit.Conclusion Simultaneous treatment of phlegm and stasis can prevent AS through improving abnormal blood fat,increasing fibrinolytic activity,inhibiting platelet aggregation,and improving microcirculation.
Effects of lumbrokinase on cerebral circulation in experimental rabbit

Abstract:OBJECTIVE To study the effects of lumbrokinase on cerebral circulation(cerebral blood flow and cerebral vascular resistance) in experimental animal of rabbit.METHODS 40 experimental rabbits were divided into 4 groups randomly according to the weight:(1)normal control group,(2)low dose lumbrokinase group,(3)high dose lumbrokinase group,(4)nimodipine group.We determine the cerebral blood flow by electro-magnetic flowmeter,and compute the cerebral vascular resistance,measure the mean arterial blood pressure and heart rate.Each changes of these targets on 0,5,15,30,60 min were recorded. RESULTS The cerebral blood flow increased remarkably and the cerebral vascular resistance decreased on 30,60 min in high dose lumbrokinase group. CONCLUSION Lumbrokinase can increase the cerebral blood flow and decrease the cerebral vascular resistance,so that it can improve the cerebral circulation.

Abstract:Objective To study the expression of plasminogen activator inhibitor type-1(PAI-1) and nitri-coxide (NO) in animal modes in order to investigate the relationship of fibrinolysis system and vascular endotheli-um function in atherosclerosis. Methods Male white rabbits were randomly divided into the normal diet group and the high-cholesterol diet group. The number of each group was 8. Blood was collected from the marginal vein of the ear in 0,8 and 16w respectively in two groups. Concentrations of PAI-1 and NO were measured. After 16w,the rabbits were put to death,aortas were harvested for pathologic morphology observation,immunohistochemistry analysis of PAI-1 expression was proformed. Results There were not obviously differences of PAI-1 and NO in the normal diet group and the high-cholesterol diet group in 0w (P0. 01),PAI-1 in the high-cholestrol diet group increased obviously in 8w(P0. 01). PAI-1 in 16w than 8w(P0. 01). NO had no difference in 8w with 0w(P 0, 01) and decreased in 16w were vigner (P0. 05). Atherosclerosis was formed by pathologic morphology observation. The expression of immunohistochemistry analysis of PAI-1 in cholesterol diet group was higher than control (P0. 01). Conclusion Atherosclerosis was accompanied by fibrinolysis system and vascular endothelium disorder and the unnormal expression of PAI-1 and NO may be one of mechanism of atherosclerosis.
Experimental Study on Serum Fibrosis Biomarkers in Hepatic Fibrosis Rabbit Model

Abstract:Objective To explore the relationship between serum fibrosis biomarkers and stages of hepatic fibrosis. Methods Forty-five rabbits were randomly divided into experiment group(n=36) and control group(n=9).Hepatic fibrosis models were established by intraperitoneal injection of carbon tetrachloride(CCl4) in experiment rabbits.Serum fibrosis biomarkers(hyaluronic acid,type Ⅲ procollagen,laminin,type Ⅳ collagen) and pathology were observed respectively at the 6th,9th and 12th week after the first injection.According to pathological grade and stage,the rabbits were grouped. Results Thirty rabbits completed the detection,including 9 cases of normal liver,6 cases of mild liver fibrosis,7 cases of moderate liver fibrosis and 8 cases of severe liver fibrosis.With the aggravation of hepatic fibrosis,hyaluronic acid,type Ⅲ procollagen,laminin,type Ⅳ collagen were increased gradually.All those four biomarkers were positively correlated with histopathological staging(P0.05),especially type Ⅳ collagen(r=0.718,P0.05). Conclusion Serum fibrosis biomarkers are effective indices to evaluate the severity of hepatic fibrosis.Ⅳ-C is the best for the diagnosis of hepatic fibrosis in the four biomarkers.
Expression of tissue factor, tissue factor pathway inhibitor in the atherosclerotic plaque on femoral artery of New Zealand rabbits

