Abstract:In this report, a mitochondria-targeted probe based on an efficient platform constructed by pyridinium-coumarin and 2-dicyanomethylene-3-cyano-4,5,5-trimethyl-2,5-dihydrofuran (TCF) with high selectivity and sensitivity toward SO2 derivatives was presented. As far as we know, pyridinium-coumarin is first used as a donor moiety to construct a FRET system. Compared with our previous work, it showed better solubility, higher biocompatibility and faster response speed. It also presented low detection limit (80.5 nM), excellent energy transfer efficiency (98.2%) and low cytotoxicity. Moreover, the probe could detect exogenous and endogenous bisulfite in living cells as well as in mitochondria successfully.

Abstract:Bisulfite (HSO3-)/Sulfite (SO32-) is widely used as a food additive, but excessive use often leads to serious consequences, so the detection of HSO3-/SO32- is of great importance. In this paper, a novel 1,4-diethylpiperazine-modified coumarin-benzopyran derivative (probe QLP) has been synthesized and characterized. In PBS (10 mM, pH = 7.4), QLP displays good selectivity and is sensitive for HSO3-/SO32- over various analytes with fluorescent "OFF-ON" rapid responding (2 min), long-wavelength emission (600 nm), and a detection limit of 177 nM. With the treatment of HSO3-/SO32-, the color of the QLP solution obviously changes from blue-green to yellow, and the fluorescent color of QLP changes from colorless to amaranth. The fluorescence-enhanced mechanism is qualitatively evaluated by density functional theory calculations using the CAM-B3LYP/6-31G (d) method, which reveals that the photoinduced electron transfer leads to the fluorescence emission of the QLP-SO3H adduct. Importantly, nontoxic QLP can be used to detect HSO3-/SO32- in sugar, natural water samples, and living cells and localized to the mitochondria and monitor the mitochondrial HSO3-/SO32- level.

Abstract:A new FRET-based ratiometric fluorescent probe (DSPT) for the detection of bisulfite was developed by the dansyl-piperazine-benzothiazolium conjugate platform, in which C=C conjugated with benzothiazolium was recognition unit and benzothiazolium moiety was mitochondria-targeted unit. The C=C double bond group would undergo Michael addition reaction with bisulfite, triggering significant fluorescence ratio increasing which was linear to the concentrations of bisulfite. The probe can be used in real sample including dry white wine and tap water. Besides, the test strips for bisulfite were prepared. Moreover, cell imaging results indicate that this probe is cell-membrane permeable and mitochondria-targetable, and can be applied to monitor both exogenous and endogenous bisulfite in ratiometric fluorescence imaging. Abstract:In this study, we report a fluorescent probe (CBT) based on coumarin and benzothiazole moieties for detecting SO2 derivatives (HSO3-/SO32-) in a ratiometric fluorescence manner. Probe CBT exhibited desirable properties such as a high specificity, a rapid response, good water solubility and a low detection limit. The Michael addition reaction was proved to be the sensing mechanism of CBT. Moreover, fluorescence imaging of cells indicated that probe CBT with low cytotoxicity can be used to monitor the endogenous SO2 derivatives in pathological liver cells (HepG2) but not in the normal liver cells (L-02), indicating that probe CBT can be applied to distinguish the two kinds of cells. Fluorescence co-localization studies proved that this probe shows an excellent mitochondria-targeting activity. Therefore, probe CBT has great potential to be applied in exploring the role of the SO2 derivatives in biology.

Abstract:A new mitochondria-targeted fluorescent probe RBC, constructed using a coumarin moiety which was selected as the donor and a benzothiazole derivative as the acceptor, for SO(2)derivatives (HSO3-/SO32-) was presented. The probe designed on a new FRET platform showed high selectivity and a low detection limit. Importantly, the probe could respond to HSO3-/SO(3)(2-)within 35 s. Furthermore, the probe could target mitochondria and was successfully used for fluorescence imaging of endogenous bisulfite in HepG2 with low cytotoxicity, which significantly assisted in cancer diagnosis.