Abstract:AIM: To observe the regulatory changes of tissue factor(TF),tissue factor pathway inhibitor-1(TFPI-1) and tissue factor pathway inhibitor-2(TFPI-2) associated with the formation of atherosclerotic plaque on femoral artery of New Zealand rabbits.METHODS: New Zealand rabbits were fed with high cholesterol diet and received femoral artery balloon injury to establish moderated atherosclerotic plaque animal model.These rabbits were classified into 8 weeks group,10 weeks group and 12 weeks group.The expressions of factor TF,TFPI-1 and TFPI-2 in femoral artery were detected by immunohistochemical methods at 8th,10th and 12th week.The positive areas of the slice were analyzed by semi-quantification system.Fluorescence quantification PCR(FQ-PCR) was used to detect the copies of mRNA of TF,TFPI-1 and TFPI-2 in each group.RESULTS: The expressions of TF,TFPI-1 and TFPI-2 determined by immunohistochemical methods were increased from 8th,10th to 12th week,respectively.In each group,the quantity of mRNA of TF and TFPI-1 at the 8th,10th and 12th week was soared,respectively.In each group,the quantity of mRNA of TFPI-2 did not change in early phase,but decreased at 12th week.CONCLUSION: In the process of the atherosclerotic plaque formation in rabbit femoral artery model,the expressions of TFPI-1 and TFPI-2 proteins are increased.However,the expression of TPFI-1 or TFPI-2 could not stave off the development of plaque formation.

Abstract:PurposeTo investigate the thrombolytic effect of nattokinase and the main functional mechanism.MethodsObserve the nattokinase′s influence on the rabbit′s urgent thrombosis in vivo, the change of coagulation time, prothrombin time, activiated partial thrombopastin time and the fibrinogen fig content of the blood concretion system by pharmacological experiment.ResultsNattokinase could resist the forming of thrombosis, influenced the endogenous and extrinsic coagulation system, and it possessed direct thrombolytic effect.ConclusionNattokinase has strong fibrinolytic activity.
Long-Term Expression of Human Factor-Ix Cdna in Rabbits

Abstract:In this study, rabbits were used as a model for gene therapy for hemophilia B. Human factor IX cDNA was transferred to cultured normal rabbit skin fibroblasts (RSF) by a recombinant plasmid (pCMVIX) or retrovirus (XL-IX or N2CMVIX) constructed in our laboratory. Infected fibroblasts capable of synthesizing and secreting high levels of biologically active human factor IX protein were selected and embedded in a collagen matrix. The latter was surgically implanted into rabbits as autografts or allografts. Human factor IX protein was detected in the plasma of all the grafted rabbits, and its expression has been maintained for more than 10 months at the time of writing. In addition, we have improved and simplified the method of implantation from surgically grafting the tissue-like matrix to the injection of the infected cell-collagen mixture subcutaneously. Using the latter method, human factor IX in rabbits injected with RSF-N2CMVIX reached a peak of 480 ng/ml plasma, and its expression has continued for more than 3 months at the time of writing. We suggest that the simplified method of transplantation by subcutaneous injection would offer an effective and acceptable approach to somatic cell gene therapy and may be practical for human trials.

Abstract:Object To study the anticoagulant activity of recombinant hirudin (rH) made in China and to provide the experimental basis for the development of hirudin. Methods Thrombin time (TT), prothrombin time (PT) and activated partial thromboplastin time (APTT) were determined by coagulometer for rabbit blood samples collected before and after iv rH. The percent of lengthening above coagulation time parameters was calculated and used to investigate the dose and time effect relationship of rH. Results The TT and APTT were prolonged remarkably by rh, while PT not within the test range, in a dose dependent manner, and the action duration was about 60 min. Conclusion Domestic rH (N Ile 1 Thr 2 r hirudin) has obvious anticoagulant effects. Its potency is much higher than heparin, and identical to that of Refludan (N Leu 1 r hirudin), which was an authorized rH product made in Germany.