Abstract:Sulfur dioxide, an air pollutant, is easily hydrated to sulfites and bisulfites and extremely harmful to human health. On the other hand, endogenous sulfur dioxide is the fourth gasotransmitter. In view of the above, it is worth developing an effective method for the detection of these compounds. In this paper, a novel colorimetric fluorescent probe (Hcy-Mo), based on hemi-cyanine, for bisulfites is reported. Hcy-Mo shows excellent selectivity for bisulfites over various other species including cysteine, glutathione, CN-, and HS-, and undergoes 1,4-addition reactions at the C-4 atom of the ethylene group. The reaction can be completed in 30 s in a PBS buffer solution and displays high sensitivity (limit of detection is 80 nM) for bisulfites. Test paper experiments show that the probe can be used for bisulfite detection in aqueous solutions. In addition, Hcy-Mo exhibits excellent cell permeability and low cytotoxicity for the successful detection of bisulfites in living MDA-MB-231 cells and in living mice, implying that this probe would be of great benefit to biological researchers for investigating the detailed biological and pharmacological functions of bisulfites in biological systems.

Abstract:We have developed a new ratiometric fluorescent probe composed of a coumarin-merocyanine dyad based on the FRET mechanism. The probe showed clear dual-emission signal changes in blue and red spectral windows upon addition of H2S in a dose dependent manner under a single wavelength excitation. The probe targeted mitochondria with high selectivity and sensitivity toward H2S.

Abstract:A new type of pentamethine cyanine dye with bromo group at the central position Cy5-Br was de-signed and synthesized, and the structure is not reported as far as we know. Cy5-Br can identify SO2 deriva-tives bisulfite and sulfite with a good linear response. Abstract:A near infrared fluorescent probe YSP for sulfite was synthesized, in which a julolidine fused with a pyran-2-one was employed as the fluorophore and the vinyl activated by an indole salt as the receptor. The introduction of julolidine and indole salt strengthens the electron push-pull effect of the probe and allows it to absorb (597 nm) and emit (681 nm) in red wavelength region. The addition of sulfite to the C˭C bond led to prominent blue-shifts in both absorption (171 nm) and emission (165 nm) spectra, which made it possible for colorimetric and ratiometric fluorescent detection of sulfite. NMR titration results illustrated that the determination of sulfite is a two-step process: nucleophilic addition of sulfite to the unsaturated carbon of C˭N in indole ring followed by intramolecular rearrangement through a four-membered ring to form adduct-B with shorter absorption wavelength. In addition, the cationic feature of YSP enables the probe to be specifically localized in mitochondria, and it could ratiometric bioimaging sulfite in living HepG-2 and L929 cells.

Abstract:Sulfur dioxide (SO2), one of active sulfur species (ASS), plays important roles in various physiologies and pathological processes. Hence, it is urgent to monitor the fluctuation of mitochondrial SO2 in vivo. In this work, a novel mitochondria-targeting probe CZ was designed and synthesized based on carbazole derivative, which showed that probe was employed to the detection of SO2 with high selectivity, rapid response as well as naked eyes. Besides, an excellent linear relationship (R-2 = 0.9977) was observed and the detection limit was calculated as low as 0.50411M (3 sigma/k). Surprisingly, with addition of SO2 derivative, fluorescence intensity ratios (F-475 nm/F-607 nm) of probe CZ increased by 1104-fold ranging from 0.007 to 7.73. Owing to its biocompatibility, probe CZ was also successfully employed for monitoring of mitochondrial SO2 by fluorescence confocal microscopy.

Abstract:A coumarin-benzopyrylium (CB) platform has been developed for the colorimetric and fluorogenic detection of bisulfite. The proposed probe utilizes coumarin as the fluorophore and positively charged benzopyrylium as the reaction site. The method employs the nucleophilic addition of bisulfite to the benzopyrylium moiety of CB to inactivate the electron-deficient oxonium ion. The driving force for photo-induced electron transfer is considerably diminished, thereby promoting the emission intensity of the coumarin fluorophore. The fluorescence intensity at 510nm is linear with bisulfite concentration over a range of 0.2-7.5M with a detection limit of 42nM (3). CB shows a rapid response (within 30s) and high selectivity and sensitivity for bisulfite. Preliminary studies show that CB has great potential for bisulfite detection in real samples and in living cells.