Abstract:Thrombotic diseases have been considered major causes of death around the world. Treatments with thrombolytic drugs, such as recombinant tissue-plasminogen activator, urokinase, and streptokinase, are reported to have a life-threatening bleeding tendency. On the contrary, lumbrokinase, identified from Lumbricus rubellus , is specific to fibrin and does not cause excessive bleeding. It possesses fibrinolytic activity and activation of plasminogen to dissolve fibrin. Hence, the purification of fibrinolytic protein monomer from earthworm and antithrombotic evaluation and investigation of mechanisms are needed. In this study, a novel fibrinolytic protein EPF3, with strong fibrinolytic activity, was purified from Pheretima vulgaris by ion exchange and size exclusion chromatography. SDS PAGE, bottom-up proteomics analysis, de novo sequencing, and circular dichroism (CD) analysis were carried out for identification and characterization of it. EPF3, with a molecular weight of 25136.24 Da, consisted of 241 amino acids and contained various forms of secondary structures, including α-helix (3.9%), β-sheet (42.8%), β-turn (21.2%), and random coil (32.1%). It was a trypsin-like serine protease and stable at pH 7.0 to 11.0 and below 40°C. EPF3 was confirmed to possess an antithrombotic effect by ex vivo clot lysis test and fibrinogen-thrombin time (Fib-TT) assay. The three-dimensional structure of EPF3 was predicted by SWISS-MODEL. Molecular docking analysis predicted that EPF3 could directly interact with antithrombotic target proteins (fibrin, fibrinogen, and plasminogen), which was further confirmed by further studies. The antithrombotic mechanism of EPF3 was clarified to be outstanding direct fibrinolysis, fibrinogenolytic activity, and certain activation of plasminogen. EPF3 possesses the potential to be developed into a promising antithrombotic agent.

[Study on Function of Decoction for Invigorating the Kidney and Improving Blood Circulation on Rabbits Blood Stasis Model].

Abstract:OBJECTIVE:To evaluate the effect of decoction for invigorating the kidney and improving blood circulation to thrombosis and pathology on rabbit blood stasis model. METHOD:Thirty rabbits were ramdomly divided into normal group, model group, high dose group, low dose group and Xue Shuan Ning group. Tissue-type plasminogen activator (t-PA), plasminogen activator inhibitor (PAI), fibrinogen (Fbg) and D-dimer (DD) were investigated after those rabbits had been treated. One rot was solected randomly from each group to observe pathological changes. RESULT:There were significant differences in t-PA, PAI, Fbg and DD between normal group and other groups is very obvious (P < 0.01) . Between groups of high dose low dose Xue Shuan Ning and model, the statistical differeces were significant, as well as between groups of high dose, low dose and Xue Shuan Ning groups (P < 0.05). However, there was no statistical difference between high dose group and high dose group (P > 0.05). The pathological changes in model group were most serious, those in Xue Shuan Ning were less serious. There were slight pathological changes in high dose group and low dose group. CONCLUSION:Models ware made successfully. High dose group and low dose group have stronger effect on thrombosis than Xue Shuan Ning group.

Abstract:Tissue factor (TF) is a cell-surface transmembrane receptor which initiates the extrinsic blood coagulation cascades. The extracellular domain of TF is the functional domain in blood coagulation. Therefore the soluble tissue factor (sTF) which contains only the extracellular domain of TF was prepared by technique of recombinant DNA and studied the substitution of rabbit brain powder in prothrombin time test. The cDNA gene of soluble tissue factor was amplified by the method of RT-PCR from the template of human placental total RNA and proved by DNA sequencing, then expressed with E. coli. The product of the gene expression was formed inclusion body in E. coli. The inclusion body was dissolved in urea system and refolded by dilution method. Ion exchange chromatography and gel filtration were used in purification of r-sTF. r-sTF could be applied to initiate blood coagulation existing of phospholipids. The results indicated that the recombinant soluble tissue factor could be substituted rabbit brain powder in prothrombin time test.