Abstract:It is well known that excessive levels of sulfur dioxide and its derivatives are connected to diverse diseases. Therefore, developing highly sensitive probes to detect and monitor sulfite in living cells is important for the diagnosis of disease and the study of biochemical processes in vivo. In this report, two zero cross-talk ratiometric fluorescent probes were synthesized (CA-ID-MC and CA-BI-MC), which were derived from carbazole-indolenine π-conjugated system for effective detection of sulfite in living cells. Observably, CA-BI-MC exhibited the largest emission shift of 157 nm from 617 to 460 nm with the addition of various concentrations of sulfite, which is beneficial for high-resolution imaging of the sulfite. CA-BI-MC also exhibits high sensitivity and low cytotoxicity. More importantly, this probe successfully located mitochondria and sensed the sulfite in HeLa cells caused by exogenous stimulation.

Abstract:The content of sulfur dioxide (SO2) in mitochondria is related to various physiological and pathological processes. The irregular viscosity in mitochondria would result in mitochondria disorders. In this research, a novel ratiometric two-photon fluorescent probe (MBCB) based on carbazole framework has been strategically designed and synthesized for dual-detection of mitochondrial SO2 derivatives and viscosity. The results show that probe MBCB is capable of responding to SO2 derivatives and viscosity using dual-color imaging. MBCB can not only recognize SO2 derivatives utilizing a ratiometric change of fluorescent intensity (I-434( nm)/I-600 (nm)), but also shows a high sensitivity and selectivity for monitoring SO2. On the other hand, the fluorescent intensity ratio (I-567 nm/I-415 (nm)) of MBCB increased with the increment of viscosity and exhibited a good logarithmic linear relationship between the ratio and viscosity (R-2 = 0.99). Satisfactorily, probe MBCB has a good targeting ability to mitochondria and a low cell cytotoxicity, and can be successfully applied to detect HSO3- in living cells and in vivo. Furthermore, the probe can visualize the change of mitochondrial viscosity induced by nystatin.

Abstract:We have developed the first two-photon colorimetric and ratiometric fluorescent probe, BICO, for the detection of bisulfite (HSO3−) in aqueous solution. The probe contains coumarin and benzimidazole moieties and can detect HSO3− based on the Michael addition reaction with a limit of detection 5.3 × 10−8 M in phosphate-buffered saline solution. The probe was used to detect bisulfite in tap water, sugar and dry white wine. Moreover, test strips were made and used easily. We successfully applied the probe to image living cells, using one-photon fluorescence imaging. BICO overcomes the limitations in sensitivity of previously reported probes and the solvation effect of bisulfite, which demonstrates its excellent value in practical application.

Abstract:When SO2 is inhaled through the respiratory tract into the body, it can be easily hydrated to sulfite (SO32-) and bisulfate (HSO3-) ions. They are widely used as essential preservatives for foods, beverages, and pharmaceutical products. Exposure to high doses of bisulfate induces a large number of respiratory diseases relevant to lung cancer, cardiovascular diseases, and many neurological disorders, as they involve in various physiology and pathological processes in mitochondria cell apparatus. In this work, a new fluorescent probe CY-SO2 for bisulfate/sulfite based on hemicyanine dye is reported, which can used in solution detection with 56-fold fluorescence ratio (F-467 nm/F-380 nm) enhancement, fast response (completed within 90 s) and excellent sensitivity (DL 2.67 nM). The effect have been exhibited in real sugar samples tests. CY-SO2 also displayed fluorescent imaging of breast cancer cells (MCF-7) successfully, demonstrating that the probe is a novel mitochondria-targeted ratiometric fluorescent probe to image cellular SO2 derivatives.

Abstract:Bisulfite plays crucial roles in diverse physiological processes. Therefore, the efficient detection of bisulfite is very important. In this study, we report a colorimetric and ratiometric fluorescent probe (CPT) with a large Stokes shift (162 nm) for bisulfite (HSO 3 − ) based FRET mechanism. The probe can quantitatively detect HSO 3 − with low detection limit (45 nM) and high specificity over other common anions and biothiols. A nucleophilic addition reaction was proposed for the sensing mechanism, which was confirmed by HRMS spectra. The test strips of the probe were made and used easily. Moreover, probe CPT was used to ratiometric fluorescent imaging of exogenous and endogenous HSO 3 − in living cells.
